1.Establishment and Biological Characteristics of Two Newly Human Hepatocellular Carcinoma Cell Lines LXY-1 and XMS-1

Objective In order to provide experimental model for the research of Human hepatocellular carcinoma(HCC), characterize two newly human hepatocellular carcinoma cell lines established from fresh tumor tissues of two patients with HCC infected HBV.Methods The two cell lines were analyzed by morphology, double time, isoenzyme pedigree(LDH), AFP、HBsAg detection, karyotype analysis, heterotransplantation utilizing hematoxylin and eosin、ELISA、flow cytometry、Southern blot and so on. Results The two cell lines have been in continuous culture over 50 passages, which showed typical epithelial-like cells in morphology; the doubling time of cell population were found to be 19.5~59.2 hours,which showed aneuploidy and revealed chromosome average number ranging from 50 to 105. Hepatitis-B-virus(HBV)DNA was integrated in the genomes of XMS-1.No lines produced alpha-fetoprotein(AFP) and HBsAg at the protein level.Their LDH isenozyme spectrum were similar to those of embryonic liver and clinical hepatoma with the increase of the percentage of LDH-1(H-type); Cell cycle analysis showed that the ratio of cells in S、G2-M phase increased for the both cell lines compared with healthy human peripheral blood lymphocyte; LXY-1 produced solid tumors after subcutaneous heterotransplantation into nude mice and the tumor formation ratio of heterotransplantation were 100%.Conclusions The two cell lines maintained the biologic characteristics as human hepatocellular carcinoma, and will serve as a model for further studies of HCC.
2.Relationship between the Expression of H19 and IGF-Ⅱ Imprinted Gene in Newborn Placenta and the Birth Body Mass

Objective To study the expression of the imprinted gene H19 and IGF-Ⅱ in newborn placenta,and to discuss its influence on the birth body mass of the neonate. Methods The fresh placental tissues from full-term newborn (without trimester of pregnancy complica-tion and placenta and funic abnormality) with normal,high and low birth body mass (12,10 and 8 samples respectively)were collected. The expression of imprinted gene H19 and IGF-Ⅱ mRNA in the placenta were estimated by reakime fluorescence quantitative PCR Results The ex-pression of H19 mRNA in the placenta was negative correlation to the birth body mass (r =-0.403,P = 0.027).The expression of of IGF-H mRNA was positive correlated to the birth body mass (r = 0.444,P = 0.014). The H19 mRNA expression level in the high birth weight neonates (0.21 0.31) was significantly lower than that in the low birth body mass neonates (1.51 2.04)(P= 0.013). But the expression level of IGF-Ⅱ mRNA in the high birth body mass neonates (2.67±3.41) was significantly higher than that in the low birth body mass neonates (0.39±0.33)(P =0.013). Conclusion The expression of H19 and IGF-Ⅱ mRNA was significantly different in the placenta of normal,high and low birth body mass newboms. These two genes may be related to the birth body mass,and there may be some realation-ship between these two genes.
3.Evaluation the significance of body mass index in diagnosing macrosomia in human neonate

