1.B-mode Ultrasonic Diagnosis of Complications in Hepatic Hydatidosis

Hydatidosis is the largest parasific disase in human being. The patients often contract hydatidosis in the childhood. The slow growth of hydatid cysts in the organ involved may lead to a protracted course of disease. In the early stage of the diseases, there are no distinct subjecive symptoms. Early diagnosis is difficult by routine examination. The complications of hydatidosis causing serious damage to the organs may often lead to sudden death. It is essential to make an early and correct diagnosis and give treatment. B-mode ultrasonography not only detects the location, dimension and chracteristics of hydatid cysts but also shows the pathological changes of the various complications caused by hydatidosis and faciliates to select the best program of operation. B-mode ulttasonography is the method of choice in the diagnosis of hydatidosis. In this series of 931 patients with hepatic hydatid disease, the diagnostic accuracy rate of B-mode ultrasonography reached 98. 8%.
2.A Decontamination Method for Surgical Instruments

OBJECTIVE To explore an effective cleaning method to improve the quality of sterilization,in order to control infections in the hospitals. METHODS To adopt orthogonal experimental forms of L_9(3~4) and take water temperature,cleaning agent type and cleaning method as experimental factors,in this way to investigate decontamination method for surgical instruments,using TOSI indicator cards and Browne STF load device to test the effectiveness of cleaning results. RESULTS It showed that by KQ-4200SY medical digital full-automatic ultrasonic cleaner with washing water at 33-35℃ an optimal cleaning result would be obtained. CONCLUSIONS It is practical to clean surgical instruments by KQ-4200SY medical digital full-automatic ultrasonic cleaner,and by TOSI indicator cards and Browne STF used as test method.
3.Function and Modulation of Coronary Endothelium in Isolated lschemic-reperfused Rat Hearts

Isolated perfused rat hearts were subjected to regional ischemia followed by reperfusion and then tested for responsiveness to acetylcholine (Ach) and nitroglycerin (NTG). The effects of L-Arginine (L-Arg) and NG-monomethyl-L-Arg (L-NMMA) on the endothelium-dependent (ED) and independent (EI) vasodilator were also observed. It was found that both 15 and 30min ischemia followed reperfusion resulted in a decrease in ED vasodilation of Ach. ED vasodilation gradually renewed after 90min reperfusion in the 15min ischemic group, not in the 30min ischemic group. No significant decrease in EI vasodilation of NTG in all groups. L-Arg accelerated the renewing of ED vasodilation of Ach in the 15 min ischemia-reperfusion group, not in the 30min ischemia-reperfusion group. L-NMMA was an inverse action. The results indicate that brief ischemia followed by reperfusion can induce coronary endothelium dysfunction which may be related to the decrease of endothelium derived relaxing factor.
4.Diagnosis and classification of hepatic echinococcosis by ultrasonography

Objective To make an early and correct diag-nosis of hepatic echinococcosis.Methods A total of 1092 patients with hepatic echinococcosis underwent operation in our hospital between 1984 and 1995. Of these paitents, 427 (39.1%) were cases with complications, including secondary infection, rupture, obstructive jaundice, anaphylactic shock, disseminated implantation with resultant multiple echinococcosis and portal hypertension. The ultrasonic examination has been generally used in clinical practice for comprehensive evaluation which can dectedct the preoperative diagnostic accuracy rate.Results B-mode ultrasonography can be used not only to detect the location, dimension and pathological characteristics of hydatid cyst but also to show the pathological changes of various complications caused by echinococcosis. Seven patterns specific ultrasonic scanning images were revealed. In this series the diagnostic accuracy rate reached 98.8%.Conclusions Ultrasonic examination is harmless to human body, and has been widely used in combination with immunological tests in clinical and epidemiological studies to detect the asymptomatic parasite carriers in early stage and to improve the preoperative diagnostic accuracy rate.
5. Dedifferentiated liposarcoma of extremities: a clinicopathologic analysis

