1.Effects of community smoking cessation supervision on lung function and quality of life in old COPD patients

Objective To investigate the effects of community smoking cessation supervision on lung function; acute exacerbation frequency and quality of life in elderly patients with chronic obstructive pulmonary disease (COPD). Methods Forty-five COPD patients were divided into the intervention group(23 cases) and the control group(22 cases).The control group was given routine drug treatment.While the intervention group was given community smoking cessation supervision based upon routine treatment.The variation of lung function, quality of life (SGRQ inventory) and acute exacerbation times during the year were compared. Results The variation of lung function and quality of life improved greatly after in-tervention group compared with those before intervention, P<0.05. Conclusions Community smoking cessation supervision was a feasible measure to improve lung function and quality of life and reduce acute exacerbation in COPD patients.
2.Transcriptional activation function of intracellular domain of EPOR and G-CSFR protein in yeast: a comparative study

Objective To investigate the feasibility of searching for proteins which interact with intracellular domain of hematopoietic growth factor receptors using yeast two-hybrid system. Methods RT-PCR method was performed to amplify the genes of intracellular domains of G-CSF receptor and EPO receptor in NFS-60 and BET-2 cells of mice. The genes were cloned into yeast expression plasmid pGBKT7 vector,and then transformed into yeast AH109. The yeast proteins were isolated and analyzed with Western blotting. Transcriptional activation was analyzed by the ?-galactosidase colony-lift filter assay. Results The intracellular domains of G-CSF receptor and EPO receptor genes were successfully cloned into pGBKT7 vector. The results of Western blotting assay showed that both proteins were expressed in the yeast cells. The ?-galactosidase colony-lift filter assay demonstrated that G-CSF receptor alone had no activity of transcriptional activation,while the EPO receptor alone could activate transcription. Conclusion The findings suggested that intracellular domain of G-CSF receptor could be used as a bait to find interacting proteins using yeast two-hybrid system,while that of the EPO receptor could not. Therefore the system could not be applied to all hematopoietic factor receptor.
3.Preparation of monoclonal antibody against human lung cancer and purification of its antigen by the immunoaffinity chromatography

Objective:To produce the mcAb specifically reacting with lung cancer and to purificate its antigen.Methods:The mice were immunized with A549, the mcAb 2B9 was screened by indirect cell ELISA and immunohistochemistry, and its antigen was purificated by immunoaffinity chromatography.Results:A mcAb was obtained, which could react to lung cancer but very little or not to normal lung tissue and other caner tissues, and the antigen of the mcAb was purificated from the cell lysate.Conclusion:A mcAb which can react to lung cancer have been obtained and its antigen was purificated, they may be useful on clinic for diagnosis and prognosis of lung cancer.
4.Population analysis as a useful method for isolation of herogeneous methecillin-resistant staphylococcus aureus

Objective To develop a new method for the isolation of heterogeneous methecillin-resistant staphylococcus aureus(hetero-MRSA)and study its application in clinical practice.Methods Both the methods of disc agar diffusion and population analysis were used to detect hetero-MRSA from 60 strains of methecillin susceptible staphylococcus aureus(MSSA) which were identified by VITEK AMS.Results Twelve subclones of hetero-MRSA were isolated by population analysis,while only 2 subclones were identified by disc agar diffusion.Conclusion Population analysis is an effective method for the isolation of hetero-MRSA,and its detection rate is higher than that of disc agar diffusion.The heterogeneous methecillin resistance of MSSA should not be overlooked by medical workers.
5.Feasibility of transplanting bone marrow mesenchymal stem cells into coronary artery

Objective To investigate the retainage rate,distribution,and emigration of the bone marrow mesenchymal stem cell(BMMSC) after transcoronary infusion and to further evaluate the feasibility of injecting BMMSCs into coronary artery.Methods BMMSCs were isolated,purified,expanded,and labelled with CM-DiI.The infarcted SD rat hearts were removed and perfused with Langendorff apparatus.The cells were injected into the aortic root and the fluid returning from the coronary system was collected and the labelled cells in the coronary effluent were quantitated with flowcytometry.At the same time,left ventricle function was recorded to evaluate the safety of this approach.In vivo study,the cells were then injected into clamped ascending aorta through a catheter inserted through the left ventricle into the aortic root.The hearts were harvested at different time points after cell transplantation to obtain the direct evidence of distribution and emigration of the implanted cells.Results Only 3%~5% of transplanted cells returned into the right ventricle and more than 90% cells retained in the heart after beinginjected into aortic root of Langendorff model of infarcted hearts of SD rats.Left ventricle function did not deteriorate after cell transplantation.The labelled cells were entrapped within the coronary capillary immediately after cell infusion,mainly in the normal area.After 24 hours some cells migrated through the capillary wall into interstitium of the heart.One week later we found that most survival cells located at the infarcted area and the border zone.Conclusion The majority of BMMSCs delivered by transcoronary infusion retained in the heart.BMMSCs can penetrate the vessel wall and home back to the interstitial compartment and the injured area in a few hours.
6.Shift handover experience of ICU nurses:a qualitative study