Objective We compared the difference of diagnosing macrosomia using the body mass index (BMI)and body mass,so as to investigate whether BMI play an important role in the diagnosis and management of macrosomia in our clinical work.Methods We analysed 5522 newborns (without any maternal complication)delivered in Shengjing Hospital of China Medical University from Jan.2004 to Apr.2009,all of them were full term,singleton and with the birth body mass larger than 2500 g,among them 4989 were in the group with body mass <4000 g,that was 2510-4000 g.533 cases were in the group of body mass ≥4000 g.By both body mass and length,we got the BMI.According to statistical receiver operating characteristic curve(ROC),we determined the cutoff of BMI for diagnosing macrosomia,in addition the sensitivity and specificity of it. Using this newly gotten BMI cutoff as a method to diagnose macrosomia and analyse the results.Results (1)When the newborns with birth length 40-43 cm.the mean birth body mass was(3010 ±351)g,BMI was(17.0 ±2.7)kg/m2;the newborns with birth length 48-51 cm,the mean birth body mass was(3450 ±313)g,BMI was(13.2±1.4)kg/m2;newborns with birth length 56-60 cm,the mean birth body mass was(4332 ±456)g,BMI was(12.5 ±1.3)kg/m2,The longer the birth length,the larger the birth body mass,while the less BMI.(2)Determined by ROC curve,the BMI value could be used to diagnose macrosomia was 14.2 kg/m2.with sensitivity of 78.4% and specificity of 85.0%, the area of under curve was 0. 892. (3) By the BMI cutoff ( 14. 2 kg/m2 ), 111 macorsomia with birth body mass ≥4000 g were not macrosomia any more (20. 8%, 111/533 ),422 still were macrosomia (79.2% ,422/533) ; while for those birth body mass <4000 g, 728 were macrosomia determined by this BMI cutoff ( 14. 59%, 728/4989 ), 4261 were still not macrosomia ( 85.41%, 4261/4989 ). Using BMI cutoff 14. 2 kg/m2 to diagnose macrosomia, within the group of birth body mass ≥4000 g, their birth length in macrosomia and non macrosomia was (52. 2 ± 1.8) cm and ( 55.6 ± 1.3 ) cm respectively, the difference was significant (P <0. 01 ) ;while within the group with body mass <4000 g, the birth length of macrosomia and non-macrosomia was (49.0 ±2. 2) cm and (50. 8 ±2. 2) cm respectively,the difference was significant as well (P <0. 01 ). The whole incidence of macrosomia was 20. 83% (1150/5522) determined by this BMI cutoff. Conclusions Birth body mass and BMI in determining macrosomia show some bias, and birth length relates with this difference, which suggests birth length maybe play an important role in determine the macrosomia. We suggest it is very necessary to use BMI≥ 14. 2 kg/m2 in the diagnosis and management of macrosomia.
4.The Study of Oral Health-Related Quality of Life (OHR-QOL) among 471 Elderly Patients with Chronic Periodontitis in Kunming City

Objective To research the relationship between quality of life among the elderly with chronic periodontitis (CP) and the severity of CP.Methods A total of 471 elderly patients seeking treatment in the affiliated Stomatology Hospital of Kunming Medical University from September 2013 to January 2014 and their companions were investigated using the questionnaire according to the Chinese version of the Oral health Impact Profile-14.Those who diagnosed with CP were separated into two groups according to Community periodontal index (CPI) through periodontal examination.Group A was diagnosed with the early CP,and Group B was diagnosed with the late CP.Database was established by Epidata and statistical analysis was performed by SPSS.Results A total of 237 elderly patients with early CP and 139 with late CP were recorded.The mean OHIP-14 scores of the former were 14.36 ± 6.67,and the score of the later were 15.65 ± 7.12 relatively.The two groups who lived through negative effects were compared.The results suggested in the 14 items of OHIP-14.The statistical significance existed in the following 4 items,obvious pain appeared,uncomfortable when eating anything,unsatisfied with own diet,become easy to lose one's temper to others (P<0.05).Conclusion OHR-QOL of elderly patients with the late CP is lower than that of those with the early CP.
5.Value of elastography in predicting central lymph nodes metastasis of papillary thyroid microcarcinoma

Objective To investigate the value of elasticity score and elasticity strain ratio (SR) in predicting the central lymph nodes metastasis of papillary thyroid microcarcinoma (PTMC). Methods This study conducted a retrospective analysis of ultrasonography ifndings of 236 thyroid lesions from 208 patients. All of them underwent surgery in Ruijin Hospital of Shanghai Jiao Tong University from June 2012 to March 2013. Totally 82 PTMC from 82 patients were included in this study. Among them 12 patients with 12 PTMC had central lymph nodes metastasis. The elasticity score and SR were compared between metastasis and non-metastasis groups, and the receiver operating characteristic (ROC) curve of SR was calculated. Results The differences of PTMC elasticity score between two groups showed no statistical signiifcance (χ2=5.00, P=0.08). The average SR of the group with central lymph nodes metastasis is 2.44±0.61, which is higher than the group without lymph nodes metastasis whose average SR is 1.67±0.42. Differences are statistically signiifcant (t=5.5, P=0.00). The area under curve of SR-ROC curve was 0.847. Using 2.01 as cutoff of SR, the sensitivity, speciifcity, accuracy of predicting central lymph nodes metastasis was 83.3%, 81.4%, 78.6%respectively. Conclusion The SR but not elasticity score could be a meaningful indicator for predicting lymph nodes metastasis.
6.Measurement of ABO blood group antibody levels using renal cortical tubular epithelial cells of cynomolgus monkey

