1.Laser treatment for premacular hemorrhage

Objective To evaluate the effective method for treatment of premacular hemorrhage. Methods In 36 cases (36 eyes) of premacular hemorrhage (2PD),25 cases were treated with medicine, and 11 cases were treated with laser surgery and medicine. Results In 25 patients treated with medicine, the average time of vision recovery was 24 days and the average time of clearance of vitreous hemorrhage was 40 days; in the other 11 patients treated with laser surgery and medicine,10 were treated successfully, and in these 10 patients, the average time of vision recovery were 5 days and the average time of clearance of premacular hemorrhage were 11 days in 10 cases which were operated successfully with laser in 11 cases. Conclusions Laser surgery of inducing preretinal hemorrhage to vitreous body is an effective method for treatment of premacular hemorrhage to relieve the impaired central vision.

3.Effect of tetramethylpyrazine on Nissl bodies in cerebral neurons of rats with epilepsy

BACKGROUND: Tetramethylpyrazine has the protective effect against the central nervous system injury. The structural changes in Nissl bodies were regarded as a marker of neuron injury.OBJECTIVE: To investigate the effect of tetramethylpyrazine on the structure and the quantity of Nissl bodies of cerebral neurons in rat with epilepsy.DESIGN: A comparative study.SETTING: It was conducted at the Physiological Department of Medical School of Xianning College.MATERIALS: From September 2004 to March 2005, it was completed at the Anatomy Department of Tongji Medical College, Huazhong University of Science and Technology. Forty healthy SD rats aging 3-4 months, weighing (250±50) g and regardless of their gender, were selected.METHODS: Rats underwent anesthesia and craniotomy. Then their cerebral cortex were exposed for placing BL-410 Experimental System of Biological Function (TME, China) to record the bilateral EEG of the brain and the seizure in rats with penicillin-induced epilepsy group and the 10 mg/kg tetramethylpyrazine group, 20 mg/kg tetramethylpyrazine group and 40 mg/kg tetramethylpyrazine group. In control groups, the brains of rats were taken out at 1 hour after craniotomy. In penicillin-induced epilepsy group, their brains were taken out at 1 hour after penicillin-induced epilepsy. In 10 mg/kg tetramethylpyrazine group, 20 mg/kg tetramethylpyrazine group and 40 mg/kg tetramethylpyrazine group, after stable penicillin-induced epilepsy, tetramethylpyrazine was injected intraperitoneally at a dose of 10 mg/kg, 20 mg/kg and 40 mg/kg, respectively.When the greatest protective effect of tetramethylpyrazine appeared, the rats' brains were taken out. Brain sections were sliced. Nissl bodies were stained by thionine staining. Under light microscope, structures of Nissl bodies were observed and the images of Nissl bodies were quantitatively analyzed by HPIAS-1000 high acuity color pathologic diagram-writing analyzing system. In each group, the average absorbency of 15 fields was regarded as the average absorbency of Nissl bodies in that group.MAIN OUTCOME MEASURES: In all the groups, the structure and the quantity of Nissl bodies of cortical neurons in rats were studied.of the structure of Nissl bodies in external granular layer cells and external pyramidal layer cells. In control group, multi-layer blue-stained and clumplike or granule-like Nissl bodies could be observed. In penicillin-induced epilepsy group, Nissl bodies were completely resolved and disappeared. In 10 mg/kg tetramethylpyrazine group, Nissl bodies were partly or completely resolved. In 20 mg/kg tetramethylpyrazine group, the quantity of Nissl bodies was significantly increased as compared with those in penicillin-induced epilepsy group and the 10 mg/kg tetramethylpyrazine group. In 40 mg/kg tetramethylpyrazine group, the quantity and the structure of of the average absorbency of stained Nissl bodies in external granular layer cells and external pyramidal layer cells in rat cortex among all the groups.In penicillin-induced epilepsy group, the average absorbency were dramatically lower than that in control group (0.033±0.002, 0.756±0.035, t=4.93,P < 0.01). In 20 mg/kg tetramethylpyrazine group and 40 mg/kg tetramethylpyrazine group, the average absorbency were significantly higher than that in penicillin-induced epilepsy group (0.435±0.011, 0.658±0.029, t=2.98,5.32, P < 0.01). In 40 mg/kg tetramethylpyrazine group, the average absorbency were similar to that in control group (t=1.75, P > 0.05).CONCLUSION: Tetramethylpyrazine can significantly elevated the concentration of Nissl bodies of neurons in rats with epilepsy. Changes in the structure and quantity of Nissl bodies of cerebral neurons may be closely associated with seizure, and tetramethylpyrazine can restore the structure and the quantity of Nissl bodies of neurons, regulate their functions and hereby, inhibit seizures.
4.Optimization of Preparation Technology of Compound Sodium Lactate Injections for Improvement of Clar-ity

