1.Correlation analysis on beta amyloid protein and expression of amyloid precursor protein gene in 32 patients with Alzheimer disease for 3 years in Yimeng mountainous area
Chinese Journal of Tissue Engineering Research 2006;10(6):173-175
BACKGROUND: Deposition of beta-amyloid protein is the main cause of Alzheimer disease. Overexpression of β-amyloid precursor protein gene (βAPP-gene) or mutation of it at certain part can be related with the attack of Alzheimer disease.OBJECTIVE: To observe the relationship of Alzheimer disease and β-amyloid protein with β APP-gene in Yimeng mountainous area.DESIGN: A case-control analysis study.SETTING: Department of Neurology, Linyi Municipal People's Hospital.PARTICIPANTS: Thirty-two Alzheimer disease inpatients were enrolled at the Department of Neurology, Linyi Municipal People's Hospital affiliated to Linyi Medical Professional College between January 2001 and December 2003. Thirty elderly patients who were treated with surgery of noncraniocerebral injury and were of the same sex and the similar age were regarded as the members of the control group [score of Mini Mental State Examination (MMSE) was over 27 points].METHODS: Level of β-amyloid protein in cerebrospinal fluid was detected with enzyme-linked immunoadsordent assay (ELISA). Expression of βAPP-gene was measured with the technic of fluorescent quantitative polymerase chain reaction.RESULTS: A total of 62 cases were involved in the result analysis. ①The level of β-amyloid protein in cerebrospinal fluid in the Alzheimer disease group was higher significantly than that in the control group[(13.63±9.34), (6.75±5.37)μg/L,t=3.354, P < 0.01]. ② The expression ofβ APP in cerebrospinal fluid in the Alzheimer disease group was higher significantly than that in the control group [(1 624±298), (1275±269)copy/μL,t=4.42, P < 0.01]. ③ The level of β-amyloid protein was posit ively correlated with the expression of β APP-gene in the Alzheimer disease group (r=0.44,P< 0.01).CONCLUSION: The level of β-amyloid protein and the expression of β APP-gene in cerebrospinal fluid of Alzheimer disease patients in the Yimeng Mountainous area increase. Besides, the higher the level of β-amyloid protein, the higher the expression of β APP-gene is, and the severer the degree of dementia is. It indicates that Alzheimer disease has closely relation with β-amyloid protein and β APP-gene. But the expression of the β APP-gene has no specificity in the diagnosis of Alzheimer disease.

Objective To investigate the impact of high plasma LDL-C level with or without metabolic syndrome(MS) on the incidence of stroke in Chinese adults. Methods Totally 42 626 subjects (25 -75 years old) from Chinese National Health and Nutrition Survey in 2002 were stratified four groups based on plasma LDL-C level: < 2. 00 mmol/L group, 2. 00 -2. 50 mmol/L group, 2. 51 -3.31 mmol/L group, and ≥ 3.32 mmol/L group. The prevalence of MS (with 2005 International Diabetes Federation criteria) and stroke and the risk factors of stroke were compared among the four groups. Results ( 1 ) The prevalence of MS and stroke increased with rising of LDL-C level. The prevalence of MS in LDL-C≥3. 32 mmol/L group increased 2. 5 times (7, 9% vs 20. 1% ) as compared with that in LDL-C < 2. 00 mmol/L group and the prevalence of stroke increased 4. 2 times(0. 5% vs 2. 1% ), all P <0. 01. (2) In subjects with similar LDL-C level, the prevalence of stroke was significantly higher in a subgroup with MS than that without (P <0. 01 ). (3) After adjustment for age, sex and smoking, logistic regression analysis showed that both LDL-C level and MS were positively associated with the development of stroke; the odds ratio (OR) was 2. 35 and 3. 15 ( P < 0. 0001 ), respectively. (4) Compared with the subgroup of LDL-C < 2. 00 mmol/L without MS, OR for stroke in the subgroups of LDL-C 2. 00 -2. 50 mmol/L, 2. 51 -3. 31 mmol/L, and ≥ 3. 32 mmol / L without MS was 1.03, 1. 89, and 2.08, whereas the OR for stroke in the subgroups with MS and similar level of LDL-C was 4. 38, 5.23 and 6. 15 ; this indicated that the risk of stroke in subjects with MS increased by 3 - 4 times compared with subjects without ( P < 0. 0001 ). Conclusion Both high LDL-C level and MS are independent risk factors of stroke, but the risk of stroke will be further increased in the presence of high LDL-C level plus MS. It is suggested that combined intervention therapy of LDL-C and MS will play an important role in the prevention of stroke.

