1.galE mutant of Campylobacter jejuni exhibits a changed lipooligosaccharide structure

Objective:To explore if galE knock-out mutant of Campylobacter jejuni(CJ) have a changed lipooligosaccharide(LOS) structure.Methods:Lipooligosaccharide from parental strains(CJ HB9313) and mutant strains were isolated and purified.The purified LOS preparations from parental and mutant strains were resolved by Tricine SDS polyacrylamide gel electrophoresis(PAGE),analyzed by silver staining and Western blot,tested for reaction with the ganglioside-binding ligands of cholera toxin(CT).Results:Silver staining showed that the parent strain expressed a LOS molecule of around 5 5 kD,whereas the galE mutant strain expressed a molecule of 4.5 kD.The Western blot showed that the LOS,expressed by galE mutant strain,no longer reacted with antiserum raised against CJ HB9313.Compared with parent strain,the mutant lost reactivity with CT.Conclusion:These results indicate that LOS molecule of galE mutants is truncated and removes epitopes that react with antiserum and ganglioside-like structures.The absence of these structures in the galE mutants may enable the development of a safe,live-vaccine without the possibility of inducing an immune response to host gangliosides.
2.Efficacy of alprostadil injection combined the octreotide for acute severe pancreatitis and influence on the serum levels of TNF-α, IL-6, IL-18, occludin

Objective To research the efficacy of alprostadil injection combined the octreotide for acute severe pancreatitis and influence on the serum levels of tumor necrosis factor-α(TNF-α),interleukin-6,interleukin-18,occludin.Method 68 cases of patients with acute severe pancreatitis from September 2013 to February 2017 in our hospital,according to the treatment method group,34 cases in each group,control group treatmented by octreotide,the research group based on the control group treatmented by alprostadil injection,both groups was treated for five days.Clinical curative effect,alleviate clinical symptoms time,TNF-α,IL-6,IL-18,occludin,and adverse reactions occur was compared between two groups.Results The total effective rate of research group was higher than the control group(97.07%vs.79.41%,P<0.05).Bowel sounds,stomach ache,body temperature and serum amylase remission time of research group was shorter than control group(P<0.05).TNF-α,IL-6,IL-18 of research group was lower than the control group(P<0.05),the occludin of research group was higher than the control group(P<0.05).The adverse reactions was no differences between the two groups.Conclusion The exact effect of alprostadil injection combined the octreotide for acute severe pancreatitis,improve serum levels of TNF-α,IL-6,IL-18,occludin.
3.Analysis of processing principle of Fructus Schisandrae Sphenantherae by steaming with wine

AIM: To study the processing principle of Fructus Schisandrae Sphenantherae steamed with wine. METHODS: The content of volatile oil was measured by means of volatile oil measuring method and identified by TLC. The total lignans was determined by spectrophotometry and the water-soluble extractive was measured by determination of extractive method. RESULTS: The volatile oil content in Fruetus Schisandrae Sphenantherae by steaming with wine decreased by 35.3%, TLC showed that a new spot was added; the lignans content had an increase of 28.9% and the water extractive was 2% less than in crude drug. CONCLUSION: Tranditional process is reasonable.
4.Changes of subfamilies of mitogen-activated protein kinases in recovery process of acute renal ischemia/reperfusion injury

AIM: To investigate the change and the possible role of mitogen-activated protein kinase(MAPK) subfamilies in early recovery process following acute renal ischemia/reperfusion injury. METHODS: Ischemia/reperfusion renal injury model was made by placing an atraumatic vascular clamp in renal pedicel. The morphologic change was observed by transmission electron microscope. The proliferating cell nuclear antigen(PCNA) positive renal cells were detected by immunohistochemistry. Extracellular signal-regulated kinase (ERK) and c-Jun NH 2-terminal kinase(JNK) activity was assayed by specific substrate phosphorylation with immunoprecipitation. RESULTS: After acute ischemia, microvilli in renal tubular cells appeared again after 2h of reperfusion. At the same time, the PCNA-positive cells were initially increased. ERK activity decreased at 45 min of ischemia, and completely recovered at 5 min of reperfusion. JNK activity was not influenced by ischemia, but increased at 5 min of reperfusion, reaching its maximal activity at 20 min of reperfusion, and prolonged within 2h reperfusion. CONCLUSION: After renal ischemia/reperfusion injury, the early recovery of renal tubule damage was related to the changes of MAPKs in which the increase of JNK activity might be more important.

