1.Investigation on the Impact of Hospital Violence on Medical Students

Objectives:To investigate the impact of doctor-attacking on medical students’ occupation atti-tude. Methods: We conducted a sample survey towards different major of Wenzhou Medical University by using self - made questionnaire. Reults: The occupation attitude of interviewee is not so bad, the confidence about the occupation is really deep down and the occupational sense of mission worries a lot of people. Doctor-attacking cases damage the honorable sense of occupation, even threatens basic personal security. The spread passive emotion by Network media , as well as the varied choices of occupations, expand the influence of hospital violence. Conclusions: The fre-quent occurrences of doctor-attacking cases weaken the occupational attitude of medical students and became the obstruc-tion to be a doctor,countermeasures should be taken from multiaspect of society,media,schools and teachers through a va-riety of ways.
2.Effects of Motivational Nursing on negative emotions and quality of life in patients undergone thyroid ;cancer surgery

Objective To explore the effect of motivational nursing intervention on anxiety, depression and quality of life in patients who had undergone thyroid cancer surgery.Methods A total of 78 patients who had undergone thyroid cancer surgery in general surgery department of the First People′s Hospital of Guangyuan were recruited for our study. These patients were randomly selected for control group or observation group, with 39 cases in each group. Patients in control group received routine nursing, whereas those in observation group were treated with motivational nursing together with routine nursing.Results After nursing, the anxiety score in the observation group was significantly lower than the control group (P<0.01). The depression scores in observation group was lower than control group, and the difference was statistically significant ( P<0.05) . Scores of factors related to the function of QLQ-C30 in observation group were significantly higher than those in control group, and the difference was statistically significant (P<0.01). The satisfaction to nursing in observation group was higher than that in control group, and the difference was statistically significant (P<0.05).Conclusions Motivational nursing can significantly alleviate patients′ anxiety and depression in those who had undergone thyroid cancer surgery, as well as improve quality of life and nursing satisfaction.
3.Analysis of immunity function of infants delivered by human immunodeficiency virus infected mothers

Objective To explore the physical development and immune function of infants without human immunodeficiency virus(HIV)infection who were delivered by HIV_infected mothers. Methods Two hundred and ninety_seven infants delivered HIV_infected mothers in Guangxi province from January 2008 to November 2011 were selected as observation group. According to whether infants had HIV infection or not,the children were further divided into the HIV_infection group and the infants in the non_HIV infection group according to the presence or absence of HIV infection,and the infants in the non_HIV infection group were divided into the antiretroviral drug(ART)treatment group and the non_ART treatment group according to whether the mother had used ART during pregnancy. Ninety_one healthy children born at the same time were selected as the healthy control group. The physical examination,T lympho_cyte subgroup analysis and humoral immunity test were performed on all infants. Results The weight and body length at birth of infants born from HIV_infected mothers were all significantly lower than those in the healthy control group [(2. 86 ± 0. 49)kg vs.(3. 15 ± 0. 52)kg;(47. 05 ± 2. 20)cm vs.(50. 01 ± 2. 58)cm],and the differences were sta_tistically significant(t﹦2. 652,2. 247,all P〈0. 05). Conclusions The humoral immune func_tion of the non_HIV infected infants delivered by HIV_infected mothers is not significantly affected,but the physical development at birth and cellular immune function are significantly affected. ART during pregnancy is not a major factor in the limitation of physical development at birth. Therefore,the nutrition support for the infants delivered by HIV_in_fected mothers and prevention of infection are especially necessary clinically.
4.Progress in research on the effects of high mobility group box 1 in pediatric rheumatic diseases

High mobility group box 1 is a kind of non-histone protein which expressed in the nucleus of eukaryotic cells.Which can promote inflammatory reaction and participate in the positive feedback loop of the production of cytokines after translocated outside the cells.As a result, it′s closely related to the pathogenesis of various autoimmune and inflammatory diseases.Recent studies have revealed that high mobility group box 1 may serve as a serum marker of Henoch-Sch?nlein purpura nephritis and lupus nephritis, and a predicator of the prognosis of juvenile idiopathic arthritis and intravenous immunoglobulin resistance in Kawasaki disease.
5.Research Status of Nanomaterial Medical Device and Discussion on Biological Evaluation

In recent years,China has made great progress in basic nanomedicine,nanotoxicology and nanobiology research.Nanotechnology has been continuously applied in biomaterial and medical device,more and more medical devices applying nanomaterials are developed and manufactured.In order to gain more comprehension and accurate understanding of the research and industrial development in nanobiomaterial medical devices,this study reviewed the common nanomaterial in medical devices and the regulatory situation of nanomaterial medical devices at home and abroad,and discussed the current challenges in biological evaluation of nanomaterial medical devices,with a view to providing ideas for the safety evaluation and research of related products.
6.Construction of CD19-CAR retroviral vector and modification of its transduction of human T-lymphocytes.

OBJECTIVETo improve the MigR1-CD19-CAR (chimeric antigen receptor) that contains a single chain variable region (scFv) which targeted to CD19 through a retroviral vector transduction efficiency of T-lymphocytes.
METHODSInsert the CD19-CAR fragment into the retroviral vector (MigR1) through recombinant DNA technology, after transfecting plat-A packaging cell lines, viral supernatant was collected to transduce K562 cell line and activated human T-lymphocytes. We used flow cytometry to determine the transduction efficiency and RT-PCR to confirm the transcription of CD19-CAR gene. The ability of the transduced T cells to produce IFN-γ and TNF-α in a CD19-specific manner was measured in an enzyme-linked immunosorbent (ELISA) assay.
RESULTS(1)Using MigR1-CD19-CAR retroviral vector to produce the high titer retrovirus. (2)MigR1-CD19-CAR transduction efficiency of K562 cell line was significantly higher than human T-lymphocytes (P<0.01). (3)120 min centrifugation could significantly improve transduction efficiency of T-lymphocytes to (54.5±14.6)%. (4)Transduction efficiency could be improved by deciding transduce time according to T-lymphocytes proliferation fold in vitro individually, and the highest transduction efficiency in the study was 69.3%. The CD19-CAR gene sequence was transcripted specificly with high efficiency. (5) IFN-γ and TNF-α released by CD19-CAR transduced T-lymphocytes significantly increased to (13 230±1 543) pg/ml and (4 217±211) pg/ml when coculture with CD19-K562 cells.
CONCLUSIONWe have successfully constructed a second generation CAR which targeted to CD19 through a retroviral vector called MigR1 (MigR1-CD19-CAR). Deciding transduce time according to T-lymphocytes proliferation fold in vitro individually and 120 min centrifugation could improve the CAR transduction efficiency of T-lymphocytes. RT-PCR confirmed that the CD19-CAR gene was specificly transcripted with high efficiency. IFN-γ and TNF-α released by CD19-CAR transduced T-lymphocytes significantly increased when activated by target cells.
