1.Establishment of Mouse Models with Kidney-positive Deficiency with Antisense Oligonucleotide-loaded Bio-degradable Polylactic Acid Microspheres

OBJECTIVE:To investigate the feasibility of mouse models establishment with kidney-positive deficiency with anti-sense oligonucleotide-loaded biodegradable polylactic acid microspheres. METHODS:Three-week old mice were randomized into groups A,B,C,D and E,with 6 mice in each group,where the mice in group A were given subcutaneous injection 0.2 ml nor-mal saline,and those in groups B-E were given subcutaneous injection 0.1,0.2,0.4 and 0.8 mg each mice of antisense oligonucle-otide-loaded biodegradable polylactic acid microspheres. After the administration,the weights and activities of mice in all groups were observed;and one month later,their livers,adrenal glands and brains were collected to detect the expression of glucocorti-coid receptor (GR) by real-time quantitative polymerase chain reaction (RT-PCR). RESULTS:All mice except those in group A were found to have kidney-positive deficiency symptoms such as developmental delay,light weight,less activities and gathering to-gether. Compared to group A,other groups showed lower GR expressions in liver,adrenal gland and brain. There was statistically difference (P<0.05). CONCLUSIONS:Antisense oligonucleotide-loaded biodegradable polylactic acid microspheres can inhibit GR expression,and the mice whose GR expression is inhibited show symptoms of kidney-positive deficiency,which provides a re-ference for the establishment of mouse models with kidney-positive deficiency.

Echinoderm microtubule associated protein like 4-anaplastic lymphomakinase ( EML4-ALK) rearrangement is another driver-mutation in non-small-cell lung cancer (NSCLC) besides epidermal growth factor receptor (EGFR).Accu-rate identification of EML 4-ALK lung adenocarcinoma is essential for the selection of appropriate therapy .Immunohisto-chemistry (IHC), fluorescence in situ hybridization (FISH), and polymerase chain reaction (PCR) are the main three assay methods for clinical practice .Recently, researchers have done lots of work on the exploration of detection , diagnosis, and treatment prediction of carcinoma based on serum proteomics .In this paper, recent developments in these fields are reviewed.
4.Telomerase activity in peripheral blood mononuclear cells from senile patients with pneumonia.

To investigate the changes of the activity of telomerase in peripheral blood mononuclear cells (PBMCs) from senile patients with pneumonia, the telomerase activity was examined before and after the stimulation of phytohemagglutinin-M (PHA-M) in PBMCs from 10 control subjects (group A), 12 non-senile patients with pneumonia (group B) and 9 senile patients with pneumonia (group C). Also observed was the proliferative response of these PBMCs to PHA-M. The results showed that, both with or without the stimulation of PHA-M, the values of telomerase activity in PBMCs from group C patients (A values: pre-stimulation, 0.43 +/- 0.04; post-stimulation, 0.63 +/- 0. 03) were significantly lower than those in PBMCs from both group A patients (A values: pre-stimulation, 0.65 +/- 0.05; post-stimulation, 1.26 +/- 0.13; P<0.001, respectively) and group B patients (A values: pre-stimulation, 0.63 +/- 0.03; post-stimulation, 0.93 +/- 0.03; P<0.05, respectively). The results of MTT test showed that the proliferative activity of PBMCs in group C patients (A value: 0.35 +/- 0.03) was also significantly lower than that in group A patients (A value: 0. 55 +/- 0.04; P<0.05) and group B patients (A value: 0.46 +/- 0.03; P<0.05). These results indicate that the telomerase activity decreases in senile patients with pneumonia, which may be one of the mechanisms for the weakened immune function in those patients.
5.Expressions of miRNA141 and miRNA375 in serum samples from prostate cancer

To explore the diagnostic values of miRNA141 and miRNA375 for prostate cancer.The expressions of miRNA141 and miRNA375 in serum samples from 30 prostate cancer (PCA) patients,40 benign prostatic hyperplasia patients and 50 normal controls were detected by quantitative reverse transcription-polymerase chain reaction (qRT-PCR).The expression of miRNA141 in tissue samples from prostate cancer was 13.4±7.2 folds of that from benign prostatic hyperplasia while the expression of miRNA375 in tissue samples from prostate cancer was 28.4 ± 10.3 folds of that from benign prostatic hyperplasia.The differential expression of miRNA141 and miRNA375 in sera of prostate cancer suggested that miRNA141 and miRNA375 might be potential tumor markers in clinical practice.
6.Clinical study on the onset of systemic lupus erythematosus in 20 postmenopausal patients

Objective To study the clinical characteristics of the onset of systemic lupus erythematosus(SLE) in postmenopausal patients. Methods The clinical features and prognosis of 20 SLE patients whose onset was at least one year after menopause,and 70 patients with the onset during normal menstruation and 43 male patients between 1992 and 2003 were comparatively analyzed. Results The onset of SLE was seen after menopause in 20 out of 699 (2.9%) female patients. The average age at the onset of the disease was 55 years with a peak from 50 to 59. The common clinical manifestations were arthritis,lassitude,fever,alopecia,malar rash,cardiac impairment and weight loss,while laboratory abnormalities were high ESR,decreased C3,ANA≥1∶80,hypergammaglobulinemia and positive anti-RNP antibody. 7.Expression of Th1/Th2 inlfammatory cytokines in rat treatment model of ulcerative colitis

