1.Mental Health of Teachers and the Relationship Between Mental Health and Achievement Motivation

Objective: To study the relationship between mental health and achievement motivation of teachers.Methods: 322 high school teachers completed SCL-90 and Achievement Motivation Scale.Results: The percent of high-score teachers with at least one SCL-90 subscale scored more than 3 is more than 5%.There is obvious correlation between the score of SCL-90 and the AMS's.Conclusion: The high level of achievement-seeking or the high level of failure-avoiding is harmful to the mental health of teachers.
2.Research advances in tooth agenesis[WT5”BZ]

Tooth agenesis constitutes one of the most common developmental anomalies in man. Oligodontia is defined as congenital absence of six or more teeth. Based on the studies of our team in cooperation with Peking University Center for Human Disease Genomics in the past five years, this article reviews the current research progress in clinical phenotypes and case collection, epidemiological investigation and etiological genetic studies of oligodontia. The symptoms of oligodontia were classified into syndromic and non-syndromic according to the occurrence of tooth agenesis with or without systemic developmental defects. As for the advancement of theories and techniques of molecular genetics, a number of gene mutations have been identified to be the direct etiological factors causing some specified diseases, especially those with developmental defects. Here, this article summarized the outcomes of molecular genetic study of some cases we collected. Of the systemic oligondontia patients, a new four-base-deletion mutation in PITX2 was identified in a large kindred with typical symptoms of Rieger Syndrome; four different gene mutations in ED1 casing X-linked hypohidrotic ectodermal displasia were found in five nucleus families. Compared with the former, non-syndromic oligodontia has more genetic heterogeneity rather than some specific virulence gene. PAX9 and MSX1 are the identified genes associated with family tooth agenesis without systemic syndrome. Also, in our research, three gene mutations in CBFA1 were detected in four cleidocranial dysplasia families, which is a systemic developmental disease including the symptoms of tooth eruption abnormality and accessory teeth.
3.Separation and Drug Resistance Anaiysis of Pseudomonas Aeruginosa in Patients With Lower Respiratory Trace Infection

Objective To understand Present condition of separation of Pseudomonas aeruginosa and its drug-resistant tendercy in patients with lower respiratory trace infection in our hospital.Methods To make a statistical analysis of durg sensitive tests of 182 strains of Pseudomonas aeruginosa form thos samples of patients with lower respiratory trace infection detected from October 2005 to August 2007.Results 1031 strains of pathogenic bacteria were separated from 1409 sample of lower respiratory trace tract,and pseudomonas aeruginosa 182 strains and 17.65% of them are PA,results of drug sensitive test indicate that Paare resistant to gentamycin CN SMZ CTX GAT etc.Conclusion The infection and drug resistance of pare wery severe now,it isbeneficial to rational use of drugs to keep abreast of current drug-resistant condition.
4.Expression of T cell immunogloblin and mucin domain-containing molecule 3 mRNA and its relationship with asthma

Objective To detect the expression of T cell immunogloblin and mucin domain-containing molecule 3 (Tim-3) mRNA in peripheral blood mononuclear cells (PBMC) isolated from asthmatic children and asthmatic murine,and to explore the role of Tim-3 in bronchial asthma.Methods Recruiting 97 cases asthmatic children,47 children with acute asthma exacerbation(acute attack group),50 children with asthma remission (remittent group)and 42 children as the healthy control group.The Tim-3 mRNA expression was measured by using reverse transcription-polymerase chain reaction(RT-PCR) method.The levels of interleukin-6 (IL-6) and transforming growth factor-5 (TGF-β)were determined with enzyme-linked immunosorbent assay (ELISA),and to analyze the correlation between Tim-3 mRNA and the level of IL-6.The asthmatic murine models were randomly divided into 2 groups:normal control group and asthma group.There were 10 mice in each group.PBMC in each group were collected.Expression of Tim-3 mRNA was detected by using real-time PCR.The level of CD4 + IL-17 + and CD4 + CD~ + FoxP3 + which reflected Th17 and regulatory T cells(Treg) expression respectively were detected by using flow cytometry,and the correlation between Tim-3 and Thl7/Treg level was analyzed.Results Expression of Tim-3 mRNA in children with acute asthma exacerbation was significantly higher than those in remission stage and healthy control group (all P < 0.05).The expression of Tim-3 mRNA in children with asthma remission was significantly higher than that in healthy control group (P < 0.05),and the level of IL-6 in children with acute asthma exacerbation was significantly higher than those in remission stage and healthy control group (all P < 0.05).In the murine model of asthma group,the level of Tim-3 mRNA in PBMC was significantly higher than that in normal control group (P < 0.01),and in asthma group CD4 + IL-17 + expression was significantly higher than normal control group but CD4 + CD25 + FoxP3 + was significantly lower than that in normal control group (all P < 0.05),and the ratio of Th17/Treg increased in asthma group compared with normal control group (P < 0.05).Tim-3 mRNA expression in PBMC was correlated positively with CD4 + IL-17 + level and Th17/Treg ratio respectively (all P < 0.05),but was correlated negatively with CD4 + CD25 + FoxP3 + level(all P < 0.05).Conclusions The high level expression of Tim-3 mRNA in asthmatic children participated in the airway inflammation,which is associated with IL-6,and animal experiments indicating that Tim-3 is closely related with the imbalance of Th1 7/Treg,indicated that Tim-3 may play a certain role in asthma.
5.Cloning and Sequence Analyses of Genome of Swine Hepatitis E Virus(HEV) HN-JY40 Strains Isolated from Henan Province.

