1.Determination of Epigoitrin in Radix Isatidis by Solid PhaseExtraction-Quantitative Nuclear Magnetic Resonance Spectroscopy

The method of quantitative nuclear magnetic resonance spectroscopy (qNMR) for determination of epigoitrin in Radix Isatidis was established based on solid phase extraction (SPE).The twice ultrasonic extraction method using pure water was used for fully extracting epigoitrin in sample, and then the extraction was enriched and concentrated by poly-Sery MCX SPE cartridge.The effect of sample pretreatment and qNMR experimental conditions was investigated.The qNMR experiment conditions were selected using DMSO as solvent, calibrated 2,3,5-triiodobenzoate as internal standard, and P1(pulse width)=14.1 μs, d1(pulse delay time)=5 s, NS(number of scan)=256.The .1H-NMR peaks of δ 5.365-5.399 (H-7b, d, 1H) of epigoitrin were chosen as the quantitative peaks.Method validation was performed including precision (intra-day precision RSD was 0.5%, and the inter-day precision was 0.8%), linearity (correlation coefficient r>0.9991), LOD (0.05 mg/g, standard curve method) and LOQ (0.19 mg/g, S/N≥150).The recoveries of the SPE-qNMR were 97.4%-101.7%.The result showed that the method was stable, accurate and reliable.With this method the epigoitrin in a real Radix Isatidis was determined to be <0.19-1.26 mg/g.SPE combining with qNMR could extend the application field of qNMR, especially in the detection of low-content component in complex samples.
2.Effect of curcumin on expression of p-ERK and p-CREB in spinal dorsal horn and dorsal root ganglion in type 2 diabetic neuropathic pain in rats

Objective To evaluate the effects of curcumin on the expression of phosphorylated extracellular signal-related kinase (p-ERK) and phosphorylated cAMP response element binding protein (p-CREB) in the spinal dorsal horn and dorsal root ganglion (DRG) in type 2 diabetic neuropathic pain (DNP) in rats.Methods Type 2 diabetes mellitus was induced by high-fat and high-sucrose diet and intraperitoneal streptozotocin (STZ) 35mg/kg,and confirmed by fasting blood glucose level≥ 16.7 mmol/L in male Sprague-Dawley rats.Type 2 DNP was confirmed by the mechanical withdrawal threshold (MWT) and thermal withdraw latency (TWL) measured on day 14 after STZ administration < 80% of the baseline value,and the rats with type 2 DNP were randomly divided into 3 groups (n =27 each):type 2 DNP group (group DNP),curcumin group (group Cur) and solvent control group (group SC).Curcumin and corn oil 100 mg/kg (25 mg/ml) were injected intraperitoneally once a day for 14consecutive days starting from 14 days after administration of streptozocin in Cur and SC groups,respectively.Another 27 normal rats were served as control group (group C) and were fed with common forage.MWT and TWL were measured at 3,7 and 14 days after curcumin injection (T1 3),and the lumbar segment 4-6 of the spinal cord and DRGs were removed at the same time for determination of the expression of p-ERK and p-CREB (by Western blot).Results Compared with group C,MWT was significantly decreased,TWL was shortened,and the expression of p-ERK and p-CREB in spinal dorsal horn and DRGs was up-regulated at T1-3 in DNP and SC groups,and at T1 in Cur group (P < 0.05).Compared with group DNP,MWT was significantly increased,TWL was prolonged,and the expression of p-ERK and p-CREB in spinal dorsal horn and DRGs was down-regulated at T2,3 in Cur group (P <0.05).There was no significant difference in the MWT,TWL and expression of p-ERK and p-CREB between DNP and SC groups (P > 0.05).Conclusion Curcumin can attenuate type 2 diabetic DNP by inhibiting up-regulation of the expression of p-ERK and p-CREB in the spinal dorsal horn and DRG in rats.
3.Pharmacokinetic study of inosiplex tablets in healthy Chinese volunteers by hyphenated HPLC and tandem MS techniques