Objective To investigate the possibility of measuring ABO blood group antibody levels using renal cortical tubular epithelial cells (RCTECs) of cynomolgus monkey.Method The primary RCTECs were isolated from cynomolgus monkey kidneys and identified by Western blotting and flow cytometry (FACS).FACS was applied to detect the expression of ABO histo-blood group antigens on RCTECs,the binding of blood group antibodies in human serum to RCTECs,and to compare the difference of measuring ABO blood group antibody levels between using human erythrocytes and RCTECs as target cells.Result The majority of cells derived from the kidney cortex were renal tubular epithelial cells.39.90%-73.80% of RCTECs were found to express the ABO histoblood group antigens with the capability to bind the human blood group antibodies specifically.The expression level of ABO histo-blood group antigen on RCTECs was not influenced by long-term cryopreservation,and the percentage of B antigen positive cells was 39.90% before frozen storage and 39.65% after a 10-month cryopreservation respectively.The normal serum and allergic serum presensitized to synthetic A antigen-keyhole limpet hemocyanin (KLH-A) of cynomolgus monkey were used to compare the abilities of RCTECs and human erythrocytes to detect antibody titers.No agglutination was observed in RCTECs test group.The mean fluorescence intensity (MFI) values of antibody were highest when the dilution fold was lowest (1 ∶ 16) and gradually decreased with increased serum dilution in both serum groups.MFI fell towards baseline value at 1 ∶ 128 dilution in normal serum group while at 1∶8 192 dilution in allergic serum group.Between 1 ∶ 16 and 1∶8 192 dilutions,MFIs of normal serum group were all lower than those of allergic serum.In human red cells test group,obvious agglutination appeared at high concentrations of antibodies,and MFIs reached the peak at 1∶64 dilution in normal serum group and at 1∶32 dilution in allergic serum group,but fluctuated irregularly thereafter.Between 1∶64 and 1∶512 dilutions,MFIs of normal serum group were all higher than those of allergic serum.Data above showed that MFI values in RCTECs group could reflect the levels of blood group antibodies more exactly.Conclusion The RCTECs of cynomolgus monkey express ABO tissue-blood group antigens and would be more suitable to be used to measure ABO blood group antibody levels by FACS.
7.Therapeutic Observation of Acupuncture in Treating Angioneurotic Headache Based on the Diagnostic Technique of Electric Meridian Detection

Objective Based on the diagnostic technique of electric meridian detection, to observe the clinical efficacy of acupuncture in treating angioneurotic headache.Method Totally 106 patients with angioneurotic headache were randomized into a treatment group and a control group, 53 cases in each group. The treatment group received acupuncture based on the electric meridian detection, while the control group received medication treatment. The hemorheology indexes [plasma viscosity (PV), erythrocyte sedimentation rate (ESR)] and McGill Pain Questionnaire (MPQ) [Pain Rating Index (PRI), Visual Analogue Scale (VAS), and Present Pain Intensity (PPI)] were observed before and after intervention, and the clinical efficacies were compared. Result The total effective rate was 88.7% in the treatment group versus 71.7% in the control group, and the between-group difference was statistically significant (P<0.05). After intervention, the hemorheology indexes and MPQ scores were significantly changed in both groups (P<0.01,P<0.05).Conclusion Acupuncture based on electric meridian detection is an effective method in treating angioneurotic headache.
8.Dexmedetomidine alleviates lung ischemia-reperfusion injury through CHOP pathway in mice

