1.Effect of two kinds of drainage operation on the short-term curative effect, mortality and complications risk in patients with single giant bacterial liver abscess

Objective To investigate the effect of two kinds of drainage operation including incision and drainage of abdominal operation and percutaneous puncture and drainage operation under ultrasound guidance on the short-term curative effect, mortality and complications risk in patients with single giant bacterial liver abscess. Methods Eighty patients with single giant bacterial liver abscess were selected, and all patients were treated with conventional therapy. Among them, 40 patients underwent incision and drainage of abdominal operation (control group), and 40 patients underwent percutaneous puncture and drainage operation under ultrasound guidance (observation group). The short-term curative effect, operation time, time of returning to normal body temperature, time of returning to normal white blood cell, hospitalization time, mortality and incidence of postoperative complication were compared between 2 groups. Results There were no statistical differences in total effective rate and mortality between control group and observation group: 92.5%(37/40) vs. 97.5%(39/40) and 5.0%(2/40) vs. 7.5%(3/40), P>0.05. The operation time, time of returning to normal body temperature, time of returning to normal white blood cell and hospitalization time in observation group were significantly lower than those in control group:(44.52 ± 5.68) min vs. (98.59 ± 12.70) min, (3.25 ± 0.74) d vs. (4.86 ± 1.32) d, (5.20 ± 1.35) d vs. (7.42 ± 1.79) d, (16.94 ± 3.61) d vs. (25.52 ± 5.28) d. The incidence of postoperative complication was significantly lower than that in control group:10.0%(4/40) vs. 27.5%(11/40), and there were statistical differences (P<0.05). Conclusions Two kinds of drainage operation in the treatment in patients with single giant bacterial liver abscess possesses the same clinical efficacy for short-and long-term curative effect; but percutaneous puncture and drainage operation under ultrasound guidance application can efficiently alleviate the clinical symptoms and signs, accelerate the recovery process of disease and be helpful to reduce the risk of postoperative complications.

In this study,we constructed 2 eukaryotic expressing plasmids namely pcS2?S and pFP,encoding HBV preS2+S envelope protein and human IL 2/IFN ? fusion protein, respectively. The double plasmid fusion protein was used as an adjuvant in preparation of a treatment type HBV gene vaccine. To investigate its feasibility for use as a therapeutic DNA vaccine, we've evaluated the immune response after injection into healthy mice, HBV transgenic(Tg) mice, New Zealand rabbits and Rhesus monkeys. 3.CELLULAR IMMUNE RESPONSE INDUCED BY DNA VACCINE IN BOTH HEALTHY AND HBV TRANSGENIC MICE

The peptides from HBV cytotoxic T lymphocyte(CTL) epitope were used to either stimulate or impact the immune effector (E) or CTL target (T) cells respectively, in order to investigate the cellular immune response induced by DNA vaccination in both healthy and HBV transgenic (Tg) mice. It was found that HBV DNA based immunization could induce CTL activity in healthy BALB/c mice, with intensity correlated with the E/T ratio and IFN ? secretion level in its supernatants. 4.Efficacy of high-dose ambroxol in elderly patients with aspiration pneumonias

Objective To investigate the efficacy of high-dose ambroxol in elderly patients with aspiration pneumonias. Methods Totally 62 patients aged 65 years and over with aspiration pneumonia were enrolled into 2 groups:conventional-dose group intravenously given ambroxol 60 mg/d (n=30) and high-dose group received 270 mg/d (n=32).The times of remission and fever disappearance,rate of adverse effects,average duration of hospitalization,rates of mortality and recurrence within 2 months were observed. Results There were no significant differences in clinical charecteristics between the two groups.The times of remission and fever disappear ance and average duration of hospitalization were lower in high-dose group than in conventional-dose group [(3.2±0.6)d,(54.2±19.5)h,(12.7±4.1) d vs.(3.8±1.1)d,(66.5±18.4)h,(13.5±3.1)d,t=2.11,2.36,2.04,all P<0.05].No differences were found in rates of mortality and recurrence within 2 months between the two groups (6.3%,3.1% vs.10.0%,13.3%,x2=0.01,0.87,both P>0.05).Adverse effects did not appear in the two dose groups. Conclusions High-dose ambroxol is efficient and safe for aspiration pneumonia in elderly patients.

Objective To investigate the roles of Arsenic Trioxide to the lupus nephritis in MRL/ lpr mice and the effect on the expression of TGF-pl in the renal tubule. Methods 45 MRL/lpr mice were chosen for such experiment, then separated in 3 different groups, including arsenic trioxide ( ATO) group, cyclophosphamide (CYC) group and sodium chloride (NS) group. Urine of these mice was kept after the treatment to detect the urine protein levels. The pathological changes and the expression of IgG were observed , and the complement 3 of the nephridial tissue as well as the TGF-pl were detected by immunohisto-chemistry. Result The levels of urine protein in ATO group and CTX group were lower than NS group after the treatment ( P <0.05). The expression of IgG along the glomerular mesangium and capillary loop in ATO and CTX groups was less than NS group ( P <0.05). The expression of C3 had no significant difference within three groups ( P > 0.05). The glomcrulus spherulous cells and the integral of activity in ATO group and CTX group were less than that in NS group ( P <0.05). TGF-β1 wildly presented in the epithelial cells endochylema of the nephric tubule, but it was rarely seen in glomcrulus in NS group. The expression in the nephric tubule of ATO group was less than NS group (20.28 ± 1. 90 vs 68. 23 ± 2. 87, P < 0.01) , and the expression in the nephric tubule in CTX group was less than that in NS group (23. 26 ± 1.71 vs 68. 23 ±2. 87, P <0. 01). Conclusion TGF-pl may take part in the immunopathogenesis of lupus nephritis in MRI/lpr mice. ATO can relieve the inflammation of nephric tubule by down regulation the expression of TGF-β1. ATO can also relieve the tissue damage of kidney in lupus nephritis through reducing the inflammation of glomcrulus and interstitial substance,as well as the expression of the immunocomplex.