Objective:
To study the clinicopathologic features of dedifferentiated liposarcoma of extremities.
Methods:
Nine cases of dedifferentiated liposarcoma of extremities diagnosed at Beijing Jishuitan Hospital from 2009 to 2017 were selected. The histological features of cases of dedifferentiated liposarcoma of extremities were evaluated by HE and immunohistochemistry, together with the clinical and radiological features. Flourescence in situ hybridization(FISH) was used to detect MDM2 amplification.
Results:
They were located in the thigh (6 cases), calf (2 cases) and buttock (1 case). There were six females and three males. Patients′ age ranged from 61 to 79 years (mean 68 years). Histologically, there were two components, well-differentiated liposarcoma and dedifferentiated sarcoma with or without transitional lesions between them. The histology of dedifferentiated liposarcoma included undifferentiated sarcoma and fibrosarcoma. Heterologous elements such as bone and cartilage were present in two cases. Immunohistochemical study showed the tumor cells expressed vimentin, CDK4 and p16. MDM2 were positive in 6 cases (6/9) and p53 was positive in one case(1/9). CKpan was positive in the epithelioid differentiation area. S-100 protein was positive in the well-differentiated liposarcoma component. FISH showed the amplification of MDM2(6/9).
Conclusions
Dedifferentiated liposarcoma of extremities is very rare. The diagnosis should be combined with the histological characteristics and immunohistochemical results and differentiated from the other tumors and tumor-like lesions.
6.Primary intraosseous Rosai-Dorfman disease: a clinicopathological analysis of fourteen cases

Objective:To investigate the clinicopathological characteristics, histogenesis, immunophenotypes and molecular genetic features of primary intraosseous Rosai-Dorfman disease (RDD) for improving diagnostic accuracy and differential diagnosis.Methods:This retrospective study included 14 RDD cases diagnosed from January 2009 to January 2019 at Beijing Jishuitan Hospital, China. The immunohistochemical staining for S-100, cyclin D1, CD1a and CD207 expression was analyzed. The BRAF V600E and KRAS mutation analyses were performed using the Scorpions amplification refractory mutation system (ARMS) fluorescence quantitative PCR.Results:There were 6 female and 8 male patients, aged from 2 to 64 years (mean 31.4 years). All of the 14 cases occurred in the bone without lymph node disease, while one patient developed additional lesions within vertebra and nasal cavity. Radiographically, the lesions were lytic with sclerotic margins. Histologically, the lesions percolated through the medullary cavity in an infiltrative fashion and alternating hyper- and hypo-cellular regions of histiocytic clusters (seen as alternating dark and light zones at low magnification). Large histiocytes also showed emperipolesis. Some cases had areas of fibrosis and dense lymphoplasmacytic infiltrates. There were vasculitis and an increased number of plasma cells in the cases involving multiple sites. One case showed concurrence of RDD and Langerhans cell histiocytosis(LCH) with inconspicuous increase of Langerhans histiocytes. Immunohistochemical staining showed that the large histiocytes were positive for S-100, CD68 and CD163 in all cases. The nuclear immunoreactivity for cyclin D1 was observed in 13 of the 14 cases. S-100, CD1a and CD207 were positive in the case with concurrence of RDD and LCH. ARMS-PCR results showed that BRAF V600E mutation was observed in the cases with concurrence of RDD and LCH, while there were no KRAS mutations (7/7). Follow-up information was available for 12 patients and ranged from 9 to 49 months. Three of the 12 patients experienced recurrences after the first surgery.Conclusions:Primary intraosseous RDD is rare, and its concurrence with LCH is a very rare phenomenon. Its clinical symptoms, imaging, and pathological manifestations need to be distinguished from other bone lesions. The molecular detection of BRAF V600E and the nuclear expression of cyclin D1 mutations can be used for the diagnosis and differential diagnosis of RDD.
7.Preventive and therapeutic effects and mechanisms of Caryopteris incana in a rat model of primary dysmenorrhea with cold coagulation and blood stasis syndrome

