1.Non-viral vector-mediated high efficiency expression of manganese superoxide dismutase gene in mouse liver
Chinese Journal of Pathophysiology 2000;0(11):-
AIM: To investigate the transfection efficiency of mouse liver with non-viral vector containing manganese superoxide dismutase (Mn-SOD) gene. METHODS: The eukaryotic expression vector, gWiz/Mn-SOD, encoding human manganese superoxide dismutase was constructed. The plasmids of gWiz/Mn-SOD were mixed with cationic lipids, followed by injection into mice via branch of superior mesenteric vein, to induce Mn-SOD over-expression in murine liver detected by RT-PCR, Western blotting, SOD activity and immunohistochemical staining. RESULTS: gWiz/Mn-SOD transfection resulted in the obvious expression of exogenous Mn-SOD mRNA and protein in hepatic tissues at 8 hours after injection, and elevated mitochondria SOD activity 8.4 times in transfected hepatocytes than that in non-transfected cells at 72 hours after injection. It was showed that nearly 70% of mouse hepatocytes was obviously Mn-SOD positive after transfection. CONCLUSION: High expression efficiency of Mn-SOD gene in mouse liver is achieved safely by injection of gWiz/Mn-SOD and cationic lipid mixture into branch of superior mesenteric vein. [

Chinese Journal of Physical Medicine and Rehabilitation 2011;33(2):128-131
Objective To predict the prognosis after acute cerebral infarction using a combination of indicators. Methods Two hundred and seventeen patients with acute cerebral infarction admitted from October 2005 to December 2008 were studied. Logistic regression analysis of the data from 112 of the patients admitted from October 2005 to March 2007 was used to select 20 indicators for study. The indicators were combined into prognostic indexes using a multi-layer perception (MLP) neural network (NN) model. Data on the subsequent 105 patients were usedto appraise the model. Results The agreement of the prediction results of the NN model with the real recovery observations was rated as "excellent" in 39 cases, "OK" in 27 and "bad" in 32. The sensitivities were 95.1%,87.1% and 96.9% respectively. On average, the differences between the predicted results with the NN model andthe real recovery were not significant. Conclusion The NN model delivered good precision in predicting the outcome of acute cerebral infarction and it is worthy of further investigation.

Chinese Journal of Digestive Surgery 2008;7(3):206-208
Objective To determine the impact of exogenous vascular endothelial growth factor (VEGF) on rat liver function after partial liver transplantation. Methods Male SD rats which had undergone 30% partial liver transplantation were divided into VEGF treatment group (n=60) and control group (n=60). Animals in each group were injected with solution of VEGF or saline, respectively. Serum ALT, AST, and hyaluronic acid (HA) levels were assessed at 0, 12, 24, 72, 168 hours after operation. Results Serum ALT, AST and HA levels were significantly decreased at 72 hours after transplantation in VEGF treatment group compared with controls (t =4.681, 4.252, 2.853, P<0.05). In accordance with liver function, treatment with VEGF minimized the damage to the liver sinusoidal endothelial cells and decreased the infiltration of inflamatory cells. Conclusion Administration of VEGF may improve the liver function of rats after partial liver transplantation by maintaining the integrality of the sinusoidal structure and reducing the inflamation reaction.
