1.Role of TLR2 in AngⅡ-induced cardiac fibrosis in hypertensive mice

Objective To study the role of TLR2 in AngⅡ-induced cardiac fibrosis in hypertensive mice .Methods Eighteen wild C57 mice were randomly divided into blank control group ,AngⅡgroup and TLR2 blocking group (6 in each group) .A hypertension model was established by sub-cutaneous infusion of AngⅡthrough a mini-pump for 7 days .Cardiac fibrosis in mice was observed with immunohistochemical staining .Results The area of cardiac fibrosis was significantly larger and the expression levels of collagen Ⅰ and TGF-βwere significantly higher in Ang Ⅱgroup than in blank control group (P<0 .05) .The area of cardiac fibrosis was 71 .2% smaller ,the expression level of collagen Ⅰ was 75 .5% ,and the expression level of TLR2 was 77 .7% lower in TLR2 blocking group than in Ang Ⅱgroup (P<0 .05) .Conclusion TLR2 is involved in Ang Ⅱ-induced cardiac fibrosis in hypertensive mice .
2.Clinic feature and treatment of ossification of cervical posterior longitudinal ligament associated with developmental spinal stenosis

3.Effect of aspirin on uterine hemodynamics in patients with early recurrent spontaneous abortion

Objective·To study the hemodynamic parameters of uterine at midluteal phase in patients with early recurrent spontaneous abortion (ERSA) and the effect of aspirin on them. Methods·Transvaginal color Doppler ultrasonography was used to measure the parameters of uterine blood flow and the endometrial thickness at midluteal phase of 271 women with ERSA (ERSA group) and 66 women without a history of recurrent spontaneous abortion (control group). Then ERSA group were administered with aspirin 50 mg/d orally for 2 months and other individualized treatment, and the effect of aspirin on parameters of uterine blood flow and the early pregnancy outcome of them were observed. Results·At midluteal phase, the endometrium was significantly thinner in ERSA group than that in control group. Pulsatility index (PI) of endometrial blood flow and mean PI (mPI), mean resistance index (mRI), and mean systolic/diastolic ratio value (mS/D) of uterine arteries were statistically significantly higher in ERSA group in comparison to control group (P<0.05). Following aspirin treatment, resistance to uterine blood flow reduced significantly in ERSA group (P=0.000), and the endometrial thickness increased in the patients with endometrial thickness less than 7 mm (P=0.000). Only 163 ERSA patients were re-examined by transvaginal color Doppler ultrasonography after aspirin treatment, among whom 136 women was pregnant after individualized treatment. Among these pregnant patients, 97 cases were pregnant for more than or equal to 12 weeks, while 10 cases aborted during the first 12 weeks, and the early pregnancy outcomes of the other 29 cases were still unclear. Conclusion·In comparison with normal fertile women, ERSA patients have significantly higher resistance to uterine blood flow and thinner endometrium. Aspirin can improve uterine blood perfusion, which improves early pregnancy outcome.
4.Investigation of depression and anxiety among patients with recurrent spontaneous abortion and analysis of the influence factors

Objective · To investigate the status of depression and anxiety in patients with recurrent spontaneous abortion (RSA) and the possible influence factors and to provide theoretical support for further psychological intervention. Methods · RSA patients and women with no history of RSA were invited to complete a questionnaire, including basic information, Self-Rating Depression Scale (SDS) and Self-Rating Anxiety Scale (SAS). All data were analyzed by SPSS. Results · Of all 1064 cases included in this study, 725 were RSA cases, 217 cases with one spontaneous abortion and 122 in control group with no history of spontaneous abortion. Our results showed that both RSA patients and patients with one spontaneous abortion have significantly higher SDS and SAS scores than control group. Furthermore, non-pregnant RSA patients with lower education level, lower household income and 3-5 years of marriage have significantly higher levels of depression and anxiety. Patients with multiple miscarriages (≥4), history of induced abortion and no live birth, score significantly higher in SDS. Conclusion · Whether pregnant or not, RSA patients are much easier to become depressive and anxious, which may be associated with education level, household income, length of marriage, numbers of pregnancy losses and previous live birth. Women with one spontaneous abortion also show a significant higher tendency of depression and anxiety. These patients should be given proper psychological intervention if necessary.

6.Expressions of Tim-3 and CD4+and CD8+T cells in peripheral blood from patients with coronary heart dis-ease

Objective To investigate the expressions of T cell immunoglobulin and mucin-domain contain-ing moleculesfamily-3(Tim-3)and CD4+ and CD8+ T cells in peripheral blood from patients with coronary heart disease (CHD). Methods 51 CHD patients were divided into two groups:stable angina pectoris group (27 patients)and acute coronary syndromes group(24 patients). Another 25 healthy subjects confirmed by coronary angiography were selected as a control group. Peripheral blood was drawn on admission. Enzyme-linked immunosor-bent assay was used to detect the concentration of Tim-3. Flow cytometry was applied to detect the expressions of CD4+and CD8+. Results As compared with the healthy control group ,the concentration of Tim-3 and the propor-tion of CD8+ in stable angina pectoris group and acute coronary syndrome group were reduced ,and those in acute coronary syndrome group were lower. The differences were statistically significant (P < 0.05). As compared with the healthy control group,the proportion of CD4+ and the ratio of CD4+/CD8+ of stable angina pectoris group and acute coronary syndrome group were increased ,while those in acute coronary syndrome group were higher. The differences were statistically significant(P<0.05). Conclusions At the onset of CHD,the concentration of Tim-3 and the proportion of CD8+ in peripheral blood are reduced ,but the proportion of CD4+ is increased. The more severe the disease,the greater changes the values.
7.A case of severe acute respiratory distress syndrome caused by inhalation injury

