2.Clinical effect of addition and subtraction of Xiaojiyinzi prescription in the treatment of bleeding after bipolar vaporization of prostate for benign prostatic hyperplasia

Objective To explore the effect of addition and subtraction of Xiaojiyinzi prescription in the treatment of bleeding after bipolar vaporization of prostate for benign prostatic hyperplasia.Methods From June 2013 to June 2017,90 patients with benign prostatic hyperplasia who underwent bipolar electrovaporization in the Traditional Chinese Medicine Hospital of Wenling were selected in this study.The patients were divided into observation group and control group according to a random number table,with 45 cases in each group.The observation group was givenaddition and subtraction of Xiaojiyinzi prescription,while the control group was treated with finasteride orally.The clinical effects of the two groups were compared and analyzed.Results The amount of bleeding in the control group at 3 days,5 days and 7 days after operation were (13.69 ± 1.27) mL,(8.37 ± 1.23) mL,(6.13 ± 0.58) mL,respectively,which in the observation group were (9.22 ± 1.18) mL,(6.12 ± 0.98) mL,(3.98 ± 0.33) mL,respectively,there were statistically significant differences between the two groups (t =17.297,P =0.000,t =9.597,P =0.000,t =21.613,P =0.000).The amount of bleeding in both two groups decreased with the prolongation of time,and the differences were statistically significant between each two moments (all P < 0.05).After treatment,the I-PSS and NIH-CPSI scores in the control group were (13.22 ± 2.95) points and (13.64 ± 4.22) points,respectively,which in the observation group were (8.11 ± 1.35) points and (7.88 ± 3.06) points,respectively,which were lower than those before treatment,and the differences were statistically significant(t =12.286,P =0.000,t =14.359,P =0.000,t =21.041,P =0.000,t =25.989,P =0.000).The scores in the observation group after treatment were lower than those in the control group,and the differences were statistically significant between the two groups (t =8.217,P =0.000,t =7.413,P =0.000).Conclusion Hemorrhage is the most common complication of transurethral resection of prostate in elderly men,which seriously interferes with postoperative recovery.The addition and subtraction of Xiaojiyinzi prescription can reduce bleeding and improve postoperative recovery.
3.AIF Distribution and Expression in CuItured SpiraI GangIion Neuron Injured by GIutamate

Objective The study aimed to explore the relationship between AIF related pathway and the inju-ring of cultured SGNs (spiral ganglion neurons)by glutamate toxicity,and to find AIF expression and distribution changes in SGNs.Methods SGNs of 40 newborn rats within 3 day were obtained and cultured in vitro.Cultured cells were divided into four groups:the normal control group,10 mM,20 mM and 40 mM glutamate injured group, separately.After 48 h hours culturing,optical microscopy,immune fluorescence staining and real-time fluores-cence quantitative PCR were used to observe the morphology,AIF distribution,and AIF,calpain,Caspase3 expres-sion changes in SGNs in vitro.TUNEL was used to verify the cell apoptosis.ResuIts Noticeable morphological chan-ges and cell apoptosis were occurred in 20 mM glutamate group,with AIF nuclear translocation.AIF gene expression was significantly higher than normal after glutamate administration (P<0.05);moreover,calpain gene expression increased(P<0.05);but caspase3 expression was not statistically significantly increased in all glutamate treated groups (P>0.05). ConcIusion In the process of cultured SGNs injured by glutamate,AIF participated in the cell apoptosis.Noticeable cell apoptosis were occurred in 20 mM glutamate group with AIF nuclear translocation.Calpain up-expression also contributed to excitatory neurotransmitter injury on SGNs,but Caspase 3 had no obvious effects.
4.Antibacterial mechanism of silver nanoparticles:a dose-dependent promotion of cell apoptosis

