1.Significance of screening chromosome 22q11.2 deletion syndrome in the congenital cardiovascular abnormalities

Chromosome 22q11.2 deletion syndrome,also called DiGeorge syndrome or Velo-CardiacFacial syndrome,has all expansive phenotype involving essentially every organ and system,such as cardiovascular abnormalities,abnormal face,immunodeficiency,even psychiatric illnesses,and etc.Fluorescence in situ hybridization analysis test for the microdeletion from chromosome 22 at the q11.2 band is the comqrmed diagnostic method So far,it has not been known thoroughly in China and there has not been a normative screening system yet.Close relations between the microdeletion and congenital cardiovascular abnormalities especially conotruncal cardiac defects and arcus aortae abnormalities have been shown in reported cases.This review will describe the 22q11 DS and how to screen it in the congenital cardiovascular abnormalities so that it Can be diagnosed early and managed properly.which will benefit the patients and their later generations.

EZH2 is the catalytic subunit of polycomb repressive complex 2 (PRC2),which represses gene expression by catalyzing lysine 27 of histone H3.It is overexpressed in prostate cancer,breast cancer,bladder cancer,gastric cancer and several other cancers.There is a close correlation between overexpression of EZH2 and progression of malignancy,invasion and migration of cancer cells.With a thorough understanding of the function,up and down stream regulatory mechanism and clinicopathological features,EZH2 will be expected to as a target and provide a new way for the treatment of cancers.
3.Clinical characteristics of basal cell nevus syndrome (BCNS) :24 cases report

砄bjective: To summarize the clinical characteristics of basal cell nevus syndrome (BCNS) and the treatment experience of cysts in jaws. Methods: 24 patients of BCNS from Mar. 1981 to Sept. 1999 were retrospectively investigated. Results: Of all the cases, the mean age for the first examination was 17 years old. The respective incidence of the main symptoms were multiple nevi in 9 cases (37.5%), palmar and/or plantar pits in 7 (29.2%), spine and/or ribs deformity in 17 (70.8%), intracranial calcification in 19 ( 79.2% ), specific facial profile in 16(66.7%). The cysts of jaws were the main causes for early examination, which were characterized by their multiple and preadolescent occurrences. The cysts were surgically removed and the postoperative recurrence rate of the cysts was 22.0%. Conclusion: Much attention should be paid to the cysts of jaws, the main symptoms, and cause of the physiological and psychological impairment in the patients of BCNS.
4.Sanitary Investigation of Rural Drinking Water in Xiamen

Objective To investigate the hygienic quality of drinking water in rural areas of Xiamen and to provide scientific bases for the improvement of drinking water quality and protection of human health.Methods The well water samples were sampled randomly according to the proportion and the water quality was analysed and the incidences of water-borne diseases were investigated in Xiamen rural areas.Results Water sources for drinking water in most parts of Xiamen rural areas are shallow-layer ground-water.As being polluted by surface ranoff water,the indices of total bacte-rial count and coliform bacteria in the well water were heavily over the standard with the over standard rates61.6%and85.1%respectively.The contents of fluoride in drinking water were at the higher levels(the highest one was1.6mg /L)in some villages and still there were more villages where the iodine contents in drinking water was at the lower level.The iodine content in well water,spring water and tap water was11.1,2.5and4.45?g /L respectively,in which the unqualified rate was48.3%,90%and58.1%respectively.The incidence of dental fluorosis in school-age children was70%in the high fluoride areas and the incidence of endemic goiter was20.9%in the low iodine areas.Conclu-sion The main hygienic problem of drinking water in rural areas of Xiamen was biological pollution and lower iodine contents and higher fluoride contents in water in some areas.In order to eliminate the water-borne diseases,the most important rural sanitary measures need to be taken:improving the sanitary conditions around the wells,repairing the well wall s and using deep ground-water or drinking water and developing centralized water supply in the areas with unbal-anced health-related microelemeuts in drinking water if possible in some villages.
5.Expression of Ezrin protein and c-met in human esophageal squamous cell carcinoma and their clinical significance

