1.Study on influence of compatibility on amygdalin in Mahuang Decoction by HPLC

Object To set up the quantitative method of amygdalin in decoctions prepared from various combinations of the recipe named Mahuang Decoction by HPLC and to determine the change of amygdalin contents in different decoctions and study the influence of compatibility on the contents of amygdalin in decoctions. Methods By L 8(27) orthogonal design and statistic analysis (SPSS 10.0), eight decoctions with Semen Armeniacae Amarum were prepared and analyzed, their amygdalin contents were determined by HPLC. Results The method was simple, accurate, and sensitive and was not interfered by other constituents in the prescription, it can be used to determine the contents of amygdalin in Mahuang Decoction. Influence of Herba Ephedrae, Ramulus Cinnamomin, and Radix Glycyrrhizae on the contents of amygdalin in decoctions were insignificant. Interactions between two of three separately were insignificant too. Conclusion The content of amygdalin in Mahuang Decoction is not markedly influenced by Herba Ephedrae, Ramulus Cinnamomin, and Radix Glycyrrhizae.
2.Effects of human leukocyte antigen-G on proliferation and invasion of JEG-3 cell line

Objective To investigate the effect of human leukocyte antigen-G(HLA-c)on the growth and invasion of JEG-3 cell line and the role of HLA-G in the onset and development of pre-eclampsia.Methods The experiment was composed of three groups:groups of transfection,negative control and blank control.which corresponded to groups of HLA.G siRNA transfection,negative siRNA transfection and no transfection HLA-G overexpressed choriocarcinoma cell line JEG-3 was used.The role of HLA-G in JEG-3cell monolayer was examined by RNA interference technology using HLA-G specific small interfering RNA (siRNA).Expression of HLA.G was detected by reverse transeriptase-polymerase chain reaction and western blot analysis.Changes of cell cycle,apoptosis,proliferation and invasion were respectively detected by methvl thiazolyl tetrazolium(Ma r).flow cytometry assay and transwell test.Results (1)The mRNA and protein levels of HLA.G control group and blank control group were 0.0013±0.0014.0.0163 ±0.0007 and 0.1923 ±0.0384.0.2184 ±0.0153,respectively,which were both significantly different(P<0.05);the number of negative transfcction group was 0.1606±0.0133 and 0.2020±0.0132.which had no significant difference compared with blank control group(P>0.05).(2)The integral absorbance(IA)valUCB of the HLA-G transfecfion group and blank control group were 0.44±0.04 and 0.75±0.13 respectively.which was significantly different(P<0.01);the/A value of negative control group was 0.69±0.10.which was not significantly different compared with blank group(P>0.05).(3)The ratios of G2/M and S phase cells in transfection group were(10.9±2.2)%and(58.6±0.8)%respectively,significantly different compared with the blank control group[(15.4±1.9)%and(52.9±2.3)%respectively;P<0.01].(4)The ratio of early apoptosis cells in transfection group[(14.5±2.7)%]Was significantly increased compared with neg~ive[(5.3 ±1.1)%]and blank control group[(4.7±0.6)%;P<0.01].(5)The invasion number of transfecfion group and blank control group were 121±12 and 452±17 respectively.with a significant eclampsia by regulating proliferation and invasion of trophoblast.

4.14-3-3 tau regulates invasiveness of BeWo cells via ERK1/2 pathway

AIM: To investigate the potential role of 14-3-3 tau in trophoblast cells on invasiveness. METHODS: 14-3-3 tau expression was detected in first-trimester villi, deciduas and human trophoblastic cell line (BeWo) by immunohistochemistry. Small interference RNA (siRNA) targeting 14-3-3 tau was transfected into BeWo cells. The effects of down-regulated 14-3-3 tau on invasion of human trophoblasts cell line BeWo were examined by matrigel invasion assay, and the transcription, translation of E-cadherin and snail were estimated by RT-PCR or Western blotting. U0126 was used to detect the extracellular-signal related kinase 1/2 (ERK1/2) function on down-regulation of 14-3-3 tau induced cell invasion. RESULTS: 14-3-3 tau was detected in the invasive trophoblastic cells in the first trimester villi and that invaded to the deciduas. BeWo cells also expressed 14-3-3 tau. Down-regulation of 14-3-3 tau increased the invasive cell-number of BeWo, as well as the expression of snail, and inhibited E-cadherin. U0126 inhibited the enhanced invasiveness in these cells induced by the down-regulation of 14-3-3 tau. CONCLUSION: 14-3-3 tau may regulate the invasiveness of human trophoblastic cells through ERK1/2 signaling pathway.
5.Postoperative complications of endovascular repair of abdominal aortic aneurysms in poor-risk patients

Objective To review the complications after endovascular aneurysm repair (EVAR) of high-risk abdominal aortic aneurysms in our medical center.Methods Fifty eight elective high-risk EVAR cases from January 2008 to December 2013 were analyzed retrospectively.Results The technical success rate was 96.6% (56 of 58 patients).The overall complication rate in perioperative period was 32.8% (19/58).Eleven systemic complications (19.0%),16 primary endoleaks (27.6%),7 access of site complications(12.1%) were observed.The follow-up rate was 86.2% (51/58).During the follow up period,the mortality was 7.8% (4/51),the re-intervention rate was 7.8% (4/51).The overall complication rate was 29.4% (15/51),including systemic complications (2/51),secondary endoleak (9.8%) and graft-related complications (11.8%,6/51).Conclusions Endoleak,graft-related complications continue to be the main causes of re-intervention in high-risk aneurysm patients after EVAR.
6.Effects of different depths of anesthesia with sevoflurane on cerebrovascular autoregulation in infants

