1.Intervention Effects of Pollen Typhae Total Flavone on INS-1 PancreaticβCells Damage Induced by Palmitic Acid

Objective To investigate the effects of Pollen Typhae total flavone (PTF) on INS-1 pancreaticβcell damage induced by palmitic acid ( PA) . Methods INS-1 pancreatic β cells were given long-term induction with PA to establish the impaired cell model, and then were intervened with PTF. Cell viability was determined by tetrazolium salt ( XTT) colorimetry. Results PA impaired the viabilities of INS-1 pancreatic β cells in concentration- and time-dependent manner, and PTF improved the impairment of INS-1 pancreatic β cells induced by PA in concentration -dependent manner. Moreover, PTF showed better improvement on the impairment when the INS-1 pancreatic β cells were impaired more seriously by PA. Conclusion PTF has effects on ameliorating the impairment of INS-1 pancreaticβcells induced by PA for long time.
2.Effects of continuous infusion of vascular endothelial growth factors with mini-osmotic pumps on survival rate of autogenous fat grafts in rats

BACKGROUND: Above curative dose, there is a dose-dependent correlation of the survival rate of fat transplantation with vascular endothelial growth factor (VEGF), the curative dose of VEGF determines the tissue volume of transplanted fat. OBJECTIVE: To investigate the changes of the blood supply rebuilding and the survival rate of the fat transplant in the microenvironment of continuous infusion of VEGF using osmotic pump. DESIGN, TIME AND SETTING: A cellular morphology observational experiment was performed from September 2007 to October 2008 in Department of Plastic Surgery at the Affiliated Hospital of Guangdong Medical College. MATERIALS: Ninety healthy SD rats were used to establish autologous fat transplantation models. Human VEGF was produced by American ONCOGEN Corporation. Type Alzet2002 mini-osmotic pump was produced by American Alzet Corporation. METHODS: Ninety rats were equally divided into 3 groups randomly. The operation control group was only processed into autologous fat transplantation; Experimental control group received autologous fat transplantation and continuous infusion of 2 000 ?L physiological saline using osmotic pump at a speed of 0.5 ?L/h for 14 days; Experimental group received autologous fat transplantation and continuous infusion of 2 000 ?L physiological saline containing 1 ?g VEGF using osmotic pump at a speed of 0.5 ?L/h for 14 days. MAIN OUTCOME MEASURES: At the 7th, 14th, 28th, 42nd, 64th days postoperation, the graft’s survival volumes were measured using mini-scale test tube; the cell morphology changes were observed with hematoxylin-eosin stain; The newly born microvessels on experimental group and control groups were researched with immunohistochemistry staining. RESULTS: At 14 days postoperation, the graft’s survival volumes of experimental group was higher than operation control group and experimental control group (P 0.05). At each time point, the vascular density of the fat transplant in the experimental group was higher than that in other groups (P 0.05). CONCLUSION: VEGF can speed up vasifaction and raise fat graft survival significantly.
3.The clinical observation to the consociation treatment that losartan potassium combined with enalapril used in treating Ⅳ phase diabetic nephropathy

Objective To observe the curative effect that losartan potassium combined with enalapril used in treating Ⅳ phase diabetic nephropathy(DN). Methods Divided 74 Ⅳ phase DN suffers into 3 groups randomly.Losartan group:taken losartan potassium lablets 100 mg once every day orally,enalapril group:taken enalapril tablets 10 mg once every day orally,consociation group:taken the two drugs above ai one time with the same dosage.The treatment in the 3 groups were all above 3 years.monitoring the 24-hour albuminuria and SCr back and forth the treatment.Results It worthed statistics meaning that the 24-hour albuminuria in the 3 groups all declined(P<0.05).The effect of consociation group did better(P<O.01).In term of SCr,it declined in the consociation group worthed statisties meaning only.Conclusion The consociation application with losartan potassium and enalapril can control the proteinuria of Ⅳ phage DN suffers efficiently as well as defer process of kidney diseases.
4.Sorafenib ameliorates renal fibrosis through inhibition of TGF-β/Smad pathway

