1.Protective Effects of Neferine on Lipopolysaccharide-induced Human Umbilical Vein Endothelial Injury

Objective:To investigate the protective effects of neferine on lipopolysaccharide ( LPS)-induced human umbilical vein endothelial cells ( HUVECs) injury. Methods:The optimum inducing concentration of LPS was screened out through pretests and used for the model establishment of HUVECs damage. CCK8 was used to detect the influence of neferine at different concentrations on LPS-induced human umbilical vein endothelial cells ( HUVECs) injury. Nitric oxide ( NO) content was measured by the Griess Reagent method. The nitric-oxide synthase (NOS) activity was assessed by the commercially available kits. Results:The inhibitory rate of HU-VECs was 54. 50% detected by CCK8, which induced by LPS at the concentration of 100 μg·ml-1(P<0. 01). Neferine at the con-centrations of 0. 3-5. 0μmol·L-1 could increase the cell viability in a concentration-dependent manner, while it inhibited the cell pro-liferation at the concentration of 10 μmol·L-1(P<0.05). Neferine could reverse the situation, and the NO release was increased and the tNOS/iNOS activity was increased induced by LPS (P<0. 05). The results shown by the inverted microscope suggested that the floating dead cells were decreased, the cell shape was basically sound and tightly packed with the concentration increase of neferine (0. 3-5. 0μmol·L-1) in a concentration-dependent manner. Conclusion:The results show that neferine has protective effects on HU-VECs injury induced by LPS, and the mechanism may be related with the decrease of intracellular levels of NO and NOS activity.

The review focused on its pharmacological effects and pharmacokinetic characters reported in recent years,which can be helpful to the further researches on nuciferine.
3.Intrahepatic biliary cystadenocarcinoma of 17 cases

Objective To explore the clinical features, treatment and prognosis of intrabepatic biliary cystadenocarcinoma (IBC). Methods We retrospectively analyzed clinical data of 17 patients with pathologically confirmed IBC, treated between January 2002 and September 2007. Results Of the 17 patients, 4 were men and 13 were women, with a mean age of 49.3 years (range 45 -68). Serum level of AFP was normal in all patients. CA19-9 was abnormally elevated in only two patients and the other two had abnormal serum level of CEA. Seven cases underwent radical resection, 7 underwent complete tumor resection combined with removal of tumor thrombi in the bile duct, 3 received palliative surgery. On histopathological examination, 10 were biliary cystadenocarcinoma,2 were biliary cystadenoma with partial eanceration,2 were papillary cystadenocarcinoma,2 were mutinous papillary cystadenocarcinoma and 1 was of mixed cystadenocarcinoma and hepatocellular carcinoma. According to tumor differentiation, the number of well, moderate and poor-differentiated group was 7, 3 and 7 cases respectively. The medality of surgery (β= - 0.692, P = 0.01) and tumor differentiation (β = - 2.041, P = 0.007) effected the prognosis significantly. Conclusions IBC occurs mainly in elderly women. CA19-9 examination does not help in the establishment of diagnosis of IBC. The occurrence of tumor thrombosis in common bile duct doesn't necessarily indicate poor prognosis, hence a IBC patient will still have a satisfactory prognosis should the primary tumor be completely resected and tumor thrombi removed.
4.SIMULTANEOUS DETERMINATION OF THIAMINE, RIBOFLAVIN, NIACIN AND PYRIDOXINE IN FOODS BY HPLC

A high performance liquid chromatographic assay has been developed for the simultaneous determination of four water soluble vitamins,thia min, ribo-flavin, uiacin and pyridoxine, in cereal products and fresh vegetables. For hydrolyzing sample in 0.4 mol/L HCL, autoclaving at 120℃, 15 psi for 30 minutes, a ?Bondapak column (300mm ? 3.9mm, Waters Co), a mobile phase of methanol-water (30:70) (0.005 mol/L heptanesulfonic acid) and a flow rate of 1.0ml/min gave the most satisfactory separation of the four vitamins. A double channel detector was used: three vitamins (B1, B2 and B5) were detected by UV spectrophotometry (254 nm) first, pyridoxine (B6) was detected by fluorometry (EX 290 nm, EM 390 nm) afterwards. Detection limits were 2, 5, 5 and 2ng, linear ranges were 5-40 ng, 5-50 ng, 5-40 ng and 5-50 ng for B1, B2, B5 and Be, respectively. Recoveries were 91.3%-102% (B1), 103%-105% (B2), 92%-100% (B5) and 99%-100% (B6), comparing with reference method and checking with food composition tables, very satisfactory results were obtained by this method.
5.Effects of glycyrrhizin on the proliferation of ASMC induced by fetal calf or histamine in rats