BACKGROUND:Silver nanoparticles (AgNPs) show strong antibacterial effect and are not easy to have drug resistance. But the antibacterial mechanisms of AgNPs have not been wel developed.
OBJECTIVE:To explain the antibacterial mechanisms of AgNPs.
METHODS:We investigated the influence of Ti, TiO2 and TiO2 containing AgNPs onEscherichia coliand Staphylococcus aureus by bacterial inhibition ring test. Escherichia coli was cultured in LB liquid medium with 0, 5, 10 mg/L AgNPs. We measured the absorbance value of bacterial culture. DNA gel electrophoresis was used to study the effect of AgNPs onEscherichia coliDNA. Then we researched the character of apoptosis on Escherichia coli by Annexin V and PI staining, using flow cytometry.
RESULTS AND CONCLUSION:The inhibiting effect of Ti and TiO2 onEscherichia coliandStaphylococcus aureus was not obvious. But the inhibition rings of TiO2 containing AgNPs to bacteria appeared. The absorbance value of Escherichia coliculture was reduced whenEscherichia coliwas co-cultured with AgNPs. And this decrease tendency was in direct proportion with AgNPs concentration. AgNPs reduced the amount of DNA of Escherichia coli and this tendency was directly proportional with AgNPs concentration. TheEscherichia coli apoptosis rate induced by AgNPs was increased and this tendency was positively correlated to the AgNPs concentration. These results indicate that AgNPs can induce bacterial apoptosis to influence the growth of bacteria.
5.Expression and clinical significance of bone morphogenetic protein 3 in hi-lar cholangiocarcinoma tissues

[ ABSTRACT ] AIM: To investigate the expression and clinical significance of bone morphogenetic protein 3 (BMP3) in hilar cholangiocarcinoma tissues.METHODS: Thirty cases of hilar cholangiocarcinoma specimens were col-lected.The expression of BMP3 at mRNA and protein levels in the tumor tissues and paracancerous tissues was detected by real-time PCR and Western blot.The hilar cholangiocarcinoma paraffin-embedded specimens (n=103) were collected. The protein expression of BMP3 was determined by immunohistochemical method, and the relationship of BMP3 protein ex-pression with clinical pathological characteristics was evaluated.RESULTS:In the 30 patients with hilar cholangiocarcino-ma, the expressions of BMP3 protein and mRNA in 22 cases of tumor tissues were significantly decreased compared with the adjacent normal tissues.The results of immunohistochemistry showed that 87 cases were negative and 16 cases were weakly positive in all 103 cases of hilar cholangiocarcinoma.The expression of BMP3 protein was associated with the tumor TNM staging, lymph node metastasis and tumor differentiation (P<0.05).CONCLUSION:BMP3 gene might be inhibited in human hilar cholangiocarcinoma.The down-regulation of BMP3 gene might be associated with the carcinogenesis and devel-opment of hilar cholangiocarcinoma.
6.A comparative study of two hyperthyroidism models induced by Yersiniaenterocolitica and Euthyrox

Objective To provide a good evaluation for research the hyperthyroidism drugs , two kinds of hyperthyroidism animal models were copied , and compared the similarities and differences of their formability and sustainability.Methods Euthyrox-induced model: The rat was taken Euthyrox (50μg· 100g-1 rat weight) for 21 d. And then the blood and urine were collected in 1 d, 7 d and 14 d after last time oral administration of Euthyrox;Yersiniaenterocolitica-induced model:the rats were injected Yersiniaenterocolitica (YE) in 0、5、10、15、20 (IOCV).And then the blood and urine were collected in 5d, 15d and 25d after last IOCV of YE.Results Euthyrox-induced model:T3, T4, FT3, FT4 were increased significantly , but returned normal 1 week later in the model group , PCA showed that model group deviated from the blank group , but recovered 2 weeks later; Yersiniaenterocolitica-induced model: T3 and T4 were increased significantly in 15 and 25 d, TSH significant raised in 25 d, the model group deviated from the blank group in PCA, and didn’t return to the blank group in 15d or 25d.Conclusion Euthyrox-induced modelhas good formability butrecovered quickly , which is suitable for prevention of drug delivery on efficacy evaluation; Yersiniaenterocolitica-induced model with good formability and sustainability could be used for treatment of drug delivery on efficacy evaluation .
7.Design and application research of PE right ventricular catheter for mice