Objective To explore the preventive and therapeutic effects of Caryopteris incana decoction in a rat model of primary dysmenorrhea with cold coagulation and blood stasis syndrome.Methods Sixty healthy specific pathogen-free SD female rats were randomly divided into six groups of ten rats each:normal group,model group,ibuprofen group,C.incana high-dose group,C.incana medium-dose group,and C.incana low-dose group.All groups except the normal group were treated with cold stimulation combined with estradiol benzoate and oxytocin to establish a rat model of primary dysmenorrhea with cold coagulation and blood stasis syndrome.On the fifth day of modeling,the rats were intragastrically administered the study drugs for 10 days.Their symptoms were observed and recorded.The writhing response and hemorheological indices were measured.The serum levels of TXB2,6-keto-prostaglandin F1α(6-Keto-PGF1α),estradiol(E2),and progesterone(PROG)were measured.Additionally,the levels of prostaglandin F2α(PGF2α),prostaglandin E2(PGE2),nitric oxide(NO),and calcium(Ca2+)in the uterine tissues were measured.The organ indices of the uterus and ovary were calculated,and histopathological changes were observed.Results Compared with the normal group,the rats in the model group showed obvious symptoms of cold coagulation and blood stasis syndrome and writhing reaction.The morphology of uterus and ovary showed obvious hyperplasia,inflammation,edema and other lesions.The plasma viscosity,packed cell volume and whole blood viscosity were significantly increased(P<0.01).The serum levels of thromboxane B2 and E2 and the E2/PROG ratio were significantly increased(P<0.01),the levels of 6-Keto-PGF1α and PROG were significantly decreased(P<0.01).The uterine index and ovarian index were significantly increased(P<0.01).The levels of PGF2α and Ca2+and the PGF2α/PGE2 ratio in uterine tissue were significantly increased(P<0.01),while the levels of PGE2 and NO were significantly decreased(P<0.01).Compared with the model group,Caryopteris incana significantly improved the symptoms of model rats,improved the morphological lesions of the uterus and ovary,prolonged the latency time of the writhing reaction,and reduced the number of writhing episodes(P<0.01);significantly reduced the plasma viscosity,packed cell volume,and whole blood viscosity(P<0.01);significantly reduced the serum levels of TXB2 and E2 and the E2/PROG ratio,increased the serum levels of 6-Keto-PGF1αand PROG,and reduced the uterine and ovarian indices(P<0.01,P<0.05);significantly reduced the levels of PGF2αand Ca2+and the PGF2α/PGE2 ratio in uterine tissue(P<0.01,P<0.05);and significantly increased the levels of PGE2 and NO in the uterine tissue(P<0.01).Conclusions Caryopteris incana decoction can effectively improve the clinical symptoms of primary dysmenorrhea in rats with cold coagulation and blood stasis syndrome,and it has a good control effect.Its mechanism may be correlated with the levels of TXB2,6-Keto-PGF1α,E2,and PROG in serum and PGF2α,PGE2,NO,and Ca2+in uterine tissue.
8.Reassessment of EWSR1 gene rearrangement-negative undifferentiated round cell sarcomas in bone and soft tissues by fluorescence in situ hybridization

Objective:To unravel the CIC rearrangement sarcomas and BCOR-CCNB3 sarcomas from EWSR1 rearrangement-negative undifferentiated round cell sarcomas in the bone and soft tissues.Methods:Twenty-eight cases of EWSR1 rearrangement-negative undifferentiated round cell sarcomas of bone and soft tissues, tested for CIC rearrangement and BCOR rearrangement by fluorescence in situ hybridization and related immunostaining were analyzed, and some of the BCOR rearrangement cases were verified by reverse transcription-polymerase chain reaction.Results:Five of 28 (17.9%) tested cases were positive for CIC rearrangement and six (21.4%) for BCOR rearrangement. Histopathologically, CIC rearrangement sarcomas comprised nodular aggregates of round to polygonal cells, containing hyperchromatic nuclei, prominent nucleoli and moderate cytoplasm, with focal variable necrosis and myxoid stroma. BCOR-CCNB3 sarcomas mostly comprised diffusely arranged, round to oval to short spindly cells with angulated nuclei, vesicular chromatin, inconspicuous nucleoli and interspersed vessels. Immunohistochemically, five of six BCOR-CCNB3 sarcomas showed CCNB3 immunostaining, which could be helpful for diagnosis . Two patients with CIC rearrangement sarcoma died of the diseases in seven months and twenty-two months. One patient with BCOR-CCNB3 sarcoma died of the diseases in forty-six months. Conclusions:Overall, 39.3% of the EWSR1 rearrangement-negative undifferentiated round cell sarcomas are CIC rearrangement sarcomas and BCOR-CCNB3 sarcomas. Molecular testing is helpful for diagnosis.
9.Expression and diagnostic value of DDIT3 immunohistochemical antibody in 53 cases of myxoid liposarcoma

