Group A rotaviruses (RVs) are major pathogens associated with acute gastroenteritis in young children and animals worldwide. VP4 is responsible for interaction with the host and viral attachment. Recent study showed that the distal portion of rotavirus (RV) VP4 spike protein (VP8*) is implicated in binding to human histo-blood group antigens (HBGAs), which is new cellular receptors on rotavirus, Published in Nature and Journal of Virology in 2012. The paper describes advances in correlation between rotaivrus and HBGAs, summarizes the main achievements has gotten, Clarify the significance of study on Rotaivrus and HBGAs.

2.Dosage Regimen Design of Aminophylline Using Pharmacokinetic Parameters

OBJECTIVE: To design the dosage regimen of aminophylline using reported pharmacokinetic parameters (mean group value). METHODS: The calculation of oral maintain dose, the infusion rate of intravenous drip maintain dose and the loading dose of intravenous drip were introduced. RESULTS: Different dose were required for different pathophysiological conditions insteading of aminophylline 200 mg three times a day (p.o.) for every patient. CONCLUSION: It is recommended that all hospitals which are unable to determine plasma concentration should design dosage regime of aminophylline using pharmacokinetic parameters.
3.The characteristics of PCNA expression in hypertrophic scars and chronic ulcers and implication of the expression.

OBJECTIVETo investigate the characteristics of PCNA expression in hypertrophic scars and chronic ulcers and to discuss its relation to their formation.
METHODSThe expressive quantity and sites of PCNA were detected with the immunohistochemical SP method.
RESULTSPCNA was expressed in all samples. The expressive quantity in hypertrophic scars was higher than chronic ulcers(P < 0.01). The expressive sites of all samples were in the nucleus of fibroblasts and capillary endothelial cells.
CONCLUSIONSThe expressive quantity of PCNA was more in hypertrophic scars and less n chronic ulcers. The quantitative difference of expression between hypertrophic scars and chronic ulcers may be correlated to their formation.

This study assessed the effects of leukemia-related protein 16 (LRP16) on the regulation of pancreatic functions in mouse insulinoma (MIN6) cells. Cells with down-regulated expression of LRP16 were obtained by a shRNA interference strategy. Insulin content and glucose-stimulated insulin secretion (GSIS) were examined by radioimmunoassay. Western blotting was applied to detect protein expression. Glucose-stimulated sub-cellular localization of PDX-1 was immunocytochemically determined. Cell proliferation and apoptosis were detected by flow cytometry. Our results showed that LRP16 regulated insulin content in MIN6 cells by controlling expression of insulin and insulin transcription factors. LRP16 gene silence in MIN6 cells led to reduced cell proliferation and increased apoptosis. The observation of phosphorylation of serine-473 Akt and the localization of PDX-1 to the nucleus under glucose-stimulation exhibited that LRP16 was a component mediating Akt signaling in MIN6 cells. These results suggest that LRP16 plays a key role in maintaining pancreatic β-cell functions and may help us to understand the protective effects of estrogen on the functions of pancreatic β-cells.

7.Associations of SUMO4 polymorphisms with tacrolimus concentrations in Chinese renal transplant recipients.

The study aims to investigate the associations of SUMO4 polymorphisms with tacrolimus concentrations in Chinese renal transplant recipients. Blood samples and clinical data were collected from 132 renal transplant recipients with tacrolimus treatment. CYP3A5*3 genotypes were detected by PCR-RFLP, and SUMO4 (rs237024, rs237025) genotypes were detected by Sequenom® MassARRAY system. SUMO4 rs237024 and rs237025 genotypes were in complete linkage disequilibrium (D' = 1). The dose-adjusted concentration of tacrolimus in SUMO4 rs237024A-rs237025A (GA-GA +AA-AA) carriers was considerably higher than that in GG-GG carriers (P < 0.05). After stratification by CYP3A5*3 genotypes, SUMO4 rs237024A-rs237025A carriers (GA-GA+AA-AA) had a higher dose-adjusted tacrolimus concentration than that in GG carriers in CYP3A5 expresser (P < 0.05). The results illustrated that SUMO4 rs237024 and rs237025 polymorphisms were associated with tacrolimus concentrations, and the test of these genotypes may be useful for individualized medicine of tacrolimus.

Rotavirus is the leading causal agent of severe acute gastroenteritis in children aged <5 years. A specific pharmacologic agent for the treatment of rotavirus-infected children is lacking. In China, only the Luo Tewei oral vaccine (Lanzhou Institute of Biological Products, Shanghai, China), which is produced from Lanzhou lamb rotavirus vaccine (LLR), is available. Studies have hypothesized that the genotype of LLR is G10P[12], To identify the genotype of LLR by reverse transcription-polymerase chain reaction, we showed that the VP7 and VP4 genotypes of LLR were G10 and P[15], respectively, based on sequencing, alignment and phylogenetic analyses. In conclusion, we identified the genotype of rotavirus strain LLR to be G10P[15].

9.Synthesis and characterization of PEG-b-(PG-g-PEI) for gene delivery.

OBJECTIVETo synthesize a biodegradable non-viral gene carrier with a high transfection efficiency and a low cytotoxicity.
METHODSPoly(ethylene glycol)-block-(poly(L-glutamic acid)-graft-polyethylenimine) was prepared via ammonolysis of poly(ethylene glycol)-block-poly (γ-benzyl L-glutamate) with the low-molecular-mass polyethylenimine (600 Da). The synthesized copolymer was characterized by 1H nuclear magnetic resonance spectroscopy and gel permeation chromatography. The polyplex micelle from PEG-b-(PG-g-PEI) and plasmid DNA (pDNA) was studied using dynamic light scattering, zeta-potential measurements, and gel retardation assay. The in vitro cytotoxicity and transfection efficiency of PEG-b-(PG-g-PEI) were tested by MTT assay and luciferase assay in HEK 293T cells using PEI (25 kDa) as the control.
RESULTSPEG-b-(PG-g-PEI) could efficiently condense DNA into nanosized particles with positive surface charges when the N/P ratio of polymer and DNA was above 5:1. The zeta potential of the polyplexes was about 25 mV, and the particle size was 120 nm at a N/P ratio of 10. The cell toxicity and gene transfection evaluations showed a lower cytotoxicity and a higher gene transfection efficiency of the copolymer than PEI 25000 in HEK 293T cells.
CONCLUSIONSThe polymer can be used as a potential non-viral gene carrier for gene therapy.

10.mRNA and protein expression of transcription factor c-fos in burned rats and their effects on wound healing.

OBJECTIVE: To explore the expression of mRNA and its protein in burned rats and their effects on burn wound healing. METHODS: A partial-thickness burn of 30% total body surface ar ea was created on the back of 40 Wistar rats. In situ hybridization and immunohi stochemical methods were used to evaluate the location and the amount of the c-fos mRNA and its protein in normal skin and the burned skin, respectively, at 3 h, 6 h, 1 d, 3 d, 7 d and 14 d after burn. RESULTS: Under a light microscope, both the expression of c-fo s mRNA and its protein could be found in the normal skin, but their induction le vels were much higher in the burned skin. The level of fos protein expression reached peak at 3 h after burn while that of c-fos mRNA reached peak at 6 h aft er burn. CONCLUSIONS: The expression of c-fos can be induced by burns. And the peak level expression of c-fos mRNA comes later than that of c-fos p rotein. It indicates that the action of fos protein is induced by post-translat ional modification of pre-existing fos molecules.
