Aim To study the effect of As_2O_3(arsenic trioxide) on the expressions of telomeric repeat binding factor1,2 and telomeric stability of human gastric cancer MGC803 cells, and explore the mechanism of cell apoptosis. Method MGC803 cell growth inhibition was measured with MTT assay. Apoptosis was analyzed with flow cytometry. Influence on chromosome distal end was analyzed with chromosome end-end fusion analysis. Expressions of TRF1 and TRF2 were determined with Western blot analysis. Results MTT assay showed that As_2O_3 clearly inhibited the growth of MGC803 cells, depending on time and dosage. The apoptosis rates were significantly higher than those of the control group in a concentration and time-dependent manner. These changes were not found in the control group. After disposal with 5 ?mol?L -1 As_2O_3, chromosome fusion rate was obviously increased in 48 h. After 48 h of disposal with 5 ?mol?L -1 As_2O_3, the TRF1 of MGC803 cells was up-regulated while TRF2 was down-regulated. Conclusion As_2O_3 induced chromosome fusion and MGC803 cells apoptosis through up-regulating expressin of TRF1 and down-regulating expressin of TRF2.

Objective To evaluate the role of the diffusion kurtosis imaging(DKI)in the differential diagnosis of breast lesions. Methods Seventy five breast lesions(32 benign and 43 malignant)in 72 patients confirmed by histopathology were studied. All patients underwent 3.0 T MR examinations, including T1WI, T2WI, T2WI-spectral adiabatic inversion recovery, 4b diffusion-weighted imaging, and dynamic contrast-enhanced MR imaging(DCE-MRI). Data were post-processed by mono-exponential and diffusion kurtosis models for quantitation of ADC, apparent diffusion for non-Gaussian distribution(D), and apparent kurtosis coefficient(K). All breast lesions were described with the classification by breast imaging report and data system(BI-RADS). Lesions with BI-RADS class 4B or above were rated as malignancy. Independent sample t test was used to compare the ADC, D, and K value differences between benign and malignant lesions . ROC analysis was performed to assess the role of ADC, D, K value, and BI-RADS in the differential diagnosis of breast lesions. The morphological characteristics, time-signal curve(TIC)type, and other differences between benign and malignant lesions were analyzed with Chi-square test. Results ADC and D values were significantly lower in malignant than in benign lesions(P<0.01). Conversely, K value was significantly higher in malignant lesions than in benign ones(P<0.01). The shape of the benign and malignant breast lesions, edge, enhancement mode, TIC, and BI-RADS classification difference had statistical significance(P<0.05, respectively). The areas under the ROC curve of ADC, D, K, DCE-MRI, and DCE-MRI combined with K value were 0.857, 0.884, 0.949, 0.806, and 0.958, respectively. DCE-MRI combined with K value had the highest diagnosis efficiency. At a cutoff value of K= 0.856, the sensitivity and specificity were 83.7% and 93.8%, respectively. Conclusions DKI model showed higher diagnostic efficiency than that of traditional DWI model. DCE-MRI combined with K value can increase the diagnostic efficiency in breast lesions.

OBJECTIVETo investigate celluar toxicity and antibacterial property of nano-silver coating modified denture base and to provide a theortical basis for clinical application.
METHODThe samples were divided into coating group and denture base group. Samples in coating groups were treated with silver ion sputtering and were further divided into the 10, 20, 40, 60, 80, 100 s subgroups according to their sputtering time. Surface microstructure of samples in coating groups and denture base group were observed by scanning electron microscopy (SEM), and composition of material surface were determined with energy dispersive spectrum (EDS) analysis. Methyl thiazolyl terazolium (MTT) method was used to analysis L929 cells proliferation rate and cell toxicity grade while the coating groups (n = 6) and group test specimens (n = 6), negative control (polyethylene, PE) (n = 6) and positive control group (PVC-org.Sn) (n = 6) were co-cultured with L929 cells. The antibacterial properties of coating groups were investigated by using the bacterial membrane adhering method with Streptococcus mutans and Candida albicans.
RESULTSSEM observation showed that the surface of control group specimens were smooth, whereas dense and uniform distributed nano silver particles were visible on the surface of coating group specimens. The silver particles were round, and the diameter size was 10 nm. MTT assay showed that after being co-cultured with specimens in control group, L929 cell had higher proliferation rate than 100% at 24, 48, 72 h, and cytotoxicity were graded 0. The membrane adhering method results showed that all of bacterial colonies were found in control group, but without anything at the coating groups.
CONCLUSIONSDeposition of nano silver on the denture base surface by ion sputtering method is a viable method. There's no cytotoxicity of series Ag-coating material. Nano-silver modified denture base material benefits L929 cell surface proliferation and has antibacterial action.
