1.The HRCT findings of pleural invasion of lung adenocarcinoma presenting as mGGN

Objective To investigate the high resolution CT (HRCT) findings of pleura invasion of lung adenocarcinoma (stageⅠ) presenting as mixture ground glass nodule (mGGN).Methods 20 cases of lung adenocarcinoma presenting as mGGN were confirmed by pathology.The correlation of those cases with pleural adjacent relation and the relation between the maximum diameter and pleural invasion were analyzed retrospectively.Results (1)The classification of mGGN adjacent to the pleura relationship:type Ⅰ,typical and atypical pleural retraction sign,10 cases of this type did not show pleural invasion;type Ⅱa,the mGGN contacted pleural for ground glass opacity,4 cases of this type showed no pleural invasion;type Ⅱb,the mGGN contacted pleural for solid elements,6 cases of this type were with pleural invasion.(2)The difference of the maximum diameter of mGGN in groups pleural invasion and non-invasion was not significant statistically (P=0.694).Conclusion The mGGN contacted pleural for solid composition is a reliable sign in diagnosis of pleural invasion.
2.Expression of MMP-1 and MMP-8 in periodontal tissues in rats with periodontitis

Objective:To investigate the expression of MMP-1 and MMP-8 in periodontal tissues in rats with periodontitis at different stages of inflammation with varied severity. Methods:Periodontitis was induced by silk ligature. P.gingivalis(Pg) or Pg with F.nu were used to induce the varied severity of periodontitis in 40 rats. 14 and 28 days after periodontitis induction the animals were sacrificed, periodontal tissues were immunohistochemically stained by antibody of MMP-1 and MMP-8 respectively. Results:MMP-1 and MMP-8 were both strongly positive in gingival epithelia cells and fibroblasts in periodontal ligament in rats with periodontitis.Higher expression of MMP-1 and MMP-8 was observed at 14 d than at 28 d(P0.05). Conclusion:The expression of MMP-1 and MMP-8 varies in different stage of periodontitis. MMP-1 and MMP-8 may play an important role in development of periodontitis.
3.CT and MRI features of testicular solid lesions

Objective To analyze the imaging features of testicular solid lesions.Methods The CT and MR findings of 18 cases of testicular solid lesions confirmed by pathology were retrospectively reviewed.The location,attenuation/signal,size,shape,edge,and enhancement patterns of lesions were analyzed.Results In all of the 18 cases,5 cases were seminomas,1 of them was huge with punctate calcification,4 of them had low signal separations in T2WI.Lymphomas were in 4 cases,and all of them were primary diffuse large B cell lymphoma;T2WI was priority with low signal.Solitary fibrous tumor was in 1 case,and the attenuation was lower than that of muscle.Embryo in sinus tumor were in 1 case,and the attenuation/signal was unhomogeneous;the continued enhancement liking grid was obviously seen after contrast injection.Nonspecific inflammatory masses were in 4 cases with involving epididymis in 2 cases,and the attenuation/signal was unhomogeneous.Tuberculous mass were in 3 cases,and 1 case with points of calcificationwas on the edge of lesion,2 cases characterized by short T2 signals.Conclusion It is possible to make an accurate diagnosis by the combination of clinical data with imaging features in most of the testicular solid lesions.
4.Optimization of Prescription of Flaxseed Lignans Sustained-release Tablets by Central Composite Design-response Surface Methodology

Objective To optimize the prescription of flaxseed lignans sustained release tables by central composite design-response surface methodology.MethodsWith HPMC, EC and starch dosage as factors, and flaxseed lignans in 2, 6 and 12 h of cumulative release as evaluation indexes, central composite design-response surface optimization method was used to conduct prescription optimization experiments, and optimized prescription analysis was carried out.Results The optimal prescription of flaxseed lignans sustained release tables was as following: HPMC dosage was 43%; EC was 26%; starch content was 17%. Optimized index forecast values were very close to the observed values. In vitro release test of three selected optimal formulations indicated that there existed high approximation between the observed and estimated values.Conclusion It shows that the established model is suitable for flaxseed lignans sustained release tables, which can be used in the optimization of the prescription of flaxseed lignans sustained release tables.
5.Research Progress in Animal Models of IBS-D Disease Combined with Liver Stagnation and Spleen Deficiency

At present, TCM treatment of diarrhea-predominant irritable bowel syndrome (IBS-D) is based on the combination of disease differentiation and syndrome differentiation. Therefore, the establishment of IBS-D of liver stagnation and spleen deficiency syndrome combined with animal model as a combination of traditional Chinese and Western medicine innovation theory has become increasingly concerned about, and gradually become a new direction for the development of TCM experimental animal model. This article reviewed the research progress in IBS-D liver and spleen deficiency syndrome in recent years, discussed the establishment of IBS-D liver stagnation and spleen deficiency animal model and research ideas for the treatment of IBS-D, and provided references for mechanism research of TCM treatment for IBS-D and research and development of new medicine.
6.Effects of probiotics combined with dietary intervention on glucose and lipid metabolism and pregnancy outcome in gestational diabetes mellitus

