1.Study on the Level of Interleukin-6 mRNA of Placentae and Amnion in Severe Pregnancy-Induced Hypertension

Objective To investigate the immune changes of interleukin-6(IL-6) of placentae and amnion in severe pregnancy-induced hypertension(PIH) and fetal growth retardation(FGR). Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect IL-6 mRNA levls of placentae and amnion. Results The level of IL-6 mRNA in placentae and amnion in severe PIH group (0.71?0.07,0.81?0.02) are significantly lower than those in normal group(P0.05). Conclusions IL-6 is involved in the immune injury in severe PIH. The decreased levels of IL-6 in placentae and amnion may be the cause of fetal growth retardation

Around the core issue how to evaluate and test hypothesis, starting from the concept of hypothesis and scientific hypothesis, learning form recent progress and understanding at home and abroad, we have analyzed the type of scientific hypothesis, test methods and pre-mortem and post-mortem evaluation criteria in depth, established scientific vision and discipline ruler, explored the awareness level of the Chinese medicine hypothesis, pointed out the problems and gaps. Whereby, we have analyzed and demonstrated brain regulating five internal organs concept of wholism scientific hypothesis, and proven it belongs to the scientific hypothesis. We were confident that these works mentioned above would lead the theoretical reform changing Chinese medicine hypothesis from traditional description to the nature clarification.
3.Role of protein kinase C in rat myocardial cells with anoxia-reoxygenation injury preconditioned with sufentanil

Objective The protein kinase C ( PKC) is an essential signaling substance in the early protection of cells in pre-conditioning.This study was to investigate the role of PKC in the myocardial cells of the rat model of anoxia -reoxygenation (A-R) in-jury preconditioned with sufentanil . Methods Primary myocardial cells isolated and cultured for 5 days were allocated to a control , an A-R, a sufentanil preconditioning ( SF) , and a phorbol+sufentanil preconditioning ( PMA +SF) group.The A-R injury model was established with cultured myocardial cells from neonatal rats , which were preconditioned with sufentanil at the concentration of 0.000 3 μmol/L in the SF group or phorbol followed by sufentanil 10 minutes later in the PMA +SF group.Then the proliferation of the cells was detected , the optical density of the Cx 43 protein observed by immunofluorescence confocal technology , the apoptosis rate of the cells determined by flow cytometry, and the total Cx43 proteins calculated by Western blot. Results Cell proliferation was signifi-cantly increased in the A-R, SF, and PMA+SF groups as compared with the control (P<0.05), higher in the SF and PMA +SF than in the A-R group (0.498 0 ±0.0432 4 and 0.7240 ±0.1234 vs 0.325 8 ±0.023 5, P<0.05), and in the SF than in the PMA+SF group (P<0.05).Flow cytometry showed significantly increased rates of early , late, and total cell apoptosis in the A -R ([4.96 ±0.59], [18.77 ±0.92], and [23.73 ±0.51]%), SF ([5.86 ±0.38], [10.37 ±0.38], and [16.23 ±0.32]%), and PMA+SF group ([5.71 ±0.58], [5.54 ±0.43], [11.24 ±0.62]%) as compared with the control (P<0.05), remarkably lower in the SF and PMA +SF than in the A-R group (P<0.05), and the late and total cell apoptosis rates markedly lower in the SF than in the PMA+SF group (P<0.05). Conclusion Sufentanil has a protective effect and PKC agonists combined with sufentanil may add to the effect on myocardial cells in A -R injury.
4.Determination of Arsenic and Its Species in Dry Seafood by High Performance Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry

Arsenic and its species in dry seafood products(DSPs) Kelp, Sargassum fusiform, Laver, Enteromorpha prolifera, were systematically determined by high performance liquid chromatography-inductively coupled plasma mass spectrometry(HPLC-ICP-MS). It was found that the of arsenic species in botanic DSPs are three unknown arsenicals(U_2, U_3 and U_4), which are speculated as arsenosugars and take approximate 63.5% 92.7% of all arsenic species detected. These arsenosugars were indentified as dimethylarsinic(DMA)-glycerol ribose, DMA-phosphate ribose and DMA-sulfate ribose by their accuracy molecular weight 329.0599, 483.0738 and 409.0162, using direct confirmatory of HPLC-TOF-MS. The quantitative analysis of these arsenic species shows that although the total arsenic exceeds the National Standard value, most of the marine plants DSPs are safe to eat. The main component in most marine plants is DMA-sulfate ribose(51.1%-80.3%), while the main component of Kelp is DMA-phosphate ribose(48.9%). The sum of those three arsenosugars was 63.5%-92.7% of peak areas in all arsenic species detected.
5.Clinical Observation of Ai Luo Kang Acupoint Application for Simple Obesity

