Objective To explore the effect of auricular plaster therapy on associated symptoms and quality of life in patients with constipation induced by strong opioid drugs for pain. Methods A total of 60 patients selected were randomly divided into the intervention group and the control group with 30 cases in each group. The intervention group was treated with auriculotherapy based on the pattern/syndrome differentiation after detecting the auricular sensitive acupoints' distribution in patients with opioid-induced constipation after receiving opioid therapy.BFI,self-made scale on symptoms accompanied by constipation and PAC-QOL were used to collect data about constipation symptoms and the level of quality of life before or after the intervention, and still with a subset of the intervention group followed for more than two weeks. Results The BFI and PAC-QOL scores of the intervention and control group were lower after intervention, and the decline score of the two groups showed significant difference. But for the self-made scale, the score of each item was slightly lower compared with that before treatment, which showed no statistically significant difference between groups.A follow-up of 2 weeks showed that the long-term efficacy of auricular plaster therapy was significantly lower than before, but was less desirable than short-term.Conclusions Scientific and reasonable choice of acupoint prescription according to syndrome differentiation can effectively improve symptoms accompanied by constipation and patients' quality of life.

Objective To compare the effects of upstream versus downstream tirofiban on tissure level perfu-sion and troponin Ⅰ release in high-risk non-ST-segment elevation acute coronary patients treated with percutaneous coronary intervention. Methods We randomized 138 non-ST-segment elevation acute coronary syndrome patients un-dergoing PCI to receive upstream(in the coronary care unit) and downstream(just prior to PCI) tirofiban. We com-pared the effects between the two drug regimens on tissue-level peffusion using Thrombolysis In Mycardial Infarction (TIMI) fram count, the TIMI myocardial perfusion grade(TMPG) and cardiac tropinin Ⅰ (cTn Ⅰ) release before and after PCI. Results There was no significant difference between two groups in TIMI 3 flow(P>0.05). High percent-age of TMPG 3 perfusion were observed in upstream tirofiban group than in downstream tirofiban group (P < 0.05). Post-procedual cTnI elevation was significantly less frequent among patients in the upstream group (P < 0.05). The cTnI level after PCI was significantly lower with upstream tirofiban compared with downstream tirofiban group (P<0.05). Conclusion Among high-risk non-ST-segment elevation ACS patients treated with an early invasive strategy, upstream tirofiban is associated with improved tissue-level perfusion and attenuated myocardial damage.

Objective: To study the effects of UV treatment of different zirconia surfaces on the function of MC3T3-E1 osteoblasts. Methods: Disks of zirconia-based materials with smooth(S) and rough(R) surfaces were treated and with and without UV light(15 W) for 48 h,respectively. MC3T3-E1 cells were cultured on zirconia disks and divided into 4 groups: smooth group(S),UV treated smooth group(S-UV),rough group(R) and UV treated rough group(R-UV). Cell proliferation was assessed by MTT 3 and 5 days after culture. Alkaline phosphatase(ALP) activity was determined 3 and 7 days after culture. Cell mineralization was observed by Alizarin red dye staining 21 days after culture. The mRNA expression of OCN and OPG were examined by RT-PCR 7 and 14 days after culture. Results: Proliferation and mineralization of MC3T3-E1 cells in R-UV group were more than in other groups(P < 0. 05). R groups showed higher ALP activity than S groups at day 7. R-UV treatment increased the mRNA expressions of OPG and OCN in a time-dependent manner(P < 0. 05). Conclusion: Ultraviolet light-treated zirconia surface may promote the proliferation, mineralization and the expression of OCN and OPG of MC3T3-E1 cells.

Objective To investigate the impact of cancer-associated fibroblasts(CAFs)on immunotherapy and liver metastasis in colorectal cancer(CRC).Methods The single-cell sequencing data(GSE205506)of CRC patients with mismatch repair deficiency(MMRd)were downloaded from the gene expression omnibus database, and R software was used to preprocess the original sequencing data and establish the umap of fibroblast subpopulations, with each subpopulation named based on signature genes.GraphPad was used for the statistical analysis of the proportion of each fibroblast subpopulation, and the key subpopulations with significant differences were analyzed among CRC patients before and after PD-1 immunotherapy,as well as between the patients with pathological complete response(pCR)and those without pCR(non-pCR)after treatment. The analysis of differentially expressed genes and the gene pathway enrichment analysis were performed for the key subpopulations. The TCGA database was used to perform a prognostic and survival analysis of the signature genes of key CAF subpopulations, and RNA sequencing data were used to score and calculate the proportion of key CAF subpopulations in the primary lesions of CRC patients with liver metastasis. The independent-samples t test was used for comparison of normally distributed continuous data between two groups;the Kaplan-Meier method was used to plot survival curves, and the log-rank test was used to calculate survival rates.CellPhoneDB software was used to analyze the receptor-ligand interaction between fibroblast subpopulations and tumor cells, and in vitro cell experiments were used to validate the effect of NRG1,a key ligand molecule,on the migration and invasion abilities of CRC cells. Results After PD-1 immunotherapy for CRC patients, there was a significant reduction in the proportion of F6_MMP1+CAFs(P<0.001),which was only observed in patients achieving complete remission after immunotherapy.F6_MMP1+CAFs were upregulated,as well as the genes and signaling pathways associated with tumor migration and invasion, and in addition, there was a significant increase in F6_MMP1+CAFs in the tumor tissue of CRC patients with liver metastasis(P<0.000 1).As a ligand,NRG1 expressed by F6_MMP1+CAFs interacted with ERBB3 receptor expressed by tumor cells, and the in vitro experiments confirmed that NRG1 promoted the migration and invasion abilities of tumor cells by activating the ERBB signaling pathway(P<0.05).Conclusion F6_MMP1+CAFs may affect the efficacy of PD-1 immunotherapy in CRC patients and play an important role in promoting liver metastasis in CRC.F6_MMP1+CAFs,along with NRG1 that is produced by them and can promote tumor metastasis, can be used as potential therapeutic targets and prognostic markers for CRC.