Objective:To optimize the preparation technology of compound sodium lactate injections for the improvement of clari-ty. Methods:The orthogonal test was adopted to screen the best preparation conditions using four influencing factors including the de-carburization temperature of the concentrated solution (A), pH value of the concentrated solution (B), decarburization time of the concentrated solution ( C) and the filling temperature of the injections ( D) with 3 levels for each, and the clarity of the injections as the index. Results:The best preparation conditions were as follows:the decarburization temperature was 60℃, pH value of the con-centrated solution was 6. 5, the decarburization time was 20minute and the filling temperature was 50℃. Conclusion: The optimal process can improve the clarity of compound sodium lactate injections.
5.Half-dose verteporfin and half-fluence photodynamic therapy for chronic central serous chorioretinopathy

Objective To compare the efficacy and safety of half-dose verteporfin and half-fluence photodynamic therapy in chronic central serous chorioretinopathy (CSC).Methods The medical records of 42 patients (42 eyes) with chronic CSC were retrospectively reviewed.According to the difference of drug dose and laser parameters in the course of PDT,the patients were divided into two groups:half-dose group and half-fluence group.Half-dose group:22 patients (22 eyes) received half-dose verteporfin(3 mg· m-2)and standard PDT(83 s,50 J · cm-2).Half-fluence group:20 patients (20 eyes) received full-dose verteporfin (6 mg · m-2) and half-fluence PDT (42 s,25 J · cm-2).Patients were followed up at 1 month,3 months,6 months after PDT.The outcome measures was the proportion of eyes with complete resolution of subretinal fluid(SRF),the changes in best corrected visual acuity (BCVA) and central retinal thickness(CMT).Results At 6 months after PDT,22 eyes(100%) that received half-dose PDT showed complete resolution of SRF,19 eyes(95%)that received half-fulence PDT showed complete resolution of SRF.There was no statistical difference between two groups (P > 0.05).At 6 months after PDT,the mean BCVA improved 7.2 letters in half-dose group and 6.7 letters in half-fluence group.There was no statistical difference compared with the improvement of BCVA between two groups(P > 0.05).In half-dose group,mean baseline central retinal thickness was (351 ± 90) μm,which was decreased to (178 ±55) μm after 6 months,respectively (P < 0.05).In half-fluence group,mean baseline central retinal thickness was (322 ± 96) μm,which was decreased to (181 ± 47) μm after 6 months,respectively (P < 0.05).None of the patients developed retinal pigment epithelium atrophy and CNV.Conclusion Half-dose verteporfm PDT and half-fluence PDT are effective and safe in the treatment of chronic CSC.
6.Influence of ligustrazine on neuronal structure in brain of epileptic rats