Objective To observe the therapeutic efficacy of Ai Luo Kang acupoint application in treating simple obesity. Methods Eighty simple obesity patients were randomized into a treatment group and a control group, 40 in each group. The treatment group was intervened by Ai Luo Kang acupoint application, while the control group was taken as a blank control. Thirty days later, the two groups were compared in the change of body weight and waist circumference. Results After intervention, the body weight and waist circumference were significantly changed in the treatment group (P<0.01). There were significant differences between the two groups in comparing the body weight and waist circumference after intervention (P<0.05). Conclusion According to the relevant criteria of the Evaluation Procedures and Methods of the Function of Health Care Products (DB22-T397-2006), Ai Luo Kang acupoint application is effective in losing weight in simple obesity.
6.Determination of Eight Typical Lipophilic Algae Toxins in Particles Suspended in Seawater by Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry

The particles suspended in seawater have great influence on pollutant migration and transformation in marine environment, while the lipophilic algae toxins enriched by the particles suspended in seawater will lead more serious toxicity to marine filter feeders. In this study, a new method was developed for the simultaneous determination of eight lipophilic algae toxins in suspended particles by ultra performance liquid chromatography-tandem mass spectrometry ( UPLC-MS/MS ) . After extracted with methanol by ultrasonic-assisted extraction, the sample was separated on an Acquity UPLC BEH C18 column (50 mm×2. 1 mm, 1. 7 μm) using gradient elution of acetonitrile and water containing 5 mmol/L ammonium acetate as eluent modifiers. The qualitative and quantitative analyses were carried out by electrospray ionization ( ESI) tandem mass spectrometry in multiple reaction monitoring ( MRM) mode. Under the optimal conditions, satisfactory precision (relative standard deviations (RSD≤14. 1%), recoveries (83. 8%-110. 4%) and detection limits (2. 9-103 pg/g) of the method were achieved. Good linearity (R2≥0. 99) was also obtained for all studied analytes. Then, the method was applied to determine the amounts of the eight lipophilic marine toxins in authentic suspended particle samples collected from Qingdao near-shore area. Pectenotoxin 2 ( PTX2 ) was detected in the samples from Shilaoren beach and No. 3 bathing beach with concentration ranges of 717 and 790 pg/g, respectively.
7.Preparation and Properties of Resveratrol-poloxamer 188-solid Dispersion

OBJECTIVE:To prepare Resveratrol-poloxamer 188-solid dispersion(RES-P188-SD)and study its dissolving char-acteristic in vitro and antibacterial activity. METHODS:With P188 as the carrier, solvent method was used to prepare RES-P188-SD. With the mass ratio of drug to carrier,melting temperature and mesh number as the observed factors,and solubility and yield of RES as the observed indexes,L9(34)orthogonal test was designed to optimize the preparation technology,and compre-hensive score of observed indexes were analyzed. The verification test was carried out. After RES-P188-SD was prepared by the op-timal technology,basket method was used to determine its dissolution rate and then its accumulative dissolution rate was calculat-ed,scanning electron microscopy to analyze phase characterization and cylinder-plate method to determine antibacterial activity. RE-SULTS:The optimal technology was as follows as the mass ratio of drug to carrier of 1∶10,melting temperature of 70 ℃ and mesh number of 80. For the sample prepared by the optimal technology,average solubility was 0.51 mg/ml (RSD=1.96%,n=3),average yield was 91%(RSD=0.64%,n=3),average 15 min accumulative dissolution rate was up to 83%(RSD=0.69%, n=3);the drug in amorphous form was homogenously distributed in the carrier;RES-P188-SD could inhibit Staphylococcus aure-us and Escherichia coli. CONCLUSIONS:The optimal technology is stable and feasible,by which RES-P188-SD has been pre-pared successfully,providing reference for increasing the solubility,dissolution rate and antibacterial activity of RES.
8.Preparation and functional identification of human high mobility group box-1 protein

Objective To clone the cDNA encoding human HMGB1, express it in E. coli, and identify its biological activity. Methods Human HMGB1 cDNA was amplified by RT-PCR and cloned into vector pUC19. After sequence analysis, the cDNA was ligated into prokaryotic expression vector pQE-80L and induced by IPTG to express HMGB1. The protein was purified with Ni~(2+)-NTA chromatography and polymyxin B affinity column. To identify the function of purified protein, the product was co-cultured with THP1 cells. Results Recombinant expression plasmid pQE-80L/HMGB1 was constructed successfully. After purification, the protein purity reached 96%. The recombinant HMGB1 stimulated THP1 to secrete TNF-? . Conclusion The highly purified HMGB1 was obtained successfully, which showed biological activity. These results lay the foundation for further research on the function of human HMGB1.
9.The application effect of ketamine and remifentanil in pediatric ophthalmic laryngeal mask placement general anesthesia surgery