Objective To investigate the effect of sorafenib in ameliorating renal fibrosis and its possible mechanisms.Methods Rats were subjected to unilateral ureteral obstruction (UUO ) and intragastrically administered sorafenib.NRK-52E cells were treated with transforming growth factor-β1 (TGF-β1)and sorafenib. HE staining was used to visualize renal fibrosis.α-SMA and E-cadherin expressions in kidney tissue and NRK-52E cells were performed using immunofluorescence.The cell cycle of NRK-52E cells was determined by flow cytometry analysis.Smad3 and p-Smad3 protein expressions in NRK-52E cells were detected by Western blot analysis. Results HE staining showed that kidney interstitial fibrosis,tubular atrophy,and inflammatory cell infiltration in the sorafenib-treated UUO groups were significantly decreased compared with the vehicle-treated UUO group (P<0.05).Compared with those in UUO and TGF-β-stimulated NRK-52E groups,the expression of a-SMA decreased but E-cadherin expression increased in the UUO kidneys and NRK-52E cells of the sorafenib-treated groups (P<0.05).After 24 h stimulation with TGF-β1 5 ng/mL,the number of cell cycles arrested in G0/G1 phase was significantly increased and the number of cells that entered G2 ,S phase decreased (P<0 .0 5 ).Compared with that in TGF-β-stimulated NRK-52E groups, p-Smad3 decreased in the sorafenib-treated groups (P<0.05). Conclusion Our results suggest that sorafenib may be useful for the treatment of renal fibrosis through suppressing TGF-β/Smad3 signaling.
5.Correlation study of DWI types and degree of neurologic impairment in acute ischemic stroke patients with atrial fibrillation

Objective To explore the correlations of diffusion-weighted imaging (DWI) types and the degree of neurologic impairment in acute ischemic stroke patients with atrial fibrillation.Methods DWI images and National Institutes of Health Stroke Scale(NIHSS) of 186 patients with acute ischemic stroke patients with atrial fibrillation were collected retrospectively.The correlation of DWI features and NIHSS was analyzed.Results On DWI,all acute ischemic stroke patients with atrial fibrillation presented high signal intensity.Single cortex-subcortical infarction mostly appeared in the anterior circulation(94,50.5%);Multi-infarction commonly occurred in the posterior circulation(18,13.0%);The neurological deficit scores of subcortical-cortex infarction in the left anterior circulation(16.75±7.10) were higher than that in the right side(13.50±5.70)(P<0.05).The neurological deficit scores of cortex-subcortical infarction in the posterior circulation (6.38±2.03) were significantly lower than that in the multi-infarction (16.77±8.90) (P<0.05).Conclusion DWI types are valuable for etiological diagnosis in ischemic stroke.Combination with NIHSS score could provide a basis for clinical individual treatment programs selection and prognostic evaluation.
6.Efficacy and safty of preventive antibiotics for stroke associated pneumonia in patients with acute stroke: a Meta analysis

Objective To determine if preventive antibiotics is effective in stroke associated pneumonia in patients with acute stroke.Methods Medline (January 1950 to January 2017),EMBASE (January 1974 to January 2017),Cochrane Library (January 2009 to January 2017),CNKI (January 1979 to January 2017)and Wanfang data (January 1998 to January 2017) were searched for randomized controlled trial comparing preventive antibiotics with placebo/blank controls for stroke associated pneumonia in patients with acute stroke.The included studies were screened out strictly based on the criterion of inclusion and exclusion.The quality of included studies was evaluated and the data were extracted by two researchers independently.RevMan 5.1 was used for Meta analysis.Results A total of 4 studies involving 3894 patients were included.The results of Meta-analysis indicated that there was no significant difference in the incidence of stroke associated pneumonia between preventive antibiotics and control groups (OR=0.96,95%CI:0.72-1.29,P=0.810);and there were no statistically significant differences in mortality (OR=1.05,95%CI:0.88-1.25,P=0.570) and good outcome (modified Rankin scale ≤ 2,OR=1.02,95%CI:0.89-1.17,P=0.780).There were no serious adverse reactions related to the studied drugs in 4 studies.Conclusion Preventive antibiotics could neither reduce the incidence of stroke associated pneumonia nor decrease the mortality or improve the proportion of good outcome.
7.Establishment of HPLC Fingerprint of Schisandra sphenanthera and S. chinensis and Analysis of Chemical Pat- tern Recognition

OBJECTIVE:To establish HPLC fingerprint of Schisandra sph enanthera and S. chinensis,and to analyze chemical pattern recognition. METHODS :HPLC method was adopted. Using schizandrin A as reference ,HPLC fingerprints of 10 batches of S. sphenanthera and S. chinensis (N1-N10,S1-S10) were drawn. Similarity Evaluation System of TCM Chromatographic Fingerprint(2012 edition)was adopted for similarity evaluation to determine the common peaks. SPSS 20.0 and SIMCA 14.1 software were used for HCA ,unsupervised madel of PCA ,supervised model of OPLS-DA. Using variable importance projection (VIP)value greater than 1 as the standard ,the differential markers that affected the quality of S. sphenanthera and S. chinensis were screened. RESULTS :S. sphenanthera and S. chinensis were identified 32 and 33 common peaks ,respectively. The similarity of 10 batches of S. sphenanthera and 10 batches of S. chinensis were all higher than 0.9,and the similarity of S. sphenanthera and S. chinensis was 0.05. A total of 19 characteristics peaks were identified ,among which five common peaks were identified as schisandraol A ,schisandraol B ,schisantherin A ,schizandrin A and schisandrin B by reference. HCA results showed that N 1-N10 were clustered into one category ,and S 1-S10 were clustered into one category ,of which N 1,N3,N8,and N 9 were clustered into one category ,and the rest were clustered into one category ;S1,S3,S6,and S 9 were grouped together ,and the rest were grouped together. The results unsupervised model of PCA showed that the cumulative variance contribution rate of the first two principal component factors was 87.20%. Supervised model of OPLS-DA showed that schizandrin A ,schisandraol A ,schisantherin A and schisandrin B were the differential markers that affected 、the quality of S. sphenanthera and S. chinensis (VIPs were 2.29,2.24,1.73,1.48,respectively). 8.Effect of Tongxinluo Capsules on Use of Anti-ischemic Drugs in Patients with Chronic Coronary Syndrome of Qi Deficiency and Blood Stasis: A Multicenter, Prospective Cohort Study