8.Studies on ultra-dry storage of Salvia miltiorrhiza seeds

Objective To study the optimum ultra-dry method and moisture at different storage time for Salvia miltiorrhiza seeds and find the principle of storability.Methods S.miltiorrhiza seeds were dried by silica gel at room temperature and by the oven at constant temperature 50 ℃ to obtain various moisture content before stored sealed at room temperature.The optimum ultra-dry method and the optimal moisture were evaluated by measuring the germination rate,germination tendency,and vigor index,etc.Soluble sugar and MDA content were measured to investigate the seed storability.Results Desiccation by silica gel was more proper than by oven;ultra-dry storage of seeds has obvious advantages at the early stage,but with the prolong of the storage time,the advantages decreased;The optimal moisture for S.miltiorrhiza seeds storage at room temperature is about 7.5%;Seed storability is closely related to soluble sugar content in the seeds.Conclusion S.miltiorrhiza seeds can be ultra-dry stored to preserve germplasm resources.
9.Expression of specific proteins of neural cells in rat's cultured amniotic epithelial cells

BACKGROUND: It has been suggested that amniotic epithelial cells (AECs) express almost all of the markers of neural cell and secret a lot of neurotrophic factors and neurotransmitters. If AECs could substitute neural cells, its neurotrophic effect will bring promising prospect in treating neuron injuries and degenerative neural disease.OBJECTIVE: To detect specific proteins of neural cells in rat's cultured AECs.DESIGN: Repeated measurement design.SETTING: Second Clinical Medical College , Jilin University; Department of Histology & Embryology, School of Basic Medical Science, Jilin University.MATERIALS: This experiment was conducted at the Department of Histology & Embryology, School of Basic Medical Science, Jilin University from October 2004 to October 2005. The rat amniotic epithelial tissue was mechanically peeled from an embryonic 12 to 14days Wistar rats. Mouse anti Nestin was purchased from Chemicon Co.,and anti-ChAT rabbit anti-NSE and anti-NT-3 antibodies from Wuhan Boshide Company. Mouse anti-Musashi antibody was donated by Pro.Okano.METHODS: AECs were dissociated and purified from the amnion of pregnancy 12-14 day rats. AECs were treated with trypsin for 5 minutes,then cultured in DMEM/F12 medium at a humidified atmosphere of 0.05 volume fraction of CO2 in air at 37 ℃. Cells were inoculated at a concentration of 5×109 cells/L in culture flask. After 3 days, cells were inoculated onto poly-lysine-treated 35 mm culture Petri dish at a density of 1 × 108 cells/L for immunocytochemically staining. The cells were fixed with 40 g/L paraformaldehyde for 20 minutes. Immunocytochemical staining method was used to detect the expression of microtubule-associated protein-2 (MAP-2),neuron specific enolase(NSE), glial fibrillary acidic protein (GFAP) and choline acetyl transferase(ChAT).MAIN OUTCOME MEASURES: ① Morphological observation of rat'AECs at different culture time. ② Expression of specific protein of neural cells in rat' cultured AECs.RESULTS: ① After cultured for 24 hours, the AECs were flat and presented fibroblast-like morphology. 3 to 5 days later, cell bodies were well stacked; AECs had a big and round nucleus and were connected with each other by flourishing dendrites. ② Immunocytochemical staining results after culture for 4 days showed that AECs expressed Nestin, ChAT,NSE, Musashi, MAP-2, GFAP.CONCLUSION: AECs are homologous to neural cells in morphology, and it may be a new cell source to treat nervous system disease.
10.Construction, expression and immunological protection of DNA vaccine of pVAX1-oipA of Helicobacter pylori

Objective:To investigate the effect of DNA vaccine expressed Helicobacter pylori(Hp) oipA on protecting against Hp infection.Methods:The ORFs of Hp oipA had been inserted into the eukaryotic expressing vector pVAX1 and SGC-7901 cells had been transfected with recombinant plasmid pVAX1-oipA. The expression of oipA had been detected by RT-PCR and ELISA. After extracted and purified, pVAX1-oipA had been injected into BALB/c mice through muscles of right leg one time each week for three weeks(100 ?g each mouse). pVAX1 blank and normal saline had been used as the control groups. Titer of antibodies had been detected by ELISA two months after the last immunization. Based on the confirmation of immunological response in the pVAX1 groups, mice had been given orogastric challenged with live Hp Sydney strain three times(0.5?108/0.5 ml each mouse). Four weeks after challenge, mice had been sacrificed. Histological change and the colonization of Hp in the gastric mucosa had been detected by urease test, the culture of Hp, and electronic microscopy.Results:SGC-7901 cells transfected with pVAX1-oipA had expressed corresponding production at the level of transcription and translation. Immunized mice had been induced anti-oipA antibodies. After challenged with bacterium, as contrast to immunized mice groups injected with pVAX1-oipA, pVAX1 blank, normal saline, the positive rate of urease test of gastric mucosa was 0(0/10), 90%(9/10), 100%(5/5)respectively,the positive rate of cultures of Hp was 20%(2/10), 90%(9/10), 100%(5/5)respectively. Histological findings: the different degree of erosion had been observed in control group, but 80%(8/10)of gastric mucosa were normal in immunized mice.Conclusion:oipA DNA could induce effective immune response in protection against Hp infection.
