Objective To predict as well as bioinforrmatically analyze the target genes of has-miR-451.Methods miRBase, miRanda, TargetScan and PicTar were used to predict the target genes of hsa-miRNA-451.The functions of the target genes were demonstrated by Gene Ontology and pathway enrichment analysis.P＜0.05 was set as statistically significant.Results 18 target spots of hsa-miRNA-451 were predicted by 3 databases or prediction software at least.The functions of the target genes were enriched in proliferation and development of epithelial cells and regulation of kinase activity (P＜ 0.05).Pathway analysis showed that transforming growth factor-beta signaling pathway, mitogen-activated protein kinase signaling pathway, epidermal growth factor signaling pathway, Wnt signaling pathway and mammalian target of rapamycin signaling pathway were significantly enriched (P＜0.05).Conclusion hsa-miRNA-451 might be involved in various signaling pathways related to proliferation and development of epithelial cells.

Objective To compare the axial length (AL) measured by Lenstar and contact A-Scan in the patients with idiopathic macular hole and study the correlation between the difference of the two measurements and the foveal thickness measured by optical coherence tomography (OCT).Methods Twenty-seven eyes of 26 idiopathic macular hole patients (IMH group) and 27 eyes of 25 patients with mild cataract (control group) were enrolled in this study.Foveal thickness was measured with 3D OCT.The AL was measured by Lenstar and contact A-Scan,and the consistency of the two measurements was determined by Bland Altman analysis.The correlation between the difference of the two measurements and foveal thickness was analyzed by Pearson correlation analysis.Results Mean foveal thickness of IMH and control eyes were (372.85±60.02) μm and (243.44±22.50) μm,respectively.The difference between the foveal thickness of the two groups was highly significant (t=-10.490,P＜0.001).In the IMH group,the AL measured by Lenstar and contact A-Scan were (23.20± 1.12) mm and (23.18± 1.13) mm,respectively,the difference between the two measurements was not statistically significant (t =-0.549,P =0.588),whereas in the control group,the AL was (23.41 ± 0.72) mm by Lenstar and (23.33 ± 0.74) mm by contact A-Scan,the two measurements were significantly different (t=-4.832,P＜0.001).However,no correlation was found by Pearson correlation analysis between the difference of the two measurements and the foveal thickness in either IMH or control group (r=0.181,-0.141; P＞0.05).Conclusions Although there is no difference of axial length measurements using Lenstar and contact A-Scan in IMH eyes,in clinical measurements the results of two instruments should be taken into comprehensive consideration.

Objective To study the compressive education on nursing students in operation room.Methods 84 nursing students in practice were randomly divided into the observation group and the control group with 42 students in each group.The two groups were in a month to complete the teaching syllabus internship period.The control group received conventional teaching,on the basis of conventional teaching,the observation group was given compressive education.After the end of internship,nursing skill score,comprehensive quality,operation doctors'satisfaction degree were compared between two groups.Results After the compressive education,the nursing skill score,comprehensive quality,operation doctors'satisfaction degree in the observation group were significantly higher than those of the control group.Conclusions According to the source of pressure in the operation room,compressive education should be given to the nursing students and make them complete the practice in a relaxed and delight state.This can also reflect the humanistic care in teaching,which is worthy of being popularized in teaching in operation room.

Objective To evaluate the apphcation of wireless mobile infusion system in the outpatients and emergency patients.Methods A total of 566 outpatients and emergency patients with infusion were divided into the observation group and the control group according to the order of infusion.The observation group with 297 cases was used wireless mobile infusion system,and the control group with 269 cases was used traditional infusion method.The reception time,infusion waiting time and satisfaction for nurses' job of patients between two groups were compared.The satisfaction for job of nurses among different work models were compared,too.Results The reception time,infusion waiting time of patients in the observation group were less than those in the control group:(2.07±0.26) min vs.(3.17±0.26) min,(9.89±0.31) min vs.(13.49± 0.21) min,and there were significant differences,P＜0.01.The satisfaction for nurses' job of patients in the observation group were better than those in the control group,x2=8.921,7.064,5.251,and there were significant differencs,P＜0.05.The satisfaction for job of nurses in the observation group was better than that in the control group,and there was significant difference,P＜0.05.Conclusion Wireless mobile infusion system can improve work efficiency,guarantee the safety of infusion,improve patients and nurses' satisfaction.

