1.Characterization and clinical significance of the Smith antigen

Systemic lupus erythematosus(SLE) is an autoimmune disease characterized by the production of autoantibodies against a wide spectrum of nuclear, cytoplasmic,and cell membrane autoantigens.Among them,the anti-Smith antibodies are specific for SLE and thus have been included as a criteria for diagnosis.They are not only a diagnostic marker but also a reliable measure of disease activity in SLE,we intend to describe the biological functions and clinical significance of the Smith antigen and the detection of anti-Smith antibodies.
2.The opportunity choice of surgical procedure for deep venous thrombosis of lower extremity

10 days.All the patients received operations.Results:The prognoses of 80.49% patients in the early and middle stages were well ,the fadeaway of edema was evident,on the contrary,the prognoses of 83.33% patients of the later stage were worse,the fadeaway of edema was slow.Conclusion:The patients of LEDVT should receive operation as early as possibly in 10 days,the patients should receive conservative therapy after 10 days.
3.Detection of alpha-fetoprotein messenger RNA in peripheral blood of patients with hepatocellular carcinoma

Objective:To determine whether alpha-fetoprotein(AFP) messenger RNA(mRNA),a specifically hepatocyte-expressed gene,could be a marker of metastasis in hepatocellular carcinoma. Methods:Peripheral blood samples were obtained from 30 patients with hepatocellular carcinoma (HCC).A sensitive reverse transcription polymerase chain reaction( RT-PCR) technique was used to detect the human AFP mRNA in the blood.Results:Of the 30 HCC patients,14(46.7%) patients had positive AFP mRNA.The presence of AFP mRNA correlated with the TNM clinical stages,the size of tumor,extrahepatic metastasis or with portal vein carcinothrombosis(P0.05).Conclusion:The positive of AFP mRNA in the peripheral blood can indicate the presence of hematogenous metastasis in patients with HCC.
4.Effect of vascular endothelial growth factor-C on invasive capability and proliferation of human cholangiocarcinoma cells

Objective: To investigate the effect of vascular endothelial growth factor-C (VEGF-C) on invasive capability and proliferation of human cholangiocarcinoma cells. Methods: The effect of VEGF-C on FRH0201 was assayed by MTT. The cycle pattern and apoptosis was assayed using flow cytometry. The effect of VEGF-C on homotypic adhesion and metastasis in FRH0201 was examined with 3H-TdR infiltration and Boyden chamber. Results: VEGF-C could enhance the proliferation of FRH0201 in a dose and time dependent manner. And VEGF-C could inhibit cell apoptosis significantly. After cultured for 2 hours with 1, 5, 10ng/ml of VEGF-C, there were more cells in the lower chamber than the control group. After 60, 90, 120 minutes induction by 1ng/ml、5ng/ml、10ng/ml, the cells showed significantly lower homotypic adhesion than that of the control group. Conclusions: VEGF-C could enhance the proliferation of FRH0201 and inhibit cell apoptosis. VEGF-C could decrease homotypic adhesion of FRH0201 and might be a cause of the metastasis.
5.Effect of antisense oligodeoxynucleotide targeting survivin on hilar cholangiocarcinoma cell line FRH-0201

Objective To investigate the effect of antisense oligodeoxynueleotide targeting sur-vivin (survivin ASODN) on hilar cholangioearcinoma cell line FRH-0201 depressing the expression of survivin. Methods Survivin ASODN was transfected into hilar eholangiocarcinoma cell line FRH-0201by liposome. Morphologieal changes were observed by inverted phase contrast microscope. RT-PCR and Western blot methods were performed to detect the expressions of survivin mRNA and protein re-spectively. The changes in cell apoptosis were detected by flow cytometry. Results The expression of survivin mRNA and protein was significantly decreased in survivin ASODN group than that in the con-trol group(mRNA: 0.51-t-0. 03 vs 0. 82-t-0.02,P%0. 05~protein: 1.82-t-0.16 vs 3. 08--t-_0. 27, P-Q 0.05). The morphologieal apoptotic changes were observed and the apoptosis rate was increased (11.50+1.49% vs 0.39-+-0.08~, P%0.05). Conclusion Survivin AS()DN can induce hilar cholan-gioeareinoma cell line FRH-0201 into apoptosis by decreasing the expression of survivin.
6.Effect of cholecystokinin on intestinal interstitial cells of Cajal and small intestinal motor function in acute pancreatitis rats

Objective To investigate the effect of cholecystokinin (CCK) on rat intestinal interstitial cells of Cajal (ICC) and the small intestinal motor function during experimentally induced acute pancreatitis (AP).Methods 20 male SD rats were divided with a random number table into model group (n =15) and control group (n =5):the 15 rats in the model group were further divided into 3 subgroups,i.e.0,20,40 μg/kg CCK sub-groups.L-omithine was intraperitoneally injected to induce pancreatitis,while normal saline was injected in the control group.24 hours after establishing AP model,CCK was administered intraperitoneally every day for 5 consecutive days.The rats were sacrificed by cervical dislocation 30 minutes after gavage with 2 ml ink.Length of the small intestine which have become black and that of the whole small intestine were measured.Intestinal sections were studied by confocal microscopy after immunofluorescence with specific antibodies.The number of positive cells was compared among the groups.Results In the control group,the number of ICC in the myenteric plexus and deep muscular plexus was (21.16 ± 3.19) /field and (17.20 ± 1.75) /field,respectively;in the AP group was (5.00 ± 1.05) /field and (4.52 ± 1.05) /field,respectively;in the AP + CCK20 group was (10.76 ± 2.09) /field and (9.84 ± 1.68) /field,respectively;in the AP + CCK40 group was (13.72 ±2.97) /field and (12.40 ± 1.81) /field,respectively;the number of ICC in the AP group were significantly lower than those in the control group (P =0.001).The ratio of transmission distance in small intestine in the control,AP,AP + CCK20,and AP + CCK40 groups was 0.71 ± 0.05,0.54 ± 0.07,0.68 ± 0.10,and 0.74 ± 0.07,respectively.The intestinal motor function was reduced in the AP group,demonstrated by shortened transmission distance,which was improved after the administration of CCK (P =0.003).Conclusions Pancreatitis can cause ICC damage,reduce ICC number,destroy ICC network and the small intestinal motor function.CCK can mitigate the damage to ICC and protect the integrity of ICC network,thereby improving intestinal motor function.
7.Clinical Observation on Treating Menopause Syndrome with Acupuncture Therapy

