1.Meta analysis on the relationship between 5-hydroxytryptamine 2A receptor gene polymorphism T102C and schizophrenia

Objective To assess the association of 5-hydroxytryptamine 2A (5-HT2A) receptor gene polymorphism with schizophrenia by Meta analysis.Methods Literatures on the relationship between 5-HT2A receptor gene polymorphism T102C and the Chinese schizophrenia in EMBASE,PubMed,CBM,CNKI and Wanfang Med Online were taken as research objects.Screening the relevant data according to the included and excluded standard,then,Software of RevMan 5.0 was employed to do Meta analysis.Meanwhile,we inspect the literatures publication bias.Results Twelve literatures were included,collecting 4 728 individuals,enrolling 2 135 for the schizophrenia group and 2 593 for the control group.The research conforms to Hardy Weinberg equilibrium (all P>0.05).The Meta analysis results showed that there was no significant association between 5-HT2A receptor gene polymorphism and schizophrenia including the allele model,recessive model and codominant model genotypes (all P> 0.05).The results of Dominant genetic model I2 =45% with moderate heterogeneity,but we used subgroup analysis to remove the author Ni's data,there were significant difference analysis (I2=9%,Z=2.01,P<0.05).Conclusions 5-HT2A receptor gene T102C site C allele is recessive genes,CC may be a healing factor,there needs evidence-based study provides further evidence.
2.Application value of Calot triangle hollowing-out maneuver in laparoscopic cholecystectomy

Objective To investigate the application value of Calot triangle hollowing-out maneuver in laparoscopic cholecystectomy (LC) for preventing bile duct injury.Methods The retrospective cross-sectional study was conducted.The clinical data of 537 patients who underwent LC in the Dexing People's Hospital between January 2011 and December 2015 were collected.The tissues in Calot triangle were hollowed out,and cystic ducts were cut off and then gall bladders were resected.Observation indicators:(1) operation situations:anatomy of the Calot triangle and operation time;(2) postoperative recovery situations:postoperative complications and bile duct injury;(3) follow-up situation.The follow-up using outpatient examination and telephone interview was performed to detect the survival of patients and occurrence of cholangitis up to May 2016.Results (1) Operation situations:of 537 patients with LC,anatomical relation among cystic duct,common hepatic duct and common bile duct (three-duct relation for short) could be seen in 165 patients without dissection,and three-duct relation cannot be seen in other 372 patients.Of 372 patients,16 were operated on with the gallbladder open due to the difficult dissection of Calot triangle,7 were converted to open surgery due to local severe adhesion and unclear structure,1 was converted to open surgery due to intraoperative varices induced bleeding in Calot triangle,and other 348 patients underwent successful LC using Calot triangle hollowing-out maneuver.Operation time was 15-190 minutes,with an average time of 28 minutes.(2) Postoperative situations:2 patients were complicated with biliary colic pain,showing stones in the distal common bile duct via magnetic resonance imaging scans,and then received endoscopic sphincterotomy (EST);3 had subxyphoid puncture hole infection,1 had a small amount of postoperative bleeding due to hepatocirrhosis,3 had pulmonary infection,and they were improved by symptomatic treatment;1 had chylous fistula and were improved through drainage and low fat diet intake for 1 week;2 with mild bile leakage was improved through peritoneal drainage.No bile duct injury was detected.(3) Follow-up situation:348 patients were followed up for 12-18 months,with a median time of 16 months.During the followup,348 patients with follow-up had survival without manifestation of cholangitis.Conclusion Calot triangle hollowing-out maneuver could effectively prevent bile duct injury in LC.
3.Observations on the Efficacy of Intermittent Liver-nourishing and Mind-regulating Acupuncture in Preventive Treatment of Migraine

Objective To investigate the clinical efficacy of intermittent liver-nourishing and mind-regulating acupuncture in preventive treatment of migraine. Method One hundred and twenty migraine patients were randomly allocated to treatment and control groups, 60 cases each. The treatment group received intermittent liver-nourishing and mind-regulating acupuncture and the control group took flunarizine hydrochloride capsules orally. The migraine score, the visual analogue scale (VAS) score and the headache severity rating score were recorded in the subjects before treatment and at the end of treatment and the 4-week, 8-week, 12-week, 16-week and 20-week follow-ups. The Self-Rating Anxiety Scale (SAS) score and the Self-Rating Depression Scale (SDS) were recorded before and after treatment.Result After one course of treatment, the total efficacy rate was 98.3% in the treatment group and 89.5% in the control group; there was a statistically significant difference between the two groups (P<0.05). There was a statistically significant difference in the migraine score between before treatment and at the end of treatment in the two groups (P<0.05) and between the two groups at the same follow-ups (P<0.05). There was a statistically significant difference in the VAS score between before treatment and at the end of treatment in the two groups (P<0.05) and between the two groups at the same follow-ups (P<0.05). There were statistically significant differences in the SAS and SDS scores between before treatment and at the end of treatment in the two groups (P<0.05) and between the two groups after treatment (P<0.05).Conclusion Intermittent liver-nourishing and mind-regulating acupuncture has marked efficacy in preventive treatment of migraine. It can increase the total efficacy rate and reduce the number of relapses.
4.Expression of p53 in breast cancer and regulatory effects of 5-aza-2′-deoxycytidine on p53