In the present study, the genomic sequence characteristics of HN-JY40 strains of the hepatitis E virus (HEV) isolated from pigs in Henan Province, China, were analyzed and the evolutionary relationship between HN-JY40 and other sequenced strains examined. The whole genome of HN-JY40 was sequenced and analyzed by reverse transcription-polymerase chain reaction, 3' rapid amplification of cDNA ends (3' RACE) and 5' RACE. Bioinformatic analyses were carried out with Megalign, Expasy, clustal x, and MEGA 4 software. The genome of HN-JY40 was 7 223 bp in size upon removal of polyA sequences. Sizes were 9 bp and 69 bp at 5' and 3' noncoding regions, respectively. The genome of HN-JY40 was predicted to contain three open reading frames (ORFs): ORF1 (5 124 bp) encoding 1 707 amino acids; ORF2 (2 025 bp) encoding 674 amino acids; ORF3 (345 bp) encoding 114 amino acids. Phylogenetic-tree analyses indicated that HN-JY40 is a typical type-IV virus that belongs to a new subgenotype of HEV genotype 4. We sequenced and analyzed the whole genome of HN-JY40. This strategy elicited the genomic characteristics of the HEV isolated from pigs in Henan Province as well as the evolutionary relationships between HN- JY40 and other HEV isolates from pigs. We revealed that the ORF1 of HN-JY40 (153-432 nt) and human HK 104-2004 had high similarity, which offers molecular evidence for uncovering the interspecies transmission of the HEV.

6.Effects of health management of stomatology on the marginal bone resorption of minimally invasive implant in the elderly

Objective To evaluate the effects of health management of stomatology on the marginal bone resorption of minimally invasive implant in the elderly.Method Tottally 54 implants were placed in 37 elderly patients of missing teeth in maxillary molar areas and the patients were randomly divided into health management group and control group.Patients in the group of health management were guided to master the proper techniques of oral hygiene practices before and after the implantation and periodontal maintenance were carried out carefully until 1 year after loading of restoration.In the control group, oral hygiene was applied. X-ray and CBCT examinations were made and implant marginal alveolar bone loss (MBL) was measured 3, 6, 9, 12 months after loading with Planmeca PROMAX3D software. Besides, 3, 6, 9, 12 months after loading, improved plaque index (iPLI), modified bleeding index (mBI) and probing depth (PD) were also recorded. SPSS 15.0 was applied to analyze the data. Result Differences in diastolic and systolic blood pressure and changes in heart rate in patients during the intraoperative period were (8.86 ± 0.31) mmHg(1 mmHg=0.133 kPa), (19.18 ± 0.22) mmHg, (20.19 ± 1.84) times/min, which were significantly lower than those of the control group values [(16.55±1.08) mmHg, (28.31±2.30) mmHg, (20.19±1.84) times/min] (P<0.05 or 0.01);at 12 months after the load around the implant in oral health management group, the values of MBL and mSBI were 2.6 (2, 4), 2.1 (2, 4), which were significantly better than control group [4.3(3, 7), 4.3(3, 7)] (P<0.01);simultaneously, there was significant difference in the values of MBL and PD between experimental group (1.42 ± 0.03, 0.77 ± 0.24) and control group (1.59 ± 0.04, 0.54 ± 0.17) (P<0.01). Conclusion Well-performed health management of peri-implant could reduce the MBL and keep healthy soft tissue environment around implants.
7.Relationship between methylation status of Rap1 GTPase activating protein and colon cancer

Objective To explore the methylation status of Rap1 GTPase activating protein (Rap1GAP) promoter in colon cancer, and to provide the oretical basis and research direction for the early diagnosis, targeted therapy, anti-multidrug resistance of colon cancer and so on. Methods The paraffin embedded specimens of 33 patients with colonic adenocarcinoma diagnosed by pathology were analyzed from Department of Pathology of Xinzhou City People′s Hospital from January 2010 to September 2014, including 19 males and 14 females, and aged 41-72 years old. The paraffin embedded specimens of 16 patients with colonic adenoma were enrolled, including 9 males and 7 females, and aged 34-58 years old. 13 normal tissues from the tumor distal margin (from the tumor > 15 cm) were selected. Quantitative methylation specific PCR (q-MSP) was applied to detect methylation level of Rap1GAP gene promoter. The methylation level differences of Rap1GAP gene promoter region among 3 groups or between different clinicopathologic factor subgroups were compared. Results The methylation rates [median (interquartile range)] of Rap1GAP promoter were 65.43 % (50.35 %), 21.37 % (8.39 %) and 17.43 % (15.71 %) in colonic adenocarcinoma group, colonic adenoma group and adjacent normal tissue group, respectively. The methylation rate of colonic adenocarcinoma group was significantly higher than that of colon adenoma group or that of adjacent normal tissue group (P< 0.05). The methylation rates of Rap1GAP promoter in colonic adenocarcinoma were not correlation with age, sex, differentiation and the stage of TNM [ male vs. female: 42.74 % (70.44 %) vs. 21.98%(80.00%);≤60yearsoldvs.>60yearsold:36.26%(62.62%)and26.23%(76.42 %);well-differentiated vs. moderately/poorly-differentiated: 21.98 % (40.32 %) vs. 42.74 % (74.20 %); TNM Ⅰ-Ⅱ vsⅢ-Ⅳ: 25.31 % (48.27 %) vs. 36.26 % (75.55 %); all P> 0.05]. Conclusion The methylation status of RAP1GAP promoter maybe associate with genesis and development of colon cancer, which might be used as a target for early diagnose of colon cancer.
8.Analysis of treatment model for stroke in 1200 elderly inpatients