A 54-year-old male patient with severe acute respiratory distress syndrome caused by inhalation injury was admitted to the First People's Hospital of Lianyungang City on June 26 th, 2022. After admission, the patient received invasive mechanical ventilation (driving pressure-guided ventilator parameter setting) combined with prone position treatment immediately, but his condition continued to deteriorate. Five hours after admission, the patient received veno-venous extracorporeal membrane oxygenation (VV-ECMO) supporting treatment, treatment based on ultra-protective lung ventilation strategy combined with prone position ventilation for more than 12 hours per day. At the same time, pulse contour cardiac output monitoring technology was used to monitor cardiac index and extravascular lung water index to guide volume management, and fiberoptic bronchoalveolar lavage was performed for several times. After that, the patient was successfully weaned from VV-ECMO and ventilator, and then discharged from hospital successfully. During follow-up of one year after the injury, the patient showed no obvious respiratory symptoms, and his lung function was basically normal.
8.Quantitative determination of sibricose A5 and sibricose A6 in Polygalae radix.

OBJECTIVETo establish the method for quantitative determination of sibricose A5 and sibricose A6 in Polygalae Radix by HPLC.
METHODThe ultrasonic extracting method was applied in sample pre-treatment. The HPLC procedure was performed on the chromatographic column of Agela Promosil C18 (4.6 mm x 250 mm, 5 microm), the mobile phase was acetonitrile-0.1% phosphoric acid water solution (10:90). The detection wavelength was 330 nm and flow velocity was 1 mL x min(-1). The column temperature was 30 degrees C.
RESULTThe method has good linearity in the ranges of 0.0087-0.0694 g x L(-1) (r = 0.9993) for sibricose A5, 0.0090-0.0723 g x L(-1) (r=0.9991) for sibricose A6. The average recoveries of sibricose A5 and sibricose A6 were 101.7%, 97.87%, with the RSD of 1.7%, 1.6%, respectively.
CONCLUSIONThe method was simple, quick accurate and reliable. It is appropriate for the quantitative determination of sibricose A5 and sibricose A6 in Polygalae Radix.

9.Effects of triptolide exposure during pregnancy/lactation on the reproductive system of male offspring in rats

OBJECTIVE To investigate the effects of triptolide (TP) exposure during pregnancy and lactation on the reproductive system development and function in male offspring of rats, providing a reference for medication safety during pregnancy and lactation. METHODS Pregnant rats were randomly divided into control group (12 rats, normal saline) and T1-T4 groups [12, 13, 14, 17 rats that received TP at 200, 400, 600, and 800 μg/(kg·d) respectively]. They were given relevant medicine/normal saline intragastrically, once a day, until the offspring were born and naturally weaned, the intragastric administration volume of each rat was consistently 2 mL. After 60 days of feeding, reproductive organ weights and coefficients were measured in male offspring, testicular and epididymal histology and sperm morphology were observed. Sperm motility, sperm count, and serum levels of gonadotropin-releasing hormone (GnRH), follicle stimulating hormone (FSH), luteinizing hormone (LH), and testosterone (T) in the epididymides were analyzed. Protein expressions of glycogen synthase kinase 3α (GSK3α), phosphorylated GSK3α (p-GSK3α), and phosphatase 1γ2 (PP1γ2) in sperm were also determined. RESULTS Compared with the control group, the testicular and epididymal weights, serum levels of GnRH and T, the relative protein expression of PP1γ2 were significantly decreased in T1-T4 groups. Additionally, in the T2 to T4 groups, there were significant reductions in the weight and coefficient of the seminal vesicle, total number of sperm, sperm concentration, sperm motility as well as relative protein expressions of GSKα, p-GSK3α in the offspring rats. Furthermore, the epididymal coefficient in the T3 and T4 groups, the testicular coefficient, mean sperm track velocity and sperm curvature velocity in the T4 group were significantly decreased (P< 0.05); the number of abnormal sperm, rate of sperm abnormality, and levels of FSH and LH in the offspring rats of the T1 to T4 groups were all significantly increased (P<0.05); in the offspring rats of the T1 to T4 groups, there was a decrease in the number of epithelial cells in the seminiferous tubules of the testes. Within the epididymal tissue, degenerative and necrotic changes in the epithelial cells were visible, accompanied by mild infiltration of inflammatory cells in the stroma. CONCLUSIONS TP exposure during pregnancy and lactation disrupts reproductive organ development, impairs spermatogenesis and sperm motility, as well as suppresses androgen synthesis in male offspring,thereby having a negative impact on the development of the reproductive system. These effects may be mechanistically linked to regulation of GSK3α, p-GSK3α and PP1γ2 protein expressions.

Metformin, the first-line treatment of type 2 diabetes, has been also increasingly used in the treatment of some skin diseases, such as psoriasis. This review summarizes therapeutic mechanisms of and clinical research progress in metformin in the treatment of psoriasis, aiming to provide ideas and methods for the treatment of psoriasis, especially psoriasis complicated by metabolic diseases.
