OBJECTIVE:To study the effect of erythromycin on the voltage-dependent K~+ current in hippocampal neurons of mice.METHODS:Isolation of mice's hippocammal neurons cells and whole-cell patch-clamp techniques were applied to study the change of voltage-dependent K~+ current of hippocampal neurons and the current-voltage curves in mice before and after treatment with erythromycin.RESULTS:Erythromycin inhibited the voltage-dependent K~+ current of mice's hippocampal neurons,and it down-regulated the peak current from(810.67?250.86)pA to(529.96?171.83)pA(P

Objective To retrospectively review the clinical data of acute oral paraquat (PQ) poisoning patients and the relationship of early serum creatinine leveles with respiratory failure , to predict and guide the clinical diagnosis and treatment .Methods Seventy-two PQ poisoning patients from the 2008 to 2012 , according to the occurrence of respiratory failure were divided into two groups , group A:patients with respiratory failure (28 cases);and group B:patients without respiratory failure(44 cases).The first serum creatinine detection early after poisoning as the basic value and the highest value within 48 hours, and the difference between the two ratios was compared .Then the sensitivity, specificity, accuracy, Youden index, receiver operating characteristic(ROC) area under the curve(AUC) were also calculated, and the ROC AUC was drawn.Results The creatinine basic value ,the highest valve and their ratio were obtained , and the difference was statistically significant .The best predictor boarder of creatinine basic value was 53 μmol/L, with forecasting moderate effect,and that of the highest value ,their ratio and the creatinine difference value was 183μmol/L, 3.39 and 113μmol/L, respectively.Conclusion The higher the early serum creatinine level rises , the higher the risk of respiratory failure increa-ses after PQ poisoning .The basic value , the highest value within 48 hours and their difference and ratio could predict the possibility of respiratory failure after PQ intoxication .

Objective To detect the expression of T cell immunogloblin and mucin domain-containing molecule 3 (Tim-3) mRNA in peripheral blood mononuclear cells (PBMC) isolated from asthmatic children and asthmatic murine,and to explore the role of Tim-3 in bronchial asthma.Methods Recruiting 97 cases asthmatic children,47 children with acute asthma exacerbation(acute attack group),50 children with asthma remission (remittent group)and 42 children as the healthy control group.The Tim-3 mRNA expression was measured by using reverse transcription-polymerase chain reaction(RT-PCR) method.The levels of interleukin-6 (IL-6) and transforming growth factor-5 (TGF-β)were determined with enzyme-linked immunosorbent assay (ELISA),and to analyze the correlation between Tim-3 mRNA and the level of IL-6.The asthmatic murine models were randomly divided into 2 groups:normal control group and asthma group.There were 10 mice in each group.PBMC in each group were collected.Expression of Tim-3 mRNA was detected by using real-time PCR.The level of CD4 + IL-17 + and CD4 + CD~ + FoxP3 + which reflected Th17 and regulatory T cells(Treg) expression respectively were detected by using flow cytometry,and the correlation between Tim-3 and Thl7/Treg level was analyzed.Results Expression of Tim-3 mRNA in children with acute asthma exacerbation was significantly higher than those in remission stage and healthy control group (all P ＜ 0.05).The expression of Tim-3 mRNA in children with asthma remission was significantly higher than that in healthy control group (P ＜ 0.05),and the level of IL-6 in children with acute asthma exacerbation was significantly higher than those in remission stage and healthy control group (all P ＜ 0.05).In the murine model of asthma group,the level of Tim-3 mRNA in PBMC was significantly higher than that in normal control group (P ＜ 0.01),and in asthma group CD4 + IL-17 + expression was significantly higher than normal control group but CD4 + CD25 + FoxP3 + was significantly lower than that in normal control group (all P ＜ 0.05),and the ratio of Th17/Treg increased in asthma group compared with normal control group (P ＜ 0.05).Tim-3 mRNA expression in PBMC was correlated positively with CD4 + IL-17 + level and Th17/Treg ratio respectively (all P ＜ 0.05),but was correlated negatively with CD4 + CD25 + FoxP3 + level(all P ＜ 0.05).Conclusions The high level expression of Tim-3 mRNA in asthmatic children participated in the airway inflammation,which is associated with IL-6,and animal experiments indicating that Tim-3 is closely related with the imbalance of Th1 7/Treg,indicated that Tim-3 may play a certain role in asthma.