Objective To study the shift handover experience of ICU clinical nurses. Method About 19 ICU nurses were enrolled in the investigation using semi-structured interviews and the results were analyzed with phenomenological analysis. Result Their shift handover experience were summarized into 6 themes, that is high recognition on the importance of shift handover, lack of standardized processes, incomplete contents of shift handover, frequent interruption, forgotten information and repeated information. Conclusions Shift handover is an important part of ICU nursing function but there are many problems in the practical operation. The nurse managers need to standard the handover process, stipulate handover contents and enhance the training on shift handover with specialist examination and positive results for the purpose of improving the quality of the shift handover.
7.Effect of Humanistic Care for Medical Care Integration for Mild Cognitive Impairment Elder Patients

Objective:To evaluate the effect of humanistic care for medical care integration on quality of life in elder patients with mild cognitive impairment. Method:Seventy patients with mild cognitive impairment in our hos-pital were randomly divided into experimental group ( n=35 ) and control group ( n=35 ) using random number ta-ble. The patients in the control group were given routine care, while those in the experimental group were given hu-manistic care for medical care integration and home visiting once a month besides routine care. All the patients were evaluated by the Montreal Cognitive Assessment Scale ( MoCA) , Activities of Daily Living ( ADL) , and the MOS Item Short from Health Survey ( SF-36 ) during hospitalization, three months and six months after hospital discharge. Result:Three and six months after hospital discharge, the scores of these three scales of all subjects in two groups were significantly higher than those during hospitalization ( P<0 . 05 ) . The scores of the subjects in the experimental group also significantly higher than those of the control group, and the differences were statistically significant (P<0. 05). Conclusion: For the elder patients with mild cognitive impairment, humanistic care for medical care integration can significantly improve the cognitive function, self -care ability and quality of life. Therefore, it is worthy using in clinical treatment.
8.Nursing risks in patients undergoing radio-frequency ablation during cardiac valve replacement

Objective To investigate the nursing risks in the patients undergoing radio-frequency ablation surgery during cardiac valve replacement.Methods The nursing risks in 110 patients undergoing radio-frequency ablation surgery during cardiac valve replacement were summarized to include disorder of water electrolyte and acid-base balance, arrhythmia,hypothermia,skin ulcer,reduction of effective circulating blood volume,infection and so on, so as to carry out the corresponding nursing measures. Results Ventricular fibrillation occurred in 11 patients,hypothermia after rewarming on cardiopulmonary bypass in 31 patients,left atrial bleeding after aortic de-clamping in 1 patient.All the complications were successfully controlled by corresponding management. The operation time ranged from 178 mins to 310 mins and average time was(245.32±34.40)mins.Conclusion The full-scale assessments and effective nursing interventions are the key points to improve the efficiency and success of cardiac valve replacement combined with radio-frequency ablation.
9.Expression of osteopontin in local tissues of patients with cervical cancer

Objective To detect the expression of osteopontin (OPN) in cervical local tissues of patients with chronic cervicitis, cervical intraepithelial neoplasia (CIN), cervical cancer. To investigate the possibility that OPN is used as tumor marker of cervical cancer. Methods The expression of OPN in 25 cases of chronic cervicitis, 18 cases of CIN and 21 cases of cervical cancer was detected by immuohistochemistry SP method. Results The positive expression rates of OPN in the chronic cervicitis, CIN and the cervical cancer had a rising trend (48 %, 77.78 % and 80.95 %, respectively) and their differences had statistical significant (P <0.05). The OPN expression level in cervical cancer had no statistical correlation with age, tumor size, clinical stage and other clinical pathological features (P >0.05). Although OPN expression level in cervical cancer wasn' t related with tumor clinical stage, positive expression rates of OPN had a gradually increased trend along with the clinical stage. Conclusion High expression of OPN is found in cervical cancer and may be related to the occurrence and development of cervical cancer.
10.Expression of CD_(44) in patients with nephrotic syndrome and its clinical significance

Objective To study the expression of adhesion molecule glycoprotein(CD_(44)) in children with primary nephrotic syndrome(NS) and its clinical significance.Methods The expression of CD_(44) in peripheral blood was detected in 28 children with NS by flow cytometry,controlled by 20 cases of healthy children.Results The CD_(44) expression level of NS in active stage was significantly higher than that of the control(P
