1.The change of MTH2 expression with aging in hippocampi of senescence-accelerated mouse P8

Objective To observe the expression of oxidized purine nueleoside triphosphatase MTH2 in the hippocampi of senescence-accelerated mouse P8(SAMP8). Methods Randomly selected SAMP8 and its control strain SAMRl aged 1-,4-,8-and 12-month and performed immunohistochemisry and Western blot analysis to explore the expression changes of MTH2 in the 2 groups of mice. Results The MTH2 protein was detected in cytoplasm and nuclei of neurons throughout the whole hippocampus of SAMRl at all ages.There were no obvious changes of SAMR1 in both CA1 and CA3 subregions,but an age-related decline of MTH2 protein in both regions of SAMP8 was found.The optieal density(OD)value of Cal hippoeampus(0.0473),CA3 hippoeampus (0.0510)were significantly less in 8-month old mice than in the 4-month old[CA1:0.0619,CA3:0.06803(all P<0.05).By immunoblot analysis,the grey values of MTH2 expression were 0.529,0.313,0.032 in hippoeampus of 4-12 months-old SAMP8 mice,and significantly Iower than in the age-matched SAMR1 mice(all P<0.05). Conclusions MTH2 deficiency may be a new factor involved in the accelerated aging and in deficits of learning and memory ability in SAMP8.
2.Detection of Nitric Oxide, Glutathione and Superoxide Dismutase in Ovarian Neoplasm

Objective: Our aim was to study the relationship between nitric oxide and ovarian neoplasm. The concentrationg of nitrite /nitrate (NO-2/NO-3),glutathione(GSH), and the activity of superoxide dismutase(SOD) were detected in ovarian tissues.Methods:A modified copper-coated cadium reduction method was used to detect NO-2/NO-3. Dithio-bis-nitrobenzoic acid (DTNB) method was used to detect the GSH concentration. The activity of SOD was measured by an ultramicroanalytic and rapid assay. All the data were obtained from 21 patients with ovarian neoplasm(10 with benign ovarian neoplasms,and 11 with ovarian carcinomas). This paper is concerned with the analysis of matched case-control studies. The carcinoma tissues and normal ovarian tissues were obtained from the same patient. We performed Paired-Samples t test to determine whether the NO-2/NO-3,GSH and the activity of SOD in carcinomas and normal ovarian tissues have significantly difference. Results:No significant difference for the concentration of NO-2/NO-3,GSH and activity of SOD was shown in the normal ovarian tissues and neoplasm tissues of benign neoplasm . The concentrations of GSH and activity of SOD in normal ovarian tissues were significantly higher than ovarian carcinomas(P<0.05). No significant difference of concentration of NO-2/NO-3 was shown in normal ovary and ovarian carcinomas. In the normal tissues of all patients, NO-2/NO-3 was positively correlated with GSH and SOD. Glutathione was positively correlated with SOD. In the ovarian neoplasms, there was no significant correlation between NO-2/NO-3 and GSH, NO-2/NO-3 and SOD,GSH and SOD.Conclusion: Nitric oxide and the other matters which attend the course of its metastasis may closely related to ovary neoplasms.
3.Effect of LGR5 knockdown by shRNA on the proliferation and drug resistance of HeLa cells

Objective To investigate the effect of leucine-rich-repeat-containing G-protein-coupled receptor 5 (LGR5 )on the proliferation and drug resistance of HeLa cells and its possible mechanism.Methods LGR5 expression was interfered using shRNA,and LGR5 knockdown HeLa cells were constructed.The effect of LGR5 on the proliferation and drug resistance of HeLa cell was evaluated by cell count,clone formation and MTT;the expressions of LGR5 and β-catenin in HeLa cells were detected by Western blot method.Results LGR5 knockdown HeLa cell line was successfully constructed;the cell growth rate and clone formation rate in shLGR5 group were markedly decreased compared to those in shCon group (P<0 .01 ).Drug resistance of HeLa to cisplatin differed significantly between shLGR5 group and shCon group (P<0.01 ).Moreover,the LGR5 knockdown inhibited the expression ofβ-catenin in HeLa cells.Conclusion LGR5 plays an important role in cell proliferation and drug resistance of HeLa cells,and its mechanism is related to Wnt/β-catenin signaling pathway.
4.Fetal external ear observation on standard sections and the best time of check-up

Objective To detect the fetal external ear observation on standard sections and the best time with ultrasonography. Methods Total 2212 normal fetuses 17-40 weeks of gestation were divided into six groups. Fetal ears were obtained on transverse-incline view at cervical vertebra level (section A) and mandible level (section B), on parasagittal view (section C) and coronal view (section D) at external ear level. Fetal ears were evaluated and analyzed statistically. Results Among 2212 normal fetuses, 3454 ears (78.07%), 1694 ears (38.29%), 3454 ears (78.07%) and 2304 ears (52.08%) were obtained on section A, B, C and D, respectively. The obtained-rate of fetal ears on section A was the same as section C at the same week of gestation, but was significant higher than that of section B and section D (P<0.05). The obtained-rate of fetal ears was statistical different between section B and section D at the groups of 25-40 weeks (P<0.01). The obtained-rate of fetal ears was negatively correlated with gestational age (P<0.01). Conclusion It is useful and easy to observe the fetal external ear with ultrasonography to detect fetal ear abnormalities. Section A and section C are recommended to be standard observation sections for fetal external ear. Section B and section D can be used as supplementary for section A and section C. The best time for fetal external ear observation with ultrasonography is 17-24+6 weeks gestation.
5.125I seed-loading stent vs. conventional stent for the treatment of middle-late stage esophageal cancer:a clinical control study