Objective To investigate the expression of Ezrin protein and c-met in esophageal squamous cell carcinoma, their significance in the progress of esophageal squamous cell carcinoma development and metastasis. Methods The expression of Ezrin protein and c-met in 50 patients with esophageal squamous cell carcinoma was studied using immunohistochemical (SP) method. The correlation of each score according to the intensity and percentage of labeled cells or intercellular substance with relevant clinical data was statistically analyzed. Results In normal esophageal squamous epithelium, the positive expression of Ezrin protein was 34 %, the positive expression of c-met was 8.0 %. In esophageal squamous cell carcinoma, the positive expression of Ezrin protein was 76.0 %, the positive expression of c-met was 58.0 %. The expression of Ezrin protein had close relation to depth of invasion and lymph node metastasis(P < 0.05). The positive rate of c-met protein expression in lymph node metastasis was obviously higher than those without lymph node metasis (P <0.05). The expressinn of Ezrin protein in c-met positive eases was obviously higher than those of c-met negative cases(P <0.05). Conclusion The test of Ezrin protein and c-met may have significant prognostic value for assessing the degree of malignancy and potential lymph node metastasis probability of esophageal squamous cell carcinoma. Ezrin protein may be a target to evaluate the tumor prognosis.
6.Evaluation value of the levels of peripheral blood CD20+ CD24hi CD38hi regulatory B cells on the prognosis of elderly patients with sepsis

Objective To explore the predicting value of peripheral blood CD20+ CD24hi CD38hi regulatory B cells (Bregs) on the prognosis of elderly patients with sepsis.Methods A prospective study was conducted. Septic patients aged > 65 years old, compliance with diagnostic criteria for Sepsis-3, admitted to emergency and emergency intensive care unit (ICU) of Shanghai East Hospital of Tongji University from April 2016 to February 2017 were enrolled. Procalcitonin (PCT), C-reaction protein (CRP) and lactate (Lac) were routinely measured. According to the worst clinical index value within 24 hours, acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) score and sequential organ failure assessment (SOFA) score were recorded. The concentrations of peripheral blood CD20+ CD24hi CD38hi Bregs were measured by flow cytometry at 1, 3 and 7 days after diagnosed in elderly patients. All patients with sepsis were followed up for 28 days and then divided into death group and survival group according to 28-day outcome. The difference of clinic data and Bregs were compared between the two groups. The significant different factors of elderly sepsis patients were analyzed by binary logistic regression analysis. The correlation between Bregs level and other indicators was analyzed by Spearman correlation. The receiver operating characteristic curve (ROC) was used to evaluate the prognosis value of Bregs in elderly patients with sepsis.Results Fifty-eight patients were enrolled in the study, with 38 male and 20 female; age of (79.91±7.97) years; 32 in sepsis group, 26 in septic shock group; 35 deaths, 28-day mortality rate was 60.3%. APACHE Ⅱ score and SOFA score in death group exhibited much higher than that in survival group (APACHE Ⅱ: 18.14±4.52 vs. 14.91±3.56, SOFA: 8.80±4.56 vs. 6.35±3.00, bothP < 0.05), the Bregs was significantly decreased at 1, 3 and 7 days in death group [cells/μL: 0.70 (0.20, 1.40) vs. 1.50 (0.70, 2.20), 0.54 (0.20, 1.00) vs. 1.42 (1.10, 2.12), 0.25 (0.10, 0.50) vs. 0.80 (0.50, 1.00), allP < 0.05]. Correlation analysis showed that the concentrations of peripheral blood Bregs at 1 day in elderly patients with sepsis was negatively correlated with APACHE Ⅱ score (r = -0.351,P = 0.007), and it was not correlated with PCT, CRP, Lac or SOFA score. It was shown by binary logistic regression that Bregs [odds ratio (OR) = 1.865,P = 0.028] and APACHE Ⅱ score (OR = 0.853,P = 0.026) were independent risk factors for elderly sepsis outcome. It was shown by ROC curve analysis that the prognostic value of the levels of Bregs at 1, 3, 7 days and APACHE Ⅱ score were higher in the elderly patients with sepsis, and the area under ROC curve (AUC) and 95% confidence interval (95%CI) were 0.842 (0.647-0.954), 0.770 (0.564-0.911), 0.888 (0.703-0.977), 0.855 (0.661-0.961), respectively, allP < 0.01. The 7-day Bregs was most powerful to predict outcome, when the cut-off value was 0.50 cells/μL, the sensitivity was 72.73% and specificity was 86.67%. Conclusions The level of peripheral blood CD20+ CD24hi CD38hi Bregs could predict the clinical outcome of elderly patients with sepsis.
7.Beneficial Effect of Vigconic VI-28 Capsule on Cyclophosphamide-Treated C57BL/6 Mice Carrying Lewis Lung Cancer Tumor

