Background and purpose:Heparanase is an endo?-D-glucuronindise that degrades the HS sdie chains of heparin sulphate proteoglycans in ECM,expressed in the human normal placenta and immune organ.Expression of heparanase significantly increases in many kinds of cancers,it plays a central and specific role in the regulation of tumor metastasis and invasion.This article evaluates the effect of heparanase gene on the proliferation,adhesion and invasion of human ovarian cancer cell line SKOV3 cells.Methods:Human ovarian cancer cell line SKOV3 was cultured in optimum medium and transfected with pCDNA3.0-HPA by Lipofectamine~(TM2000),and cells transfected with blank vector pCDNA3.0 were used as control.The expression of HPA mRNA,and HPA protein in SKOV3,SKOV3/neo,and SKOV3/HPA were detected by RT-PCR and immunohistochemical staining;cell growth kinetics and the ability of invading was detected by MTT and transwell-ECM methode;the ability of adhesion and separation among same cells were evaluated by cell-adhering and separating method;Flow cytometry was used to test cell cycle.Results:RT-PCR and immunohistochemical staining showed that over-expression of HPA mRNA and protein in pCDNA3.0-HPA transfected cells in comparison with that in the parental SKOV3 cells and the cells transfected with blank vector(P

Objective Investigate the clinicopathological characteristics of h?rthle cell tumors of the thyroid. Methods Sixteen cases of HCT were studied by the clinicopathological study and immunohistochemical and review of the literature. Results There were 13 cases of h?rthle cell adenomas, 5 h?rthle cell follicular carcinomas. Immunostains showed that almost all benign and malignant HCT expressed TG, vimentin, S-100 protein. Conclusions HCT is a potential malignant tumor with a defined pathologic feature. It should be follow up for a longer time to their biological behaviors. To distinguish between the benign and the malignant depends on traditional encapsulation, and whether there is a blood vessel invasion or metastasis.

This study examined the relationship between PDGF-induced proliferation of vascular smooth muscle cells (VSMCs) and Nur77 expression and the effect of atorvastatin on VSMC proliferation and Nur77 in PDGF-treated VSMCs. Rat VSMCs were isolated and cultured. After incubation with atorvastatin or Nur77 siRNA, the cells were stimulated with PDGF and detected for BrdU incorporation to measure the proliferation of the VSMCs. Quantitative PCR and Western blotting were used to determine the Nur77 protein and the CREB phosphorylation level, to observe their relations with PDGF-induced VSMC proliferation. Our results showed that PDGF increased the BrdU incorporation in VSMCs, suggesting that it induced the proliferation of the cells. The VSMC proliferation was associated with increased Nur77 expression and elevated CREB phosphorylation. Atorvastatin inhibited the PDGF-induced VSMC proliferation, suppressed Nur77 expression. After silencing of Nur77 gene, the PDGF-induced VSMC proliferation was decreased. It was concluded that PDGF-induced VSMC proliferation was related to the Nur77 expression and CREB phosphorylation. Atorvastatin reduced the Nur77 expression and, at the same time, inhibited the VSMC proliferation.

Transition metal oxide ZnO nanoparticles as inorganic matrix was adopted in the matrix assisted laser desorption ionization time-of-flight mass spectrometry ( MALDI-TOF-MS ) analysis of small molecule compounds including sugars and stearic acid and its performances were investigated. In addition, the feasibility of metal ion selective ionization of L-arginine by using NiO and CuO nanoparticles as matrix directly was explored initially. In the experiment, the analyte and nanoparticle suspension were mixed on the sample plate. The crystal of analyte and matrix was excited by 337 nm laser after the solvent was evaporated, and then detected under reflection mode of TOF-MS. The results indicated that ZnO nanoparticles with high UV absorption could eliminate the interference of conventional matrix, simplify the mass spectra, and enhance the peak intensity of stearic acid under negative mode. Moreover, Cu+ ion selective ionization of L-arginine was achieved in comparison of CuO and NiO as matrix for MALDI-TOF-MS analysis of L-arginine.

Objective To observe and explore the therapeutic effect of acupuncture combined rehabilitation training in treating with spasm cerebral palsy patients.Methods 128 spasm cerebral palsy patients in our hospital from February 2012 to May 2014 were selected and randomly divided into the observation group (68 cases) and the control group (60 cases).The patients of the control group were given massage,ganglioside intravenous treatment,the patients of the observation group were given acupuncture combined rehabilitation training on the basis of the control group.The total treatment was 3 months,after the treatment,the Ashworth score and the total effective effective rate of the two groups were compared.Results After treatment,the observation group of patients with Ashworth spasm rating scale significantly better than that of control group (P ＜ 0.05),the observation group of patients and the control group after treatment in patients with Ashworth spasm of rating scale were significantly better than that of before treatment (P ＜ 0.05),the treatment of patients with observation group total effective rate is significantly higher than control group (P＜ 0.05).Conclusions On the basis of the comprehensive treatment of acupuncture combined rehabilitation training therapy in patients with spastic type cerebral palsy treatment effect significantly,can significantly improve the patient' s level of limb muscle tension,worthy of clinical popularization and application.

Objective To determine the contents of geniPoside in different Parts of the fruit of Gardenia jasminoides Ellis with different maturity degree Produced in fangxian,in order to find the oPtimum harVest Periods and the medicinal Parts. Methods The maturity of gardenia fruit are diVided into four leVels according to the color from totally green to yellow and red, and then the contents of geniPoside were determined by HPLC. Results The contents of geniPoside in all four leVels of maturity reached the standard by the Chinese Pharmacopoeia. The content of geniPoside in fruits of caesious color is uP to 13. 01%. Conclusion There is a negatiVe correlation between the maturity and the content of geniPoside in the mature fruit of Gardenia jasminoides Ellis. Caesious fruit has the best maturity degree. It is reasonable to use the whole fruit as medicine according to the Chinese Pharmacopoeia.

