Journal of International Oncology 2014;41(4):254-258
Plant alkaloids have anti-tumor activities. In recent years, the anti-tumor mechanisms of plant alkaloid are demonstrated to be related to the block of tumor cell cycle, the induction of tumor cell apoptosis, the inhibition of tumor angiogenesis and multi-drug resistance. In addition, most recently, plant alkaloids are found to inhibit telomerase activity and induce autophagy. Therefore, plant alkaloids may be developed into a sort of potential antitumor drugs.

Objective To investigated the absorbed dose response of Al2O3 dosimeter in water phantom irradiated by 60Co γ spectrum source. Methods The EGSnre simulation program code DOSRZnrc was used to calculate the absorbed dose of the Al2O3 dosimeter and that of the equivalent volume of water in the corresponding position, as well as the absorbed dose conversion factor, irradiated by 60Co photon beams in a water phantom. Simulations were done for a eylindrieal geometry dosimeter (diameter 0.4 cm and height 0.1 cm) and the dosimeter was placed at the centre of the water phantom at different depths. Results The average absorbed dose conversion factor is 1.143 ± 0.006 and changes little with the depth of the dosimeter in the water phantom, and the deviation is less than 1.0 %. Conclusion The absorbed dose response of Al2O3 dosimeter irradiated by 60Co γ spectrum source is steady and is independent on the depth of the dosimeter in water phantom in this research.

[Objective]To investigate the effect of rabbit BMSCs with fibrin sealant on accelerating early tendon healing.[Methods]The BMSCs were isolated and amplified. The rabbits were divided into experimental(E) and control(C) groups. The injury model was a sharp complete transection through the midsubstance of the achilles tendon. The experimental group was implanted with composite of BMSCs and fibrin sealant,The control group was operated with only fibrin sealant.Specimens were harvested at 1,3,6,and 12 weeks for analysis, which included evaluation of gross morphology, cell tracing, histological assessment, immunohistochemistry studies, morphometric analysis, and mechanical testing.[Results]The gross morphology of the tendons showed the experimental group had lesser adhesion and better reactiveness than that in the control group. The fibrin had degraded at 3 weeks.Cell tracing showed that the labeled bone marrow derived mesenchymal stem cells remained viable and presented in the intratendinous region for at least 6 weeks, becoming more diffuse at later timepoints. At 3 weeks, collagen fibers appeared more organized and there were better morphometric nuclear parameters in the treatment group. At 6 and 12 weeks, there were no differences between the groups with regard to morphometric nuclear parameters.Biomechanical testing showed improved modulus in the treatment group as compared with the control group at 3 weeks, but not at subsequent timepoint.[Conclusion]Intratendinous cell therapy with bone marrow derived mesenchymal stem cells following primary tendon repair can improve histological and biomechanical parameters in the early stage of tendon-healing.

The influencing factors of ultrasonic extraction(UE) and ultrasonic-soxhlet extraction(USE) were analyzed based on orthogonal experimental design. The optimum analytical conditions of two methods were determined. The three different methods: UE, USE and traditional soxhlet extraction(SE) were compared by simultaneous analysis with three different soil samples: fresh contaminated soil, soil after microbial remediation and soil after phytoremediation. The optimum condition of the UE was extracting 4 times with chloroform, 15 mL extractant every time. The optimum condition of the USE method was ultrasonic for 10 min at 60 W, followed by soxhlet extraction with 125 mL extractant for 6 h. Compared the three different methods, USE method showed the best result with the highest average extraction rate of 102.98%, the UE method with average extraction rate of 99.69%, and the SE method with low average extraction rate of 97.31%. The research indicates that USE method is effective in determining soil total petroleum hydrocarbons(TPH) within shorter time and higher efficiency and can be used as a method for determining TPH content in different soils.

Background and purpose:More than 70 percent of ovarian cancer patients were diagnosed in the advanced stage. Currently the 5-year disease free survival (DFS) of stageⅢC-Ⅳovarian cancer patients wsa about 10 percent after first line chemotherapy. This study aimed to improve the 5-year DFS by three-step chemotherapy according to the mechanisms of ovarian cancer biological characteristics, cytodynamics and pharmacology. Methods:In arm A, the patients received three-step chemotherapy after primary debulking surgery, step one with paclitaxel plus carboplatin (TC regimen), every 3 weeks for 6 to 8 cycles;step two with etoposide plus cyclophosphamide, every 4 weeks for 6 cycles;step three wit carboplatin plus cyclophosphamide every eight weeks for six cycles. In control arm B, we retrospectively analysed 51 cases withⅢC-Ⅳstage ovarian cancer, who had completely response after standard chemotherapy with six to eight cycles of TC after primary surgery during 2007. We compared the 5-year DFS between the two arms. Results:The 5-year DFS of 15 cases in arm A was 80%(12/15), which was signiifcantly higher than that of arm B (5.9%, 3/51, P<0.01). Conclusion: The three-step chemotherapy after optimal debulking surgery in stageⅢC-Ⅳ can improve the 5-year DFS. This regimen is high efficacy, mild side-effect witn low cost, which deserves further exploration.

Objective To investigate four methods for establishing animal models of human breast cancer bone metastasis. Methods Thirty-two female nude mice aged 4-6 weeks were divided randomly into four groups (n=8 in each group). 5×105 MDA-MB-231 cells were injected into the body via the left second mammary fat pads (group A), the tail veins (group B), the left heart ventricles (group C) and the left tibia marrow cavities (group D), respectively. Tumor formations in situ were recorded in group A. Deaths after the injection were recorded. The surviving nude mice 49 days after the injection were subjected to pathological examination to determine bone metastasis. Results The rate of tumor formation in situ of group A was87.5 %(7/8). One mouse in group C died after the injection of MDA-MB-231 cells. The bone metastasis rate in groups A, B, C and D was zero (0/8), 12.5 % (1/8), 71.4 % (5/7) and 100 % (8/8), respectively. There was statistically significant difference in the bone metastasis rate between group A and group C, group A and group D, group B and group C; and group B and group D. Conclusion Injections of tumor cells via the breast fat pads and tail veins were not suitablemethods to establish animal models of human breast cancer bone metastasis. The bone metastasis model could be established efficiently by injecting tumor cells into the left heart ventricles or the bone marrow cavity of nude mice.