Objective To establish a simple ,low cost and stable method to detect right ventricular pressure in mice .Methods A PE-50 duct length 15 cm(outside diameter :0 .9 mm ,inner diameter :0 .5 mm) was bent on one terminal and the other terminal was inserted into a 7# syringe needle to connect to a pressure transducer .This duct was intubated into right ventricle via right external jugular vein to detect right ventricular pressure in 80 SPF grade male C57BL/6 mice .Successful cases and operation time were re-corded .Besides ,40 SPF grade male C57BL/6 mice were randomized into the control group (n=20) and chronic hyperbaric hypoxia group(n= 20) .Mice in chronic hyperbaric group were raised in a hyperbaric chamber of simulated 5 000 m high altitude for 4 weeks .The control group was raised outside the chamber simultaneously .Right ventricular systolic pressure was detected with the PE duct .Left and right ventricles were detached and weighed ,and Hermann-Willson index was calculated .Results With this PE duct ,right ventricular intubation success rate was 90% (72/80) ,the operation cost approximately 3 to 5 min each mouse from the separation of blood vessels to detect the time needed for the right ventricle waveform .right ventricular systolic pressure[(39 .52 ± 4 .34 )mm Hg] and Hermann-Willson index(0 .356 ± 0 .039)of chronic hyperbaric hypoxia group were significantly higher than that of control group [(21 .24 ± 2 .7)mm Hg and (0 .256 ± 0 .020)] ,which has significant positive correlation (P<0 .01) .Conclusion It is simple ,fast ,stable ,costing low and of high success ratio to detect right ventricular pressure with this method .
8.Influences of R2O-Al2O3-B2O3-SiO2 system glass and superfine alpha-Al2O3 on the sintering and phase transition of hydroxyapatite ceramics.

The effects of R2O-Al2O3-B2O3-SiO2 system glass and superfine alpha-Al2O3 on the sintering and phase transition of hydroxyapatite (HAP) ceramics were assessed. The results showed that alpha-Al2O3 impeded the sintering of HAP and raised the sintering temperature. When glass and alpha-Al2O3 were used together to reinforce HAP ceramics, better results could be obtained; the bending strength of multiphase HAP ceramics approached 106 MPa when 10% (wt) alpha-Al2O3 and 20%(wt) glass were used and sintered at 1200 for 1 h.

9.Microstructure and adhesion of HAP-glass-alumina composite gradient bioceramics coating.

HAP-Glass-alpha-Al2O3 gradient coating on alumina ceramics substrate was prepared by multi-layer slurry-dipping and sintering process, effects of the composition of the coating on the microstructure and adhesion strength of the gradient coating were studied. R2O-Al2O3-B2O3-SiO2 system glass with low softening temperature and thermal expansion was used in the gradient coating. The composition of the coating was varied from interface to surface layer, i.e. the HAP concentration increased and glass concentration decreased gradually from the under layer to the surface layer. A suitable amount superfine alpha-Al2O3 was added in the gradient coating to reinforce the bonding strength between the coating and substrate. The results show that the use of R2O-Al2O3-B2O3-SiO2 system glass is propitious to sintering the coating at low temperature and to combination between the coating and substrate. The addition of superfine alpha-Al2O3 can reinforce the bonding strength between the coating and substrate, the highest bonding strength is about 48.2 MPa.

10.Effect of Visceral Pain and Water Metabolism on IBS-D Model Rats by Moxibustion and Its Products

This study was aimed to investigate the effect of visceral pain and water metabolism on irritable bowel syndrome with diarrhea (IBS-D) model rats by moxibustion and its products. A total of 40 SD rats were randomly di-vided into the normal group, model group, moxibustion group, non-smoke moxibustion group, and moxa smoke group. IBS-D model was induced by colorectal distension with an inflatable balloon. The general condition of rats was ob-served. Visceral pain was assessed by abdominal withdrawal reflex (AWR) scores. The immunohistochemical method was used in the detection of claudin-1, AQP3, AQP8 and Na+/K+-ATPase protein expression. The results showed that compared with the normal group, the AWR scores, claudin-1, AQP3, AQP8 and Na+/K+-ATPase protein expres-sion in the colon were decreased in the model group (P < 0.01). The stool was loose, which was on hair around the anus. After moxibustion, non-smoke moxibustion and moxa smoke stimulation, the stool conditions of IBS-D model rats were improved. The AWR scores were decreased in different degrees (P < 0.05 or P < 0.01). Except Na+/K+-ATPase, expressions of claudin-1, AQP3 and AQP8 in colon were increased in different degrees (P < 0.05 or P <0.01). The comprehensive therapeutic effect showed that moxibustion was better than non-smoke moxibustion; and non-smoke moxibustion was better than moxa smoke stimulation. It was concluded that the combination of warm stimulation, optical radiation and moxa smoke induced best effect in improving visceral pain and water metabolism on IBS-D model rats. Among them, warm stimulation and optical radiation may play the main role.