Objective:To study the clinical effect and the endocrine mechanism of acupuncture therapy on menopause syndrome.Method:There were 47 cases with menopause syndrome,which were divided into two groups,32 cases in the acupuncture group,treated by acupuncture with main acupoins of Shenshu (BL 23),Zusanli (ST 36) and Sanyinjiao (SP 6),and 15 cases in the control group,treated by orally taking the Oryzanol.The Kupperman index of the two groups was observed before and after treatment for evaluating the therapeutic effect.The changes of blood-serum F2,T,FSH,LH of the 25 cases in the acupuncture group were tested by radioimmunoassay before and after the treatment.Results:The acupuncture therapy can relieve the clinical symptoms of menopause syndrome,and its therapeutic effect was better than Oryzanol.After being treated by acupuncture therapy,the content of blood-serum E2 obviously increased,while the contents of LH and T declined,and the FSH also had a tendency to lower,but it was not statistically significant.Conclusion:The therapeutic effect of acupuncture therapy for menopause is confirmed.The acupuncture therapy can improve the function of hypophysial-gonadal axis of the patient with menopause syndrome,which may be the main mechanism of the acupuncture therapy for treating menopause syndrome.
8.Involvement of mitochondrial pathways of apoptosis in hippocampus apoptosis in kainic acid induced epilepsy rat

Objective:To investigate the hippocampus neuron injury,mitochondrial function and mitochondrial apoptosis related protein expression in kainic acid(KA) induced epilepsy rat,and to explore the role of mitochondrial pathways of apoptosis in the neuron injury.Methods:KA induced epilepsy model was produced by injection of KA into the hippocampus.Forty SD rats were randomly divided into five groups: control and KA group(6 h,1 d,3 d and 7 d),n=32.The concentration of Glu in hippocampus CA3 area was detected by HPLC.The apoptosis of hippocampus neurons and the concentration of free calcium ion were assayed by flow cytometry.The expression of cytochrome C and caspase-9 in hippocampus CA3 area was determined by Western blot.Results: ①The concentration of Glu in hippocampus CA3 area was increased in the fourth day after KA injection.②The concentration of free calcium ion,mitochondrial membrane potential,and the number of apoptosis neurons was significantly changed after 6 h of KA injection.Seven days after KA injection,these changes were more significant.③The hippocampus neuron expressed low levels of cytochrome C and caspase-9,and the expression of cytochrome C and caspase-9 was dramatically increased in a time dependent manner which increased by 4.37and 3.20 fold 7 d after KA injection.Conclusion:Mitochondrial pathways of apoptosis involve in hippocampal neuron apoptosis in KA induced epilepsy rat.
9.Cloning and expression of urease B subunit (UreB) of Helicobacter pylori

Objective To obtain the recombinant urease B subunit (ureB) of Helicobacter pylori (Hp) and apply it in serological detection of Hp infected patients. Methods Urease B subunit gene was amplified from the complete genome of Helicobacter pylori by PCR, and cloned into the PinPoint TM Xa Ⅲ fusion expression vector, then was sequenced. The protein of urease B subunit was expressed in E.coli JM109 and purified with affinity chromatography. 113 serum samples of peptic ulcer patients were detected by ELISA combined with purified UreB protein. Results DNA sequence analysis showed the nucleic acid sequence homology of ureB gene was 96.44% and the putative amino acid sequence homology was 99.65%. The recombinant UreB protein was composed of 571 amino acid residues and kept original immunologic reaction with corresponding antibody, and its purity was over 90% after affinity chromatography. The results of ELISA associated with recombination UreB antigen showed the sensitivity and specificity was 92%, 98.5%. Conclusion The recombinant UreB protein will be of value for clinical serodiagnosis and epidemiological study of Helicobacter pylori.
10.Analysis of the usage of antibacterial drugs in 75 hernior-rhaphy inpatients during perioperative period

Objective: To evaluate the rationality of the use of antibiotic prophylaxis in the perioperative period of herniorrhaphy. Methods: Seventy-five cases of herniorrhaphy discharged in March, June and September of 2009 were collected. Parameters including indications, the types of antibacterial drugs, time of administration, solvent choice, usage and dosage, combination, etc, were assessed for the rationality according to the "the guiding principles of clinical use of antibacterial drugs"and other relevant documents. Results: The antibacterial drugs application rate was 100%, while the rationali-ty of the drug selection, the drug administration route, the single dose and the number of dosing, was 70.7%, 100% , 100% and 93.3% respectively. Sixty-two patients(82.7%)were treated with antibacterial drugs at 0.5~2 hours before surgery, and the average time course of the prophylactic use of antibiotics after surgery was 2.3 days. Conclusions: Some problems existed for the use of antibiotic prophylaxis in the perioperative period of herniorrhaphy. Surgeon should ensure the correct use of the antibacterial drugs to make the safety, efficiency, rationality, and economy for the patients.