Objective Tumor suppressor gene p53 can inhibit tumor cell growth, arrest cell cycle, and promote apoptosis.Howev-er, the effects of p53 on the pathogenesis of breast cancer have not been fully elucidated.The aim of this study was to explore the expression of p53 protein and the correlation with clinical pathologic features in breast cancer.Furthermore, the regulatory effects of 5-aza-2′-deoxycyti-dine on p53 in breast cancer cell line were also studied. Methods The expression of p53 protein in 80 cases of breast cancer and normal and adjacent tissue were determined by the immunohistochemical staining .The expressions of p53 mRNA and p53 protein in breast cancer cell line were determined by RT-PCR and Western blotting. Results The positive rate of p53 in breast cancer (41.25%) was higher than that in the normal and adjacent tissue (22.5%) (P<0.01).The expression of p53 was not significantly correlated with age, grade, stage and lymph node metastasis (P>0.05).The low expression of p53 both in mRNA and in protein levels were found in breast cancer cell line of MCF-7.The expres-sion of p53 increased after 5-aza-2′-deoxycytidine administration . Conclusion p53 is highly expressed in breast cancer , which may play an im-portant role in the development and progression of breast cancer. 5-aza-2′-deoxycytidine, up-regulating the p53 expression in breast cancer cell line, which provides the evidents for the development of therapeutic drugs for the patients with low expression of p53 breast cancer.
5.Expressions of survivin and caspase-3 in human non-small cell lung cancer and their relationship with cell apoptosis.

BACKGROUNDSurvivin, a member of inhibitor of apoptosis protein (IAP) family, can directly inhibit caspase-3 and caspase-7 activity and plays an important role in oncogenesis. The aim of this study is to investigate the expressions of survivin and caspase-3 in human non-small cell lung cancer (NSCLC), and to evaluate their relationship with cell apoptosis.
METHODSThe expressions of survivin and caspase-3 in 88 patients with NSCLC were examined by using immunohistochemical SP methods, and TNUEL method was used to detect the cell apoptosis simultaneously.
RESULTSThe positive expression rates of survivin in NSCLC tissues and normal lung tissues were 61.4% (54/88) and 13.8% (4/29) respectively, there was a significant difference between them (P < 0.01). Survivin expression in NSCLC was not related to the histologic type, pathological grade and lymph node metastasis (P > 0.05), but correlated with TNM stage (P < 0.05). The positive expression rate of caspase-3 was 89.7% (26/29) in normal lung tissues, which was higher than that in NSCLC tissues (73.9%, 65/88), but there was no significant difference (P > 0.05). Caspase-3 expression in NSCLC was associated with pathological grade (P < 0.05). The average apoptosis index (AI) of survivin-positive cases was significantly higher than that of surviving-negative ones (1.63±0.58 vs 3.29±0.76)(P < 0.05). The average AI of the caspase-3 positive cases was significantly higher than that of the caspase23 negative cases (2.42±0.59vs1.28±0.65)(P < 0.05). Expression of survivin was negatively correlated with caspase-3 (P < 0.05).
CONCLUSIONSSurvivin may play an important role in the process of carcinogenesis of NSCLC by inhibiting cell apoptosis. Moreover, survivin may prevent cell apoptosis by inhibiting caspase-3 activation.
6.Protective effect of hyperbaric oxygen preconditioning on rat syngeneic kidney transplantation

Objective To investigate the effect of hyperbaric oxygen preconditioning on syngeneic kidney transplantation in rats and to explore the potential mechanism. Methods Forty male SD rats were randomly divid-ed into control group(Sham group),kidney transplantation group(KTx group)and hyperbaric oxygen precondi-tioning group(HBO+KTx group). Kidney transplantation was performed 24 h after hyperbaric preconditioning. Rats were sacrificed 24 h after the transplantation.Serum creatinine(Scr)and blood urea nitrogen(BUN)were de-tected.The level of superoxide dismutase(SOD),malondialdehyde(MDA)and heme oxygenase-1(HO-1)in the graft was examined. Kidney pathological examination was performed after renal tissue fixed. Results Compared with the KTx group,HBO + KTx group showed lower Scr and BUN level. The MDA level was decreased and the SOD activity was increased significantly after hyperbaric oxygen preconditioning in HBO+KTx group.Renal tubu-lar necrosis was significantly reduced in HBO+KTx group.HO-1 level in HBO+KTx group was significantly high-er than that of KTx group. Conclusion Hyperbaric oxygen preconditioning can reduce lipid peroxidation,en-hance antioxidant enzyme activity and improve the level of HO-1 in graft tissue,thus attenuating kidney injury.
7.Evaluation of the clinical and radiological.efficacy of recombinant human TNFR Ⅱ -Fc combined with methotrexate in the treatment of moderate and severe rheumatoid arthritis

