1.Concentration of protoporphyrin Ⅸ in cancer tissues and blood in patients with colorectal cancer at early stage

Objective To determine the concentration difference in protoporphyrin in cancerous intestine and to discuss its application in laser-induced autofluorescence diagnosis for colorectal cancer at early stage.Methods We detected the concentration of protoporphyrin Ⅸ in 30 patients with colorectal cancer and 30 control patients, as well as that in 60 cases of intestinal tissues (30 control tissues and 30 cancer tissues).Results The concentration of blood protoporphyrin Ⅸ in patients with colorectal cancer was significantly higher than that in the controls (P<0.05). Protoporphyrin Ⅸ concentration in the cancer tissue was significantly higher than that in the control tissues (P<0.05).Conclusion That the concentration of protoporphyrin in cancerous intestine becomes abnormally high may be the material basis for spectrum intensity peak of (644.3±5.7)nm in laser-induced autofluorescence diagnosis for colorectal cancer at early stage.
2.Management of hypogastric artery in endovascular repair of abdominal aortic aneurysm

Objective To summarize our experiences of management for hypogastric artery in endovascular repair(EVR) of infrarenal abdominal aortic aneurysm(IAAA).Methods From July,1997 to March,2007,62 cases of IAAAs required special management of the internal iliac artery(IIA) during the EVR.Among them,57 cases were type C AAA who included 35 cases of unilateral common iliac artery(CIA) bifurcations and 22 cases of bilateral iliac bifurcations.The remaining 5 cases involved superior iliac bifurcations.Various techniques including simple coverage by stent-graft,embolization of the IIA trunk combined with stent-graft covering,reconstruction of the IIA and combination of the above techniques were applied according to the different conditions of the IIA involved.Results None of the 62 patients required conversion to surgical repair.Immediate post operational angiography demonstrated type Ⅰ endoleak in 6 cases(9.7%).Claudication due to gluteus ischemia occurred in 5 cases(8.1%),leg numbness in 1 case(1.6%),and severe constipation in 1 case(1.6%),but no buttock and colorectal necrosis was recorded.Conclusion It is important to preserve one of the hypogastric arteries and avoid occlusion of both hypogastric arteries in EVR of IAAA.
3.Biological Characteristics of the Human Hepatocellular Carcinoma Cell Line Transfected with Interleukin-l?Converting Enzyme (ICE) Gene

We used retroviral vector pLXSN to construct recombinant retroviral vectors with the human apoptosis gene, interleukin-l? converting enzyme (ICE). The vectors were introduced into packaging cell line PA317 by electroporation method. The G418 resistant colonies were selected, and the supematants of the colonies were used to infect the human hepatocellular carcinoma cell line SMMC7721. G418 resistant colonies of SMMC7721 were named SMMC7721-MCE and SMMC7721-neo. The results of RT-PCR analysis showed that exogenous hICE gene had successfully integrated into the genome of SMMC7721-hICE cells. The proliferation rate and tumorigenicity of cells in nude mice were examined. Our data showed that the growth rate and the tumorigenicity of SMMC7721-hICE cells in nude mice were considerablely decreased comparing with parent SMMC7721 and SMMC7721-neo. These results suggested that the retroviral vector expressing hICE gene was successfully constructed and could suppress the growth ability and tumorigenicity of human hepatocellular carcinoma cells, which provided a basis for further investigation of hICE gene therapy.
4.Polymorphisms in The 5' flanking Region of IGF-Ⅰ Gene are Associated With Cashmere Fibre Traits in Liaoning Cashmere Goats