Objective To compare the differences in curative effect between general treatment model and stroke unit model in the treatment of stroke in the elderly inpatients, to explore the standardized management model, for improving efficacy and reducing the degree of invalidity and mortality. Methods A total of 1200 cases with acute cerebrovascular disease were admitted into our department from April 2002 to April 2007. All patients were randomly assigned either into stroke unit or general ward to receive treatment, and the data of two groups were compared. The key indexes included mortality, hospitalization time, Barthel Index (BI), National Institutes of Health Stroke Scale (NIHSS) and Oxford Handicap Scale (OHS). All the patients or their family members were asked to have the Visual Analogue Scales (VAS). The data of cost and effect were analyzed with SPSS software. Results The complication rate and mortality rate were significantly lower in stroke unit group than in general ward group (P< 0.01). The scales of BI, NIHSS, OHS and VAS were significantly better in stroke unit group than in general ward group when discharged (P<0.05). The hospitalization time was shorter in stroke unit group than in general ward group (P<0.05). There was no significant difference in the costs of hospitalized patients between the two groups (P>0.05).Conclusions Stroke unit model can improve the physical activity and life quality of patients and reduce disease mortality without increasing the cost of patients.
9.Expression of Rap1 GTPase-activating protein 1, matrix metalloproteinase 2 and matrix metalloproteinase 9 in colorectal carcinoma and their significance

Objective To investigate the expression of Rap1 GTPase-activating protein 1 (Rap1GAP),matrix metalloproteinase 2 (MMP-2) and matrix metalloproteinase 9 (MMP-9),and their relation with clinical patterns in colorectal carcinoma.Methods Immunohistochemistry was used to detect the expression of Rap1GAP,MMP-2 and MMP-9 in colorectal carcinoma,villous adenoma,tubular adenoma and normal colorectal tissue,and their relationship with clinicopathological parameters was analyzed.Results The positive rate of Rap1GAP expression was 30.4 % (14/46),77.8 % (14/18),69.6 % (16/23) and 95.2 % (20/21) in colorectal carcinoma,villous adenoma,tubular adenoma,and normal colorectal tissue,respectively (x2 =30.659,P=0.000).The figures were 71.7 % (33/46),55.6 % (10/18),52.2 % (12/16) and 9.5 % (2/21) for the positive rate of MMP-2 expression (x2 =22.459,P =0.000),as well as for 76.1% (35/46),61.1% (11/18),56.5 % (13/23) and 14.3 % (3/21) for the positive rate of MMP-9 expression,respectively (x2 =22.643,P =0.000).In patients with colorectal carcinoma,the expression of Rap1GAP was correlated with tumor differentiation (x2 =5.275,P =0.022),but not sex,age,or lymphatic metastasis (all P > 0.05).The expression of MMP-2 and MMP-9 were correlated with lymphatic metastasis (x2 =6.661,P =0.010;x2 =8.475,P =0.040),but not sex,age or tumor differentiation(all P > 0.05).There was a negative correlation between expression of Rap1GAP and MMP-2,MMP-9 in colorectal carcinoma,respectively (r =-0.424,P =0.003;r =-0.294,P =0.048),but no correlation between the expression of MMP-2 and MMP-9 (r =0.101,P =0.505).Conclusions Rap1GAP,MMP-2 and MMP-9 play important roles in the malignant biological behavior of colorectal carcinoma,and the expression of Rap1GAP is negatively correlated with MMP-2 and MMP-9.The interactions among the three affect the occurrence and development of colorectal carcinoma.
10.Mutation detection in ED1 gene in hypohidrotic ectodermal dysplasia (HED) families

Objective: To detect ED1 gene mutation in three hypohidrotic ectodermal dysplasia (HED) nuclear families. Methods: Peripheral blood samples were obtained from three different families of hypohidrotic ectodermal dysplasia. Genomic DNA was extracted. Polymerase chain reaction, direct sequencing and restriction enzyme reaction were performed to identify the mutations. Results: Different missense mutation in ED1 gene were found in each family: C412G, A1201G and C1375T. Two of the mutations had not been previously reported. Conclusion: Mutations in the ED1 gene are responsible for the phenotypes of HED of the patients in the family.