7.Application of improved enema in low-dose retention enema for the treatment of ulcerative colitis

Objective To investigate the efficacy of low-dose retention enema for treating ulcerative colitis.Methods Eighty patients diagnosed with chronic recurrent ulcerative colitis were randomly divided into control group and observation group with equal number.The retention enema was used in the control group and the improved retention enema was used in the observation group. Enema was done once every night,10 days as a course of treatment in both groups.The two groups were compared in terms of liquid retention time,curative effect after two courses and patients’satisfaction degree with enema.Results The liquid retention time in the intestinal lumen in the observation group was significantly longer than that of the control group.The efficacy was significantly better than the control group.Patients satisfaction with enema in the observation group was significantly higher than that of the control group (P<0.05).Conclusions The improved low-dose liquid enema is effective in the treatment of ulcerative colitis for its liquid retention time in the intestines.It is effective,comfortable,convenient,and likely to be accepted by patients.
8.Effect of swimming and caressing on neonatal jaundice and weight

Objective To explore the effect of swimming and caressing on neonatal jaundice and weight.Methods Two hundred and forty newborns were divided equally into control and observation group with random digits table.The former received care with caressing and the latter with swimming and caressing.The two groups were compared in terms of weights and jaundice indices at birth and 7d after birth.Results Seven days after birth,the jaundice index in the observation group were lower than that of the control group and the weights of the observation group were larger(P<0.05).Conclusion Swimming and caressing are helpful for the development of neonates by C-sect and the reduction of jaundice duration.
9.Quality standard for prepared pieces of Rhizoma Panacis Majoris

AIM: To establish quality standard for prepared pieces of Rhizoma Panacis Majoris. METHODS: Traditional identification and TLC were used for qualitative analysis of prepared pieces. The contents of ginsenoside Ro and chikusetsusaponin IVa were determined by HPLC. Water,ash and acid-insoluble ash were also detected. RESULTS: The average content of ginsenoside Ro was 7. 19% ,chikusetsusaponin IVa was 3. 16% from different habitants. CONCLUSION: The established method has been proved to be simple,stable,accurate,and can be applied to the determination of ginsenoside Ro and chikusetsusaponin IVa in Rhizoma Panacis Majoris. The quality control standards for Rhizoma Panacis Majoris is normative and the result is accurate.
10.Analysis of infant-mother attachment and the related influencing factors

BACKGROUND: Attachment reflects the early social experience of infant, which plays an important role in later child development. Although most of the attachment develops between infant and mother, some may occur between infant and care giver or others who have close relationship with it.OBJECTIVE: To analyze the infant-mother attachment patterns and the factors related to infant attachment.DESIGN: It was a sampling investigation.SETTING: Department of Maternal and Child Health, School of Public Health, Sun Yat-sen University.PARTICIPANTS: Totally 75 infants and their mother were selected randomly for this study in the Department of Child Health in Guangdong Maternal and Child Health Hospital from August to October 2002.METHODS: Strange situation test (SST) was performed by 6 researchers who were trained systematically. They watched the videos collectively and classified the patterns of infant attschment based on their performance. The internal consistency coefficient was 0.90. For those inconsistent assessments, the video should be reviewed and discussed to get an accordant conclusion. In addition, a self-designed maternal questionnaire was used to investigate the general condition of the infant and the familial information.MAIN OUTCOME MEASURES: ①Distribution of infant attachment patterns. ②Analysis of the risk factors related to infant attachment.RESULTS: All the 75 infants and their mother entered the statistical analysis. ①Distribution of the infant attachment patterns: In 75 infants, secure attachment was 65% and insecure attachment was 35%, among which,insecure-indifferent type was 18%, insecure-importunate type was 13%,and insecure-disorganized type was 4%. ②Analysis of the risk factors related to infant attachment Those infants with a younger age, a poor approachability (the response to strangers, new environment and objects), a closer relationship with caregiver and a more inconsistent education from the family members, are prone to develop insecure attachment.CONCLUSION: Secure attachment is dominant in infants. The security of attachment is related to the maturity and personality traits of the infants,the relationship between infant and caregiver and the educational approach for them.