Insulin-like growth factorⅠ(IGF-Ⅰ) is essential for the growth and differentiation of hair follicles which is an important part of wool and cashmere.But there is no report on polymorphisms of the IGF-Ⅰ gene in cashmere goat,and also few candidate genes for cashmere production traits have been reported in cashmere goats.The objectives of this work were to detect the single nucleotide polymorphism(SNP) in the 5' flanking region of IGF-Ⅰ gene and to determine their association with fibre traits in Liaoning cashmere goats.The fibre traits data investigated in the experiment were combed cashmere weight,cashmere fibre length and cashmere fibre diameter.A few individuals of the Liaoning cashmere goats,selected according to phenotypic character,were used for SNPs detection in the 5'flanking region of IGF-Ⅰ gene,and four point mutation G→C(388 bp),A→G(668 bp),A→ C(719 bp),G→A(752 bp) were identified,which result in a CdxA bonding-site lack,and score increase of C/EBP site in the 5' flanking region compared with the wild type.Then different genotypes were detected in 520 Liaoning cashmere goats using create restriction site(CRS) and polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),three genotypes,AA,AB and BB were observed coded for by two different alleles A and B for each SNP,and 13 diplotypes were identified in the groups of the four SNPs.The relationships between the genotypes and combed cashmere weight,cashmere fibre length and cashmere fibre diameter were analyzed.The statistical analysis showed that animals with the genotype AA of SNP2 in Liaoning cashmere goats had the thinnest cashmere fiber diameter compared with those with the AB and BB genotype(P
5.Effect of water storage on the flexural properties of three dual-cure core buildup composite resins

BACKGROUND:Hydrothermal condition of oral environment has an effect on the mechanical properties of dental core materials.
OBJECTIVE:To evaluate effect of water storage on the weight change rates and flexural properties of three dual-cure core buildup composite resins.
METHODS: Rectangular specimens (25 mm×2 mm×2 mm) were prepared from three dual-cure core buildup composite resins (LuxaCore Smartmix Dual, Para core and Clearfil DC Core). Specimens were stored in distiled water at 37℃ for 0, 1 day, 1, 2, 3 weeks and 1 month, respectively, weighted and then subjected to three-point bending test in a universal testing machine.
RESULTS AND CONCLUSION: Weight change rates of the three composite resins increased with immersion time; at the same immersion time, weight change rates of three composite resins ranked as the folowing order: LuxaCore Smartmix Dual > ParaCore > Clearfil DC Core. The flexural strength of LuxaCore Smartmix Dual decreased with immersion time. The flexural strength of ParaCore showed no significant change during 3 weeks immersion (P > 0.05), while it showed a significant decreasing after 1 month immersion (P < 0.05). There was no significant change on the flexural strength of Clearfil DC Core in the whole immersion process(P > 0.05). The flexural modulus of LuxaCore Smartmix Dual showed a decreased trend in the whole immersion process, but it showed that there was no significant change during the first 2 weeks(P > 0.05), while a significant decrease occurred after 3 weeks and 1 month (P < 0.05). Compared with original specimens, the flexural modulus of ParaCore and Clearfil DC Core showed a decreased trend, but this change was not significant (P > 0.05); the flexural properties of three composite resins showed a fluctuant decreased trend along with weight changes increased. To sum up, Clearfil DC Core showed the best water resistance among three composite resins.

7.Effect of adenosine on perilymph glutamate concentration and compound action potentials in guinea pigs exposed to white noise

Objective:To investigate the protective effect of adenosine on cochlea in acute acoustic trauma. Methods: Guinea pigs were exposed to 118 dB SPL white noise for 30 min. During the process of exposure, artificial perilymph or artificial perilymph containing adenosine (0.1 mmol/L,1 mmol/L or 5 mmol/L) was infused into the cochlea through perilymphatic perfusion. Concentration of glutamate in perilymph and thresholds of compound action potentials (CAP) were measured before and after noise exposure. Results: After exposure, the concentration of glutamate and the mean threshold shifts evoked by click were significantly higher than those of before exposure. Concentration of glutamate and thresholds of CAP in adenosine (1 mmol/L and 5 mmol/L) treated groups were significantly lower than those of the control group. Conclusion: Adenosine can inhibit the elevation of glutamate and decrease the threshold shift of CAP after exposure to noise,which can protect cochlea in acute acoustic trauma in guinea pigs.
8.Content Determination of Aciclovir Cream by Centrifugal Partition Chromatography