Objective To investigate the prognostic factots by analyzing clinicopathologlcal characteristics of 972 cases of colonic carcinoma. Methods Clinical and pathological data of 972 patients with colonic carcinoma treated surgically were studied by univariate and multivariate analysis,survival rate was calculated by life-table method and compared by Log-rank test. Results The univariate analysis indicated that the predictors of survival were age,perioperative blood transfusion, perioperative carcinoembryonic antigen(CEA)level,tumor gross type,depth of tumor invasion,lymphatic invasion,liver metastasis,extrahepatic metastasis,local recurrence、peritoneal seeding,pathological type,TNM stage and type of lymph node clearance.The multivariate analysis indicated that the independent predictors of suivival were age,periopemtive CEA level,tumor gross type,lymphatic invasion,liver metastasm,extrahepatic metastasis,local recurrence、peritioneal seeding,pathological type,type of lymph node clearance and TNM stage. Confclusions Lymphatic invasion is the most important independent prognostic factor of patients with colonic carcinoma treated surgically.

Objective To study the effective anticoagulant components of the fermented products of mycelium from Tremella aurantialba 9800. Method To adopt system menstruum deal with fermented products of mycelium from Tremella aurantialba,then mycelium from tremella aurantialba was studied with the clotting time of the thrombogen,the whole blood clotting time in mice and thrombosis experiment in mice. Result Butanol extract of fermented products of mycelium from Tremella aurantialba behave anticoagulant function. Ethyl acetate extract and water extract also behave feeble activity. Conclusion The effective anticoagulant components of the fermented products of mycelium from Tremella aurantialb is butanol extract.

Objective To evaluate mono-and bi-exponential decay models in the diagnosis of hepatocellular carcinoma (HCC). Methods 28 patients with HCC who underwent conventional MRI imaging and diffusion-weighted imaging were collected,and all the HCC lesions were proved by operation pathology.The ADC values in mono-exponential decay model and ADCst ,ADCslow , ADCfast and ffast of the lesions in bi-exponential decay model were measured through the reconstruction of ADCmap ,ADCslow map, ADCfast map and fraction of fast ADC at a workstation.The relationships between the ADC values with low,medium and high b-val-ues and different differentiation degrees of HCC were analyzed.Moreover,the relationships between the ADCst ,ADCslow ,ADCfast , ffast values and HCC defferentiation grades were also explored.Results Significant differences in ADC value were found among differ-ent degrees of HCC differentiation in middle and high b-value groups (P <0.05).A positive correlation was found between the ADC value and the degree of HCC differentiation in middle (r=0.377,P <0.01)and high b-value group (r =0.81 5,P <0.01).There were no significant differences in ADC value among the different degrees of HCC differentiation in low b-value group.There were significant differences in the values of ADCst ,ADCslow and ADCfast among different degrees of HCC differentiation.Significant posi-tive correlations were found between the values of ADCst ,ADCslow and ADCfast and the degree of differentiation (P <0.01).There were no significant differences in the values of ffast among different degrees of HCC differentiation.Conclusion ADC value in mono-exponential decay model plays a potential role in the diagnosis of different differentiation degree of HCC with a b value ≥400 s/mm2 .Furthermore,the parameters in bi-exponential decay model,especially the values of ADCst ,ADCslow and ADCfast ,can provide new and unique information in the distinction of different degrees of HCC differentiation.

