1.Changes of Myoepithelial Cells in Mammary Atypical Hyperplasia and Breast Cancer

Objective To investigate the changes of myoepithelial cells in mammary atypical hyperplasia and breast cancer. Methods SP immunohistochemistry was used to detect actin expression in normal breast tissue, grade Ⅰ, Ⅱ and Ⅲ atypical hyperplasia and breast cancer. Electromicroscopy was used to observe the changes of ultrastructure of myoepithelial cells. Results Actin was only detected in myoepithelial cells of normal breast tissue and grade Ⅰand Ⅱ atypical hyperplasia. The positive expression rates of actin in grade Ⅲ atypical hyperplasia(70%) and breast cancer(90%) were significantly higher than that in grade Ⅱ atypical hyperplasia(10%), P
2.Clinical analysis of 47 cases with traumatic macular hole resulted from ocular contusion

Objective To analyze the clinical characteristic,treatment and prognosis of traumatic macular holes resulted from ocular contusion.Methods The clinical data of 47 cases with traumatic macular hole was retrospectively reviewed.The general condition of the patients was summarized,optical coherence tomography and multifocal electroretinogram (mfERG) were used to evaluate anatomic and functional outcomes.The patients were divided into observation group and surgery group by the treatment they received,and the prognosis was evaluated.Results Traumatic macular hole occurs mainly in male.In the observation group,the mean diameter of macular hole was(490.0±86.9) μm.During the 12 month follow up,the holes in 7 cases (33.3%) were closed spontaneously,Vision and diameters of 14 cases (57.1%) maintained stable for a long time,the vision of 1 case (3.3 %) declined mildly and the diameter of 1 case (3.3%) enlarged slightly.Visual acuity was improved significantly at last follow up (Z=-2.40,P< 0.05).The amplitudes of N1 wave of mfERG increased both in central fovea and macular area(t=13.30,5.06;P<0.05).These data suggests that the macular function was recovered well.In the surgery group,the mean diameter of macular hole was (643.3 ± 125.0) μm and statistically larger than that of the observation group (t=-4.76,P<0.05).At the last follow-up,visual acuity were not improved significantly (Z=-1.79,P>0.05).The amplitudes of N1 wave in 6 cases (23.1 %) improved merely and the difference was not statistically significant (t =1.98,P > 0.05).These data suggests that the macular function was recovered slightly only in a few patients.Conclusions A part of the patients with smaller diameters of macular holes may close spontaneously,and they may get better visual acuity.Vitrectomy may help to close the macular holes in some severe cases,but the improvement of functional outcomes is not significant.
3.Laminin and fibronectin expressions in atypical hyperplasia and breast cancer and the significance

Objective To investigate the changes of laminin(LN) and fibronectin(FN) expressions in atypical hyperplasia and breast cancer. Methods LN and FN expressions in normal breast tissue, tissues from grade Ⅰ, Ⅱ and Ⅲ atypical hyperplasia and breast cancer were detected by means of S P immunohistochemical staining method. Results In normal breast tissue, LN expression was found only in basement membrane, but FN expression in basement membrane and matrix. In gradeⅠ hyperplasia, LN expression was found in basement membrane and epithelium, but FN expression was similar to that in normal tissue. In grade Ⅱhyperplasia, more LN expression was found in epithelium, but FN expression in epithelium and matrix. 4.Combined transplantation of pancreatic islets and hepatocytes in the treatment of diabetic rats

Objective To study the effect of combined transplantation of pancreatic islets and hepatocytes on the treatment of diabetic rats and the possibility on hepatocytes inducing islet allograft tolerance. Methods Forty diabetic SD rats were randomly divided into 4 groups: normal control group,diabetic control group, pancreatic islet grafted group,and hepatocyte and pancreatic islet transplanted group. The pancreatic islets of fetal SD rats were implanted into the gastric subserosa of recipients. The alteration of diabetes and immune status were observed. Results There were 58.8% (20/34) diabetic rats cured after pancreatic islets transplantation. Objective To investigate the interrelation between nuclear DNA content and the expressions of cyclin D1 and P53 in carcinogenesis of breast carcinoma induced by 7,12 dimethylbenz(a)anthracence(DMBA) in rats. Methods Animal model of breast carcinoma was replicated by gastric perfusion of DMBA at 20 mg/100 g. Nuclear DNA content was detected with image analysis instrument and the expressions of cyclin D1 and P53 at every stage of carcinogenesis of breast carcinoma induced by DMBA in rats observed by Sp immunohistochemical staining. Results In mammary gland tissue at grade Ⅰ, grade Ⅱ and grade Ⅲ atypical hyperplasia of breast carcinoma, there was overepression of cyclin D1(3/20, 6/20 and 11/20, respectively). The overexpression of cyclin D1 was detected to be present in grade Ⅰ, grade Ⅱ and grade Ⅲ breast carcinoma (7/9, 1/4 and 0/7, respectively). The overexpression of P53 was detected to be present in grade Ⅰ, grade Ⅱ and grade Ⅲ breast carcinoma (1/9, 1/4 and 6/7, respectively). Conclusion Nuclear DNA content reflects the state of cell proliferation. Cyclin D1 overexpression participates in the carcinogenesis of breast carcinoma, which may be the early event in breast carcinoma. P53 expression is associated with the development of breast carcinoma, which may be the late event in breast carcinoma. The expression of cyclin D1 is negatively correlated with P53 overexpression.
6.Effect of different teeth arrangement methods on the masticatory efficiency of complete dentures