Objective To assess the clinical value of the implantation of 125I seed-loading stent in treating middle-late stage esophageal cancer.Methods A total of 64 patients with middle-late stage esophageal cancer,who were treated with esophageal stent implantation during the period from July 2013 to December 2015,were included in this study.According to patient's own will,the patients were divided into group A (n=28,using conventional stent) and group B (n=36,using 125I seed stent).Based on the treatment planning system (TPS) and tumor morphology,conformal comprehensive isodose distribution of 125I seeds was formulated.The success rate of stent implantation,the complication rate,the improvement rate of dysphagia,the stent patency rate,the average hospitalization days,the hospitalization expenses and the survival time were compared between the two groups.Results In both groups,the success rate of stent implantation and the improvement rate of dysphagia were all 100%.The 12-month stent patency rate of group B was evidently higher than that of group A,and the difference was statistically significant (P<0.05).No statistically significant difference in the average hospitalization days existed between the two groups (P>O.05).The mean hospitalization expenses between the two groups was statistically significant (P<0.05),with the mean medical expense in group B being 13,769.57 RMB more than that in group A.Both the mean survival time and the median survival time of group B were longer than those of group A (P<0.05).Conclusion It is safe and effective to use 125I seed stent to treat middle-late stage esophageal cancer.This technique can evidently prolong the survival time of patients,although its medical cost is higher than that of the ordinary stent.
6.Relationship between transferrin levels and function of male fertility and testicular podocyte

Objective To investigate the relationship between levels of transferring(Tf)and function of male fertility and testic-ular podocyte.Methods Semen samples from male patients with infertility and volunteers with normal infertility were collected and tested for sperm density and motility by using sperm quality analyzer.In additon to that,Tf concentrations of Tf were determined;aseptically excised rat testis and by collagenase and hyaluronidase digestion,podocytes were isolated with high purity and cultured in culture medium,then the concentration of Tf in cell culture medium was determined;Tf concentration were determined by rate nephelometry.Results Tf concentration in semen of male-infertility patients[(15±5)μmol/L]was significantly lower than that of normal-fertility volunteers[(24.5±6.5)μmol/L,P <0.01],which were positively correlated sperm count and motility(P <0.01). Tf concentrations of cell culture medium[(25 ±8)μmol/L]in which testicular podocytes were cultured were significantly higher than that of infertile rat[(15 ±6)μmol/L,P <0.01].Conclusion Determination of semen Tf levels can reflect podocyte function and be used as an indicator for the evaluation of seminiferous tubules spermatogenesis,which was valuable in the diagnosis and treatment of male infertility.
7.Expression of Th17 cells and related cytokines in peripheral blood of patients with Graves′disease

Objective:To explore the proportions of Th17 cells in the peripheral blood and levels of IL-17,IL-23 in the serum of patients with Graves′disease ( GD ) and their clinical significance.Methods: We studied 29 patients with GD ( GD group ) , and reevaluated the GD group after therapy ( euthyroid GD group ).29 gender-and age-matched volunteers were selected as the normal control ( NC group).The proportions of Th17 cells were investigated by flow cytometry.The levels of IL-23,IL-17 in the serums were measured by ELISA.The levels of FT3,FT4,TSH were determined by ECLIA and the levels of TrAb were tested by RRA.Results:There were no significant difference among 3 study groups in sex and age match ( F=0.0075 , P>0.05;χ2=0.4213 , P>0.05 ).The proportions of Th17 cells and the levels of IL-17 , IL-23 were increased in the GD and euthyroid GD patients compared with the control group (respectively,P<0.05).The proportions of Th17 cells and the concentrations of IL-23 in euthyroid GD group were significantly lower than those of GD group ( respectively , P<0.05 ) , but there were no significance in the concentrations of IL-17 between euthyroid GD group and GD group(P>0.05).Correlation analysis revealed that the proportions of Th17 cells ,and the levels of IL-17,IL-23 were positively correlated with the levels of FT3,FT4,TrAb(r=0.588 2,0.337 2,0.371 0;0.549 6,0.287 5,0.342 7;0.361 0,0.420 8, 0.330 8;P<0.05 ,for all parameters ) ,and were negatively correlated with the levels of TSH ( r=-0.319 7 ,-0.472 8 ,-0.428 2;P<0.05,for all parameters).Conclusion:Th17 cells and their related cytokines IL-17,IL-23 are highly expressed in the serum of patients with GD.Th17 cells and their relative cytokines have certain relevance with 4 thyroid function parameters of the patients with GD , which can be used as biological markers for GD.
8.The research on therapeutical effect of 4-AP-3-MeOH on chronic spinal cord injury in rats

