1.Effect of Cold Stress on Connexin43 Protein Expression With Drug Intervention in Neonatal Rats’ Myocardial Cells

Objective: To study the effects of acute cold stress on connexin43 (Cx43) protein expression with drug intervention, and cell to cell conduction with its mechanism in neonatal rats’ myocardial cells.
Methods: The primary neonatal rats’ myocardial cell culture was conducted in 4 groups. Group① , the cells were normally cultured, Group②, the cells were cultured at 4℃, Group③, the cells were cultured at 0℃and Group④, the anti-arrhythmia peptide (AAP 10) was added in Group②and Group③. The apoptosis rate of myocardial cells was evaluated by lfow cytometry assay, mRNA and protein expressions of CX43 were examined by RT-PCR and Western blot analysis, and CX43 phosphorylation product (P-CX43) was detected.
Results: Compared with normally cultured cells, the myocardial cell apoptosis rate was obviously increased by acute cold stress at 4℃and 0℃with time extension. The mRNA expression of Cx43 was decreased at varying degrees at 4℃and 0℃stimulation, the protein expression of Cx43 was decreased at varying degrees at 4℃and 0℃stimulation with time extension, and P-Cx43 level was decreased. While the APP 10 intervention may obviously elevate the protein levels of Cx43 and P-Cx43.
Conclusion: Acute cold stress could reduce the protein expression of CX43 and P-CX43, while APP 10 intervention may elevate such expression and improve the cell to cell conduction in neonatal rats’ myocardial cells.
2.Effect of PTENP1 on Proliferation and Apoptosis of Colorectal Cancer Cells and Its Molecular Mechanism

Objective To investigate the effect of PTENP1 on the proliferation and apoptosis of colorectal cancer cells and its molecular mechanism. Methods We selected 107 cases of colorectal cancer and corresponding adjacent tissues as the research objects. The expression level of PTENP1 was analyzed by fluorescence quantitative PCR. Colon cancer HT29 cells with PTENP1 overexpression (PTENP1 group) and empty vector cell line (control group) were established by lentivirus. The cell proliferation and apoptosis were analyzed by CCK8 and flow cytometry. The PTENP1 target gene was analyzed by bioinformatics and double luciferase reporter genes. 3.Pathogenesis, diagnosis and treatment of cerebral fat embolism.

In this study, we analyzed two cases of pure cerebral fat embolism and reviewed related literatures to explore the pathogenesis, clinical manifestations, diagnosis and treatment of cerebral fat embolism, improve the treatment efficiency and reduce the misdiagnosis rate. In our cases, patients fully returned to consciousness at the different times with good prognosis, normal vital signs and without obvious sequelae. For patients with the limb fractures, who developed coma without chest distress, dyspnea or other pulmonary symptoms 12 or 24 h post injury, cerebral fat embolism should be highly suspected, except for those with intracranial lesions, such as delayed traumatic intracerebral hemorrhage, etc. The early diagnosis and comprehensive treatment can improve prognosis.

4.Effects of three different iodine interventions on the speed of normalization of enlarged thyroid gland due to iodine deficiency.

OBJECTIVETo compare the effects of three different iodine interventions on the speed of normalization of enlarged thyroid gland.
METHODSSchoolchildren aged 8 - 10 years were randomized divided into one of three groups: group A was given iodized salt by researchers with an iodine concentration of 25 mg/kg; group B used iodized salt purchased from the market; and group C was similar to group B with additional intake of iodized oil capsules containing 400 mg iodine at the beginning of the study. Salt iodine content was measured bimonthly for 18 months and indicators of iodine deficiency were measured at baseline and 6, 9, 12 and 18 months thereafter.
RESULTSThe prevalence of goiter measured by ultrasound, based on the World Health Organization (WHO) body surface area reference > 97(th) percentile, was 18% at baseline and declined to less than 5% by 12 month in groups A and C respectively, and to 9% after 18 months in group B. Rates of goiter were similar by palpation or by ultrasound. The median urinary iodine was 94 micro g/L at baseline and increased in all groups to > 200 micro g/L at the 6-month follow-up.
CONCLUSIONSIn this sample of schoolchildren with initially low or moderate level of iodine deficiency, the group receiving salt with 25 mg/kg (group A) recovered from iodine deficient on all indicators after 18 months of study. However when the iodine content of salt was floating, as seen in group B, the sizes of thyroid did not yet achieve normal status by 18 months.

