1.Characteristics and clinical application of umbilical cord blood stem cells

Umbilical cord blood that is rich in hemopoietic stem cells and mesenehymal stem cells is collected in vivo and in vitro.Reclamation of stem cells plays an important role in processing umbilical cord blood;additionally,separation of monocytes also plays an important role in establishing bank of umbilical cord blood and performing umbilical cord blood stem cell transplantation.Umbilical cord blood is separated using density gradient centrifugation,hydroxyethyl starch settling process,monoclone antibody flow cytometry,and immunomagnetic bead sorting.Density gradient centrifugation combined with adherent method is used to in vitro separate and culture mesenehymal stem cells;bio-reactor is used to amplify human umbilical cord blood stem cells and progenitor cells.Under a certain condition,mesenehymal stem cells are able to differentiate into hepatocytes.Cryopreservation is mainly used to freeze umbilical cord blood.Vitrification method characterizes by sample,rapid,and high effective,thus it is an ideal method to maintain umbilical cord blood stem cells for a long term.Umbilical cord blood stem cell transplantation benefits for treating various diseases in clinic.
2.EFFECTS OF 17?-ESTRADIOL ON HYPERTROPHY IN CULTURED NEONATAL RAT CARDIOMYOCYTES

Objective To observe effects of 17?-estradiol on phenylephrine-induced hypertrophy in cultured neonatal rat cardiomyocytes and to explore its mechanisms. Methods Using cultured neonatal rat cardiomyocytes treated with different factors as a model,surface area of cardiomyocytes was measured by computer photograph analysis software;the protein synthesis rate was assayed with leucine intake method;the proto-oncogene c-fos protein expression was assessed with immunocytochemistry technique;the markers of cardiomyocyte hypertrophy,embryonic genes such as ?-MHC\,?-skA and ANP mRNA expressions were assessed with semiquantitative reverse transcription-PCR. Results 17?-estradiol inhibited obviously phenylephrine-induced increase of surface area and the protein synthesis rate of cardiomyocytes,reduced c-fos protein expression of hypertrophic cardiomyocytes,and down-regulated ?-MHC、?-skA mRNA expressions and up-regulated ANP mRNA expression.Conclusion 17?-estradiol inhibits hypertrophy of cardiomyocytes and its mechanisms might be related with the suppression of c-fos protein expression,the reversion of the embryonic switching of contractile protein genes(?MHC and ?-skA),the increase of ANP mRNA expression.
3.Exploration of Strengthening Quality Management of Drug Microbiology Laboratories

OBJECTIVE:To strengthen quality management of drug microbiology laboratory and provide reference for relevant inspection institutions. METHODS:Taking measures from the laboratory document management,personnel training,quality con-trol,quality supervision,process control and biological safety,the elements of laboratory quality management were analyzed,and effectively measures were put forward to improve the management level. RESULTS & CONCLUSIONS:The laboratory should es-tablish strict,standardized,systematic laboratory management rules and regulations,quality documents,operating procedures, work guide,etc. to achieve documented management;develop various forms of annual training and assessment programs to con-struct a professional team;strengthen internal(developing an annual internal quality monitoring plan,conducting quality control ac-tivities,regularly checking the suitability of the medium) and external (capability testing,inter-laboratory comparison) manage-ment control level,and reasonably formulate the contents and frequency by combing with actual situation. Besides,the laboratory should strengthen inspection process control,attach great importance to the biological safety management to reduce the risk of labo-ratory quality to a minimum.
4.Analysis of the Use of Antihistamine Agents in 18 Hospitals of Hangzhou During the Period 2000~2002

OBJECTIVE:To survey and objectively evaluate the present situation and trend of the antihistamine agents used in 18 hospitals of Hangzhou.METHODS:To make a survey of the use of above mentioned drugs in 18 hospitals of Hangzhou during the period 2000~2002 in respect to the sum of money for drugs and manufacturing factories.RESULTS:In comparison with 2001,the sum of money for consumption of antihistamine agents increased 17.89%in 2000,and decreased 4.58%in 2002.Joint venture and imported products accounted for 70%of share of market.Lortadine and Cetirizine were still the first choice in therapy of allergic reactions.CONCLUSION:Consumption of antihistamine agents decreased,but kinds of drugs had no obvious change.
5.Effect of quality control circle on blood pressure and metabolic syndrome in patients with newly diagnosed primary hypertension

