International Journal of Laboratory Medicine 2015;(12):1649-1650
Objective To understand correctly the hook like effect of the immune transmission and transmission turbidity in the automatic biochemical analyzer and the prevention of the hook like effect .Methods First ,it had the function set of measurement linear range of the upper limit of the instrument and the instrument automatic dilution redo ,and then ,had determination of excee‐ding the upper limit of measurement range and artificial dilution before the sample setting .Results The group average value was 2 .49 g/Lbefore the determination setting ,3 .31 g/L after the determination setting ,and 3 .33 g/L artificial dilution determination . Conclusion High dose hook effect makes the strong positive to weak positive samples of measurement by error ,even false negative results ,and high concentration sample makes low value .So the set of samples and dilution to determination of instrument ,measured values are more close to the true value .

Objective To observe the changes of bone mineral density (BMD) in young boys and girls from 6-19 years old in Beijing area. Methods 1 139 healthy young boys and girls undergoing whole body scanning with dual energy X-ray bone densitometer were divided into different groups according to sex and age. Results There were no significant difference in body height, weight, BMD and bone mineral contents (BMC) between boys and girls from 6 to 10 years old. The annual growth rates of body height, BMD, and BMC were the fastest in girls from 6 to 14 and boys from 6 to 16 years old, respectively. Thereafter, the annual growth rates of body height, BMD, and BMC didn't significantly increase in boys and girls. There were two fast-growth periods in BMD growth stage, being at 11 and 14 years old in girls, and at 12 and 15 years old in boys. The total body BMD was significantly correlated with the height in girls under 12 years (P

Objective To evaluate the trimester specific reference ranges and dynamic changes of thyroid functional indicators during gestation of healthy pregnant women. Methods According to the selection criteria,139 cases were selected as the normal pregnant population to establish a self-sequential longitudinal reference range. We collected samples eight times in every case throughout the gestation(including the 8th,12th,16th,20th,28th,36th week of prenatal, the 3rd and the 6th month postpartum) and detected the indicators of thyroid funciton.Meanwhile,301 healthy non-pregnant women of childbearing age were selected as the control group. Results The median of TSH was lowest in the first trimester, and then was increased in the second and third trimesters. The reference ranges of TSH were 0.20-4.01 mIU/L,0.43-3.97 mIU/L,and 0.57-3.96 mIU/L in the above trimesters. The free thyroxine (FT4) level from the 12th week of gestation to the 6th month of postpartum was gradually decreased and was lower than the non-pregnant level. The total thyroxine (TT4) was significantly increased at the 12th week of gestation and thereafter it was stable and returned to normal after childbirth. The free triiodothyronine(FT3) was gradually declined from the 12th week to the 28th week and returned to normal postpartum.Total triiodothyronine(TT3) was significantly increased at the 12th week and thereafter was stable, and returned to normal after childbirth. There was a significant negative correlation between TSH and FT4 in the first trimester(P<0.01).There was no correlation between TSH and TT4 during the whole gestation. There were positive correlations between FT4 and TT4,FT3 and TT3 since the 12th week of gestation (P < 0.01).Conclusions The gestational specific reference range of TSH based on the selfsequential longitudinal study was narrow, however, the upper limit was still higher than that recommended in foreign guidelines.

Objective To investigate the association between the six single nucleotide polymorphisms ( SNP),named as rs179247,rsl2101261,rs2284722,rs4903964,rs2300525,rsl7111394 in the intron 1 of thyroid stimulating hormone receptor gene (TSHR) and Graves' disease (GD).MethodsThe genotypes of the six SNPs were genotyped by Taqman probe technique on Fluidigm EP1 platform in 618 GD patients and 646 control subjects.Meanwhile,TSH receptor antibodies (TRAb) of the patients were determined.ResultsAmong the six SNPs,five S NPs were strongly associated with GD,with the most signals at rs179247_G,rs12101261_C,rs4903964 _G (P=2.85×10-10,OR=1.73,95%CI1.46-2.05;P=1.74×10-10,OR=1.73,95%CI 1.46-2.05;P=2.24×10-10,OR=1.69,95% CI 1.44-1.99 ).The results of logistic regression analysis indicated that rs12101261 and rs4903964 were main susceptibility loci of GD in the intron 1 of TSHR.rs179247_G,rs1210126 1_C,and rs4903964_G were associated with subset of the GD patients with positive TRAb (P=4.24× 10-13,p=5.48× 10-13,P =3.89×10-12 ).Conclusionrs179247,rs12101261,and rs4903964 in TSHR intron 1 were significantly associated with GD in the Chinese Han population from Bengbu city.rs12101261 and rs4903964 were the major susceptibility SNPs associated with GD.TSHR gene may play a main role of susceptibility gene in the subset of GD patients with persistent positive TRAb.

Objective:To investigate the relationship between TP53 gene mutation and overall survival (OS) time of myelodysplastic syndrome (MDS).Methods:Databases, including PubMed, Cochrane Library and Embase were searched for relevant studies published up to 20 October, 2019. The corresponding hazard ratio ( HR) and their 95% confidence interval ( CI) for OS from multivariate Cox proportional hazard models were extracted. The combined HR with their 95% CI was calculated by using fixed or random effect models. Meta-analysis was performed by using Revman 5.3 software. Results:A total of 1 033 patients from 6 studies were enrolled. Meta-analysis results showed that OS time in TP53 gene mutation group was shorter than that in wild type group for patients with MDS ( HR = 2.16, 95% CI 1.52-3.07, P < 0.01). The prognostic risk for post-transplantation patients with MDS ( HR = 2.69, 95% CI 1.63-4.43, P < 0.01) was lower compared with that for patients treated by azacitidine( HR = 2.89, 95% CI 1.37-6.08, P = 0.005). Conclusion:TP53 gene mutation is a risk factor affecting OS of MDS patients.

International Journal of Stem Cells 2022;15(2):136-143
Background and Objectives:
Circulating endothelial progenitor cells (EPCs) participate in vascular repair and predict cardiovascular outcomes. The aim of this study was to investigate the correlation between EPCs and abdominal aortic aneurysms (AAAs).
Methods:
and Results: Patients (age 67±9.41 years) suffering from AAAs (aortic diameters 58.09±11.24 mm) were prospectively enrolled in this study. All patients received endovascular aneurysm repair (EVAR). Blood samples were taken preoperatively and 14 days after surgery from patients with aortic aneurysms. Samples were also obtained from age-matched control subjects. Circulating EPCs were defined as those cells that were double positive for CD34 and CD309. Rat models of AAA formation were generated by the peri-adventitial elastase application of either saline solution (control; n=10), or porcine pancreatic elastase (PPE; n=14). The aortas were analyzed using an ultrasonic video system and immunohistochemistry. The levels of CD34+/CD309+ cells in the peripheral blood mononuclear cell populations were measured by flow cytometry. The baseline numbers of circulating EPCs (CD34+/CD309+) in the peripheral blood were significantly smaller in AAA patients compared with control subjects. The number of EPCs doubled by the 14th day after EVAR. A total of 78.57% of rats in the PPE group (11/14) formed AAAs (dilation ratio >150%). The numbers of EPCs from defined AAA rats were significantly decreased compared with the control group.
Conclusions
EPC levels may be useful for monitoring abdominal aorta aneurysms and rise after EVAR in patients with aortic aneurysms, and might contribute to the rapid endothelialization of vessels.
