2.Chemiluminescence determination of melamine with LuminoI-K3Fe(CN)6 system

A sensitive chemiluminescence(CL) method was developed for determining melamine in urine and plasma samples based on the fact that melamine can remarkably enhance the chemiluminescence of Luminol-K3 Fe(CN)6 system in alkaline medium.The determination conditions were optimized.Under optimum conditions,the chemiluminescence intensity had a good linear relationship with melamine in the range of 9.0 × 10 97.0 × 10 6 g/mL with a correlation coefficient of 0.9992.The detection limits (3σ) were 3.54 ng/mL for urine sample and 6.58 ng/mL for plasma sample.The average recoveries of melamine were 102.6% for urine sample and 95.1% for plasma sample.Melamine in samples was extracted with liquid-liquid extraction procedures and the assay results coincided very well with that determined with flow injection chemiluminescence method.The method provides a reproducible and stable approach for sensitive detection and quantification of melamine in urine and plasma samples.
3.Study on screening potential allergenic proteins from infant milk powders based on human mast cell membrane chromatography and histamine release assays

Cow's milk allergy is mainly observed in infants and young children. Most allergic reactions affect the skin, followed by the gastrointestinal and respiratory systems. Conventional diagnosis is based on po-sitive allergy studies and evaluation of parameters including IgE and IgG1 levels, acute allergic skin response and anaphylactic shock reactions. We developed a cell membrane chromatographic (CMC) method based on human mast cells (HMC-1) for screening potential allergens in infant formula milk powders (IFMP). HMC-1 cell membranes were extracted and mixed with silica to prepare cell membrane chromatography columns (10 mm × 2 mm i.d., 5 mm). Under the conditions of 0.2 mL/min flow rate and 214 nm detection wavelength, human breast milk showed no retention. However, IFMP showed clear retention. The retained fractions were collected and analyzed through matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS). Four major milk proteins, i.e., α-casein, β-casein, α-lactalbumin, and β-lactoglobulin A, were identified. Furthermore, these proteins and β-lacto-globulin B showed clear retention on HMC-1/CMC columns. To test the degranulation effects of the five proteins, histamine and β-hexosaminidase release assays were carried out. All five proteins induced HMC-1 cells to release histamine and β-hexosaminidase. Also, we established a reversed phase liquid chromatographic (RPLC) method for the determination of the five proteins in IFMP and the results showed that 90% proteins in IFMP were α-casein and β-casein. We concluded that cow's milk proteins may be potential allergens and caseins cause more β-casein allergic risk than other proteins. This con-clusion was consistent with other studies.

Literature analysis,expert consultation,case analysis and other methods were used to establish a public hospital specialty operation evaluation system suitable for high-quality development,including 2 first-level indicators of medical ability and economic operation and 13 second-level indicators.Urology surgery in the pilot hospital was taken as an example to carry out surgical operation effect evaluation.To strengthen the evaluation of public hospital specialized operation,it should pay attention to the evaluation of medical capacity,strengthen the cooperation of functional departments,and improve the supporting policies of operation,for better promoting the development of high-quality operation of hospitals.
5.Association of serum uric acid with outcome at one year in patients with acute ischemic stroke