Objective To investigate the effects of probiotics combined with dietary intervention on pe-ripheral blood glucose and lipid metabolism indicators,placental tissue insulin signaling pathway proteins ex-pression and pregnant outcome in the patients with gestational diabetes mellitus(GDM).Methods A total of 83 patients with GDM in this hospital from December 2021 to December 2022 were selected as the study sub-jects and divided into the probiotics group(probiotics combined with diet intervention,43 cases)and control group(simple diet intervention,40 cases)by the random number table method.The levels of peripheral blood glucose,lipid and insulin resistance related indicators before the intervention and in 8 weeks after the interven-tion were compared between the two groups.The histological changes of placenta were observed by HE stai-ning.The pathological indicators were compared between the two groups.The expression levels of insulin re-ceptor substrate-1(IRS-1),glucose transporter 4(GLUT4)and synaptosome-associated protein of 23 kDa(SNAP23)in placental tissue were detected by immunohistochemistry.The pregnant adverse outcomes were compared between the two groups,and the clinical efficacy of probiotics was evaluated.Results Compared with the control group,the levels of fasting blood glucose(FBG),fasting insulin(FINS),serum triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)in 8 weeks after intervention in the probiotics group were significantly decreased(P<0.05),and the level of serum high density lipoprotein cholesterol(HDL-C)was significantly increased(P<0.05).There were no significant differences in the incidence rates of poor villi maturation,thickening of dry villi arterioles and capillary filling in villi interstitial between the two groups(P>0.05).Compared with the control group,the expression levels of IRS-1,GLUT4 and SNAP23 in placen-tal tissue of the probiotics group were significantly increased(P<0.05).The incidence rates of neonatal hy-poglycemia and neonatal hyperbilirubinemia in the probiotics group were significantly lower than those in the control group(P<0.05).Conclusion Compared with simple dietary intervention,probiotics combined with dietary intervention has more advantages in improving glucose and lipid metabolism of GDM patients,moreo-ver reduces the adverse events occurrence in newborns.
7.Best time for the first wrist bandage decompression in patients with coronary artery intervention after radial artery puncture operation

Objective To explore the best time for the first wrist bandage decompression in patients with coronary artery intervention after radial artery puncture operation. Methods A total of 165 patients were randomly divided into observation group ( n=85 ) and control group ( n=80 ) . Wrist bandage decompression time was 60 min and 90 min after operation in the observation group and in the control group. We recorded blood oxygen saturation (SpO2) of operated hand and unaffected hand at 15, 30, 45, 60, 90 and 120 min of post-operation. Then, we compared the differences of vascular complication and degrees of comfort. Results The SpO2 was (94. 48 ± 2. 585)% and (95. 06 ± 2. 014)% in operated hand and unaffected hand at 15 min after operation in the observation group (t= -0. 118, P<0. 05). The SpO2 was (94. 41 ± 1. 992)% and (95. 25 ± 2. 149)% in operated hand and unaffected hand at 120 min in the control group (t= -3. 617, P<0. 01). The SpO2 was no significant difference between these six time points in operated hand (t= -0. 233, 0. 269, 0. 992,-0. 203, 0. 297, 0. 670; P>0. 05). There was no incision bleeding and radial artery occlusion in both two groups. There was 1 case upper arm swelling and 1 case ecchymoma in the observation group. There was no statistically difference between vascular complication and degree of comfort (χ2 =4. 023, 1. 418, P>0. 05). Conclusions Best time for the first wrist bandage decompression was 60 min after coronary artery intervention, which can decrease the risks of vascular complication and increase degree of comfort.
8. Fatty acid synthase interacts with signal transducer and activator of transcription 3 to promote migration and invasion in liver cancer cells

Objective:
Hepatocellular carcinoma (HCC) is one of the most common malignant tumor worldwide. Metastasis is a marker of cancer deterioration in patients with liver cancer and a major cause of death. In order to develop effective therapeutic strategies, it is urgent to study the molecular basis of liver cancer metastasis.
Methods:
Immunohistochemistry was used to detect the expression of fatty acid synthase (FASN) in HCC. Wound healing and transwell cell invasion assays was used to confirm the role of FASN in liver cancer migration and invasion. Proteins that interacted with FASN were identified using iTRAQ (isobaric tag for relative and absolute quantification). Co-immunoprecipitation (Co-IP) and cellular immunofluorescence analysis were used to assess the interaction between FASN and signal transduction and transcription activator 3 (STAT3). The expression of STAT3, p-STAT3, matrix metalloproteinase (MMP)-2 and MMP-9 was detected after FASN knockdown using Western blot method. Statistical analysis was performed using the t-test.
Results:
Immunohistochemistry showed that the expression of FASN in HCC tissue was higher than that in adjacent tissues. iTRAQ, Co-IP and immunofluorescence analysis revealed that FASN interacted with STAT3. Western blot analysis showed that the expression of p-STAT3, MMP-2 and MMP-9 decreased after FASN knockdown.
Conclusion
FASN may promote the metastasis of liver cancer by interacting with STAT3 and affecting the expression of MMP-2/MMP-9.
9.Evaluation of correlation between left ventricular changes and left atrial function in hypertrophic cardiomyopathy based on quantitative cardiac MR

