2.Pathological changes of brain tissues from contused and contralateral non-contused sides and their significance
Chinese Journal of Trauma 2014;30(8):827-830
Objective To investigate the pathological changes of brain tissues from contused and contralateral non-contused sides and their significance. Methods A total of 100 healthy adult Wistar rats were assigned to brain contusion group and sham operation group according to the random number table.Ten rats in each group were sacrificed at 1,3,6,12,and 24 hours respectively.Brain samples were collected to perform pathophysiological analysis of brain tissues and test blood brain barrier (BBB)permeability by semiquantitative immunohistochemical staining of IgG.Results There was no damage to the bilateral brain tissues in sham operation group and IgG stain was negative. In brain contusion group, angioedema characterized by the breakdown of BBB was seen in the contused side at 1 hour followed by cellular edema at 3 hours, with aggravation of both over time. Moreover, tissue necrosis, inflammatory cell infiltration, and microglia proliferation emerged at 12 hours.Besides,IgG-positive staining was seen at 1 hour, was strongest at 6 hours, and remained a high level at 24 hours. With respect to the non-injured side in brain contusion group,no pathological abnormalities and negatively stained IgG were observed at 1 hour; cellular edema and weakly positive-stained IgG were found at 3 hours; aggravated cellular edema, emergence of angioedema, and IgG strongly positive staining were noted at 6 hours;cellular edema continued aggravation, but angioedema tended to be attenuated,IgG staining weakened, and microglia proliferation were observed at 12-24 hours. Conclusion Pathological changes of brain tissues from the contused and contralateral non-contused sides are differed, which provides a basis in determining treatment choices.
3.Pathogenesis and interventions of cancer-related sarcopenia
Chinese Journal of Clinical Nutrition 2015;23(3):183-188
The pathogenesis of cancer-related sarcopenia is complicated, involving all aspects of tumor and the body's metabolism, mainly related to accelerated proteolysis and insufficient protein synthesis.Cancerrelated sarcopenia can predict antineoplastic-induced dose-limiting toxicities, increase complications, and reduce survival rate. The interventions of cancer-related sarcopenia includes nutrition, exercise, hormone therapy and other interventions.A single treatment can not reverse clinical outcome.Various interventions are still in the investigation stage,in which more evidence-based studies are needed.
4.The variation of ADC values and pathological change in non-injured side tissues of traumatic brain injury of rat
Journal of Practical Radiology 2014;(6):1026-1029,1034
Objective This study was designed to investigate the correlation between the apparent diffusion coefficient(ADC)value and pathological change of the non-injured side brain tissue of traumatic brain injury(TBI)of rat. Methods Sixty healthy adult Wist-ar rats were randomly divided into two groups:the control or TBI groups. The TBI group was divided into five sub-groups according to the different time intervals:1,3,6,12,24 h(n = 10).The animal brain of each group was scanned with MR-DWI,and the ADC value of damaged areas and contralateral non-damaged areas were measured. After that the brains were taken out at different time points after TBI.The cerebral edema and blood-brain barrier(BBB)changes in structure were examined with an optical micros-copy and transmission electron microscopy, and the IgG content in the same tissues were determined by means of immunohistochem-istry.The data was analyzed with SPSS 13.0 statistical software. Results There was no signal abnormality on MR-DWI and tissue structure in control group, and the IgG stain was negative. In the contralateral non-damaged areas of TBI group, pathological obser-vation revealed no variation at 1 h after TBI,and the IgG stain was also negative.Cellular edema was shown at 3 h and the IgG stain was slight positive. The cellular edema aggravated with time and angioedema appeared at 6 h.IgG stain was shown significantly posi-tive.At 12~24 h,cellular edema kept increasing more severe, however angioedema had a trend to mitigated along with time,IgG stain became slight and the proliferation of glial cells could observed.Compared with control group, the ADC values of the contralat-eral non-damaged areas in each group showed no significant difference (P >0.05).However,in the damaged areas of each group, angioedema appeared at 1 h and gradually aggravated, cellular edema occurred at 3 h,Both of them were aggravated with time. ADC values increased at 1 h,and then decreased until 6 h,then followed a climbing up to 24 h,showed as a “V”-shaped.Con-clusion When contused on one side brain, the contralateral non-inj ured side tissue also has pathological changes that occurs later than those on the inj ured side.Cellular edema of the tissues ap-peares first and is followed by angioedema, which lessens over time..There is no significant difference of ADC values between con-tralateral side of TBI groups and control group(P >0.05),which reveals a “pseudo-normal”phenomenon.

Chinese Journal of Biochemical Pharmaceutics 2017;37(5):353-355
Objective To investigate the clinical efficacy and mechanism of minocycline hydrochloride ointment adjuvant therapy for chronic periodontitis.Methods86 patients of chronic periodontitis who received therapy from August 2013 to June 2015 in our hospital were selected.According to random number table, those patients were divided into the experimental group (n=43) and the control group (n=43).The control group was treated on the basis of the foundation treatment with tinidazole Oral Sticking Tablets and the experimental group were treated on the base of the foundation treatment with Minocycline hydrochloride sustained-release ointment, the levels of hs-CRP, TNF-a, IL-8 and IL-10 in gingival sulcus fluid were measured before and after treatment, the indexes of PLI, PD, SBI and AL were recorded,at the same time the clinical curative effect between two groups was compared.ResultsThe effective rate of the experimental group(95.35%) was higher than the control group(81.40%),the difference was statistically significant (P<0.05);Compared with before treatment, two groups general situations and inflammation factors changed after treatment, plaque index, periodontal pocket probing depth, gingival sulcus bleeding index and adhesion levels of the experimental group were lower than the control group after treatment, the PD, SBI, AL and PLI in the experimental group were significantly lower than that of the control group, and the hs-CRP, TNF-a and IL-8 levels were lower than the control group, IL-10 was higher than the control group after treatment(P<0.05).ConclusionMinocycline hydrochloride ointment is well for chronic periodontitis, which can effectively relieve periodontal chronic inflammation, improve clinical efficacy, the underlying mechanism may be related to the reduction of IL-8 and TNF-α levels in gingival crevicular fluid.