Objectives:To evaluate the effect of different teeth arrangement methods on the masticatory efficiency of complete dentures.Methods: Three complete dentures for each of ten edentulous patients were made respectively by using maxillary teeth arrangement,mandibular teeth arrangement and comprehensive teeth arrangement methods.After three months of wearing,the masticatory efficiency of the three complete dentures was tested by light absorption method,and was compared to that of dentulous subjects.Results:The masticatory stroke and masticatory efficiency in denture wearing group were fewer and weaker than those in dentulous group in either bilateral mastication or unilateral mastication. Good masticatory efficiency was gained after three months of wearing the complete dentures made by the three different teeth arrangement methods.There were no significant difference in the masticatory stroke and masticatory efficiency of complete dentures made by mandibular teeth arrangement and comprehensive teeth arrangement methods,but they were both higher than those made by maxillary teeth arrangement method.Conclusions:The complete dentures made by mandibular teeth arrangement method and comprehensive teeth arrangement method are feasible for restoration of masticatory efficiency of endentulous patients.
7.Comparative study of mammogram and magnetic resonance imaging on diagnosis of breast ductal carcinoma in situ with or without microinvasion

Objective To compare the sensitivity and diagnostic features of mammogram (MG) and magnetic resonance imaging (MRI) on diagnosis of breast ductal carcinoma in situ (DCIS) with or without microinvasion (DCIS-MI).Methods From Jan 2012 to Nov 2013,results of MG and MRI from 72 cases of DCIS or DCIS-MI were retrospectively analyzed.Results The sensitivity of MG was 52.8% (38/72).The sensitivity of MRI was 87.5% (63/72),among those 76.2% (48/63)lesions presented as non-mass-like enhancement.Sensitivity of MRI was significantly higher in DCIS-MI than DCIS (84.6% vs 100%,P =0.027).Logistic regression analysis showed calcifications was an independent factor influencing the sensitivity of MG (OR =23.785,P < 0.001).Conclusions The sensitivity of MRI is higher than MG for the diagnosis of DCIS and DCIS-MI.
8.Analysis of protein expression profiling in a mouse model of oxygen-induced retinopathy

Background Retinal neovascularization (RNV) is one of the major causes of blindness worldwide,but the pathogenic mechanism of this disease remains unclear, and therapeutic modalities need to be improved.Therefore,it is necessary to identify ocular protein markers with significant expression changes during RNV, thereby providing novel therapeutic targets for neovascular retinopathies.Objective This study was to investigate retinal vessel morphological characteristics and protein expression profiling in a mouse model of oxygen-induced retinopathy (OIR).Methods Forty four C57BL/6J mouse pups were randomly divided into normal control group and OIR group at postnatal day 7 (P7).The mice in the normal control group were raised under the normal air for 10 days.The mice of the OIR group were exposed to (75±2)% oxygen for 5 days.The mother mice were alternated between the two groups every day.The mice of the OIR group returned to normal air at P12.Fluorescein isothiocyanate-dextran (FITC-dextran) was retrobulbarly injected at P17 mice from both groups, and the retinal flatmounts were prepared after fixation.The FITC-dextran-labeled retinal vessels were observed and quantified;the paraffin sections of eyeballs were processed for hematoxylin and eosin staining,and the number of pre-retinal vascular cell nuclei was quantified.The total proteins were extracted from the eyes, and the expression profiling was analyzed by a customized protein array and verified by Western blot and ELISA.Results The retinal flatmounts labeled with FITC-dextran showed that the peripheral retinal microvessels in the OIR group were tortuous, disorganized with neovascular buds,and the vascular obliteration was prominent in the center of retina.Contrastly,the vessels were smooth,organized, and evenly distributed in the normal control group.The percentage of vascular obliteration area in the OIR group was (25.53±2.16)% ,which was significantly higher than (0.66±0.36)% in the normal control group (t=-27.61 ,P< 0.01).The number of pre-retinal vascular cell nuclei,as revealed by hematoxylin and eosin staining, was (28.41 ± 3.97)/slide in the OIR group, which was substantially higher than (0.16±0.31)/slide in the normal control group (t =-54.42,P<0.001).Protein array showed that 10 out of the 62 examined pro-inflammatory, pro-angiogenic cytokines exhibited more than 1.5-fold expression changes,including 3 up-regulated cytokines and 7 down-regulated cytokines;4 cytokines showed more than 2-fold expression changes,in which 3 cytokines were down-regulated and 1 cytokine was up-regulated.The differential expressions were verified by Western blot and ELISA.The expression trends of platelet factor 4 (PF-4), vascular endothelial growth factor A (VEGF-A), selectin P (SELP), vascular cell adhesion molecule 1 (VCAM-1) ,soluble tumor necrosis factor receptor Ⅱ (sTNF-RⅡ) and chemokine C-X-C motif ligand 16 (CXCL16) were consistent with those revealed by protein array.PF-4,VEGF-A and SELP were up-regulated,and the other 3 were significantly down-regulated (all at P<0.05).Conclusions Differential expression patterns of the cytokines,including PF-4,VEGF-A, SELP, VCAM-1, sTNF-RⅡ and CXCL16, are identified between normal and OIR mouse eyes.These differential expression patterns suggest that under the condition of OIR,the platelet system is activated,and proinflammatory factors are down-regulated.PF-4 might become a new target for VEGF-independent therapeutic strategy against RNV.
9.Inhibitory effects of silencing EZH2 gene expression on cell proliferation of cholangiocarcinoma QBC939cell line and its mechanisms

