Objective Porphyromonas gingivalis(Pg)is one of the main putative pathogens of periodontal disease,espcially of adult periodontitis. This in vitro study was aimed at observing the inhibition actions of 3 kinds of antimicrobial drugs on Pg. Methods 106 stains of Pg were isolated from subgingival plaques with adult periodontitis. The agar diffusion test was used to detect the inhibition actions of metronidazole,clindamycin and tinidazole on 106 stains of Pg. Results Their MIC was respectively:0. 25mg/L,0. 6rng/L. The MIC90 of tinidazole＜MIC90 of clindamycin. The results showed:all of metroidazole, tinidazole and clindamycin can be used effectively against Pg,clindarnycin and tinidazole showed more potent antibacterial activity than metronidazole. The different concentration of Pg-liquid (l05 CFU/ml, 106 CFU/ml and 108 CFU/ml) had not influenced the results. Couclusion This study is significant to antibiotical therapy of periodontal diseases.

This study aimed to investigate the method of conserving the mice skin for long time in vitro,and forty clean male KM mice of 8-week old were divided into two groups by random digit table,namely vitrified skin transplantation group(n =30) and fresh skin transplantation group(n =10) .The skin was vitrified quickly with freezing solution EFS40 by the two-step method,and then was thawed with 0.5 mol/L glucose,which was followed by self-transplanted.The vitrified skin transplantation group was self-transplanted after the skin was vitrified for 2 weeks.A piece of skin which was smaller than the vitrified one was cut off on the other side of the spine,and the thawed skin were laid smoothly to coincide with the edge of the cut skin which was sutured then.In fresh skin transplantation group,fresh skins were prepared to put in Dulbecco phosphate-buffered saline for 15 minutes and then be transplanted to original position.The outcomes of transplantation were valuated on day 14 and day 28 after transplantation.And the results indicated that the survival rate of the fresh skin transplantation group was 100%(10/10) .The survival rate of the vitrified skin transplantation group was 77%(23/30) on day 14 and 70%(21/30) on day 28,respectively.But the surfaces of the mice skin and part of the hair follicle cells were damaged during vitrification.However,the method of vitrifying the mice skin quickly in this paper may be used to conserve the skin in vitro.

Objective To make a further study on the character and mechanism of hepatotoxic effect of microcystin(MC),which stands for the extractions of cyanobacteria Methods The enzymology and morphology effects of different concentrations of 0.1,1,10 ?g/ml extractions of cyanobacteria on primary cultured hepatocyte were observed Results It was shown that the concentrations of LDH and AST increased in the culture medium after exposure to MC at concentrations of 0.1 ,1 and 10 ?g/ml No significant changes of concentrations of AKP, GGT, ALT and GSH were observed in the culture media of tested groups.An active proliferation of cultured hepatocytes with apopotosis or necrosis were observed in the center of the proliferation foci in the treated groups.Characterisitic morphological alterations such as irregular shape,cytoplasmic cavity,plasma membrane blebs and protrusions were viewed in scanning electronic microscope as well as in cultured cell. Conclusion The extractions of cyanobacteria could promote the proliferation of hepatocyte and might influence the physiological and biochemical function and the integrity of hepatocyte

Objective To evaluate the impact of hepatitis B e antigen (HBeAg) states and hepatitis B virus DNA loads on the prognosis of chronic severe hepatitis B.Methods A total of 406 hospitalized patients with chronic severe hepatitis B in Nanfang Hospital of Southern Medical University from January 2002 to December 2007 were retrospectively analyzed.The impact of HBeAg states and HBV DNA loads on the prognosis was evaluated.The measurement data were compared by t test and rates were compared by chi square test.Results Of all the 406 patients with chronic severe hepatitis B,208 (51.2%) patients were HBeAg-positive and the remaining 198 (48.8%) were HBeAgnegative.There was no significant difference of constituent ratio of male and female,average peak value of total bilirubin and average valley value of prothrombin activity between HBeAg-positive group and HBeAgnegative group.However,the average age of HBeAg-negative patients was (46.7±12.8) years old,which was significantly higher than that (38.3±13.5) years old in HBeAg-positive group (t = 6.43,P＜0.01 )the proportion of patients with liver cirrhosis in HBeAg-negative group (67.7%) was much higher than that in HBeAg-positive group (45.7%) (X2=19.97,P＜0.01);the improved rate in HBeAg-negative group (32.3%) was significant lower than that in HBcAg-positive group (44.7%) (X2=6.56,P＜0.05).Increasing HBV DNA levels was associated with lower improved rate in both 208 HBeAg-positive and 198 HBeAg-negative patients(X2=22.98,26.04,respectively,both P＜0.01 ).Conclusions HBeAg-negative patients with chronic severe hepatitis B has worse prognosis than HBeAg-positive patients;and the prognosis is getting worse with the increasing HBV DNA level regardless of the HBeAg status.