Objective: To investigate whether Vigconic VI-28 capsule, a formulated traditional Chinese medicine containing radix Ginseng, cornu cervi pantotrichum and semen cuscutae, can assist tumor chemotherapy in a mouse model. Methods: Female C57BL/6 mice were s. c. injected with viable Lewis lung cancer ( LLC) cells (106 cells/mouse) . The mice were then treated with cyclosposphamide (Cyp, 40 mg/kg bodyweight, once every other day). One group was intra-gastrically given 2% VI-28 (0.5 ml/mouse, every other day) during the course of the chemotherapy. By day 28, the mice were sacrificed and their thymic indices and tumor indices were calculated and compared. Splenocytes were collected for analysis of their immunological status. Histological study was carried to examine the solid tumors. Results: Fourteen days after the injection of LLC cells, solid tumors developed in most of the animals, reaching 1 ~ 1.8 cm diameters by 28 th day Compared with mice of the LCC + Cyp group, thymus glands from the LLC + Cyp + VI-28 group were significantly heavier. Splenocytes of the same group responded better to ConA stimulation in vitro. Histochemical examination of the tumor tissues revealed that tumors of the Cyp + VI-28 group were better differentiated (less aggressive) than that of the Cyp group. Conclusion: Vigconic VI-28 capsule can promote recovery of immune system in mice undergoing chemotherapy and help Cyp to control the growth of tumor cells in vivo.
8.DNA vaccination encoding variable region gene of the T cell receptor induces regulatory immune response in BALB/C mice

Objective:To investigate regulatory immune response induced by DNA vaccination encoding T cell receptor V?5.2 or V?2.1 chain predominantly displayed on ovalbumin (OVA)-specific T cell clone.Methods:BALB/C mice were vaccinated with pcDNA3.1 encoding T cell receptor (TCR) V?5.2 or V?2.1 chain respectively.Using RT-PCR,transcription of the recombined plasmids was analysed. Cell proliferation was measured by 3H-TdR incorporation.CTL response was assayed by JAM test.Immuno-fluorescent assay was used to examine the anti-TCR antibody level and the number V?2 +T cells.Results:RT-PCR analysis showed that the recombined plasmids can be transcripted in vivo and in vitro.DNA vaccine with TCR variable chain effectively induced TCR-specific humoral and cellular immune response,V?2 +T cells was not depleted by V?2.1 TCR-DNA vaccination,but rather was anergy.Conclusion:Regulatory immune response can be induced by DNA vaccination encoding TCR V? or V? region in normal mice.
9.Investigation and Analysis of the Relationship of Hypertension and Diabetes with Superoxide Dismutase,Malondialdehyde,Hypersensitive C-Reactive Protein in 8 Longevity Areas in China of People Aged 40 and Over