Objective:To explore whether left ventricular interstitial fibrosis is associated with left atrial enlargement and left atrial dysfunction in patients of hypertrophic cardiomyopathy(HCM) with preserved ejection fraction.Methods:From October 2018 to September 2021, 59 HCM including 30 with enlarged maximal left artrial volume index (LAVI max), 29 with normal LAVI max and 28 age-and gender-matched controls were retrospectively enrolled. Imaging protocol included cine sequence, late gadolinium enhancement and T 1 mapping.The relationships between left ventricular mass index (LVMI), quantitative myocardial fibrosis and left atrial-related indexes were analyzed. One-way analysis of variance with Bonferroni post hoc correction or Kruskal-Wallis was performed for continuous variables. Categorical variables were assessed using the Chi-square test or Fisher′s exact test. Pearson or Spearman analysis was used for linear or monotonic nonlinear correlations. Results:The left ventricular end-diastolic volume index, left ventricular end-systolic volume index, left ventricular cardiac output and LVMI of HCM with enlarged LAVI max group were higher than HCM with normal LAVI max group and control group( P<0.05).Correlation analysis showed that LVMI correlated positively with LAVI max( r=0.780, P<0.001) and minimal left artrial volume index (LAVI min) ( r=0.816, P<0.001), extracellular volume correlated positively with LAVI max( r=0.462, P<0.001) and LAVI min( r=0.483, P<0.001),%LGE was correlated positively with LAVI max( r=0.311, P<0.05) and LAVI min( r=0.327, P<0.05),left ventricular index interstitial volume was correlated negatively with left atrial ejection fraction of reservoir ( r=-0.669, P<0.001),left atrial ejection fraction of conduit ( r=-0.472, P<0.001),left atrial ejection fraction of pump ( r=-0.518, P<0.001)and left atrial expansion index( r=-0.626, P<0.001). Conclusion:There is association between LVMI and fibrosis and left atrial enlargement and phases dysfunction in HCM with preserved ejection fraction.
10.Study on the distribution of human alanine aminotransferase isoenzyme in human tissues.

This study intends to obtain recombinant proteins of ALT1 and ALT2 isozymes by using genetic recombination technology. Monoclonal antibodies ALT1 and ALT2 with high specificity and high activity were prepared and screened (ALT1 monoclonal antibody has been successfully prepared and published). The localization, distribution and expression of ALT1 and ALT2 isozymes in human tissues were discussed. The ALT2 genes were amplified from human liver cancer cell (HepG2) by RT-PCR method. The mature ALT2 gene was subcloned into the pET32a-ALT2 prokaryotic expression vector. Its ligation product was transformed into BL21(DE3) competent cells, and transformed into competent cells to express ALT2 proteins induced by IPTG. The recombinant proteins of ALT2 were purified by nickel column (Ni⁺) affinity chromatography. Balb/c mice were immunized with recombinant proteins of ALT2. Positive serum mouse spleen cells and myeloma cells SP2/0 were selected for cell fusion. The positive cell lines were selected by indirect ELISA and subcloned by limited dilution method. Affinity chromatography was used to purify ALT2 antibodies. The expression and distribution of ALT2 in human normal tissues were detected by RT-PCR and Western blotting. Results show that the expression of ALT isoenzyme in tissues was almost the same at gene mRNA level and protein level. ALT1 is highly expressed in liver, kidney and skeletal muscle, and moderately expressed in gastrointestinal smooth muscle. ALT2 is highly expressed in fat, skeletal muscle and myocardium, and is poorly expressed in gastrointestinal smooth muscle. Immunohistochemical studies show that ALT1 is highly expressed in hepatocytes, renal medullary tubules and muscle fibers, ALT2 is highly expressed in adipocytes and myocardial cells, and ALT1 and ALT2 in gastrointestinal tissues are mainly expressed in mucosa of upper intestinal wall region. The results showed that the isoenzymes ALT1 and ALT2 were mainly expressed in the mucosa of the upper part of the intestinal wall. It is widely distributed in the tissues, providing theoretical basis for understanding the mechanism of ALT activity increase under different pathological conditions.