Objective To evaluate intraoperative tracing of sentinel lymph node(SLN) by fluorescence staining combined with dye dyeing.Methods A total of 174 patients with breast cancer were randomly divided into three groups : the group A with 57 patients receiving methylene blue (MB), the group B with 58 patients receiving indocyanine green(ICG) as the lymphatic mapping tracers,the group C with 59 patients receiving MB and ICG.The sentinel and axillary lymph node of level Ⅱ, Ⅲ was excised, followed by conventional histopathology.Results There was no significant difference among the three groups in the term of visualized detection rate (x2=2.96, P =0.241).There was statistical significant difference among three groups in the term of detected lymph nodes(F=15.34, P＜0.05).Comparing with the three groups, the number of detected lymph nodes of A and B group had no significant differences(P=0.07) ,the number of detected lymph nodes of C was higher than that of A and B group, and the difference was significant(P＜0.05).There was statistical significant difference among three groups in the term of SLN positive rate (x2 =6.75, P =0.039), and there was no significant difference among A and B group(P=0.915) ,SLN positive rate of C group was higher than than A and B group, and the difference was statistical significant (P＜0.05).Conclusion Intraoperative tracing of SLN by fluorescence staining combined with dye dyeing has the skin and subcutaneous reveal advantage.The use of ICG fluorescence and MB increases lymph node detection rate.

Objective To explore the profiles of physical activity domains among professionals with different occupational activity patterns.Methods A cluster random sampling method was used to recruit employed adults aged 30-60 years in Beijing as well as Zhejiang province.The information on the physical activity domains was collected with a revised Global Physical Activity Questionnaire (GPAQ).Results Of 3 326 professionals,about 30.79％,50.75％ and 18.46％ adults had Class Ⅰ (mostly sedentary reading or writing during work time) ,Class Ⅱ (mostly working with arms or legs with little effort) ,and Class Ⅲ (working with effort)occupational activity patterns,respectively.The median volume of leisure time activity and bicycling were all 0 Met-min/week,and no significant difference of domestic activity volume was detected among all the three classes of professionals.Of all the non-occupational activity volume among three classes of adults,domestic activity accounted for the most proportion (58.87％) performed by adults of Class Ⅱ,traffic activity accounted for the most proportion (34.2％) by those of Class Ⅲ,and leisure time activity accounted for the most proportion (12.7％) by those of Class Ⅰ.Most time was spent in sedentary behaviors by those of Class Ⅰ after hours.Conclusions Besides the generally low level of moderate activity,the diversity of the domains of physical activity among this population was evident.It is necessary to develop appropriate technology to improve physical activity for professionals with different occupational activity patterns.

Objective To investigate the treatment of inverted nipple by continual extension with S tractors made of Kirschner wires on the base of preliminary clinical obervation.Methods Under local anesthesia,with the inverted nipples being dragged out by a presutured thread or a pair of towel clips,a Kirschner wire of 10 cm long and 0.5-1.0 mm in diameter was transversly penetrated through the base of inverted nipple.Then the Kirschner wire was bended at both points of 1.5 cm distantly from the nipple center in clockwise cemicircularly.Two cemicircular Kirschner wires were folded downward hingelikely and the center axis was supported by two cemicircles.This folded S tractor pushed and supported the inverted nipple forwardly and constantly.The hinge angle was adjusted during the treatment according to the traction requirment.It was nessessary for wearing these S tractors for at least 6 months before the Kirschner wires were being taken out.During referrals,the hinge angle was revised for mentaining the extension efforts.Results We treated 32 cases of inverted nipple,total 58 moderate and severe lesions,with this method effectively.No infection was observed during the treatment.The nipple form was naturally and stably outstanding after Kirschner wires taken out.No relapses were observed during a mean 6 monthe following up.Conclusions This procedure is simple and convenient without the need of complicated surgical procedures or perplexed equipments.The nipple shaps are observed natural and stable after Kirschner wire is removed.No recurrences are observed.There are no damages to breastfeeding function.The obvious drawback of this method is its reqairing of long continuously wearing and its inconvenience.

