1.Conformational Changes of Hemoglobin Induced by Surfactants

Objective To study the effects of surfactants on conformation of hemoglobin and further to understand the potential impact of surfactants in environment on human health. Methods The changes of the conformation of hemoglobin induced by cation and anion surfactants were investigated with four methods, such as synchronous fluorescence spectra, UV, electrochemistry and atom force microscope. Results The changes of the conformation of hemoglobin induced by anion surfactant were obvious but were not by cation surfactant. Conclusion Sodium lauryl sulphate, an anion surfactant, may change the conformation of hemoglobin.

3.Study on change of thymus in different clinical period of EAE mice

Objective:To investigate the change of thymus in different period of EAE mice. Methods: The thymic index and thymocyte number at 0,10,15,20 days after EAE induced were recorded. The percent of thymic apoptosis and CD4+CD44+T cells in spleen was acquired by Flow Cytometry. The content of p53 and Bcl-2 gene was detected by PCR and electrophoretic technique. Results:The thymic index and thymocyte number correspondingly reduced at 0,10,15 days after EAE induced. Apoptotic rate of thymocyte increased at 10,15 days after EAE induced,highest at 10 days. The percent of CD4+CD44+T cells also increased at 10,15 days after EAE induced, highest at 10 days. The content of p53 increased, while Bcl-2 reduced at 10 days after EAE induced. Conclusion:The atrophy of thymus is most serious at 15 days after induction;while the apoptotic percent is highest at 10 days after induction,which have a relationship with regulation of p53 and Bcl-2. The change of thymus in EAE mice closely related with EAE attack,and the recovery of thymus precede EAE.
4.An experimental study of interventional therapy by direct puncture for the treatment of hormone-induced necrosis of femoral head

Objective To evaluate the clinical efficacy of interventional therapy by direct puncture for the treatment of avascular necrosis of the femoral head. Methods 24 experimental rabbits were undertaken and divided randomly into 3 groups, B, C groups as the models of hormone induced necrosis of femoral head, and A group as the control one. B group was followed up later with interventional therapy by direct puncture. Thromolytic agents, drugs improving microcirculation, and repairing bone tissue were infused directly into the femoral head; and then microscopic observations were conducted to study the pathology. Results The results showed: in group B, the number of vacant bony lacunae in femoral head, the average diameter of fat cells in marrow cavity and the number of blood vessel in subchondral areas showed significant improvement, approaching group A. Conclusions Treatment of femoral head necrosis through interventional method of direct puncture has commenced a solid foundation for clinical therapy.

6.Bacterial Distribution and Drug-resistance of Isolated Enterococci in Clinical Specimens

OBJECTIVE In order to supply the basis of reasonable use of antibiotics,investigation of the change in distribution and drug-resistance of isolated enterococci was made in our hospital.METHODS Enterococci were cultured,isolated and identified with ATB Expression microbe identification system.Drug-resistance was detected with ATB ENTEROC and was statistically analyzed.RESULTS A total of 1 354 strains of enterococci were isolated from 50 148 clinical specimens,the isolation rate was 2.7%.The main strain of enterococci was Enterococcus faecalis.E.faecium was the second one.The isolation rate of enterococci,which were the highest in urine specimens,was increasing in sputum specimens year by year.The drug-resistance rate of enterococci to erythromycin,rifampin,ciprofloxacin and levofloxacin was high,but it was still sensitive to vancomycin and teicoplanin.CONCLUSIONS Great changes have taken place in bacterial distribution and drug-resistance of isolated enterococci,and the drug-resistance of strains of enterococci differed greatly.To insure effective treatment and decrease the infection of enterococci in hospital,the antibiotics sensitive to enterococci should been used on the basis of drug-resistance test when the infection of enterococci was treated clinically.

Objective:Tumor size,lymphnode metastasis,hormone level and so on have been the prognosis predictors of breast cancer,but they are not ideal. This study is to investigate the influence of the expression of p27Kip1 in early breast cancer prognosis and its relationship with clinical feature. p27kip1 is an important negatively regulator of cell cycle progression and plays a central role in the pathogenesis of a member of tumors including breast cancer. In breast cancer cells, the level of p27kip1 expression usually decreases during tumor development and progression, in addition, cytoplasm mislocalization of p27kip1 has been reported, but less is known about the exact molecular mechanisms. Studies have indicated that phosphorylation is the key regulation way, several signal transduction pathways are involved in the regulation of the expression and distribution of p27kip1. To further understand the mechanism, the disparity of the interacting protein profiling between tumor cells and normal cells must be identified first. Including cyclins, cyclin-depend kinases, CRM1, jab1, SKP2, p27kip1 has various interacting molecules. There are also several interacting molecules especially for breast cancer cells. It seems that different protein profiling cause the different expression and intracellular distribution in different cell cycle phase. So, disparity of the p27kip1 protein profiling may be the main mechanism of its down-expression and mislocalization in breast cancer cells.
9.Clinical significances of sialylation level in fragment crystaline of serum immunoglobulin G

Objective To establish the method of testing immunoglobulin G (IgG) with Fc sialylation, and to investigate the clinical significance of IgG with Fc sialylation in systemic lupus erythematosus (SLE), especially in those with neuropsychiatric manifestations (NPSLE). Methods Seventy-five SLE including thirty patients with neuropsychiatric manifestations (NPSLE) and forty-five non-NPSLE patients, 30 rheuma-toid arthritis (RA) patients, 32 juvenile idiopathic arthritis (JIA) patients and 41 healthy controls were recruited in this study. Standard method of testing lgG with Fc sialylation was established by a lectin based sandwiched enzyme linked immunosorbent assay (ELISA). The levels of IgG with Fc sailylation were assayed, and its clinical significance was evaluated. Results There were no difference in the levels of serum IgG with Fc sailylation in RA group (0.82±1.81) mg/ml, JIA group (0.69±1.30) mg/ml, healthy control group (0.64± 1.09) mg/ml, and the levels of IgG with Fc sailylation in SLE group (0.12±0.17) mg/ml (P<0.01), especially in NPSLE group [(0.03±0.03) mg/ml, P<0.01] was significantly lower than that of control groups. The perc-entage of IgG with Fc sialylation in control group (4.64±5.90)% were significantly higher than that in non-NPSLE (1.88±2.16)% (P<0.01) and in NPSLE (0.29±0.47)% (P<0.01). The percentage of IgG with Fc sial-ylation was negatively associated with SLEDAI score (r=-0.43, P<0.01). Conclusion Significantly low level of serum IgG with Fc sialylation was associated with disease activity in SLE patients, especially in NPSLE patients, lgG with Fc sialylation may be a new target for therapeutic strategy.
10.Study on effect of different family nursing methods on major catheter-related complications in cancer patients with PICC

Objective To study the effect of different family nursing methods on major catheter-re-lated complications in cancer patients with peripherally inserted central catheter (PICC) during chemother-apy intermission so as to reduce the incidence of complications and prolong the indwelling time and service life of the catheter. Methods We randomly divided 213 cancer patients who were discharged from hos-pital with PICC into two groups. One group(108 cases) routinely came back to our outpatient clinic and the other (105 cases) went to the community for nursing of the catheter. Results The total incidence rate of complications of the group who came back to our outpatient clinic for nursing of the catheter was 13.8%(15/ 108) and the other group was 51.4%(54/105). There existed statistical significance between the two groups (P<0.01). Conclusions There exists correlation between different family nursing methods and catheter complication rate. We suggest patients come back to our outpatient clinic for nursing of the catheter as pos-sible as they can.
