1.Measurement of transforming growth factor beta 1 mRNA expression in renal cortex of diabetic rats using real-time reverse transcriptase-polymerase chain reaction with SYBR Green I

Expression levels of TGF-β1 mRNA in renal cortex of control group, diabetic group and taurine group measured by real-time quantitative RT-PCR were (7.0±0.8)×10 -3, (64.4±8.0)×10 -3 and (16.7±2.0)×10 -3, respectively. The corresponding results with end-point RT-PCR method were 0.28±0.12,0.58±0.16 and 0.43±0.10, respectively. Compared with end-point RT-PCR method, real-time quantitative with SYBR Green I was easier, faster, sensitive and specific.
2.Effect of matrine on PLA_2 activity of LPS-induced inflammatory rats and its mechanism

Object To study the antiinflammatory effect of matrine and its active mechanism. Methods The matrine effects on activities of secretory phospholipase A 2 (sPLA 2) in peripheral serum and cytosolic phospholipase A 2 (cPLA 2) in leucocyte were measured with Escherichia coli membrane incorporated by arachidonic acid as the substrate; the Ca 2+ level of leucocyte was determined using Frua2 AM loading method, and the rat plantar swelling test was used to examine the antiinflammatory effect of matrine. Results One hour after ip 30 mg/mL matrine, the inhibitory rate of sPLA 2 activity was (81.9?1.8)% , cPLA 2 (28.4?6.0) %, while Ca 2+ concentration was (157.10?20.56) nmol/L which was 15.3% higher than that of the control. Plantar swelling test showed that matrine had a significant anti inflammatory effect. Conclusion Matrine is a novel PLA 2 inhibitor with anti inflammatory effect.
3.A study on neuronal proliferation in the learning and memory ability impaired aged rats

4.Expression and significance of CD62P in peripheral blood and renal tissue in patients with aged primary nephrotic syndrome

Objective:To study the changes of CD62P expression in peripheral blood and renal tissue in aged patients with primary nephrotic syndrome(APNS) and to explore its effects on APNS.Methods:Peripheral blood samples and renal tissues of 36 APNS patients were collected.The number of platelets expressing CD62P was counted by flow cytomerty.Renal frozen sections were coated with fluorescent isothiocyanate conjugated monoclonal anti CD62P antibody.Results:Compared with the healthy control subjects,the number of platelets expressing CD62P in APNS patients was significantly higher.CD62P staining in renal tissues was found in 32 of 36 APNS patients,while the staining in renal tissues was negative in 8 control subjects.In APNS patients,the level of CD62P expression of peripheral blood was correlated with that of renal tissue and both of the levels of CD62P expression in peripheral blood and in renal tissue were correlated with 24 h urine protein respectively.Conclusion:The expressions of CD62P in APNS patients increase both in peripheral blood and in renal tissue;CD62P involes in the pathological course of APNS;The levels of CD62P expression of peripheral blood and renal tissue can reflect the condition severity of APNS patients,thus they have important value in clinic.
5.The preparation of 99Tcm-J591 and its SPECT imaging of nude mice bearing human prostate cancer

Objective To study the binding performance of 99Tcm labeled anti-human prostatic specific membrane antigen (PSMA) monoclonal antibody J591 (99Tcm-J591) and prostate cancer cells in vitro,the biodistribution and SPECT imaging of 99Tcm-J591 in nude mice bearing human prostate cancer in vivo.Methods The monoclonal antibody J591 was labeled with 99Tcm by improved Schwarz method.Labeled antibody was purified by Sephadex G-50.The labeling efficiency and radiochemical purity were measured by paper chromatography and trichloroacetic acid method.The binding performance of J591 and prostate cancer cells was measured by flow cytometry in vitro.The nude mice bearing PSMA-positive C4-2 prostate carcinoma xenografts served as experiment group,mice bearing PSMA-negative PC3 tumors served as control group.6.2-8.5 MBq of 99Tcm-J591 (25 μg) was intravenously injected into mice.Gamma imaging was performed 2,6,12 and 24 h after injection,T/NT was calculated by ROI technique.After scanned 12 h post injection,4 mice of the experiment group and 5 mice of the control group were sacrificed and the tracer in vivo biodistribution was measured by gamma-counting,and the % ID/g was calculated.Two-sample t test was carried out to validate significant difference of %ID/g between two groups.Results The labeling efficiency and radiochemieal purity of 99Tcm-J591 were (78.9±6.2)% and (92.3±5.1)%,respectively,and the specific activity of 99Tcm-J591 was 68.7 MBq/mg.The antibody J591 and 99Tcm-J591 could strongly combine with PSMA-positive C4-2 cells in vitro,and didn't combine with PSMA-negative PC3 cells in vitro.SPECT imaging results showed that radioactive concentration was obvious in tumor 6 h post injection,the concentration scope became large and the tumor image was clear 12 h post injection.T/NT was 1.9±1.1 at 2 h,4.3±1.8 at 6 h,5.6±2.7 at 12 h,1.4±0.6 at 24 h,respectively.In the control group,no radioactivity concentration was found in tumor,and T/NT was less than 2.The biodistribution results showed that %ID/g of tumor tissue was 20.1±5.2 in the experiment group and 5.8±2.6 in the control group,and there was significant difference (t=5.37,P<0.001).No significant tracer uptake occurred in other tissues and organs between the two groups (all t< 1.98,all P>0.05).Conclusion The immunoactivity,characteristics of biodistribution and tumor targeting property of monoclonal antibody J591 show promising future and potential values in diagnosis and therapy of prostate cancer.