OBJECTIVE:To establish a method for the content determination of Aciclovir cream. METHODS:Centrifugal parti-tion chromatography(CPC)was conducted. The solvent system was hexane-acetonitrile-water(2∶1∶1,V/V/V),the injection vehicle was an aqueous solution of 5%Tween 80 and the volume was 1.0 ml;the flow rate was 5 ml/min;the wavelength was 254 nm. RE-SULTS:There was a good linear relationship between quality concentration and peak area in the range of 0.012 7-0.126 7 mg/ml (r=0.998 7). The RSD of precision,stability and reproducibility tests was all no more than 2.0% and the average recovery was 97.34%(RSD=0.90%,n=9). CONCLUSIONS:The method is with high precision and accuracy,and can be used for the content determination of principal components of Aciclovir cream.
9.Clinical characteristics of 277 cases with dengue fever in Guangzhou during an epidemic of 2006

Objective To explore the clinical and laboratory characteristics of patients with dengue fever(DF)in Guangzhou from July to September in 2006 and to improve the awareness of dengue virus infection for physicians.Methods Clinical data of 277 patients with DF admitted to our hospital in Guangzhou were retrospectively analyzed.Dengue virus was isolated from patients' blood specimens by C6/36 cell culture and typed by reverse transcriptase-polymerase chain reaction(RT-PCR)assay and gene sequencing.Results Age of the 277 patients,148 males and 129 females,averaged 36 years with a standard deviation of 17 years,ranging from 9 months to 78 years.Their most common symptoms included fever(100.0%),skin rash(85.2%),headache(69.3%),myalgia(51.3%),bone soreness (39.0%)and nausea or vomiting(39.4%),and leucopenia and/hromboeytopenia occurred in 75.5% and 62.5% of them,respectively.Elevated alanine aminotransferase(ALT)was found in 59.6%,elevated aspartate aminotransferase(AST)in 81.6%,and hypopotassemia in 53.8% of them.Serum positivity of antibody against dengue virus IgM was 90.6%,which could be detected 3 to 10 days after onset of the illness,with a mean of 6.2 days with a standard deviation of 1.9 days.Cell-cultured virus from 20 patients were identified as typed by RT-PCR and RNA sequence analyses.All patients were clinically classified as typical DF and all of them completely recovered.Conclusions DF during this epidemic in Guangzhou was caused by dengue virus-1,with typical manifestations in all the patients but more damage in the liver.Some patients demonstrated hypopotassemia,but no dengue hemorrhagic fever or dengue shock syndrome was found.All the patients experienced a good prognosis with timely diagnosis and treatment.
10.Expression of human long-acting FSH in CHO cell and its bioactivity in vivo.

Follicle-stimulating hormone (FSH) is a pituitary glycoprotein hormone that is essential for the development of ovarian follicles and testicular seminiferous tubules. The relatively short half-life of FSH in vivo requires daily injections for more than 10 days that is inconvenient and possibly contribute to the stress perceived by the patients. The goal of the present study was to increase FSH glycosylation, in order to develop a long-acting recombinant FSH. The cDNA of native alpha and beta subunit of human FSH was linked by a sequence with two N-linked glycosylation sites, and the resulted DNA was inserted into pcDNA3.1 vector to generate a recombinant vector of pcDNA3.1-FSH. The pcDNA3.1-FSH was linearized and transfected into CHO-K1, positive transformants were selected by G418 and confirmed by PCR and Western blotting. A single chain recombinant FSH was expressed, with molecular weight of about 49 kDa. The recombinant FSH expression level in CHO-K1 cell strain in serum-free culture was 3 mg/L. Single injection of this recombinant FSH could induce folliculogenesis and ovulation in rats, the efficacy was similar with the commercially available FSH preparation (Folltropin-V) administrated 8 times consecutively. The results suggested a long-acting FSH was produced successfully.