BACKGROUND: Ligustrazine inhibits discharge of cerebral hippocampal neuron, penetrates blood-brain barrier effectively after absorbed in the body and is distributed extensively in cerebral cortex, brain stem, striate body, hippocampus, cerebellum and midbrain.OBJECTIVE: To probe into the influence of ligustrazine and its different concentrations after abdominal injection on cerebral cortical neural cell structure in epileptic rats induced by penicillin.DESIGN: Randomized control experiment.SETTING: Physiological Department of Xianning Medical College.MATERIALS: The experiment was performed in Anatomy Department of Tongji Medical College of Huazhong University of Science and Technology from September 2004 to March 2005. Forty healthy SD rats of clean grade were employed, of either sex, mass weighted varied from 200 to 250 g.They were randomized into 5 groups, named operation control, penicillininduced epilepsy group and ligustrazine groups of 10 mg/kg, 20 mg/kg and 40 mg/kg, 8 rats in each group.METHODS: After anesthetized, the cranium was opened to expose cerebral cortical record region. BL-410 biofunctional experimental system was used to record brain electricity bilaterally and epileptic discharge of cerebral cortex in penicillin-induced epilepsy group and ligustrazine groups of 10 mg/kg,20 mg/kg and 40 mg/kg. In the control, 1 hour after anesthesia and craniotomy, cerebrum was collected. In penicillin-induced epilepsy group, 1 hour after induction, cerebrum was collected. In ligustrazine groups of 10 mg/kg,20 mg/kg and 40 mg/kg, after penicillin-induced epileptic discharge was stable, ligustrazine of 10 mg/kg, 20 mg/kg and 40 mg/kg was injected abdominally successively, and cerebrum was collected when the most remarkable inhibition was achieved. Brain tissue section was prepared separately, with HE staining, the observation was done under optic microscope.MAIN OUTCOME MEASURES: Structure changes in cerebral cortical neural cells in rats of each group.In the control, the morphological structure of cerebral cortical neural cell alternations on cerebral cortical neural cell structure, karyopykosis, plasmarrhexis and vacuolar structure, but there was no Nissel bodies in cytomarrhexis, vacuolar structure and decreased Nissel bodies in cytoplasm with the control, there were decreased vacuoles in neural cell, increased cytoplasm and few Nissel bodies in cytoplasm and cell structural morpholcontrol, karyon was big, round and light stained; clot-like Nissel bodies were visible and cell structural morphology was in tendency to be normal.CONCLUSION: In penicillin-induced epilepsy, morphological structure of cerebral cortical neural cell in rats is abnormal. Tetramethylpyrazine of various dosages may improve at different degrees morphological structure of cerebral cortical neural cell, especially significantly at high dosage, by which, its inhibition on epileptic discharge in rats is achieved.
7.Analysis of current situation and influencing factors on the knowledge, attitude and practice of creative credit system in a medical university

Objective To understand the situation of college students' awareness about innovative credit, analyze the influencing factors, in order to provide a scientific basis for constructing innovation credits and promoting the double strategy. Method The undergraduates of a medical university who were still at school in April 2015 were selected, and by using a stratified cluster sampling method, 450 students were chosen to be conducted by self-administered questionnaire survey to understand the cognitive status of in-novation of credit system in college students. Chi square test was used to compare the passing rates of students in different demographic characteristics, and the relationship between scores was analyzed by linear regression analysis. Result A total of 439 valid questionnaires were collected , and the total scores of innovative knowledge, attitudes and behaviors of 439 undergraduates were 15.95% (70/439), 36.44% (160/439) and 32.80%(144/439) respectively. In the part of knowledge innovation credits, girls' passing rate was higher than boys ( χ2=4.010,P=0.045. In the part of attitude, high grade group students had more positive attitude than the low grade group of students ( χ2=6.227,P=0.0013). In the part of behavior; higher grade students with innovative credits had higher pass rates than those in lower grades (chi, 2=7.781, P=0.005), and boys had a higher rate of passing than girls ( χ2=6.658,P=0.010). The total score of knowledge was positively linear with the total score of attitude and behavior. Conclusion College Students' awareness of innovation credit rate is low, but the innovative attitude and behavior is positive. The higher the awareness rate is conducive to the cultivation of attitude and behavior, so it is necessary in medical colleges to carry out innovation of credit and promote the cultivation of innovative consciousness.
8.Multifocal electroretinogram and optical coherence tomography characteristics of macular edema secondary to retinal vein occlusion