Inosiplex is a compound formulation composed of inosine and p-acetaminobenzoic acid (PABA) salt of N,N-dimethylamino-2-propanol (DIP). This study was to investigate the clinical plasma pharmacokinetic properties of DIP and PABA after single and multiple oral doses of inosiplex tablets in healthy Chinese volunteers. The established LC/MS/MS method for plasma DIP determination had a linear range of 0.02-10 mg/mL, and the HPLC method for plasma PABA determination had a linear range of 0.05-40 mg/mL. Linear pharmacokinetic characteristics were found with single oral doses of 0.5, 1.0 and 2.0 g. No obvious accumulation effects were observed for DIP and PABA.
4.Effect of Electromyography Feedback Functional Electrical Stimulation on Plantar Pressure under Walking in Stroke Patients

Objective To evaluate the therapeutic effects of electromyography feedback functional electrical stimulation (FES) on plantar pressure of stroke patients. Methods 18 subjects were included in this study, who were with drop foot resulting from stroke, but could partly dorsiflex the paretic limb's ankle (Manual Muscle Test >2). They were tested on the RS-footscan plate before and after stimulation. The plantar pressure data of paretic foot and walking velocity before and after stimulation were compared. Results The initial touchdown points of most paretic foot were changed from Meta (metatarsal) 5 to heel; the time of initial touchdown of heel medial and heel lateral significantly improved, while that of Meta 3, Meta 4 and Meta 5 significantly delayed; the peak force, impulse and contact area of the heel significantly improved after stimulation (P<0.05). At the same time, the time of initial touchdown of heel medial and heel lateral of nonparetic foot significantly improved (P<0.05). Conclusion Electromyography feedback functional electrical stimulation can improve the gait pattern and the stability of patients with stroke.
6. Influence of occupational aluminum exposure on cognitive function and glutamate receptor protein expression in workers

Objective:
To investigate the influence of occupational aluminum exposure on cognitive function and glutamate receptor protein expression in peripheral blood lymphocytes in workers and the possibility of glutamate receptor being used as a biomarker for cognitive impairment in aluminum workers.
Methods:
From October to December, 2014, cluster sampling was performed to select 121 workers in aluminum electrolysis workshop as exposure group and 231 workers in thermoelectric workshop and logistics department as control group. Mini-Mental State Examination, clock drawing test, digit span test (DST) , verbal fluency test (VFT) , and Fuld Object-Memory (FOM) Evaluation were used to analyze cognitive function. Graphite furnace atomic absorption spectrophotometry was used to measure plasma aluminum level as an exposure indicator. Enzyme-linked immunosorbent assay was used to measure the content of glutamate receptor proteins in peripheral blood lymphocytes, including the subunits of N-methyl-D-aspartate receptor NR1, NR2A, and NR2B and metabotropic glutamate receptor 1 (mGluR1) . The correlation between cognitive function indices and the content of glutamate receptor proteins was analyzed.
Results:
There was no significant difference in plasma aluminum level between the control group and the exposure group (132.52±80.40 μg/L
7.The Hemostatic Effect and Safety Evaluation Tests of Soluble Hemostatic Gauze

Objective The aim of this paper is to study the hemostasis and security of soluble hemostatic gauze in the rabbit liver hemorrhage model.Methods After making the rabbit liver hemorrhage model, the control group used sterile gauze to stop bleeding, the positive control group used TISTAT to stop bleeding, the test group used soluble hemostatic gauze to stop bleeding. Hemostasis, blood loss and animal clinical symptoms were measured. Liver and kidney parameters, along with histopathology were recorded and analyzed. Transmission electron microscopy examination was also done. Results The blood loss is cut back and hemostasis is shortened in the test group. Other tests have no difference with the control group.Conclusion No toxic effects on rabbit are found in the test group. The hemostatic effects have no difference with positive control group.
8.Effects of pre-treatment on Cu2+ absorption of Penicillium janthinellum strain GXCR.