Objective: To investigate the expression of Th1/Th2 inflammatory cytokines IL-2, interferon-γ (IFN-γ), IL-4, and IL-10 in rat treatment model of dextran sulfate sodium (DSS) -induced ulcerative colitis. Methods: Forty Sprague Dawley (SD) male rats were divided into a normal group, a colitis model group, a sulfasalazine(SASP)-treatment group (SASP group) and a Jiechangning-treatment group (Jiechangning group) (each groupn=10). Disease activity index (DAI) and colorectal histological damage scale were assessed. The expression levels of cytokines IL-2, IFN-γ, IL-4, and IL-10 in the serum and the colon mucosa tissues were detected by enzyme-linked immuno sorbent assay (ELISA) and real time polymerase chain reaction (RT-PCR).
Results: Compared with the colitis model group, the DAI and colorectal histological damage scale were decreased in the Jiechangning group (bothP<0.05), but there was no obvious difference compared with the SASP group (P>0.05). There was no signiifcant difference in IL-2 expression both in the serum and the colon mucosa tissues before or after the treatment in various groups (P>0.05). Compared with the colitis model group, IFN-γ level both in the serum and the colon mucosa tissues was decreased in the SASP group and the Jiechangning group, with significant difference in the serum (bothP<0.05), but there was significant difference in the colon mucosa tissues only in the Jiechangning group (P<0.05). The serum IL-4 level in the SASP group and the Jiechangning group was increased compared with that in the colitis control group, with significant difference only in the Jiechangning group (P<0.05). There was no difference in IL-4 level in the colon mucosa tissues whether treated or not (P>0.05). IL-10 level both in the serum and the colon mucosa tissues in the SASP group and the Jiechangning group was increased compared with that in the colitis model group, with significant difference (allP<0.05). There was no difference in the expression level of cytokines IL-2, IFN-γ, IL-4, and IL-10 both in the serum and the colon mucosa tissues between the SASP group and the Jiechangning group (allP>0.05).
Conclusion: DSS can break the balance of Th1/Th2 expression in the colon. Jiechangning enema can ameliorate DSS-induced acute experimental colitis in rats by decreasing IFN-γ level and increasing IL-10 level both in the serum and the colon mucosa tissues to regulate the Th1/Th2 balance and improve immunity.
8.Experience in the diagnosis and treatment of 31 patients with primary presacral tumor

ObjectiveTo explore the diagnosis and surgical treatment techniques for the patients with primary presacral tumor.Methods Data from 31 patients with primary presacral tumor admitted into our Department for Surgery from Jan. 1999 to Jar. 2009 were retrospectively analyzed.All patients underwent surgical treatment,among them,10 received per-sacroiliac or per-perineum approaches to remove the tumors,18 receivedper-abdomentumorectomy, and3receivedcombinedper-peritoneumandper-perineum tumorectomy.Results Patients visited the doctor while exhibiting the compression symptoms by the the tumors.Final diagnosis could be reached with the help of rectal touch,B-mode ultrasound,computerized tomography,or magnetic resonance imaging.Complete resection was performed in 28 patients.Partial resection was performed in 3 patients.Fractionated resection was successfully performed in 2 patients with chronic infection or sinus tract.ConclusionThe primary presacral tumor should be surgically resected once final diagnosised and without surgical contraindications.Pelvic plexus should be preserved and presacral hemorrhage should be prevented.The tumor should be resected as complete as possible.
9.Effect on the Content of Peoniflorin in Radix Paeoniae Alba for Different Storing Time after Harvesting.

Objective To determine the content of paeoniflorin with different medicinal parts in Radix Paeoniae Alba and offer a reference for collection and processing. Methods The content of paeoniflorin was measured by HPLC method. Combining air drying with weight relief condition, the content of paeoniflorin was calculated in fresh Radix Paeoniae Alba. Results There was the highest content of paeoniflorin in Radix Paeoniae Alba in 24~48 hours after collection, and the another increasing of the content was founded 120 hours after collection. Conclusion Radix Paeoniae Alba should be processed in 48 hours after collection.
10.Assaying telomerase activity of urine exfoliated cells and its clinical significance

Objective: To determine the value of telomerase activity in urine exfoliated cells as clinical indicator of tumor presence, stage, and recurrence. Methods: The techniques of TRAP-PCR and TRAP-sliver staining were employed to detect telomerase activity in 73 patients with TCC before operation (study group) and 20 benign urothelial cancer patients (control group) and 21 normal individuals (normal group). Cytologic results were obtained simultaneously. The bladder tumor specimens were obtained from 73 patients during operation, and histologically evaluated for tumor content and grade. Results: Positive rate of telomerase activity detection in TCC patients (80.8%,59 of 73) was significantly higher than that of cytological examination (20.5%, 15 of 73). Positive telomerase activity was not found in 17 of 20 in control group and none was found in normal group. Analysis of the distribution of abnormalities with tumor stage revealed greater detection of high pathological stage (T 2-T 4) (89.7%, 26 of 29) compared with low stage (Tis-T 1) (75%, 33 of 44). Conclusion: Detection of telomerase activity in urine exfoliated cells may be a sensitive and effective marker in the diagnosis of TCC.