Objective:To investigate the association of serum uric acid (SUA) with the outcome in patients with acute ischemic stroke (AIS) at one year after onset.Methods:Patients with AIS admitted to the Department of Neurology, Dagang Hospital, Tianjin Binhai New Area were included retrospectively. According to the modified Rankin Scale (mRS) score at 1 year after onset, patients were divided into a good outcome group (0-2) and a poor outcome group (>2). They were also divided into a survival group and a death group based on their survival status at 1 year after onset. The clinical baseline data and laboratory tests were compared. Multivariate logistic regression analysis was used to determine the associations of SUA with poor outcome and death in patients with AIS. Results:A total of 651 patients were enrolled, including 430 males (66.1%) aged 67.5±11.7 years. Four hundred and fourteen patients (63.6%) were in the good outcome group, and 237 (36.4%) were in the poor outcome group. There were 568 patients (87.3%) in the survival group and 43 (16.7%) in the death group. Univariate analysis showed that there were differences in age, atrial fibrillation, history of stroke or transient ischemic attack, baseline National Institutes of Health Stroke Scale (NIHSS) score, and pre-admission mRS score between the poor outcome group and the good outcome group. The homocysteine, SUA, white blood cell count, and creatinine in the poor outcome group were higher than those in the good outcome group, while the red blood cell count and hemoglobin were lower than those in the good outcome group (all P<0.05). There were differences in age, history of ischemic heart disease, atrial fibrillation, history of stroke or transient ischemic attack, baseline NIHSS score, pre-admission mRS score, and stroke etiology classification between the survival group and the death group. Hemoglobin and triglycerides in the survival group were higher than those in the death group, while the white blood cell count and creatinine were lower than those in the death group (all P<0.05). Multivariate logistic regression analysis showed that SUA was an independent risk factor for poor outcome in patients with AIS (odds ratio 1.004, 95% confidence interval 1.001-1.006; P<0.01), but there was no independent correlation with death. Conclusion:Higher SUA is an independent risk factor for poor outcome at one year after onset in patients with AIS.
6.Measurement and analysis of hand-transmitted vibration of vibration tools in workplace for automobile casting and assembly

Objective To investigate the features of hand-transmitted vibration of common vibration tools in the workplace for automobile casting and assembly.Methods From September to October,2014,measurement and spectral analysis were performed for 16 typical hand tools (including percussion drill,pneumatic wrench,grinding machine,internal grinder,and arc welding machine) in 6 workplaces for automobile casting and assembly according to ISO 5349-1-2001 Mechanical vibration-Measurement and evaluation of human exposure to hand-transmitted vibration-part 1:General requirements and ISO 5349-2-2001 Mechanical vibration-Measurement and evaluation of human exposure to hand-transmitted vibration-Part 2:Practical guidance for measurement in the workplace.Results The vibration acceleration waveforms of shearing machine,arc welding machine,and pneumatic wrench were mainly impact wave and random wave,while those of internal grinder,angle grinder,percussion drill,and grinding machine were mainly long-and short-period waves.The daily exposure duration to vibration of electric wrench,pneumatic wrench,shearing machine,percussion drill,and internal grinder was about 150 minutes,while that of plasma cutting machine,angle grinder,grinding machine,bench grinder,and arc welding machine was about 400 minutes.The range of vibration total value (ahw) was as follows:pneumatic wrench 0.30~11.04 m/s2,grinding wheel 1.61 ~8.97 m/s2,internal grinder 1.46~8.70 m/s2,percussion drill 11.10~14.50 m/s2,and arc welding machine 0.21~2.18 m/s2.The workers engaged in cleaning had the longest daily exposure duration to vibration,and the effective value of 8-hour energy-equivalent frequency-weighted acceleration for them[A (8)] was 8.03 m/s2,while this value for workers engaged in assembly was 4.78 m/s2.The frequency spectrogram with an 1/3-time frequency interval showed that grinding machine,angle grinder,and percussion drill had a high vibration acceleration,and the vibration limit curve was recommended for those with a frequency higher than 400 min/d.Conclusion The workers who are engaged in cleaning,grinding,and a few positions of assembly and use grinding machine,angle grinder,internal grinder,and percussion drill are exposed to vibrations with a high vibration acceleration and at a high position of the frequency spectrum.The hand-transmitted vibration in the positions of cutting,polishing,and cleaning in automobile casting has great harm,and the harm caused by pneumatic wrench in automobile assembly should be taken seriously.

8.The eEffects of Pixu Yihao Fang on small intestinal GLUT1 of small intestine in spleen-deficient rats with functional dyspepsia rats with spleen deficiency