Objective To evaluate the effects of different depths of anesthesia with sevoflurane on cerebrovascular autoregulation in infants.Methods Twenty pediatric patients,of ASA physical status Ⅰ,aged 1-3 yr,undergoing elective hypospadias plasty surgery,were enrolled in the study.Single tube laryngeal mask was inserted after anesthesia was induced with 6% sevoflurane inhalation.Caudal block was performed with 1 ml/kg of 0.2% ropivacaine.Anesthesia was maintained with sevoflurane inhalation.The end-tidal sevoflurane concentration was adjusted to 1.8%,2.5%,3.3% and 4.0%,and each concentration was maintained at this level for 15 min.The cerebral blood flow was collected from the middle cerebral artery immediately before adjusting the next concentration to record the Doppler spectrum and transient hyperemic response ratio (THRR) was measured.Results THRR at different depths of anesthesia with sevoflurane was larger than 1.09,and was within the normal range.THRR was significantly lower when the end-tidal concentration was 2.5%,3.3% and 4.0% than that obtained when end-tidal concentration was 1.8%.No significantdifference was detected in THRR between 2.5% and 3.3 %.THRR was significantly lower when the end-tidal concentration was 4.0 % than that obtained when the end-tidal concentration was 2.5% and 3.3%.Conclusion Although the inhibitory effect on cerebrovascular autoregulation provided by sevoflurane anesthesia provides no obvious clinical significance,it shows statistical significance in infants.
7.Preeclampsia alters the expression of HPA axis related genes in the hippocampus of offspring rats

Objective To explore the effect of preeclampsia on the expression of hypothalamic-pituitary-adrenal (HPA) axis related genesin the hippocampus of adolescent offspring rats.Methods The pregnant Sprague-Dawley rats were randomly divided into two groups to be given 125 mg · kg-1 ·d-1 L-NAME or 1 mL pure saline by daily injection from day 14 to delivery.Serum corticosterone (CORT) levels in neonatal offspring rats were detected by ELISA.The mRNA and protein levels of glucocorticoid receptor (GR),corticotropin releasing hormone (CRH),CRH receptor 1 (CRHR1) and IL-6 in the hippocampus of 8-week offspring rats were determined by real-time PCR and Western blot,respectively.Results Compared with normal neonatal rats,the serum CORT concentration of neonatal rats with preeclampsia was increased (P<0.05).Compared with normal adolescent offspring,the levels of GR,CRH and CRHR1 mRNA and protein were increased in the hippocampus of adolescent rats with preeclampsia (P<0.05),accompanied by increased IL-6 mRNA and protein (P<0.05),as a downstream inflammatory gene of the GR.Conclusions We found that adolescent SD rats exposed to preeclampsia showed alteration in the expression of HPA related genes in the hippocampus,which played a role in the impairment of learning and memory in children.

9.Percutaneous Transluminal Angioplasty and Stent Placement for the Treatment of Long Segmental Atherosclerotic Occlusive Disease of Lower Limb

Objective To evaluate the clinical effects of endovascular angioplasty and stenting for the treatment of TASC B,C arteriosclerotic occlusion(ASO) of lower extremity.Methods After the identification of the occlusion by angiography via femoral artery access by Seldinger method,the occluded arteries (mean length 7.5cm; range 1~20 cm) in 40 patients(47limbs) were treated by percutaneous transluminal angioplasty and stent placement. Results The primary recanalization was successful in 45 of 47 limbs. The technical successful rate was 96%. Thirty one self-expanding stents were deployed. PTA without stent placememt was performed in 16 limbs(of them,11 were blow knee).The ankle brachial pressure index (ABI) increased from (0.36?0.14) preoperatively to (0.82?0.15) on the 7th postoperative day. All patients were follow-uped for 1 to 48 months. Stents were occluded in 4 patients. Restenosis was observed in 1 limbs. All of the rest remained patent in the follow-up period.Conclusion Endovascular angioplasty and stenting can be an effective method for the treatment of long segmental (including below-knee) arteriosclerotic occlusion of lower extremities.
10.Influence of different image processing methods on mensuration of cardiac function in myocardial perfusion imaging

Objective To evaluate the influences of FBP and OSEM (with different iteration numbers or subset numbers) on cardiac function parameters with MPI,and to identify a suitable clinical method.Methods The MPI of 66 patients (33 males,33 females,age range: 18-77 years) from January 2012 to December 2014 were retrospectively analyzed.The imaging data was reconstructed with FBP and 5 different OSEM (4-16 subsets and 2-4 iterations) in order to evaluate if there was any difference of cardiac function parameters calculated by QGS between the different image processing methods.In addition,the results were compared with those of cardiac ultrasonography.One-way analysis of variance was used for data analysis.Results The statistical differences of LVEF,EDV,and ESV(F values: 10.73,4.89 and 5.97,all P<0.05) were found among 6 reconstructed methods.The values of LVEF were (66.14±11.12)%,(75.05±12.10)%,(70.09±11.27)%,(66.88±10.38)%,(64.97±10.25)%,and (62.58±9.84)%.Those of EDV were (77.32±27.58),(67.97±27.56),(75.10±27.89),(81.03±28.11),(84.94±29.07),and (89.98±29.71) ml,and the ESV values were (28.71±10.04),(19.71±16.51),(25.13±17.66),(29.01±18.47),(32.10±19.63),and (35.83±20.41) ml respectively.The cardiac function parameters measured by OSEM (with 2 iterations,12 subsets) were much similar to that measured by cardiac ultrasonography.Conclusion Compared with other 5 processing methods of MPI,the OSEM (with 2 iterations,12 subsets) method may be more suitable for practical clinical application.
