1.The changes of calmodulin and cAMP in rabbit myocardium during ischemia and reperfusion

This study was performed on rabbits to examine the changes of calmodulin (CAM) and cAMP in the myocardium during ischemia and reperfusion by radioimmunoassay method. The result showed the levels of CaM and cAMP increased at 20 min of ischemia. Although these two parmeters were unchanged at 40 min of ischemia as compared with the changes at 20 min, they were changed during reperfusion at different times of ischemia. At 20 min ischemia following 10 min reperfusion, the content of CaM decreased, the content of cAMP increased. On the basis of 40 min ischemia, reperfusion induced opposite changes: CaM increased, cAMP decreased. The possible mechanism of these changes was discussed.

To explore the origin and prevention of myopia from disfunction of internal organs and obsrtuction of channels.According to the research of ancient literature on relationship between internal organs/channels and myopia,and in the combination with research on neuroelectrophysiology,hemorheology molecular biology and clinic study,the feasibility of treatment on myopia from dysfunction of internal organs and obstruction of channels were analyzed.Dysfunction of internal organs and obstruction of channels played an increasingly important role in the occurring and developing of myopia.In the treatment of myopia,the role of internal organs and channels must be emphasized.
3.Developing of internal mandible distracter and the animal experimental research

ObjectiveTo introduce the progressing history of DO technique,and report the characteristics of the home-made type MS-1 internal mandibular distracter, and the effects of application of it in animal experiment. Methods15 Mongrel dogs were subjected to be lengthened of the unilateral or bilateral mandibles using the home-made type MS-1 mandibular distracter, and the regular X-ray ex aminations were made. ResultsAbout (20±5)mm in length of the andible had been achieved by the DO (distraction of osteogenesis)technique using the device. The accuracy of the distracter on lengthening would be attained to 98 % of the expectancy. The mineralization of the new bone fulfilled the gap zone about 5 weeks after the distraction had been finished during the consolidation period. ConclusionThe MS-1 type internal mandibular lengthening device has its special advantages on ap plication of cranio-maxillofacial surgery with a great expectancy.
4.Expression of ICAM-1 and VCAM-1 in human chronic renal allograft rejection

To study the mechanism of human chronic renal allograft rejection, kidney tissues were taken from 16 patients with chronic renal allograft rejection and from 5 healthy subjects, and underwent the frazed section staining for ICAM-1 and VCAM-1 to anti-ICAM-1 and anti-VCAM-1 respectively by using immunohistochemistry(ABC).The results showed that there were differ-ent distribution of ICAM-1 and VCAM-1 expression in nomal kidney and renal allograft during chronic rejection.It was suggested that ICAM-1 and VCAM-1 might play an important role in the pathogenesis of human chronic renal allograft rejection.
5.Determination of Psoralen and Isopsoralen in Bushen Jiannao Soft Capsule by HPLC

Objective To establish a method for determination of psoralen and isopsoralen in Bushen Jiannao Soft Capsule. Methods The HPLC separation was performed on Diamonsil-C18 (250 mm?4.6 mm, 5 ?m) column with a mixture methanol-water (48∶52) as the mobile phase, at a flow rate of 1.0 mL/min. Detection wavelength was at 246 nm, and column temperature at 40 ℃. Results The linear response of psoralen and isopsoralen ranged from 0.010 08~0.252 ?g (r=0.999 8) and 0.009 14~0.228 5 ?g (r =0.999 9). The average recovery was 98.91% (RSD=1.74%) and 99.93% (RSD=1.27%) respectively. Conclusion The method is simple, accurate, and suitable for the determination of the psoralen and isopsoralen in the preparations.

7.Expression of TET2 and DNMT3A in peripheral T cell lymphoma and their significance

Purpose To investigate the expression of TET2 and DNMT3A in patients with peripheral T cell lymphoma (PTCL) and the relationship to immunophenotypes of PTCL.Methods Using a panel of immunohistochemical markers (CD3,CD4,CD10,BCL-6,CXCL-13,CD30,ALK),all cases of PTCLs were further divided into four groups,including angioimmunoblastic T cell lymphoma (AITL),peripheral T cell lymphoma,not otherwise specified (PTCL-NOS),anaplastic lymphoma kinase negative anaplastic large cell lymphoma (ALK-ALCL) and anaplastic lymphoma kinase positive anaplastic large cell lymphoma (ALK + ALCL).The expression of TET2 and DNMT3A in 89 cases of PTCL was detected by immunohistochemical analysis.Results 89 cases were divide into four subtypes,AITL (36/89),PTCL-NOS (26/89),ALKALCL (18/89),and ALK + ALCL (9/89).Immunohistochemistry staining revealed higher cytoplasmic expression of TET2 and DNMT3A in AITL than that of in PTCL-NOS and ALCL (P < 0.05).And the nuclear expression of DNMT3A in patients with AITL was higher than that of PTCL-NOS and ALCL (P < 0.05).The cytoplasmic expression of TET2 was positively related with both cytoplasmic and nuclear expression of DNMT3A in patients with AITL (P < 0.05).Conclusion TET2 combined with DNMT3A could be used as markers in AITL diagnosis,which could provide new strategy for AITL diagnosis.
8.Relationship between unintentional injury and health risk behaviors among adolescents