Objective To assess the effect of quality control circle on blood pressure and metabolic syndrome in patients with newly diagnosed primary hypertension. Methods Review health examination in 132 cases of newly diagnosed patients with primary hypertension as the control group, routine health education was given. The 128 cases of patients with newly diagnosed primary hypertension as the intervention group and the prospective study was done.The quality control group of quality control circle was set up in order to improve treatment adherence for patients with newly diagnosed primary hypertension, and the nursing intervention was implied for the intervention group for 6 months. Blood pressure and metabolic syndrome were compared between the two groups. Results The compliance rate was improved from 56.3%(72/128) to 90.6%(116/128) after intervention in the intervention group. There was significant difference before and after intervention (χ2=62.57, P<0.01).The systolic blood pressure, diastolic blood pressure, triglyceride (TG), fasting blood glucose (FBG) and waist circumference were (151.6±11.3) mmHg (1 mmHg=0.133 kPa), (98.6±12.1) mmHg, (1.98±0.36) mmol/L, (6.37±1.25) mmol/L, (92.88±6.90) cm respectively before intervention in the intervention group and were (132.6±14.3) mmHg, (82.5 ± 12.5) mmHg, (1.53 ± 0.17) mmol/L, (5.34 ± 1.06) mmol/L, (83.29 ± 5.47) cm respectively after intervention. There was statistically significant difference before and after intervention in the intervention group(t=7.11-12.79, P<0.05). The systolic blood pressure, diastolic blood pressure, TG, FBG and waist circumference were (150.4 ± 10.6) mmHg, (99.4 ± 10.8) mmHg, (1.94 ± 0.39) mmol/L, (6.52 ± 0.29) mmol/L, (91.37±6.03) cm respectively before intervention in the control group and were (145.3±13.7) mmHg, (92.6± 10.9) mmHg, (1.86 ±0.31) mmol/L, (6.02±0.27) mmol/L, (93.86±6.52) cm respectively after intervention. There was significant difference in systolic blood pressure and diastolic blood pressure before and after intervention in the control group (t=3.38, 5.09, P<0.05). But the difference was not statistically significant in TG, FBG and waist circumference (t=1.85, 1.27, 1.93, P>0.05). The difference of systolic blood pressure,diastolic blood pressure,TG, FBG and waist circumference between two groups was statistically significant after the intervention (t=6.77-12.84,P<0.05). The intervention group was significantly better than the control group. Conclusions The quality control circle may improve treatment adherence and it is helpful to improve the metabolic syndrome and it has high clinical application value for the treatment of primary hypertension.
6.Expression of IL-12 family subunit genes in mice C6 glioma cells

Objective:To observe the expression of IL-12 family subunit genes by real-time quantitative PCR in mice C6 glioma cells,construct the basis of the brain glioma research on IL-12 family in the future.Methods:Mice C6 glioma RNA was abstracted and reversed transcription cDNA.The mice C6 glioma cells mRNA expression influence of IL-12 family subunit genes was compared and analyzed by real-time quantitative PCR.Results: In mice C6 glioma cells, high expression abundances in IL-23a, IL-12a, midlde expression abundances in EBI3, IL-27, low expression abundance in IL-12b.Conclusion: IL-12 families are closely related to the occurrence and development of glioma,IL-12,IL-23 are regarded as the most potential anti-glioma cytokines among them,research de-velopments will bring a new way of brain glioma immune therapy.
7.125I seed-loading stent vs. conventional stent for the treatment of middle-late stage esophageal cancer:a clinical control study

Objective To assess the clinical value of the implantation of 125I seed-loading stent in treating middle-late stage esophageal cancer.Methods A total of 64 patients with middle-late stage esophageal cancer,who were treated with esophageal stent implantation during the period from July 2013 to December 2015,were included in this study.According to patient's own will,the patients were divided into group A (n=28,using conventional stent) and group B (n=36,using 125I seed stent).Based on the treatment planning system (TPS) and tumor morphology,conformal comprehensive isodose distribution of 125I seeds was formulated.The success rate of stent implantation,the complication rate,the improvement rate of dysphagia,the stent patency rate,the average hospitalization days,the hospitalization expenses and the survival time were compared between the two groups.Results In both groups,the success rate of stent implantation and the improvement rate of dysphagia were all 100%.The 12-month stent patency rate of group B was evidently higher than that of group A,and the difference was statistically significant (P<0.05).No statistically significant difference in the average hospitalization days existed between the two groups (P>O.05).The mean hospitalization expenses between the two groups was statistically significant (P<0.05),with the mean medical expense in group B being 13,769.57 RMB more than that in group A.Both the mean survival time and the median survival time of group B were longer than those of group A (P<0.05).Conclusion It is safe and effective to use 125I seed stent to treat middle-late stage esophageal cancer.This technique can evidently prolong the survival time of patients,although its medical cost is higher than that of the ordinary stent.