Objective To investigate the effect of antisense telomerase reverse transcriptase (TERT) on 5-hydroxytryptamine (5-HT) induced proliferation and cell cycle of pulmonary artery smooth muscle cells (PASMCs). 8.Correlation between mothers reactions to teens exposure to intimate footage and junior high school students viewing of porn

Objective:
To explore the association between mothers reactions to adolescents exposure to intimate shots and pornography use of junior high school students, and to provide references for family communications on sexual topics in similar scenarios.
Methods:
A total of 1 760 students from grade 6 to 8 in three junior high schools in a certain district of Shanghai were selected by cluster sampling. Self-filled questionnaires on tablets were used to collect data.
Results:
The proportion of junior high school students who watched pornography was 26.36%(464), and that of boys (29.83%,270/905) was higher than of girls (22.69%,194/855). Most junior high school students believed that mothers reactions to adolescents exposure to intimate shots would be “taking the opportunity to educate teenagers not to do this” (30.54%) or “avoiding adolescents watching intimate shots” (42.24%). The results of multivariable analysis showed that comparing with junior high school students who believed that mothers would “watch together without making any comments ”, students who believed that mothers would “take the opportunity to educate teenagers not to do this”(OR=0.43, 95%CI=0.29-0.64) and “avoid adolescents watching intimate shots”(OR=0.56, 95%CI=0.39-0.80) were at lower risk of viewing pornography (P<0.01).
Conclusion
There is a significant correlation between junior high school students pornography use and mothers possible reactions to adolescents exposure to intimate shots. Family sex education and parental monitoring should be emphasized, and parents awareness of communicating sexual issues with adolescents should be strengthened.
9.Heterosexual romantic expectation of early adolescents and its influencing factors

Objective:
To explore the heterosexual romantic expectation in early adolescence(10-14 years)and its influencing factors, and to provide a reference for instructing children of early puberty to establish a positive interpersonal relationship(including heterosexual relationship) and improving their healthy development.
Methods:
Stratified cluster sampling method was adopted to recruit students of grade 6-8 in three middle schools of Shanghai during November to December, 2017. Students were surveyed anonymously using Computer Assisted Self-Interview approach via the tablets. The collected data included the information on demographic, families, peers, use of the media, community cohesion and heterosexual romantic expectation. The ttest, Chi-square test, ANOVA and multiple linear regression were used to explore the relationships between potential factors and heterosexual romantic expectation.
Results:
The mean score of heterosexual romantic expectation was 2.55. Score of the boys was higher than that of girls (2.62 vs. 2.47, t=2.65, P<0.05). There was no statistically significant difference between the scores of the ages before 12 years old (10-11: 2.34, 12: 2.28), but a significant increase of the scores along the age after 12 years old(13: 2.69, 14: 3.05). Multiple linear regression suggested that respondents with older age, poorer caregiver-child relationship, higher proportion of friends of the opposite sex, more friends that thought having boyfriends/girlfriends was important, more TV/movie use and lower community cohesion had higher score of heterosexual romantic expectation of early adolescents(P<0.05).
Conclusion
The early adolescents had relative conservative views on the heterosexual romantic relationships of adolescents of their own age while the boys were more permissive than girls. The heterosexual romantic expectation improved significantly after 12 years and may be influenced by factors of family, peers, media and community.
10.Gender stereotype of early adolescents and its influencing factors

Objective:
To explore the gender stereotype in early adolescence and the potential influencing factors.
Methods:
Stratified cluster sampling method was adopted to recruit students of grade 6-8 in three middle schools of Shanghai during November to December, 2017. Students were surveyed anonymously using Computer Assisted Self-Interview approach regarding geder stereotype and associated factors.
Results:
Average gender stereotype score was (3.32±0.75), with boys(3.43±0.75)higher than the girls(3.20±0.72)(t=6.37, P<0.05). The "agreement" proportion was highest among gender stereotypes items which reflect personality characteristics such as "it’s important for boy to be strong and determined" "girls should be like a lady" and "girls were expected to be submissive"(60.99%,50.79%,43.95%), and the "agreement" proportion among boys increased with age, while decreased in girls. Boys who had sisters or recent bullying behaviors showed more gender stereotype[β(95%CI)=0.19(0.05-0.33), 0.39(0.12-0.67)]. Girls whose parents allowed to go to opposite sex homes alone had less gender stereotype, however, the gender stereotype became stronger when girls had more female peers[β(95%CI)=-0.22(-0.41--0.03),0.04(0.00-0.07)].
Conclusion
The young adolescents, especially boys, show certain level of gender stereotype. The normal heterosexual friendships and safe campus environments would help promote young adolescents to establish equal gender beliefs.