Objective To learn about the antibiotic resistance status of methicillin resistant coagulase negative staphylococcus(MRCNS),and to investigate the distribution and resistant feature of different staphylococcal chromosomal cassette mec(SCCmec) genotypes of children in Anhui,so as to guide clinical medication.Methods Resistance phenotype screening was conducted in coagulase negative staphylococcus,which were isolated from clinical strains in children in Anhui from 2010 to 2014 each year in September.MecA gene was detected by using PCR method in order to collect MRCNS.Minimal inhibitory concentrations (MIC) of 16 antibiotics were determined by adopting agar dilution method.Vacomycin-resistant strains were identified with population analysis and the Brain Heart Infusion vancomycin screen agar dilution method recommended by Clinical and Laboratory Standards Institute in 2013.Van gene and SCCmec types were detected by using PCR method.Results A total of 148 MRCNS strains were detected through the resistance phenotype screening and the detection of mecA gene.There were methicillin resistant staphylococcus epidermidis,methicillin resistant staphylococcus haemolyticus,methicillin resistant staphylococcus hominis,and other kinds of MRCNS,and the proportions of them were 44.59％ (66/148 cases),25.68％ (38/148 cases),19.59％ (29/148 cases) and 10.14％ (15/148 cases),respectively.The analysis of antibiotic resistance showed the antimicrobial resistant rates of MRCNS to Penicillin,Cefoperazone,Cefotaxime,Ceftriaxone,lmipenem and Meropenem were all 100％,to Erythromycin and Azithromycin,Ciprofloxacin,Clindamycin,Gentamicin,Lewofloxacin,Rifampincin,Chloramphenicol,Teicoplanin and Vancomycin were 92.57％,97.98％,83.78％,79.05％,43.24％,35.81％,24.32％,8.78％,2.03％ and 0.68％,respectively.There was 1 heterogeneous Vancomycin-resistant strain,which was resistant to both Vancomycin and Teicoplanin (with MIC 32.00 mg/L and 64.00 mg/L).No vanA,vanB,vanC1 or vanC2/3 gene was detected from heterogeneous Vancomycin-resistant strain by PCR.Ⅰ to Ⅴ SCCmec genotypes were detected from 148 MRCNS strains,and the major SCCmec type was SCCmec type Ⅲ,which was followed by hybrid type.Three subtypes of SCCmec type Ⅳ were identified,including Ⅳa,Ⅳc and Ⅳd.There were 148 MRCNS strains that showed different resistant phenotypes to various antibiotics.Conclusions The MRCNS strains of children in Anhui province showed multiple resistance to antibiotics.It should be on alert when heterogeneous Vaneomycin-resistant strain appeared.There were several different SCCmec types among several kinds of MRCNS,and SCCmec Ⅲ genotype was the major epidemic isolate.There was no significant correlation between the different resistance rates of non-β-lactamase antibiotics and SCCmec genotypes in MRCNS.

Objective To examine the therapeutic effect of C Ⅱ TA inhibition in collagen-induced arthritis(CIA),using a delivery system tailored to target C ⅡTA gene by small interfering RNA (siRNA).Methods Mice with collagen-induced arthritis were injected intravenously with C Ⅱ TA siRNA.The clinical score was monitored for up to 4 weeks after treatment.The severity of inflammation of mouse joint was evaluated by histological examination.Real-time PCR was used to determine the cytokine mRNA expression.Cytokine production was measured by ELISA from serum.T cell proliferation was examined by MTT method.Results IFN-γ and IL-17 were elevated in CIA mice,but were iuhibited significantly by C Ⅱ TA siRNA either prevention or intervention of autoimmune arthritis.Collagen specific T cell proliferation was significantly suppressed.Increased level of IL-4 by T cells was observed in C Ⅱ TA siRNA treated group compared with that of control group.Conclusion Our findings indicate that systemic RNAi-mediated C Ⅱ TA gene silencing is effective in the treatment of CIA and regulateds the balance of Th1/Th2 differentiation.

Objective To discuss the standard of endoscopic diagnosis of duodenal white spot syndrome and to test its accuracy by pathological diagnosis standard. Methods A total of 6,995 patients undergoing gastroscopy were detailedly examined through gastrofiberscope or electrogastroscope and endoscopic diagnosis were made by the standard of endoscopic diagnosis of duodenal white spot syndrome. Then, pathological examination was done to test and verify the accuracy of the standard of endoscopic diagnosis. Results There were 533 patients suffering from duodenal white spot pathological changes, accounting for 7.62%. And duodenal mucositis was confirmed by pathological findings in all 90 cases of the patients undergone their biopsy at the same time. Conclusions The appropriate standard of endoscopic diagnosis can improve the detection rate of duodenal white spot syndrome and there is a high coincidence rate between endoscopic diagnosis and pathological diagnosis.