Objective To investigate the inhibitory effects of silencing expression ofEZH2 gene on the cell proliferation of human QBC939 cells and its mechanisms. Methods The targeting siRNA was designed and trans-fected into QBC939cells. The expressions of EZH2 mRNA and protein were detected by real-time qPCR and west-ern blotting,respectively. The ability of cellproliferationwas analyzed by MTT assay and plate clone formation assay. Cell apoptosis and cycle percentage weremeasured by flow cytometry. Cell senescence was assessed byβ-galactosi-dase dyeing.The expressions of H3K27me3,P14ARF,P16INK4a,P53,P21 and E2F1 proteinwere determined by West-ern blotting.Results Compared with the control group ,the expressions of mRNAand protein were significantly elevat-ed in experimental group. The ability of cellproliferation in the experiment group was significantly down regulated , which could also cause a rise of G1/S phase ,but not a marked variation of apoptosis rate. Silencing EZH2 would induce a obvious senescence phenotype in QBC939 cells. EZH2-siRNA transferredcould also down-regulate the expressions of H3K27me3 and E2F1 protein,while up-regulating the expressions of P14ARF,P16INK4a,P53 and P21 protein in QBC939 cells.Conclusions Silencing EZH2 could induce a significant inhibition on cell proliferation of QBC939 cells,the mechanism of which may be associated with the senescence pathway regulation.
10.Low-frequency fluctuation amplitude analysis of resting-state fMRI for functional brain response differences between acupuncture and moxibustion at Zusanli (ST 36) in patient with functional dyspepsia

Objective: To compare and analyze functional brain response characteristics by applying acupuncture or moxibustion to Zusanli (ST 36) in patients with functional dyspepsia (FD) and investigate the differences of central action mechanism resulting from acupuncture or moxibustion. Methods: A total of eligible 24 FD cases were divided into two blood-oxygen-level dependent (BOLD) sequences for functional magnetic resonance imaging (fMRI) scan. The amplitude of low frequency fluctuation (ALFF) analyses were conducted on the data of location phase, structure phase, resting state before acupuncture/moxibustion, working state during acupuncture/moxibustion and resting state after acupuncture/moxibustion using Data Processing Assistant for Resting-State fMRI (DPARSF) software. Results: Acupuncture and moxibustion produced significant differences in functional brain response. The working state during acupuncture/moxibustion mainly decreased ALFF values in the right supramarginal gyrus, right superior parietal lobule, right frontal gyrus, upper right occipital lobe, right precuneus and right cingulate gyrus. At the same time, it increased ALFF values in the left cerebellum, right caudate nucleus, right cerebellum and left inferior gyrus. The differences during the resting state after acupuncture/moxibustion were significantly smaller than the working state in intensity and size. It mainly resulted in decrease in ALFF values in the right postcentral gyrus and right supramarginal gyrus and increase in ALFF values in the left precuneus, orbital part of inferior frontal gyrus and right cerebellar peduncles. Conclusion: Needling and moxibustion at Zusanli (ST 36) can produce significant differences in immediate functional brain response.