Objective To survey the status of clinical isolates multi-drug resistant Enterobacteriaceae produ-cing KPC carbapenemase in Huizhou city of Guangdong Province .Methods 81 strains of multi-drug resistant Enter-obacteriaceae were collectded from 2009 to 2011 in Huizhou region.Modified Hodge test(MHT) was used to screen stains which may produce carbapenemase , modified three-dimensional test was used to detect ESBLs , AmpC and MBLs.Polymerase chain reaction (PCR) and gene sequencing methods were performed to confirm KPC genotypes . Results Of 81 strains multi-drug resistant enterobacteriaceae ,13 of which had passitive results were screened by modified Hodge test ,including 7 strains of Enterobacter cloacae ,4 strains of Klebsiella pneumonia and 2 strains of Escherichia coli;By modified three-dimensional test ,9 strains produced MBLs ,2 strains produced ESBLs ,1 strain pro-duced AmpC ,1 strain produced ESBLs and AmpC at the same time .KPC was amplified from a strain of Klebsiella pneumonia which produce ESBLs by PCR ,KPC-2 genetype was identified by gene sequencing .Conclusion Klebsiel-la pneumonia with KPC–2 carbapenemase has emerged in huizhou city ,but the production of metallo-beta-lactamase is the main cause that Enterobacteriaceae resist to carbapenemase drugs .

Objective To detect the expression of T cell immunoglobulin mucin 3 (Tim-3) and its relationship with Th17/Treg cytokine isolated from peripheral blood mononuclear cells (PBMCs) and broncho-alveolar lavage fluids (BALF) cells in the asthmatic murine model and to investigate the role of Tim-3 in the occurrence and development of the asthmatic inflammation. Methods The asthmatic murine model was established by the method of ovalbumin (OVA) injection and inhalation. The mice were randomly divided into 2 groups: the normal control and the asthma group. PBMCs and BALF cells in each group were collected. Expression of Tim-3 mRNA was detected by the real-time PCR assay. Levels of CD4+IL-17+ and CD4+CD25+FoxP3+, reflecting expressions of Th17 and Treg respectively , were detected by flow cytometry. The correlationbetween Tim-3 and Th17/Treg level was analyzed. Results Contrast with the normal control group, the expression of Tim-3 mRNA in PBMCs and BALF cells in the asthma group increased significantly (P < 0.01). OVA sensitization and challenges resulted in the increased CD4+IL-17 production and the decreased CD4+CD25+Foxp3+in PBMCs and BALF cells compared with those in the normal control group (both P < 0.05). The ratio of Th17/Treg significantly increased in asthma group (P < 0.05). Tim-3 mRNA expression in PBMCs and BALF cells was positively correlated with CD4+IL-17+ level and Th17/Treg ratio , respectively , and was negatively correlated with CD4+CD25+Foxp3+ level. Conclusions Tim-3 mRNA level in PBMCs and BALF cells in the asthma group was increased , indicating that Tim-3 might take part in the occurrence and development of asthmatic inflammation and has a very close relationship with the Th17/Treg imbalance.

Background and purpose:Metabolism change is one of the main characteristics of the tumor de-velopment. Many studies have conifrmed that cytosolic acetyl-CoA synthetase 2 (ACSS2) plays a critical role in hydro-carbon metabolism of cancer cells. This study aimed to explore the effect of ACSS2 on cellular proliferation, apoptosis and migration of A549 cells by RNA interference.Methods:The ACSS2 interference fragment ACSS2-siRNA and neg-ative control were designed and synthesized for RNA interference followed by the transient transfection in non-small cell lung cancer (NSCLC) cell line A549. Real-time lfuorescence quantitative polymerase chain reaction (RTFQ-PCR) was used to detect ACSS2 mRNA expression. Methyl thiazolyl tetrazolium (MTT), lfow cytometry and wound healing assay were used to detect cell proliferation, apoptosis rate and migration.Results:The expression of ACSS2 mRNA was signiifcantly decreased after transfection with the interference fragment ACSS2-siRNA in NSCLC cell line A549. The proliferation and migration activity of ACSS2-siRNA treated cells were decreased significantly compared with the control group. The apoptosis rate, especially the early apoptosis, was increased..Conclusion:Knockdown of the ACSS2 expression in NSCLC cell line A549 can signiifcantly inhibit the cell proliferation, migration ability and pro-mote the apoptosis rate, especially early apoptosis. This study indicates that ACSS2 may contribute to the progression of human lung adenocarcinoma and may have the potential to serve as a novel therapeutic target.
