1.Pathogenesis and interventions of cancer-related sarcopenia
Chinese Journal of Clinical Nutrition 2015;23(3):183-188
The pathogenesis of cancer-related sarcopenia is complicated, involving all aspects of tumor and the body's metabolism, mainly related to accelerated proteolysis and insufficient protein synthesis.Cancerrelated sarcopenia can predict antineoplastic-induced dose-limiting toxicities, increase complications, and reduce survival rate. The interventions of cancer-related sarcopenia includes nutrition, exercise, hormone therapy and other interventions.A single treatment can not reverse clinical outcome.Various interventions are still in the investigation stage,in which more evidence-based studies are needed.

International Journal of Cerebrovascular Diseases 2016;24(3):225-229
The etiology of ischemic stroke in young adults is extensive, and cardiogenic embolism and cervicocephalic arterial dissection are the most common. In recent years, the proportion of atherosclerotic ischemic stroke has increased because of the increased incidence and inadequate control for the traditional vascular risk factors in young adults. However, even after a detailed and comprehensive examination, there is stil about 1/3 unknown etiology.

Objective To compare CT perfusion changes of patients 1 month after living liver transplantation with healthy volunteers. Methods Twenty perioperative patients without complications and 25 healthy volunteers underwent dynamic multi-slice CT scan. On each targeted slice, liver was divided into 3 regions of interest (ROI) according to hepatic venous drainage, i.e. anterior segment area drained by middle hepatic veins (MHV) which was most likely congested, risk area drained by both MHV and RHV which was posterior to anterior segment, reference area drained by RHV. Perfusion parameters and relative perfusion difference were calculated. Results The relative perfusion differences of anterior segment and risk area in study group were as follows: BF 44.56±43.19 vs 25.41±27.49 [ml/(min〖DK〗·100 g)]; BV 3.91±2.45 vs 2.15±1.86(ml/100 g); PVP 41.50±39.15 vs 22.66±23.82 [ml/(min·100 g)]. Significant differences of RPDs of BF, BV and PVP were found in anterior segment (P<0.0001) and risk area (P<0.01) compared with control group. Conclusion In perioperative period, there are hemodynamic differences in hepatic segments drained by RHV and MHV. BF, BV, PVP of anterior segment decreases obviously.

AIM:To investigate the effect of donor bone marrow derived dentritic cell (DC) treated with B7 - 1, B7 - 2 antisense oligonucleotide on mouse heart allografe survival time and its mechanism. METHODS: There were 7 groups of C57BL/10J (B10) mouse bone marrow DCs which were treated by 400 nM antisense oligonucleotide target to B7 -1, B7 -2 mRNA (AS B7- 1/2), B7- 1 mismatch oligo control ,B7- 2 mismatch control(mASB7- 1/2), lipofeetamine only and non-treatment, respectively. Each group of DC were named as ASB7- 1 DC, ASB7- 2 DC, mASB7 - 1 DC, mAS B7 - 2DC, and Lipo DC, respectively. RESULTS: Flow cytometer results shown that AS B7- 1/2 can inhibit B7- 1 (CD80)and B7- 2 (CD86) molecule express on DC surface, while control groups have no effects. To observe their tolerogenicity in mouse cardiac allograft model, B10→C3H heterotopic heart transplantation were performed. Recepients were received 2 x 106 of DC injection 7 days before transplantation. Results showed that both AS B7 - 1 DC and AS B7 - 2 DC can prolong mouse cardiac allograft survival time to (18.6 + 0.89) days and (23.67 + 10.73) days, respectively, compared with IL - 4 DC [ (6.22 + 0.97) days ( P < 0.01 ) ]. Two mismatch control groups can slightly prolong while oligo DC has no effect. For understanding its mechanism, each group of DC was used as stimulator to stimulated C3H spleen T cell. Results suggested that AS B7 - 1DC and AS B7 - 2 DC had less allo - stimulate function, including MLR and generation CTL and IL - 2 production than IL - 4 DC but control groups have no effect. CONCLUSION: Donor bone marrow derived DC treated with AS B7 - 1 oligo and AS B7 - 2 oligo expressed lower level of CD80 and CD86, respectively. These cells can induce allogeneic T cells anergy in vitro and markedly prolong mouse heart allograft survival time in vivo.

BACKGROUND: Studies have shown that isogeneic splenocyte transplantation can induce immune tolerance and prolong liver graft survival under the effect of immunosuppressant. It is reported that bone marrow mesenchymal stem cells (BMSC) transplantation can also prolong the liver graft survival.OBJECTIVE: To observe the antirejection effect on liver grafts by simultaneous transplantation of isogeneic BMSC and spleen tissue in liver transplant rats.METHODS: Recipient Lewis rats were randomly divided into 4 groups: in acute rejection group, DA rats as the donor and Lewis rats as the recipients for liver transplantation; in cyclosporine A (CsA) group: recipients received CsA after liver transplantation. In the BMSC group, recipient Lewis rats received simultaneous BMSC transfusion and liver transplantation. In the spleen tissue group, the recipient Lewis rats received DA rat liver, spleen tissue and isogeneic BMSC transplantation. The serum cytokine levels were determined, and liver pathological changes and survival were observed postoperatively. The level of chimerism in the spleen of Lewis rats were also observed.RESULTS AND CONCLUSION: The blood levels of alanine aminotransferase, aspartate aminotransferase and total bilirubin interleukin-2 (IL-2) and interferon gamma of spleen tissue group were significantly reduced compared with the other groups (P< 0.05), but IL-6 and IL-10 were increased (P< 0.05), and donor positive cells were increased at 30 days (P< 0.05). Liver pathology revealed a mild acute rejection in liver graft of CsA and BMSC groups, severe acute rejection in acute rejection group, but no rejection in spleen tissue transplantation group. Results indicate that isogeneic BMSCs transfusion combined donor spleen tissue transplantation can induce allograft tolerance in liver transplant rats.