Exosomes are vesicular bodies secreted by living cells containing proteins and RNA, and play an important role in the process of physiology and pathology.Pancreatic cancer is one of the common gastrointestinal tumor with high morbidity and mortality.As a hotspot in oncology, exosomes have potential values in the research of development, diagnosis and treatment of pancreatic cancer.This article reviewed the role of exosomes in pancreatic cancer.
9.Application of diffusion-weighted magnetic resonance imaging in precise radiotherapy for esophageal carcinoma

ObjectiveTo explore the application of diffusion-weighted magnetic resonance imaging (DWMRI) in precise radiotherapy of esophageal carcinoma.MethodsThirty-seven patients with biopsy proven esophageal cancer from March 2010 to January 2011 were included.To delineate the gross tumor volume (GTV) using CT and DWMRI images,each patient was examined by DWMRI and CT scan using the same position before radiotherapy.To compare the maximum diameters and volumes of tumor between CT and DWMRI. The tumor lengths measured by esophagogram,esophagoscope,CT and DWMRI were compared.ResultsTumor lengths under esophagogram,esophagoscope,CT and DWMRI were 5.70 cm,6.06 cm,7.97 cm and 5.79 cm respectively. The lengths between CT and esophagogram,CT and esophagoscope,CT and DWMRI had statistical significance respectively (F=4.88,P=0.003).The maximum diameters of tumor shown on CT and DWMRI were 3.79 cm and 3.81 cm respectively ( t =-0.32,P=0.751 ).The GTV were 45.75 cm3 and 38.05 cm3 in CT and DWMRI respectively (t=5.30,P =0.001 ).53 lymph nodes were assessed positive on both CT and DWMRI.DWMRI excluded 25 positive lymph nodes assesed by CT; also confirmed 15 negative lymph nodes excluded by CT,6 of which were paraesophageal lymph nodes.The addition of DWMRI information altered the clinical stage in 6 patients.ConclusionsTumor lengths measured on DWMRI and esophagogram had the optimal approximation.It was easy to find paraesophageal lymph nodes via DWMRI.With the addition of DWMRI information,the target range and clinical stage were alerted in some patients.
10.The evaluation of prognosis and investigation of clinical staging for esophageal carcinoma treated with non-surgical methods

ObjectiveTo analyze the prognosis of 784 patients according with clinical staging of esophageal carcinoma treated with non-surgical methods,investigate the predictive value and deficiency of the clinical staging.MethodsFrom July 2003 to January 2009,784 patients with esophageal carcinoma received 3DCRT treatment.The prescribed doses ranged from 50 Gy-70 Gy with median dose of 60 Gy,1.8-2.0 Gy/fraction,1 fraction/day,5 fractions/week.65 patients received prescription dose of<60 Gy and all the others'≥60 Gy.All the patients were divided into subgroups according to different T,N and TNM stages.Therapeutic effect was evaluated.ResultsThe follow up rate was 97.1%,503 patients were followed up for more than 3 years and 122 were followed up for more than 5 years.The 1-,3-,5-year local control rates and overall survival rates were 77.2%,54.2%,46.5% and 69.5%,34.9%,23.9%,respectively,with median survival time of 21 months.There were significant differences of survival curves for different T stages,N stages and TNM stages.For the groups of stage Ⅰ,Ⅱ and Ⅲ,the 1-,3-,5-year survival rates were 86.4%,47.6%,45.1% ;84.7%,46.3%,36.4% and 64.0%,30.9%,19.1%,respectively ( x2 =29.34,P =0.000).There were 752 patients with squamous cell carcinoma ( 95.9% )and 32 patients with non-squamous cell carcinoma (4.1% ),the median survival time were 21 and 16 months,respectively ( x2 =4.44,P =0.035 ).There were significant difference of survival rates for the subgroups whose GTV volume ≤20 cm3,20 -40 cm3,40 -60 cm3 and >60 cm3 (54 months,29 months,21months and 14months,x2 =68.71,P =0.000).ConclusionsThe clinical staging of esophageal carcinoma treated with non-surgical methods could predict the prognosis accurately,for patients with different pathology and GTV volumes,there were variance in the prognosis,so we advised the complement of the two factors in the draft of clinical stages.
