1.Study on variation characteristics of anti-donor specific antibodies in different species of sensitized mice after skin transplantation

Objective To compare the change features of anti-donor specific antibody (DSA) in different species of sentitized mice after skin transplantation. Methods All mice were divided into the Balb/c → C57BL/6 (6 pairs) and Balb/c → C3H skin transplantation groups (6 pairs). At d0, d2, d4, d7, d13, d17, d28, d35, d42, d49 and d56 after skin transplantation, the serum sample was prepared for detection of DSA-IgG and DSA-IgM levels. Results Moderate increase was noted in the DSA-IgG level in the sensitized mice within 1 week after skin transplantation. The IgG level was significantly increased within 1-4 week and peaked and stabilized within 4-8 week. No significant variation was observed in the DSA-IgM level at 8 weeks after skin transplantation. In the Balb/c → C57BL/6 skin transplantation group, the DSAIgG level was significantly lower than that in the Balb/c → C3H group. Statistical significance was identified in the IgG levels between two groups at d2, d17, d28, d35, d42, d49 and d56 after skin transplantation (all P<0.05). No statistical significance was noted in the DSA-IgM levels between two groups at each time point (all P>0.05). Conclusions Advancing the time of renal transplantation after skin transplantation moderately in the Balb/c → C3H group, or changing to the lower immunoreactive combination of Balb/c → C57BL/6 are aimed to establish AMR mouse models with mild rejection reaction.
2.Efficacy of facial artery perforator flap repairing procedure in patients with moderate area loss after facial tumor surgery

Objective:To investigate the value of facial artery perforator flap repairing procedure in patients with moderate area loss after facial tumor surgery.Methods:A total of 103 patients (male/female: 54/49, age: 57.5±8.2 years) with moderate area loss after facial tumor surgery from March 2015 to December 2018 were enrolled. The facial artery perforator flap was designed according to the facial wound area and the postoperative efficacy was evaluated.Results:Of the 103 patients included, scars in the donor area were mostly hidden in the nasolabial folds, and there were no complications such as eyelid valgus, angular deformity, nasal drift and facial nerve injury. Besides, 98 had flaps fully survived and 5 had cyanosis and swelling at the tip of the flap. The results of surgical evaluation were good in 98 cases and ordinary in 5 cases. After 6 to 36 months of follow-up, the color and texture of the flaps were similar to normal skin. There was no obvious scar in the donor area and the appearance was satisfactory.Conclusions:The use of facial artery perforator flap repairing procedure for the treatment of patients with moderate area loss after facial tumor surgery has the advantages of good blood supply, small donor injury, and scar concealment.

Objective To investigate serum lipid profiles in newly diagnosed patients with polycystic ovary syndrome (PCOS) using lipidomics and correlate these features with hyperinsulinemia and hyperandrogenism associated with PCOS and obesity. Methods 32 newly-diagnosed PCOS women and 34 controls were enrolled and divided into obese and lean subgroups according to the body mass index (BMI). Anthropometric, biochemical, and hormonal parameters were collected. Serum lipid profiles including phospholipids, free fatty acids (FFAs), and bioactive lipids were analyzed using GC-MS and LC-MS. Results PCOS patients, in particular, the obese ones with fatty liver, have abnormal phosphatidylcholine (PC)/lysophospholipid (LPC) metabolism. PC was increased (16∶0, 18∶0, 18∶1, 18∶2, and 20∶4), while LPC was decreased (16∶0, 18∶0, and 18∶1; all P<0.05). Serum polyunsaturated fatty acids (PUFAs), were decreased significantly, and the long chain saturated fatty acid was increased. We also found that insulin stimulated the metabolism of PUFAs, but the androgen inhibits the metabolism of PUFAs by measuring their metabolites. Conclusion PCOS patients have metabolic disorders of phospholipids and PUFAs. Insulin stimulated while androgen inhibited PUFAs metabolism.