Objective: To explore the risk factors for heart failure (HF) occurrence in patients with inferior or combining right ventricular myocardial infarction (MI).
Methods: A total of 143 patients with inferior or combining right ventricular myocardial infarction (MI) in our hospital from 2013-01 to 2015-06 were studied. Based on HF occurrence, the patients were divided into 2 groups:HF group, n=81 and Non-HF group, n=62. The risk factors related to HF occurrence as age, gender and hypertension were retrospectively studied and compared between 2 groups.
Results: Compared with Non-HF group, HF group had more patients with hypertension, female gender, higher blood levels of Hs-CRP and creatinine, more volume of lfuid input on the 3rd day of admission and the higher ratio of V4-6 ST segment depression in ECG; while obviously lower diastolic blood pressure (DBP) at admission. Multivariate Logistic regression analysis indicated that hypertension (OR=3.275, 95% CI 1.220-8.793), female gender (OR=13.236, 95% CI 3.476-50.405), Hs-CRP (OR=1.029, 95%CI 1.005-1.070), low DBP at admission (OR=0.945, 95%CI 0.911-0.979), serum creatinine (OR=1.053, 95%CI 1.029-1.078) and V4-6 ST segment depression in ECG (OR=4.118, 95%CI 1.395-12.154) were positively related to HF occurrence in relevant patients.
Conclusion: The incidence of HF has been relatively high in patients with inferior or combining right ventricular MI;hypertension, female gender, low DBP at admission, blood level of Hs-CRP, serum creatinine, V4-6 ST segment depression in ECG were the independent risk factor for HR occurrence.

Objective To investigate the effects of peptide YY (PYY) on the interdigestive migrating myoelectrlc complex (MMC) in the small intestine in vivo and explore the neural and endecrinal mechanisms of the effects. Methods Spragne-Dawley rats were supplied with a venous catheter and bipolar electrodes in the duodenum and jejunum for electromyography of stomach and small intestine in wake state. PYY, phentolamine, nitro-L-arginine (L-NNA, the inhibitor of nitric oxide synthase) and atropine were served with PYY respectively. The plasma motilin levels before and after the infusion of PYY were observed. Results At all the three recording points, PYY lengthened the drde length of MMC [from (591.90±128.98)s to (999.25±216.59)s, P<0. 01] and lowered the frequency of phase Ⅲ [from (39.28±8.40) min-1 to (22.08±3.13) min-1 , P<0.01], amplitude of phase Ⅲ [from (0. 320±0.060)mV to (0. 179±0.030)mV, P<0.01], and the portion of phase Ⅲ over the whole circle length [from (28. 61 ± 5.84)% to (15.43 ±5.16)% , P<0.01]. Phentolumine had no influence on the role of PYY. Administered L-NNA combined with PYY, the percentage of phase Ⅲ increased [(42. 09±8.67)%] compared with that of control(P<0.01) and compared with that of PYY administered alone (P<0. 01) too. Atropine combined with PYY showed stronger depressing effects on MMC. No significant difference was found between the plasma motilin levels before and after the infusion of PYY. Conclusion PYY my inhibit the interdigestive intestine motility through the none-adrenergic none-choUnergic tract, while the m-receptor tract and circulating motilin are probably not involved In the depressing effect.