Objective:To explore the application effect of ketamine and remifentanil in pediatric ophthalmic laryngeal mask placement general anesthesia surgery.Methods:A total of 68 children who underwent ophthalmic laryngeal mask placement general anesthesia surgery in Zhangjiakou Fourth Hospital from July 2020 to May 2023 were prospectively selected and they were randomly separated into a ketamine group (34 cases, anesthesia with ketamine combined with propofol) and a remifentanil group (34 cases, propofol combined with remifentanil). The mean arterial pressure (MAP), heart rate (HR), and blood oxygen saturation (SpO 2) of the two groups were compared at before anesthesia (T 1), after anesthesia (T 2), intraoperative 5 min (T 3), intraoperative 20 min (T 4) and after surgery (T 5). Meantime, the satisfaction with anesthesia among the surgeons, perioperative adverse reactions, surgical duration, recovery time and time to transfer out of the postanesthesia care unit (PACU) were compared between the two groups. Results:The MAP and HR of the remifentanil group at T 2 to T 4 were lower than before anesthesia, MAP: (72.83 ± 8.25), (74.18 ± 8.57) and (74.49 ± 9.13) mmHg (1 mmHg = 0.133 kPa) vs. (76.26 ± 9.51) mmHg; HR: (83.21 ± 6.63), (85.18 ± 7.06) and (86.81 ± 7.53) times/min vs. (89.72 ± 7.46) times/min ( P< 0.05), while the MAP and HR of the ketamine group at T 2 to T 5 were higher than before anesthesia, MAP: (81.67 ± 9.31), (85.06 ± 11.25), (86.34 ± 11.56) and (87.51 ± 9.30) mmHg vs. (77.58 ± 10.22) mmHg; HR: (95.04 ± 9.78), (97.60 ± 8.24), (98.64 ± 10.47) and (100.21 ± 8.56) times/min vs. (88.53 ± 9.15) times/min ( P<0.05), there was no difference in SpO 2 in the two groups between T 2 to T 5 and T 1 ( P>0.05), while the MAP and HR of the remifentanil group were lower than those of the ketamine group at T 2 to T 5, MAP: (72.83 ± 8.25) mmHg vs. (81.67 ± 9.31) mmHg, (74.18 ± 8.57) mmHg vs. (85.06 ± 11.25) mmHg, (74.49 ± 9.13) mmHg vs. (86.34 ± 11.56) mmHg, (75.38 ± 8.96) mmHg vs. (87.51 ± 9.30) mmHg; HR: (83.21 ± 6.63) times/min vs. (95.04 ± 9.78) times/min, (85.18 ± 7.06) times/min vs. (97.60 ± 8.24) times/min, (86.81 ± 7.53) times/min vs. (98.64 ± 10.47) times/min, (88.45 ± 9.51) times/min vs. (100.21 ± 8.56) times/min ( P<0.05), there was no difference in T 2 to T 5 SpO 2 between the two groups ( P>0.05); the satisfaction with anesthesia among the surgeons in the remifentanil group was higher than that in the ketamine group: 82.35% (28/34) vs. 58.82% (20/34), and the incidence of perioperative adverse reactions was lower than that in the ketamine group: 11.76% (4/34) vs. 32.35% (11/34); the surgical time, recovery time, and time to transfer out of PACU in the remifentanil group were all shorter than those in the ketamine group: (48.26 ± 9.54) min vs. (56.32 ± 11.86) min, (13.84 ± 4.12) min vs. (18.36 ± 5.08) min, (21.50 ± 6.14) min vs. (27.98 ± 8.62) min ( P<0.05). Conclusions:Compared with ketamine, remifentanil has a better effect in pediatric ophthalmic laryngeal mask placement general anesthesia surgery, which is more conducive to stabilizing the cardiovascular system during surgery, reducing the occurrence of adverse reactions such as perioperative restlessness, and shortening the surgical time and recovery time.
10.Effects of Autophagy-related Genes and Signal Pathways on Occurrence and Development of Oral Tumors

Autophagy is a highly conservative cellular self-protective behavior dependent on lysosomes, and can be used as an important factor in promoting or preventing cancer, and its effect is related to the type and development of tumors. A full understanding of autophagy pathway is helpful to improve the diagnosis and treatment of tumors. Studies have shown that autophagy is closely related to the occurrence and development of oral tumors. Autophagy-related genes and signal pathways play a dual regulatory role on oral tumors. This article reviews the latest progress in the regulatory mechanism and therapeutic effect of autophagy on oral tumors.