Objective To investigate the effects of Pixu Yihao Fang (PXYHF, No.1 Formula for Spleen Deficiency) on the expression of Glucose Transporter (GLUT1)protein (GLUT1) and mRNA in small intestine tissue of FD spleen-deficient rats and functional dyspepsia (FD) the intervention of Pixu I Fang.Methods 70 10-day-old male SD rat pups were randomly divided into normal control group (n=10), FD model group (single model, n=10), and spleen-deficient FD model group (double model, n=50).The normal control group received intragastric administration of 0.2 mL of 2% sucrose, while the single and double model groups were given 0.2 mL of 0.1 % iodoacetamide (IA)IA in 2 % sucrose, for 6 days.6 weeks after birth consecutive days, the Modified multiple platform method (MMPM) was also applied to establish double model for 14 days after week 6.These spleen-deficient FD rats were randomly divided into double model control group, domperidone group, PXYHF low-dose, medium-dose and high-dose dose groups, and were orally gavaged with 10 mL/(kg·d) of distilled water, 3.125 mg/(kg·d) of domperidone, 1.275, 2.55, 5.1 g/(kg·d) of PXYHF respectively for 14 days.Immunohistochemistry, Western-blot and RT-PCR methods were used to measure the GLUT1 protein expression in liver tissues.Results Compared with the normal group, average optical density of GLUT1 protein in small intestinal mucosal tissue, the expression of GLUT1 protein and GLUT1 mRNA in small intestinal tissue of double model group were decreased (P<0.05, P<0.01);Compared with double model group, average optical density of GLUT1 protein in small intestinal mucosa tissue, expression of GLUT1 protein and GLUT1 mRNA in small intestinal tissue of low-dose PXYHF were increased (P<0.01).Conclusion The expression of GLUT1 gene and protein decreased in the FD rats with spleen deficiency, and Pixu Yihao Fang could increase the expression of GLUT1 gene and protein.
9.Influence of Pixu Fang 1 (Spleen-deficiency Formula 1) on protein and gene expressions of myosin light chain in rats with functional dyspepsia-spleen deficiency pattern

Objective To investigate the intervention of Pixu Fang 1 (Spleen-deficiency Formula 1) to the protein and mRNA expressions of myosin light chain (MLC) in gastric tissue in rats with functional dyspepsia (FD)-spleen deficiency pattern(SDP).Methods SD rats (n =60,aged 7 d) were randomly divided into normal group (n =10) and FD-SDP model group (n =50).The normal group was orally given 2% sugar solution and FD-SDP model group was given 0.1% iodoacetamide sugar solution in a dose of O.2 mL each rat a day for 6 d.The FD-SDP model group was fed with normal forage until rats aged 6 weeks and additionally given modified multiple platform method (MMPM) continuously for 14 d.After modeling,FD-SDP model group was divided randomly into model group (distilled water,10 mL/kg · d),domperidone group (3.125 mg/kg · d),low-dose Pixu Fang 1 group (low-dose group,1.275 g/ kg · d),mid-dose Pixu Fang 1 group (mid-dose group,2.550 g/kg · d) and high-dose Pixu Fang 1 group (high-dose group,5.100 g/kg · d,each n =10),and all groups were given orally corresponding medicinal for 14 d.The protein and mRNA expressions of MLC in gastric tissue were detected by using immunohistochemistry technique,Western blotting and reverse transcription-polymerase chain reaction (RT-PCR).Results Compared with normal group,MLC protein expression decreased in model group detected by immunohistochemistry technique and Western blotting,and MLC mRNA expression decreased in model group detected by RT-PCR (P <0.01).Compared with model group,MLC protein expression increased in low-dose group,mid-dose group and high-dose group detected by immunohistochemistry technique (P < 0.05,P < 0.01).MLC protein expression increased in mid-dose group and high-dose group detected by Western blotting (P < 0.O1).MLC mRNA expression increased in high-dose group detected RT-PCR (P < 0.01).Conclusion There are decreases of protein and mRNA expressions of MLC in FD-SDP rats,and Pixu Fang 1 can up-regulate these expressions.
10. Tea consumption and the risk of lung cancer in Chinese males: a prospective cohort study

Objective:
To investigate the association between tea consumption and lung cancer risk in Chinese males.
Methods:
Tea consumption and incident lung cancer cases were collected on a biennial basis among males in Kailuan Cohort during 2006-2015. Up to 31st December 2015, a total of 103 010 male candidates from the Chinese Kailuan Male Cohort Study were enrolled in the present study. Cox proportional hazards regression model was used to evaluate the association between tea consumption and risk of lung cancer in males.
Results:
The age of male candidates was (51.3±13.4)years old. There were 828 810.74 person-years of follow-up and 8.91 years of median follow-up period. During the follow-up, 964 lung cancer cases were identified. In male, the rate of never cosumers, tea drinkers (<4/week) and tea drinkers (≥4/week) were 58.17%(