Objective To study the expression and clinical significance of serum levels of transforming growth factor-31 (TGF-β1) and tumor necrosis factor-alpha(TNF-α) in patients with hypertensive disorder complicating pregnancy(HDCP).Methods The serum levels of TGF-β1 and TNF-αin 50 cases with HDCP and 30 cases of normal third trimester pregnant women(control group) were detected by ELISA.50 cases with HDCP were divided into gestational hypertension group,mild preeclampsia group and severe preeclampsia group according to the severity of HDCP.Results The serum levels of TGF-β1 and TNF-α in HDCP patients were significantly higher than the control group (t =13.283,13.607,all P ＜ 0.05).The serum levels of TGF-β1 and TNF-α gradually increased with the aggravating of HDCP disease,the serum levels of TGF-β1 and TNF-αwere significantly different among the three subgroups of HDCP(P ＜ 0.05).The correlation analysis showed that the serum levels of TGF-β1,TNF-α were positively correlated with the severity of HDCP (r =0.575,0.512,all P ＜ 0.05),there was significantly positive correlation between the serum TGF-β1 and TNF-α(r =0.515,P ＜0.05).Conclusion The serum levels of TGF-β1 and TNF-α were high in the HDCP patients,the high levels of TGF-β1 and TNF-α can reflect the changes and development of HDCP disease.The abnormal levels of TGF-β1 and TNF-α may be associate with dysfunction of trophoblast cell.

Objective To explore the correlation between the serum levels of interferon (IFN)-γ,interleukin (IL)-18,IL-10 and recurrent spontaneous abortion (RSA).Methods Enzyme-linked immunosorbent assay was used to detect the serum levels of IFN-γ,IL-18 and IL-10.There were 40 patients with RSA (RSA group) and 40 normal early pregnant women (control group) enrolled in this study.After 2 months,those serum cytokines in two groups of non-pregnant state were detected again.Results Compared to control group,the serum levels of IFN-γ and IL-18 of RSA group were significantly higher (P ＜0.01),the serum level of IL-10 was significantly lower (P ＜ 0.01).Two months after termination of pregnancy,there was no significant difference in the serum levels of IFN-γ and IL-10 between the two groups.The serum level of IL-18 maintained a high level in the RSA group after termination of pregnancy.Conclusions In patients with RSA,the serum levels of IFN-γ and IL-18 were elevated,the serum level of IL-10 was decreased.The serum levels of IFN-γ,IL-18 and IL-10 were closely related with RSA.

Objective: To investigate the influence of Icarrin(ICA) on transforming growth factor ?2(TGF?2) expression by PG cells and the mechanisms of reversion of TGF?2 mediated immunosuppression by ICA.Methods: Cell proliferation and cytotoxici- ty were assayed by MTT assay. mRNA level of TGF?2 and perforin was detected by RT-PCR assay. assay.Expression of TGF?2 and IL- 2R? was analysed by folw cytometry assay. Results: ICA could inhibit the protein and mRNA expression of TGF?2 in PG cells. It could also reverse the supressed cytotoxicity of LAK, CD3AK cells by TGF?2. The IL-2R? expression on LAK and perforin gene transcription and proliferation of CD3AK inhibited by TGF?2 were also partly recovered. Conclusion: The anti-anti-tumor activity of ICA is at least partly due to its capability of inhibiting TGF?2 expression in tumor cells and restoring the cytotoxicity of immunocompetent cells inhibited by TGF?2.

Objective To investigate the distribution of human papillomavirus(HPV)in different age groups and its correlation with the degree of cervical lesions.Methods 2 031 patients with HPV screening specimens in our hospital from May 2012 to May 2015 were chosen as study subjects.To acquire patient cervical squamous columnar epithelial cells at the junction to detect HPV DNA types.HPV -positive patients used ultra -thin liquid -based cervical cytology technology (TCT)to detect.Patients with abnormal TCT detection were performed with electronic biopsy,and the diagnosis was made by pathology biopsy,and analyzed the distribution of HPV in different age groups and different degree of cervical lesions.Results In different age groups,≤20 years of age groups of patients with high -risk HPV positive rate was 19.7%,which was significantly higher than that of >20 -30 years,>30 -40 years and >40 -50 years three groups (14.4%,13.9%,15.0%)(χ2 =4.259,5.724,3.988,all P <0.05).There was no significant difference with the 50 years old group (17.1% )(χ2 =1.724,P >0.05).There was no significant difference in HPV positive rate among the ≤20 years,>20 -30 years,>30 -40 years,>40 -50 years,>50 years ages patients with high risk HPV infection group and low risk HPV infection group(χ2 =0.679,1.021,0.968,0.736, 0.668,all P >0.05).In the high -risk HPV infected group,the risk of single type infection in >20 -30 years,>30 -40 years,>40 -50 years,>50 years ages patients were significant higher than the mixed types infection(χ2 =4.213,3.894,4.256,5.330,5.666,all P <0.05).Infected patients of all ages with high -risk HPV were HPV16, HPV52,HPV58 type -based,low -risk type HPV43 type places mainly.More than 31 to 40 years age group of patients infected with high -risk HPV types in cervical lesions mainly cervicitis and 40 years and older group of patients with high -risk HPV infection incidence of invasive cervical cancer were significantly higher.Conclusion Different age groups of women for HPV genotyping is important for diagnosis of cervical lesions.