Objective To investigate the therapeutic effect of the new K+ channel blocker 4-aminopyridine-3-methanol (4-AP-3-MeOH) on chronic spinal cord injury in rats.Methods 18 adult male Sprague-Dawley rats were randomly divided equally into 3 groups as follows:4-AP-3-MeOH treatment group (A),normal saline control group (B) and sham operation group (C).T10 segment of spinal cord compression injury was applied in group A and group B.4 weeks later,the rats in group A were daily treated by injecting 4-AP-3-MeOH 1 μmol (1000 μmol/L× 1 ml) into the veins of the tails for 4 weeks.While the same volume of saline was administrated into the group B for 4 weeks.Lamina of vertebra of T10 was cut without spinal cord injury in the sham-operation group.After modeling,the locomotor functional recovery was assessed by using Basso Beattie Bresnahan (BBB) scores and inclined plane (IP) tests,and all rats were periodical inspected by the somatosensory evoked potential(SEP) and motor evoked potential(MEP) post-operatively.Results BBB scores of group A started to increase gradually and were higher than those of group B from the 6th week,but the 2 groups at each time point in BBB scores were less than group C.Until 8 weeks after surgery,the inclined-plated angles of group A and B did not show significantly difference,but the both groups were significantly lower compared with group C.Electrophysiology study found that the amplitude of SEP and MEP in group A was higher than group B from the fourth to 8 weeks,but less than in group C.Histological examination showed that the spinal cord of rats in group C were morphologically intact with a clear demarcation between the grey and white matter.The gray matter structures of rats in group A and B were gone and a big cavity appears in the center of the spinal cord injury,with varying degrees of demyelination in the white around.In addition,statistically significant differences were found in the percentage of residual myelin at the injury epicenter between group A and group B.Conclusion With a persistent demyelination change and poor motor function in chronic spinal cord injury,daily administration of 4-AP-3-MeOH can improve the sensory and motor functions.
9.Effect of group visits on health condition among follow-up patients with chronic heart failure

Objective:To explore the effect of group visits on health condition among follow-up patients with chronic heart failure.Methods:Totally 126 outpatient follow-up patients with heart failure were divided into intervention group and control group by random number table from 2018 to 2019. The intervention group consisted of 63 cases and control group consisted of 59 cases. The intervention lasted 6 months. The intervention group received group visits, while the control group received routine outpatient follow-up. Medication adherence, quality of life and heart failure related indicators were evaluated at baseline and after 6 months of intervention.Results:At 6 months after intervention, the scores of medication adherence, total quality of life, body, emotion and others dimensions of the intervention group were (5.79±1.38), (30.11±8.22), (12.65±5.53), (5.24±4.57) and (12.22±4.76) points. These scores of the control group were (5.31±1.09), (37.26±9.02), (15.87±5.21), (7.03±5.14), (14.36±5.54) points. The differences between the two groups were statistically significant ( t values were -4.581-2.161, P<0.05 or 0.01). The BNP and proportion of New York Heart Association (NYHA) I the intervention group were (180.87±174.92) ng/L and 84.1% (53/63). These indicators of the control group were (351.02±268.13) ng/L and 67.8% (40/59). The differences between the two groups were statistically significant ( t value was -4.177, χ2 value was 4.484, P<0.01 or 0.05). Conclusions:Group visits program is an effective management mode to provide intensive patient education and foster peer support, improving medication adherence and quality of life of follow-up patients with heart failure.
10.Effect of shuxuetong in preventing restenosis after intracoronary stenting.

OBJECTIVETo evaluate the effect of shuxuetong (SXT) in preventing restenosis after intracoronary stenting.
METHODSSixty-eight patients, accepted intracoronary stenting, were divided into two groups, the SXT group and the control group, both of them were treated with conventional treatment, and to the SXT group, SXT was given additionally. The condition of treated coronary artery restenosis in the two groups was compared by way of quantitative coronary angiography and a 6-month follow-up study was adopted.
RESULTSFollow-up study was completed in 43 patients (23 cases in the SXT group, and 20 in the control group). The angina recurrence rate in the SXT group (3 cases, 13%) was significantly lower than that in the control group (7 cases, 35%, P < 0.05). Quantitative coronary angiography showed the restenosis degree of operated artery in the SXT group was significantly milder than that in the control group, with the last lumen losing and index in the SXT group (0.46 +/- 0.25 mm, 24.26 +/- 8.64%) less than those in the control group (0.75 +/- 0.33 mm, 31.25 +/- 11.03%). The net gain lumen and the net gain index in the SXT group (1.23 +/- 0.30 mm, 58.96 +/- 24.68%) were greater than those in the control group (0.98 +/- 0.33 mm, 42.68 +/- 29.51%), all P < 0.05. But the restenosis rate in the two groups was insignificantly different (P > 0.05).
CONCLUSIONSXT might has some definite effect in preventing restenosis after intracoronary stenting.