Objective To investigate the changes of superoxide dismutase (SOD),malondialdehyde (MDA),hypersensitive c-reactive protein (hsCRP)in the process of age changes,in longevity areas in China,and the relationship between the above three indicators and hypertension and diabetes.Methods Residents who participated in the project of biomedical research of aging population conducted in 2014 were selected from 8 longevity Areas in China.2 467 people aged 40 and over attended the study,including 22 aged 40 and over,249 aged 60 and over,521 aged 70 and over,679 aged 80 and over,548 aged 90 and over,448 aged 100 and over.Information,including socio-demographic,were collected by self-designed questionnaire.Calibra-ted instruments were used by the clinical personnel who had worked over 3 years to manually check subjects’heath.The
fasting blood samples were collected by vacuum tube at early morning.The contents of plasma SOD,MDA,hsCRP and glu-cose were detected and compared among these elderly who were classified into different genders and different age groups.At the same time,compare the above three indicators of different subjects of hypertension,diabetes prevalence.Results In the groups aged 40 and over,60 and over,70 and over,80 and over,90 and over,100 and over,SOD were 55.55±8.82,54.49± 6.74,55.47±8.42,56.40±8.37,57.45±8.71 and 57.38±7.72 U/ml respectively in males,the difference between groups showed statistically significant (F =3.398,P <0.01)and 59.36± 10.98,56.08±7.98,57.41 ±8.54,57.38±8.18,59.16 ±7.71 and 57.90±8.66 U/ml respectively in females,the difference between groups showed statistically significant (F =2.667,P <0.01);MDA were 6.92±4.02,6.10±4.35,5.66±3.83,5.83±3.66,5.39±2.68 and 4.80±2.34 μmol/L re-spectively in males,the difference between groups were not statistically significant (F =2.198,P =0.052)and 8.93±8.20, 7.23±4.57,6.47±3.67,6.42±4.12,5.59±2.88 and 5.85±3.73 μmol/L respectively in females,the difference between groups showed statistically significant (F = 4.434,P < 0.01);positive rates of hsCRP were 11.8% (2/17),14.8% (26/176),19.3%(62/322),23.5%(77/327),26.1%(57/218)and 43.4%(33/76)respectively in males,the difference between groups showed statistically significant (χ2 =29.914,P <0.01)and 20.0%(1/5),16.4%(12/73),15.6%(31/199),20.7%(73/352),22.7%(75/330)and 28.0%(104/372)respectively in females,the difference between groups showed statistically significant (χ2 =14.078,P <0.01).In the groups of hypertension complicated with diabetes mellitus,hypertension,diabetes mellitus,non-diabetes hypertension SOD were 55.65±8.73,55.35±7.82,57.32±8.45 and 57.24±8.01 U/ml respective-ly,the difference between groups showed statistically significant (F = 3.694,P =0.011).MDA were 6.67 ±4.79,5.16 ± 2.70,6.01±3.87 and 5.66±3.15 μmol/L respectively,the difference between groups showed statistically significant (F =5.594,P =0.001);positive rates of hsCRP were 28.9%(54/187),29.5%(28/95),21.4%(275/1 288)and 21.2%(181/853)respectively the difference between groups showed statistically significant (F=8.749,P =0.033).Conclusion The re-sults indicated that the body had the ability to keep oxidation-antioxidant function stable in the process of aging.However, with the increase of age,the risk of cardiovascular disease was significantly increased,and it was more common for the aged suffering from inflammation.SODS,MDA,hsCRP are closely related to the occurrence of diabetes,high blood pressure,they were the reliable clinical indicators of oxidative stress degree.
10.Effects and mechanisms of Rapamycin on renal interstitial fibrosis in rats

Objective To discuss the intervention effects and mechanisms for Rapamycin on renal interstitial fibrosis in rats.Methods A total of 72 Wistar male rats were randomly divided into 3 groups,normal group(n =24) ,model group(n =24), treatment group (n =24).The model group and treatment group received adenine 200 mg/kg daily,and the treatment group was also given Rapamycin 5 mg/(kg·d) at the 8th day,the normal group was just given the same amount of normal saline for 6 weeks.In the end of the 2nd,4th and 6th week,8 rats in each group were sacrificed respectively.The expression of hepatocyte growth factor(HGF), transforming growth factor-β (TGF-β) and neutrophils gelatinases related apolipoprotein (NGAL) in each group were observed.The software of image analysis system was used for semi-quantitative analysis.Results HE and Masson staining results showed that the renal tubular were progressive swelling, and changed with interstitial fibrosis,atrophy and even necrosis in model group from 2 weeks to 6 weeks.The pathological changes of kidney were more ease in the treatment group compared with those in model group.Immunohistochemical staining results showed that HGF expression levels of renal interstitial tissue in model group and treatment group at the 2nd week were significantly higher than those of normal group(P <0.05), and were significantly decreased at the 4th week and 6th week (P < 0.05);HGF expression levels of renal interstitial tissue in treatment group were significantly higher than those in model group (P < 0.05).NGAL expression levels of renal interstitial tissue in model group and treatment group at the 2nd week were significantly higher than those in normal group(P <0.05) ,and were significantly decreased at the 4th week and 6th week(P < 0.05);NGAL expression levels of renal interstitial tissue in treatment group were significantly higher than those in model group (P < 0.05).TGF-β expression levels of renal interstitial tissue in model group and treatment group at the 2nd,4th,and 6th week were significantly higher than those in normal group (P <0.05) ,while TGF-β expression levels of renal interstitial tissue in treatment group were significantly lower than those in model group (P < 0.05).Conclusion Rapamycin could improve the rat kidney tissue pathology, relieve renal tubular expansion, and slow progression of renal interstitial fibrosis, and has certain protective effect to the kidney.