ObjectiveTo evaluate the clinical and radiological efficacy of TNFR Ⅱ -Fc combined with methotrexate( MTX ) in treatment of patients with moderate and severe rheumatoid arthritis.MethodsThree hundred and ninty-six RA patients were randomized into the combined treatment group,the TNFR Ⅱ -Fc only group and MTX only group.All patients were treated for 24 weeks.ACR-N,ACR20,ACR50,ACR70,DAS28-ESR and Sharp score of both hands were measured for efficacy,and the side-effects were analyzed by one-way ANOVA.Results After 24-week therapy,the ACR-N of the combined treatment group [( 12.79±9.24)%-year] was significantly improved than that of the TNFR Ⅱ-Fc only group [(9.56±11.16)%-year,P<0.05] and that of the MTX only group[(5.08±11.10)%-year,P<0.05],and the TNFR Ⅱ-Fc group was significantly improved than that of the MTX group(P<0.05).The ACR20 response rate of the combined group(80.4%) was significantly higher than that of the TNFR Ⅱ -Fc group(71.1%,P<0.05) and the MTX group(56.7%,P<0.01 ).The ACRS0 response rate of the combined group(53.6%) was significantly higher than that of the MTX group(30.8%,P<0.01 ).The ACR70 response rate of the combined group was 27.7%,which was significantly different from that of the TNFR Ⅱ -Fc group (15.8%) and MTX group (7.7%,P<0.05or P<0.01 ).DAS28-ESR in the combination group was significantly reduced than those of the TNFR Ⅱ -Fc group and MTX group,and the DAS28-ESR of the TNFR Ⅱ -Fc group was significantly reduced than MTX group.The average total Sharp score of both hands,which demonstrated the radiographic changes,was significantly reduced in the combination group than the MTX group(P=0.03).The total adverse events in the combined group(40.9%) was significantly high than that of the MTX group(28.8%,P<0.05).Conclusion TNFR Ⅱ -Fc combined with MTX can effectively control the activity of RA and radiological progress.
8.Curcumin attenuates renal ischemia reperfusion injury via JNK pathway with the involvement of p300/CBP-mediated histone acetylation

Apoptosis is proved responsible for renal damage during ischemia/reperfusion. The regulation for renal apoptosis induced by ischemia/reperfusion injury (IRI) has still been unclearly characterized to date. In the present study, we investigated the regulation of histone acetylation on IRI-induced renal apoptosis and the molecular mechanisms in rats with the application of curcumin possessing a variety of biological activities involving inhibition of apoptosis. Sprague–Dawley rats were randomized into four experimental groups (SHAM, IRI, curcumin, SP600125). Results showed that curcumin significantly decreased renal apoptosis and caspase-3/-9 expression and enhanced renal function in IRI rats. Treatment with curcumin in IRI rats also led to the decrease in expression of p300/cyclic AMP response element-binding protein (CBP) and activity of histone acetyltransferases (HATs). Reduced histone H3 lysine 9 (H3K9) acetylation was found near the promoter region of caspase-3/-9 after curcumin treatment. In a similar way, SP600125, an inhibitor of c-Jun N-terminal kinase (JNK), also attenuated renal apoptosis and enhanced renal function in IRI rats. In addition, SP600125 suppressed the binding level of p300/CBP and H3K9 acetylation near the promoter region of caspase-3/-9, and curcumin could inhibit JNK phosphorylation like SP600125. These results indicate that curcumin could attenuate renal IRI via JNK/p300/CBP-mediated anti-apoptosis signaling.

10.Clinical and biological characteristics in childhood acute myeloid leukemia with 8;21 translocation.

OBJECTIVETo investigate the clinical and biological characteristics of childhood acute myeloid leukemia(AML)with 8;21 translocation.
METHODSA retrospective analysis including clinical information, cell morphology, chromosome, immunophenotype and molecular biology was performed on 41 cases of childhood t(8;21)AML. The control group included 19 cases of AML without t(8;21) translocation detected during the same period.
RESULTSThe 41 cases of t(8;21)AML accounted for 68.3% of 60 continuous childhood AML patients. Among them, classical t(8;21) translocation was seen in 29 cases; variant t(8;21) translocation, simple 8q-, near-tetraploidy characterized by the duplication of t(8;21) translocation each came into view in 2 cases; and cryptic t(8;21) translocation was seen in 6 cases. Thirty seven cases (80.4%) belonged to M2 subtype of AML. Most of them had the morphological changes such as the leukemia cells' indent nucleus with a light stain region of perinucleus, basophilic cytoplasm, differentiation with maturation, megaloblastoid changes and nuclear-cytoplasm imbalance; the high expression of CD13 antigen; and the AML1/ETO fusion transcript in 23 cases examined by reverse transcription-polymerase chain reaction (RT-PCR) assay, including 6 cases with normal karyotype. The difference in complete remission rate between t(8;21) positive patients group and t(8;21) negative patients group was not significant in statistics (82.4% vs 75%, P>0.05). However the difference in recurring rate of the leukemia was statistically significant (10.7% vs 41.7%, P<0.05).
CONCLUSIONt(8;21)AML is the most frequent type of childhood AML. It is predominantly associated with M2 subtype of AML and has unique morphological, immunological prognostic features .