Objective To identify the effect of health risk behaviors on accidental injury of adoles-cents.Then analyze the strength and approach of the health risk behaviors. Methods Cluster sampling on various stages and strata methods was adopted. The adolescents from grade one to three in junior school students and grade one to three in senior high school students completed the questionaire. A total of 4408 students were surveyed and 4309 samples were valid. Path analysis was used to understand how factors could influence injury incidence. Results The incidence of adolescent injury was 15.53%. Statistical differences were found between genders (male 17.77%,female 13.36%,χ2=15.91, P<0.01),education levels (junior high school 17.36%,senior high school 13.99%,χ2=11.45, P<0.01) and cities ( Beijing 12.97%,Zhongs-han 17.95%,χ2=20.33, P<0.01) . Those who got injured were found with more health risk behaviors. Health risk behavior was the direct influencing factor to injury,as well as a possible risk factor to multiple injuries. Conclusion Health risk behaviors can strongly affect adolescent injury,and can be an indicator. Reducing health risk behaviors is important in adolescent injury prevention.
9.Construction of mouse MyoD vector and its expression in Escherichia coli

Objective:To identify the cDNA gene of mouse MyoD by restriction enzyme analysis, and to express the gene in Escherichia coli (E coli) using a protein expression vector.Methods: After the cDNA gene of mouse MyoD had been amplified and identified,it was inserted into expression vector pBV220 in which exogenous gene was controlled by R RP L promoters.The recombinant plasmid pBV-my was transformed into E coli DH5? and the bacteria were induced at 42 ℃ to express encoded protein.Results:The cDNA of mouse MyoD was sequenced correctly.When the engineered bacteria had been induced an anticipated 55 ku protein band from the bacteria was observed on SDS-PAGE gel and amounted to 30% of total bacterial protein. Conclusion:The cDNA of mouse MyoD has been successfully coloned and efficiently expressed in E coli.
10.Expression and significance of leptin receptor and phosphorylation of signal transducer and activator of transcription 3 in diffuse large B-cell lymphoma

Objective To investigate the expression and clinical significance of leptin receptor (OBR) and phosphorylation of signal transducer and activator of transcription (p-STAT3) in patients with diffuse large B-cell lymphoma (DLBCL).Methods Immunohistochemical analysis was used to detect the expression of OBR and p-STAT3 in 80 patients with DLBCL and 10 patients with reactive lymphoid hyperplasia (RLH).Using a panel of immunohistochemical markers (CD10,bcl-6 and Mum-1),all cases of DLBCL were further divided into two groups,GCB (germinal center B-cell-like) or non-GCB.Results Immunohistochemistry revealed high expression of OBR and p-STAT3 in 45.0 % (36/80) and 28.8 % (23/80) cases of DLBCL,respectively,and minimal straining in 100.0 % (10/10) cases of RLH (P < 0.05).Compared with GCB group (8.7 %,2/23),non-GCB group had higher p-STAT3 high expression rate (36.8 %,21/57) (P < 0.05).There was no significant difference in the expression of OBR between these two groups.Compared with clinical stage Ⅰ-Ⅱ [46.2 % (18/39) and 25.6 % (10/39)],stage Ⅲ-Ⅳ had higher OBR and p-STAT3 high expression rate [61.9 % (13/21) and 38.1% (8/21)] (P > 0.05).The expression of OBR and p-STAT3 were not correlated with age,gender,extranodal infiltrations,LDH level,B-symptoms and IPI(international prognostic index)(P > 0.05).The expression of OBR was positively related with that of p-STAT3 in DLBCL patients (r =0.232,P =0.039).Conclusion OBR could stimulate the JAK-STAT signaling pathway and induces the phosphorylation of STAT3.This may be involved in carcinogenesis and prognosis of DLBCL.