[ ABSTRACT] AIM:To explore the effect of dexmedetomidine ( DEX) on the CCAAT/enhancer-binding protein-homologous protein ( CHOP) pathway during lung ischemia-reperfusion ( I/R) in mice.METHODS:C57BL/6J male mice were randomly divided into sham operation group ( sham group) , lung ischemia/reperfusion group ( I/R group) , ischemia/reperfusion +normal saline group ( I/R+NS group ) and ischemia/reperfusion+dexmedetomidine group ( I/R+DEX group) .Dexmedetomidine was infused intraperitoneally with 25 μg/kg for 30 min prior to the ischemia period in I/R+DEX group, the normal saline was administrated with the same volume of dexmedetomidine in I/R+NS group.After fini-shed the 3 h-reperfusion period , the left lung tissues were harvested to determine lung wet/dry weight ( W/D) , the total lung water content ( TLW) , and index of quantitative evaluation for alveolar damage ( IQA) .Morphological observation and terminal-deoxynucleotidyl transferase mediated nick end labeling ( TUNEL) were applied to evaluate the structure changes and the apoptosis index (AI) of the lung tissues.The expression of CHOP and glucose-regulated protein 78 (GRP78) at mRNA and protein levels in the lung tissues was detected by Western blot and RT-PCR.RESULTS:Compared with sham group, the W/D, TLW, IQA, AI, the mRNA and protein expression of CHOP and GRP78 obviously increased, and the left lung tissues structure were damaged more obviously both in I/R group and I/R+NS group.Compared with I/R group, the W/D, TLW, IQA, AI and the protein and mRNA expression of CHOP in I/R+DEX group decreased, the injury of the left lung tissue structures induced by I/R in I/R+DEX group were also alleviated .CONCLUSION:DEX alleviates the
lung I/R injury, which may be related to inhibition of apoptosis mediated by CHOP pathway.

AIM:To explore the relationship between apoptosis in the lung tissues and lung ischemia/reperfusion injury,and to observe the effects of human thioredoxin (hTrx) on apoptosis in lung ischemia/reperfusion injury. METHODS:The single lung in situ ischemia/reperfusion animal model was used. Eighty four Wistar rats were randomly divided into control group (control),groups of ischemia for 1 h and reperfusion for different times (IR1h,IR3h,IR5h),and groups of IR + human thioredoxin treatment (IR1h + hTrx,IR3h + hTrx and IR5h + hTrx). Transmission electron microscope (TEM),terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) and immunocytochemistry techniques were used to observe apoptosis,apoptosis signal-regulating kinase 1 (ASK1) and expression of Bcl-2 and Bax in various phases of lung ischemia/reperfusion. 10.Effect of osgentide on MC3 T3-E1 pre-osteoblasts proliferation under simulated microgravity

Objective To study the effect of osgentide (OST) on proliferation of mouse preosteoblast MC3T3-E1 under simulated microgravity ( SMG ) .Methods Under normal conditions , cell proliferation was evaluated by MTT assay to screen an OST compound of an effective concentration after MC 3T3-E1 cells were treated with series OSTs .Furthermore, cell proliferation and cell cycle distribution of MC 3T3-E1 cells were analyzed after treatment with 1 nmol/L OST5 by MTT assay and by flow cytometry ( FCM) scanning under SMG .Results Under normal conditions , 1 nmol/L OST5 was able to significantly promote the proliferation of MC3T3-E1 cells (P<0.01).Under SMG, proliferation of MC3T3-E1 cells was significantly inhibited and more cells entered G 1 than under normal conditions (CN).The proportion of S phase of MC3T3-E1 cells after treatment with 1 nmol/L OST5 ( OST-SMG) for 3 d was higher than that of untreated MC 3T3-E1 cells under SMG,suggesting that OST5 could promote DNA synthesis ( P<0.05 ) .Conclusion OST5 facilitates the proliferation of MC3T3-E1 cells under SMG, which provides a basis for the use of OST5 in the prevention and treatment of bone loss relat-ed to microgravity .