Aim To investigate the effects of glycyrrhizin on the proliferation of ASMC stimulated by fetal calf or histamine in rats. Methods Cell culture of rat ASMC, MTT assay, flow cytometry and cell growth counts were used in this study. Results ①In the culture medium containing 100g?L -1 fetal calf serum, Glycyrrhizin at low concentration(6?10 -5 mol?L -1 ) stimulated the increase of A 570 in ASMC. This effect descended with increasing glycyrrhizin concentration and changed to be inhibitory at high concentration (from 384?10 -5 mol?L -1 to 1 536 ?10 -5 mol?L -1 ). In the culture medium containing 10 -2 mol?L -1 histamine, Glycyrrhizin at both low and high concentration inhibited the increase of A 570 in ASMC. ②In the culture medium containing 100 g?L -1 fetal calf, Glycyrrhizin at low concentration(6?10 -5 mol?L -1 ) stimulated the proliferation of ASMC. This effect descended with increasing glycyrrhizin concentration and changed to be inhibitory at high concentration (from 384?10 -5 mol?L -1 to 1 536 ?10 -5 mol?L -1 ). In the culture medium containing 1 g?L -1 histamine, Glycyrrhizin at both low and high concentration inhibited the proliferation of ASMC. ③In the culture medium containing 100 g?L -1 fetal calf or 10 -2 mol?L -1 histamine, with increasing glycyrrhizin concentration, the cell count in G 1 phase increase, the cell count in G 2 and M phase decrease. Conclusion ①Glycyrrhizin accelerated the proliferation of ASMC stimulated by fetal calf at low concentration, inhibited the proliferation at high concentration. ②Glycyrrhizin inhibited the proliferation of ASMC stimulated by histamine at both low concentration and high concentration.
6.Evaluation of transnasal esophagogastroduodenoscopy:a controlled trial

Objective To evaluate the safety and efficacy of ultra-thin transnasal esophagogastroduodenoscopy(EGD).Methods The patients(n=240)receiving diagnostic EGD were randomly assigned to 3 groups to undergo ultra-thin trails-nasal EGD(group A,n=82),ultra-thin trans-oral EGD(group B,n=79)and conventional trans-oral EGD(group C,n=79).The blood pressure,pulse rate,arterial oxygen saturation,myocardial oxygen consumption were monitored and recorded before and during the procedure,as well as the operation time.All patients completed a questionnaire after the procedure.Results No significant difference was found in drop of arterial oxygen saturation between three groups.Increases in pulse rate,blood pressure,rate-pressure product in group A were significantly lower than those in group B and C(P<0.05).Patients in group A experienced the lowest VAS scores of discomfort and nausea during the procedure (P<0.05).But examination time and insertion time were significantly longer in group A than those in group B and C.With the increase in number of cases,the insertion time for group A decreased gradually.Conclusion Ultra-thin trans-nasal EGD is well tolerated and has fewer hemodynamic effects.It is safe and may be the optimal route of intubation with ultra-thin scopes.

Objective To observe the effect of STK15 gene silencing on the growth of gastric cancer cell line-MKN45 in vitro and vivo. Methods STK15 expression was inhibited by RNAi techenique, STK15 protein level was detected by Western blot, the ability of MKN45 invasion in vitro was assessed by cell migration and invasion assay, the change of cell cycle distribution was detected by flowcytometry, MKN45 proliferation was measured by MTT method, and MKN45 cells treated with STK15 siRNA were transplanted subcutanuously in nude mice and their tumorgenesis ability were observed. OBJECTIVE To study the risk factors related to the infection in the patients with the breast implant and soft tissue expander postoperatively,such as the time of drainage and retro-irrigation.METHODS Thirty consecutive patients who underwent breast implants and soft tissue expander involving at least two sites and set negative pressure drainage tube were studied.For each patient with drainages at two sites,one was retro-irrigated during the early stage postoperatively,the other was not.The samples were from the skin out of the drainage tube,the drainage tube out of skin and the end of drainage after 2,4,6 and 8 days of the operation and at the time of removing the tube.Total samples were tested for bacteriology.RESULTS Samples were divided into two groups: common groups and retro-irrigated group with negative pressure drainage.A total of samples from each group for bacterial test were 170.The postive rate was 2.94% and 1.76%,respectively(P=0.474).The positive rate of bacterial test was closely related to the time at all 3 different sampling sites(P
9.Clinical significance of H-FABP,hs-CRP and Hcy in patients with atrial fibrillation and chronic heart fail-ure

Objective To investigate the level changes and clinical value of serum H-FABP,hs-CRP and Hcy in patients with atrial fibrillation(AF)and chronic heart failure(CHF).Methods 120 patients with AF and CHF were chosen as the observation group,100 patients with sinus rhythm were chosen as the control.The levels of H-FABP,hs-CRP and Hcy of the two groups were compared.H-FABP,hs-CRP and Hcy levels were also compared in the different subgroups divided according to the cardiac function.Risults Compared with the control group,the levels of H-FABP,hs-CRP and Hcy in patients with atrial fibrillation were significantly higher(P<0.05).The lev-els of H-FABP,hs-CRP and Hcy in the observation group were higher than those in the control group in the same cardiac function subgroup(P < 0.05). Conclusion The elevation of the level of H-FABP,hs-CRP and Hcy are closely related to the occurrence and maintenance of atrial fibrillation and the damage of myocardial cells,and it was related with the severity of cardiac function.

Objective:
To analyze the influence of the confidence to pre-exposure prophylaxis (PrEP) on the willingness to use PrEP in men who have sex with men (MSM) by using the path analysis model based on structural equation model approach.
Methods:
A total of 550 MSM were recruited in Urumqi by snowball method and a questionnaire survey was conducted among them. According to the professional knowledge and analysis results of the confidence and willingness to use PrEP, structural equation model (SEM) analysis method was used to construct a path analysis model.