7.Formation of cell sheet on acellular porcine corneal stroma with human corneal endothelial cells cocultured by mouse embryonic stem cell conditioned medium
Chinese Journal of Experimental Ophthalmology 2016;34(8):705-709
Background Corneal transplantation faces a great challenge because of the shortage of corneal donors and difficulty of human corneal endothelial cells (HCECs) regeneration in vitro. So the study on tissue engineering cornea is still a main topic.Previous research showed that mouse embryonic stem cell conditioned medium (ESC-CM) improved the proliferative capacity of HCECs in vitro, and acellular porcine corneal stroma (APCS) was a good saffold material. However, whether HECEs cultured by mouse ESC-CM can form cell sheet in vitro were rarely studied. Objective This study was to investigate the potential that HCECs cultured by mouse ESC-CM form a monolayer cell sheet. Methods The supernatant of ESC-CM was collected after mouse ES-E14 cells were cultured, and the cultured medium was centrifuged and mixed with 75% human corneal endothelium medium (CEM)at a proportion of 1 ∶ 3 to prepare the 25% ESC-CM system.Primary cultures of HCECs were established from explants of corneal limbal with Descemet's membrane, and the cells were identified by using reverse-transcription PCR to determine the expressions of collagen Ⅷ (Col Ⅷ) mRNA and neuron-specific enolase (NSE) mRNA in the cells.APCS was prepared by decellularization with phospholipase A2 and bicarbonate solution, and the second generation of HCECs were inoculated on the sterilized APCS at a 800/mm2 density. The morphology of the cells was observed by hematoxylin-eosin staining under the phase-contrast microscope. The expressions of zona occludens protein-1 (ZO-1)and Na+-K+-ATPase in the cell sheet were detected by immunofluorescence staining. Results The second generation of HCECs cultured with 25% ESC-CM in vitro showed the hexagon in shape with positive expressions for Col Ⅷ mRNA and NSE mRNA.Decellularization APCS was transparent, and no corneal cells were seen, the structures of corneal collagenous fibres were regular.HCECs attached closely to APCS and formed monolayer sheet 7 days after culture on the APCS with the cell density of (2 694±143)/mm2.ZO-1 and Na+-K+-ATPase were positively expressed on the HCECs sheet. Conclusions Twenty-five percent ESC-CM promotes the proliferation and maintains the normal morphology of HCECs.APCS provide a good scoffold and microenvironment for the formation of HCEC sheet. The HCEC sheet Possesses the pump function of HCECs and is a good corneal donor for transplantation.

Journal of Preventive Medicine 2024;36(2):139-142
Objective:
To understand the situation of mosquito larva breeding and adult mosquito density among schools in Yangpu District, Shanghai Municipality before the start of the fall semester, so as to provide insights into mosquito prevention and control in schools.
Methods:
A stratified sampling method was used to select four schools each from universities, primary and secondary schools, and preschool institutions as survey sites. A survey of mosquito larva breeding sites and adult mosquito density was carried out in August each year from 2021 to 2023. The mosquito larva breeding sites in small and large water bodies were investigated by visual inspection and spoon trapping methods, and adult mosquito density was investigated by human-induced stinging method. The positive rate of mosquito larva breeding sites and adult mosquito landing index were analyzed.
Results:
Totally 1 253 mosquito larva breeding sites were investigated from 2021 to 2023, and the positive rate was 4.07% (51 positive sites). The positive rates of mosquito larva breeding sites in universities, primary and secondary schools and preschool institutions were 2.36%, 4.05% and 8.30%, respectively. The top three positive breeding sites in small water bodies were tyres (6.67%), small stagnant water in green belts (6.04%) and unused containers (5.92%), and Aedes albopictus was the dominant species (97.21%). The top three positive breeding sites in large water bodies were rainwater wells (3.90%), landscape water bodies (3.45%) and sewage wells (3.23%), and Culex pipiens pallens and Aedes albopictus each accounted for 50.00%. A total of 200 adult mosquitoes were captured from 2021 to 2023, with Aedes albopictus being dominant (93.00%). The total landing index decreased and then rebounded slightly from 2021 to 2023, with the highest seen in 2021 as 1.78 mosquitoes/0.5 hours per person. The highest mean adult mosquito landing index was 1.17 mosquitoes/0.5 hours per person in preschool institutions.
Conclusions
The positive rates of mosquito larva breeding sites and landing index of adult mosquitoes among schools in Yangpu District before the start of the fall semester from 2021 to 2023 were higher than the relevant provisions of the Shanghai municipal local standards. Aedes albopictus was the dominant species. Mosquito surveillance in schools should be carried out on a regular basis and hygiene management should be strengthened.