In order to effectively increase capacity of Cu2+ absorption by Penicillium from Cu2+-containing aqueous solution and to study the mechanisms of absorption, effects of eight pre-treatment methods on Cu2+ absorption of Penicillium janthinellum strain GXCR were compared. The results showed that the efficiency of Cu2+ absorption obviously increased through pre-treatment by homogenization, homogenization-basification (NaOH), oven dry (80 degrees C), homogenization-salinification (NaCl), homogenization-detergent and homogenization-polarization (C2H6SO), but significantly decreased after acidification pretreatment with H2SO4. In comparison with the previous reports, the pretreatment in a homogenization-NaOH way could more efficiently enhance the Cu2+ absorption capacity of this fungus. Homogenization-basification (0.5 mol/L NaOH) increased Cu2+ biosorption by 47.95%. The Cu2+ absorption of the mycelia treated by homogenization-basification followed Langmuir isotherm equation, suggesting a surface absorption process. After four cycles of absorption-desorption, mycelia pretreated by homogenization-alkalization still had 70.82% of Cu2+ biosorption efficiency. Infrared reflectance analysis indicated that alkalization treatment made marked effects on molecular groups of C-H, C=O, and C=O in COOH on the mycelial surfaces, and -OH was a key Cu2+-binding group. It is therefore suggested that the Cu2+ absorption by the GXCR is likely to be a chemical absorption process through Cu2+ binding with -OH group on the mycelia.

9.Cu and Fe bioleaching in low-grade chalcopyrite and bioleaching mechanisms using Penicillium janthinellum strain GXCR.

Bioleaching of Cu and Fe in low-grade chalcopyrite using Penicillium janthinellum strian GXCR was studied. As a result, shaking bioleaching was more efficient than submerged bioleaching; Cu bioleaching was much better than Fe bioleaching; under conditions of optimum carbon source (10% sucrose, W/V), optimum nitrogen source (1.5% NaNO3, W/V), shaking bioleaching and the optimum combination of conditions (initial pH 6.0 in leaching media, 5% (W/V) 200-mesh ore and initial inocula of 3.0x10(5) conidia/mL), Cu bioleaching efficiency reached 87.31% (W/W). One of the most important factors affecting Cu bioleaching in shaking bioleaching was the initial pH in leaching media (F > F0.05). The major organic acids for Cu and Fe bioleaching were citric and oxalic acids, respectively. Low bioleaching efficiency by submerged bioleaching was due to low production of citric and oxalic acids. The mechanisms employed by the GXCR in Cu bioleaching included biochemical functions of citric and oxalic acids as well as ore crack caused by mechanical power generated from mycelial growth.

10.Evidence-based practice in the management of maternal discharge preparation services

Objective:To implement evidence-based practice of discharge preparation service in maternal and to explore the clinical effect.Methods:Based on the evidence-based continuous quality improvement framework, apply the evidence on the clinical through evidence acquisition, baseline evaluation, evidence introduction, and after-effect evaluation. Sixty women hospitalized in the obstetrics department of the Affiliated Hospital of Chengde Medical College from January to March 2021 were selected as the baseline group, and 60 women hospitalized in the obstetrics department of the hospital from April to July 2021 were selected as the evidence application group, to evaluate and compare the quality of discharge guidance for practitioners, discharge readiness level, maternal and newborn management knowledge level, incidence of maternal and newborn complications and the compliance of practitioners to evidence.Results:Finally, 10 pieces of evidence applied to clinical practice were included and 15 review indicators were translated. After the application of evidence, the quality of discharge guidance of practitioners increases from (141.83±24.66) to (166.13±10.30), the maternal discharge readiness increased from (127.12 ± 26.29) to (137.63 ± 22.59) , the score of maternal knowledge level increased from (164.58 ± 20.10) to (176.08 ± 24.25) , t values were -7.04, -2.39 and -2.83, all P<0.05. The incidence of postpartum women breast disease, postnatal constipation and newborn eczema, newborn red buttocks, newborn diarrhea, newborn jaundice decreased significantly, the difference were statistically significant, χ2 values were 5.21-7.50, all P<0.05. After the application of evidence, the implementation rate of the review indicators increased from ≤31.67% to ≥86.66%. Conclusions:The evidence-based practice of discharge preparation service in postpartum women improve the quality of discharge guidance for practitioners and compliance with evidence, promote clinical quality improvement, can be used for clinical reference.