Objective To investigate the characteristics of multifocal electroretinogram (mf-ERG) and optical coherence tomography (OCT) in macular edema (ME) secondary to retinal vein occlusion (RVO).Methods Mf-ERG and OCT were performed in 38 patients (38 eyes) of ME secondary from RVO patients and 23 normal persons (23 eyes) in the same period of the control group.OCT was used to detect macular retinal thickness (CMT) at foveal area with diameter 1 mm,mf-ERG was used to evaluate the function of macular retina and analyze the amplitude density of P1 wave and latency of P1,N1 wave from ring 1 to ring 5.Results Compared with the control group,the amplitude densities of P1 in BRVO group were obviously decreased in ring 2 (35.95 ± 17.04)nV · deg-2,ring 3(24.72 ±8.32)nV · deg-2,ring 4(19.28 ±6.38)nV ·deg-2,ring 5 (13.49-± 6.16) nV · deg-2,respectively (all P < 0.05);The latencies of P1 were prolonged significantly in ring 2 (35.74-± 3.72) ms (P < 0.05);The latencies of N1 were prolonged significantly in ring 2 (18.43-± 4.63) ms,ring 3 (18.67-± 2.86) ms (both P < 0.05).Compared with the control group,the amplitude densities of P1 in CRVO group were obviously decreased in ring 1 (81.58 ± 43.15) nV · deg-2,ring 2 (33.71 ±9.81) nV · deg-2,ring 3 (22.15 ± 9.75) nV · deg-2,ring 4 (16.65-± 6.38) nV · deg-2,ring 5 (14.18-± 4.59) nV · deg-2,respectively (all P < 0.05);The latencies of P1 were prolonged significantly in ring 2 (37.56 ± 5.55) ms,ring 3 (37.31 ± 5.22) ms,ring 4 (35.71 ±5.63) ms,ring 5 (37.30 ± 5.37) ms (all P < 0.05);The latencies of N1 were prolonged significantly in ring 1 (21.82 ±5.76) ms,ring 2(19.18 ±4.82) ms,ring3 (19.31 ±4.25)ms,ring 4 (19.05 ± 4.55) ms,ring 5 (19.43 ± 4.12) ms (all P < 0.05).There was negative correlation between CMT and amplitude densities of P1 of ring 1 in CRVO group (r =-0.576,P < 0.05).Conclusion OCT combined with mf-ERG can comprehensively reflect the change of retinal fumction and structure in ME secondary to RVO.
9.The relationship between problem behavior and neurotransmitter deficiency in adolescents.

This study examined the association of problem behavior with neurotransmitter deficiency in adolescents, which would provide new insights into behavioral problems. A total of 1259 students of the seventh grade from 4 middle schools in Wuhan city located in the central China were recruited. With the approval of school and parents, they were invited to complete the Youth Self-Report (YSR) questionnaire and Symptom Scale of Neurotransmitter Deficiency (SSND) questionnaire. Pearson's bivariate correlation analysis showed that the correlation coefficients between each subscale of YSR and SSND ranged from 0.24 to 0.61 with all P<0.01. Canonical correlation analysis indicated that anxiety/depression was interrelated with insufficiency of GABA and 5-HT; aggressive behavior was associated with inadequate GABA; famine of DA influenced the attention problems. It was concluded that neurotransmitter deficiency may cause a series of behavioral and mental problems.
10.Effects of IL-1β on expression of AQP4 and its role in attack of seizure

Objective To observe the effects of interleukin-1β (IL-1β) and pentylenetetrazol (PTZ) induced acute epilepsy and the dynamic expression of aquaporin-4 (AQP4) in hippocampus. To explore the role of IL-1β in the pathogenesis of epilepsy by regulating AQP4. Methods All rats were randomly divided into control group, IL-1β group, PTZ group, IL-1ra + PTZ group and dexamethasone + PTZ group. Observe the behavior of the rats within 60 minutes after injection and record seizure score in each group. Then immunohistochemistry and RT-qPCR were used to detect the expression of AQP4 at at 6 , 12, 24 and 36 h. Results Almost of rats in IL-1β group and PTZ group showed severe degree seizure. The rats in control group and dexamethasone + PTZ group showed no obvious seizure. The seizure of rats were more remarkable serious in PTZ group than that in the IL-1ra + pentylenetetrazole group (P < 0.05). Immunohistochemistry and RT-qPCR Show: the expression of AQP4 in hippocampus in PTZ group increased gradually after 12 h (P < 0.05), then reached in the peak after 24 h (P < 0.001). The expression of AQP4 in IL-1ra + PTZ group was lower compare with PTZ group in each time (P < 0.05). Although the expression of AQP4 in dexamethasone + PTZ group higher than the control group, it was not significantly different (P < 0.05). Conclusion The proinflammatory cytokine IL-1β break the balance of water in brain and increasing the concentration of extracellular excitatory amino acids or ions by upregulate the expression of AQP4 in order to promote the excitatory of neurons.