5. Cardioversion efficacy of nifekalan in patients with sustained atrial fibrillation after radiofrequency ablation

Objective:
To evaluate the efficacy and safety of nifekalan (NIF) on cardioversion in atrial fibrillation (AF) patients post radiofrequency ablation, and investigate the relevant factors related to the cardioversion efficacy of NIF.
Methods:
We screened patients with sustained AF rhythm after radiofrequency ablation between November 2016 and July 2018. Participants were treated with intravenous NIF 0.4 mg/kg within 5-10 minutes after ablation. We observed the adverse reaction, and monitored the rhythm, heart rate, QT interval and QTc interval before the medication and at 5, 10, 20, 120 min after the medication. According to the drug outcome of NIF, patients were divided into conversion group and non-conversion group, related factors affecting conversion efficacy were evaluated using logistic regression analysis.
Results:
(1)A total of 116 patients were enrolled in the study (63 males and 53 females, mean age was (64±18) years). Among them, 72 patients were converted to sinus rhythm, and the overall successful rate was 62.1%. There were 84 patients with persistent AF, of which 50 cases (59.2%) were restored to sinus rhythm. There were 32 patients with paroxysmal AF, 22 cases (68.8%) of them were restored to sinus rhythm. The conversion time was 1.5 to 12 (6.8±3.4)min. 6.Surveillance of Escherichia coli O157:H7 among animals in Jiangsu province in 1999.

OBJECTIVETo understand the Escherichia coli O157:H7 carrier rate of host animals and the toxic gene of the strains in different areas in Jiangsu province.
METHODSSurveillance spots were set up in different areas, to collect feces of pigs, chickens, sheep, cattle to culture for O157:H7 with immunomagnetic separation as well as detection of toxic gene of the strain with MPCR were both carried out.
RESULTSOne hundred and seventy strains of O157:H7 were separated from 1 767 feces of different animals in six spots, with a overall positive rate 9.62%. The positive rates of cattle and sheep were 19.05% and 12.01% respectively. Among 85 strains SLT1, SLT2, eaeA and hly toxic genes were detected. In which, 56.47% of the strains were positive curturely while 79.17% of them carried SLT2, eaeA and hly gene simultaneously.
CONCLUSIONThe positive rate of O157:H7 in animals and the positive rates of strains were correlated to the incidence of the area. The highest rates were seen in areas where there had been O157:H7 epidemic, followed by the areas where there were only scattered cases identified while the lowest was in areas with no patients. Data indicated that it was important to enforce the surveillance of O157:H7 in animals to better predict and control of the disease.

7.Research on the Characteristics and Ideology of Moxibustion in Mawangdui Medical Books

This article systematically studied the moxibustion methods embedded in the 15 medical books of the Mawangdui medical literature. Starting from the origin of the medical moxibustion culture in Mawangdui, this paper comprehensively analyzed the records of moxibustion. By analyzing the Classic of Moxibustion to Eleven Foot-arm Channels (《足臂十一脉灸经》) and Classic of Moxibustion to Eleven Yin-yang Channels (《阴阳十一脉灸经》) as well as Formulas for Fifty-two Diseases (《五十二病方》), which have relatively rich records of moxibustion, it is found that moxibustion involves more than 170 diseases and syndromes, covering 12 categories such as limb diseases related to meridians and collaterals, lung system diseases, and heart and brain diseases. In summary, the distinctive characte-ristics of moxibustion in Mawangdui medical books include primitive simplicity, combination of witch culture and medical practices, philosophical deduction, centripetal circulation, diverse moxibustion materials, extensive cove-rage of diseases and syndromes, taboos in moxibustion, and the integration of prevention and treatment. The ideas and applications of moxibustion mainly manifest in preventing diseases before they occur and preventing changes in existing diseases, distinguishing and treating each disease based on different causes, and combining treatment based on meridians, collaterals and comprehensive diseases.