Background Retinal neovascularization varies with the change of microRNA (miRNA) expression level.Quantitative real-time PCR (qRT-PCR) is a common method for the analysis of miRNA expression profiling.However,the housekeeping genes selected as references may exhibit differential expression levels under the distinctive experimental conditions.The accuracy of the levels of target gene expression often is affected if the selected housekeeping genes with significantly fluctuating expression as references.Determining a reference gene with stable expression level is the premise to consecutive studies.Objective This study was to compare the expression levels and stability of the frequently used reference genes for miRNA expression analysis in normally developing eyes and in eyes of a mouse model of oxygen-induced retinopathy (OIR),and select the optimal reference gene (s) exhibiting stable ocular expression under both hypoxia and normal development conditions.Methods P7,P12,P17,P37 and 8-week-old clean C57BL/6J mice were selected randomly.q-PCR was used to detect the dynamic changes of relative expressions of 5 kinds of reference genes in different ages of mice,including snRNAU6 (RNU6),5S ribosomal RNA (5s rRNA),snoRNA U68 (U68),snoRNA U70 (U70),snoRNA U49A (U49A).Other 40 P7 mice were randomized into the normal control group and the OIR group.The mice of the normal control group were fed in the normal oxygen environment,and those in the OIR group were raised in the (75-±2)％ oxygen environment for 10 days.Fluorescine was injected via ritrobulbar vein for retinal stretched preparation,and the histopathological examination of mouse retinas was performed to identified the models.The expressing difference of 5 kinds of reference genes in the retinas were detected to compare the differences of 5 kinds of genes expression between the two groups.GeNorm program was employed to compare the stability of the expressing genes.This study were approved and granted by Ethical Committee of Tianjin Medical University and met the requirements stipulated in the Association for Research in Vision and Ophthalmology Statement for the Use of Animals in Ophthalmic and Vision Research.Results Across the developmental ages,the expression levels of U68 in the eye exhibited the greatest variability,and then coming with U49A,U70,RNU6 and 5s rRNA.Significant differences were seen across the developmental ages for the expression levels of U68,U49A,U70,and RNU6 (U70:F =7.005,P =0.000 ; U68:F =10.189,P =0.000 ; U49A:F =21.134,P=0.000;RNU6:F=4.968,P=0.004).Expression of 5s rRNA showed the least variability across the developmental ages (F=2.099,P =0.107).In P17 mice of the OIR group,tortuous access vessels and non-perfusion area were found in the retinal section.Hematoxylin-eosin stain showed the neovascular sprout through across the inner limiting membrane.The relative expression of U70 exhibited the greatest variability and then were U49A,U68 and 5s rRNA in turn in P17 OIR mice,and significantly elevated expressions were found in U70,U49A,U68 and 5s rRNA genes between the OIR group and the normal control group (t =5.174,P =0.000;t =3.376,P =0.012;t =4.802,P =0.000 ;t=2.856,P=0.029).Expression of RNU6 showed the least variability between the two groups (t =2.104,P=0.065).As analyzed by GeNorm program,the stability for the five reference genes across developmental ages was 5s rRNA/RNU6＞ U70 ＞ U49A ＞ U68,and the optimal reference combination was 5s rRNA and RNU6.Whereas the stability for the OIR model was 5s rRNA/RNU6＞U68＞ U49A＞U70,and the optimal reference combination was 5s rRNA and RNU6.Conclusions Reference genes should be selected based on specific subjects and experimental conditions when qRT-PCR is used to analyze miRNA expression levels.In the OIR model,both developmental and hypoxic factors need to be considered.5s rRNA and RNU6 reference combination is the preferred option for the qRT-PCR analysis of ocular miRNA expression if available.