ObjectiveTo investigate the effect of Tongxinluo capsules on the use of anti-ischemic drugs in patients with chronic coronary syndrome (CCS) of Qi deficiency and blood stasis. MethodA multicenter,prospective cohort study was conducted,with Tongxinluo capsules intervention as the exposure factor. Patients were divided into an exposed group (combination of traditional Chinese and western medicine) and a non-exposed group (western medicine alone),and followed up for one year. The use of anti-ischemic drugs was observed on the day of enrollment and at 3,6,12 months. ResultA total of 186 patients were enrolled,with 128 in the exposed group and 58 in the non-exposed group. There were no statistically significant differences in baseline characteristics between the two groups. At the 3-month follow-up,the types of first-line anti-ischemic drugs used in the exposed group were significantly fewer than those in the non-exposed group (P<0.01),and this difference remained statistically significant at 6 months (P<0.05) but was no longer significant at 12 months. At the 3- and 6-month follow-ups,there were no significant differences between the two groups in the types of second-line anti-ischemic drugs used. However,at the 12-month follow-up,the types of second-line anti-ischemic drugs used in the exposed group were significantly fewer than those in the non-exposed group (P<0.01). At the 3-month follow-up,both groups showed a reduction in the types of first-line anti-ischemic drugs used compared to baseline (P<0.05),with a more pronounced reduction in the exposed group (P<0.05). At the 6-month follow-up,the exposed group showed a significant reduction in the types of second-line anti-ischemic drugs used compared to baseline (P<0.05),while no significant changes were observed in the non-exposed group. At the 12-month follow-up,the difference in the types of second-line anti-ischemic drugs between the exposed and non-exposed groups was statistically significant (P<0.05),while there was no significant difference in the types of first-line anti-ischemic drugs. ConclusionTongxinluo capsules can effectively reduce the use of anti-ischemic drugs in patients with CCS of Qi deficiency and blood stasis.

TSCI have dyskinesia and sensory disturbance that can cause various life-threaten complications. The patients with traumatic spinal cord injury (TSCI), seriously affecting the quality of life of patients. Based on the epidemiology of TSCI and domestic and foreign literatures as well as expert investigations, this expert consensus reviews the definition, injury classification, rehabilitation assessment, rehabilitation strategies and rehabilitation measures of TSCI so as to provide early standardized rehabilitation treatment methods for TSCI.
10.Study on quality control method of Qiguiling mixture

OBJECTIVE To establish the fingerprint of Qiguiling mixture and the method for the content determination of 4 kinds of active components such as calycosin-7-glucoside, so as to control the quality of Qiguiling mixture. METHODS The fingerprints of 12 batches of Qiguiling mixture were established by HPLC. SPSS 25.0 software was used for cluster analysis and principal component analysis, and SIMCA 14.1 software was used for orthogonal partial least squares-discriminant analysis. The variable importance in projection (VIP) value greater than 1.0 was used as the index to screen the differential components. The contents of calycosin-7-glucoside, glycyrrhizin and glycyrrhizic acid were calculated by the quantitative analysis of multi- components by single marker (QAMS) with hesperidin as the internal reference, and the results were compared with external standard method. RESULTS In the fingerprints of 12 batches of samples, 17 common peaks were identified, and the similarities were more than 0.940. A total of 4 common peaks were identified, which were calycosin-7-glucoside (peak 6), glycyrrhizin (peak 8), hesperidin (peak 12), and glycyrrhizic acid (peak 17). The 12 batches of samples could be clustered into two categories, S4, S7-S9 and S11-S12 were clustered into one category, and the other batches of samples were clustered into one category. The cumulative variance contribution rate of the six principal components was 85.840%, and VIP values of peaks 15, 14, 4, 8 (glycyrrhizin) and 9 were all greater than 1.0. The relative error between the results of QAMS and external standard method was less than 5% (n=3) for the contents of calycosin-7-glucoside, glycyrrhizin and glycyrrhizic acid. CONCLUSIONS Established HPLC fingerprint and content determination method in this study can be used for quality control of Qigiling mixture. Five components such as glycyrrhizin are the differential components.