Objective:To investigate the diagnostic value of DDIT3 immunohistochemical antibody in myxoid liposarcoma.Methods:A total of 53 patients diagnosed with myxoid liposarcoma and with DDIT3 gene rearrangement detected by fluorescence in situ hybridization (FISH) from 2010 to 2022 in Beijing Jishuitan Hospital were enrolled as the case group, and 50 patients with other tumors that needed to be differentiated from myxoid liposarcoma during the same period were enrolled as the control group. The immunochemical (IHC) expression of DDIT3 was evaluated in all cases and the specificity and sensitivity in the diagnosis of myxoid liposarcoma were calculated. The correlation between IHC DDIT3 expression and FISH detection of DDIT3 gene rearrangement was analyzed.Results:Among the 53 cases of myxoid liposarcoma, thirty-one patients were males and twenty-two were females. The patients ranged in age from 11 to 74 years, with a median age of 51 and an average age of 49.4 years. The expression rate of IHC DDIT3 expression in 53 cases of myxoid liposarcoma was 100%. No expression was found or only scattered individual cell (<10%) expression was seen in a very few cases in the. control group. There was no significant correlation between the proportion and intensity of IHC and FISH DDIT3 positive cells or the partner gene.Conclusions:The sensitivity and specificity of DDIT3 immunohistochemical antibody in the diagnosis of myxoid liposarcoma are both 100%, which are consistent with the results of FISH. It can be used as a convenient and economical diagnostic tool for myxoid liposarcoma.
10.Lipid-lowering effect of propolis in mice with Triton-WR1339-induced hyperlipidemia and its mechanism for regulating lipid metabolism.

OBJECTIVETo evaluate the therapeutic effect of propolis against Triton-WR1339-induced hyperlipidemia in mice and explore the underlying mechanism.
METHODSC57BL/6 mice were randomly divided into 7 groups (=10), including the control group, hyperlipidemia model group, fenofibrate (30 mg/kg) treatment group, and 4 treatment groups treated with low- (30 mg/kg) or high-dose (60 mg/kg) propolis HB01 or HB02. In all but the control group, acute hyperlipidemia models were established by intramuscular injection of Triton WR-1339, and corresponding treatments were administered via gastric lavage for 7 days. After the treatments, blood samples were collected for testing the levels of total cholesterol (TC), triglycerides (TG), highdensity lipoprotein-cholesterol (HDL-C), low-density lipoprotein-cholesterol (LDL-C), malondialdehyde (MDA), superoxide dismutase (SOD), alanine aminotransferase (GPT), and aspartate aminotransferase (GOT); Western blotting was used to detect the expressions of the proteins involved in lipid metabolism in the liver tissues including ABCA1, ABCG8, LDLR, and SR-B1.
RESULTSCompared with the normal control group, the mice with Triton-WR1339-induced hyperlipidemia showed significantly increased levels of TC, TG, LDL, MDA, GPT, and GOT and lowered HDL-C levels and SOD activity ( < 0.05). Treatments with fenofibrate and the 2 propolis at either low or high dose significantly reversed Triton-WR1339-induced changes in blood lipids ( < 0.05), and the effects of propolis were more potent. Triton-WR1339 injection also significantly decreased the expressions levels of ABCA1, ABCG8, LDLR, and SR-B1 in the liver ( < 0.05), and these changes were obviously reversed by treatments with fenofibrate and propolis ( < 0.05), especially by the latter.
CONCLUSIONSThe lipid-lowering effects of propolis are mediated by improving lipid metabolism and regulating the expressions of lipid transport proteins in the liver tissue.
