1.Advance in DNA tumor vaccine

DNA vaccine is one of the hot spot in the research of vaccine now. DNA tumor vaccines mainly include tumor-associated-antigens-based completed, epitope, idiotope determinants DNA vaccine,fusion DNA vaccines, RNA self-replicating vaccines, dendritic cell-based tumor vaccines etc. The recent developments are discussed.
2.Fatigue Self-assessment Scale(FSAS)

Fatigue Self-assessment Scale(FSAS)was developed on the basis of review on the study of fatigue assessment in both China and overseas countries and considering the Chinese culture background.The scale consists of 23 items and is used to assess the type and severity of fatigue(including three subscales of measuring physical fatigue,mental fatigue and the consequences of fatigue) and characteristics of fatigue(including three subscales of measuring responsiveness of fatigue to sleep/rest,the situation specifi city of fatigue and time mode of fatigue) among the people of sub-health and various diseases with fatigue.It may also be used to assess intervention effectiveness of fatigue.
3.Clinical characteristics of congenital long QT syndrome families

Objective To elucidate the clinical manifestati on s and electrocardiogram characteristics of congenital long QT syndrome families and try to find out the genotype of the long QT syndrome(LQTS) patients. Methods The routine clinical check up and ECG recordings we re done for the 3 family members. Both QT interval and QTc were measured. Diagno stic criteria for LQTS were defined by Schwartz. Results Fifteen family members were identified as with LQTS and 11 members with intermediate probability to LQTS. The clinical manifestatio ns and ECG characteristics were different from each other. Conclusion The clinical manifestations and ECG characterist ics of LQTS patients from family 1,family 2 and family 3 correspond with LQT2, L QT1 and LQT3, which is caused by HERG,KVLQT1 and SCN5A gene mutation.
4.Purification of monoclonal antibody against PGRP and its activity for small cell lung cancer in vitro

Objective To explore the effect of purification on monoclonal antibody (MAb) against PGRP by Protein A-Sepharose affinity chromatography, and to provide some based data for the purification of other antibody using the same method. Methods The ascites which include MAb was purified by Protein A-Sepharose affinity chromatography. The purity and activity of MAb was tested by SDS-PAGE and ELISA. The biological function was identified by flow cytometer and immunohistochemistry. Results The average concentration of protein in ascites before purification is 23.62 mg/ml. Before and after purification, the total protein is 148.79 mg and 146.67 mg, respectively. The recovery coefficient of protein is 98.58%. The concentration of MAb in ascites is 5.21 mg/ml averagely. The MAb purity is more than 95 %. The immunoactivity of purified antibody is higher than that of unpurified antibody. Conclusion The purity of MAb against PGRP purified by Protein A-Sepharose affinity chromatography is very high. The immunoactivity of purified antibody is higher than that of unpurified antibody. So the ProteinA-Sepharose affinity chromatography is a rapid, convenient and reliable method for the purification of MAb Against PGRP.
5.Clinical characteristics of 5 Chinese LQTS families and phenotype-genotype correlation.

In order to assess the clinical manifestations and electrocardiogram (ECG) characteristics of Chinese long QT syndrome (LQTS) patients and describe the phenotype-genotype correlation, the subjects from 5 congenital LQTS families underwent clinical detailed examination including resting body surface ECG. QT interval and transmural dispersion of repolarization (TDR) were manually measured. Five families were genotyped by linkage analysis (polymerase chain reacting-short tandem repeat, PCR-STR). The phenotype-genotype correlation was analyzed. Four families were LQT2, 1 family was LQT3. Twenty-eight gene carriers were (14 males and 14 females) identified from 5 families. The mean QTc and TDRc were 0.56 +/- 0.04 s (range 0.42 to 0.63) and 0.16 +/- 0.04 s (range 0.09 to 0.24) respectively. 35.7% (10/28) had normal to borderline QTc (< or = 0.460 s). There was significant difference in QTc and TDRc between the patients with symptomatic LQTS and those with asymptomatic LQTS, and there was significant difference in TDRc between the asymptomatic patients and normal people also. A history of cardiac events was present in 50% (14/28), including 9 with syncope, 2 with sudden death (SD) and occurred in the absence of beta-blocker. Three SDs occurred prior to the diagnosis of LQTS and had no ECG record. Two out of 5 SDs (40%) occurred as the first symptom. Typical LQT2 T wave pattern were found in 40% (6/15) of all affected members. The appearing-normal T wave was found in one LQT3 family. Low penetrance of QTc and symptoms resulted in diagnostic challenge. ECG patterns and repolarization parameters may be used to predict the genotype in most families. Genetic test is very important for identification of gene carriers.

Now syndrome study becomes the important research content in our country' TCM field,normalization of the symptoms standardization of syndromes especially becomes scientific research focal point in this field,furthermore some studies exists the conditions of symptoms short of norm and standards of syndrome differentiation discordant,as well stagnation of liver-QI with deficiency of the spleen is clinical common syndrome,so this article carried out all round analysis and investigations on the modern literature of stagnation of liver-QI with deficiency of the spleen syndrome recent ten years,to aim at analyzing the symptom characteristic and standards of syndrome differentiation on stagnation of liver-QI with deficiency of the spleen syndrome,to provide the bases of standardization and normalization of stagnation of liver-QI with deficiency of the spleen syndrome.

8.Study on elderly mental sub-health status and influencing factors of the three gorges reservoir

Objective To investigate the status of psychosomatic health on the aged in reservoir area of three gorges .Study men-tal sub-health population distribution and the related influencing factors of the aged .Methods 604 elderly of reservoir area of three gorges were investigated by using self-made general questionnaire ,Cornell Medical Index(CMI) ,Eysenck Personality Questionnaire-R Short Scale(EPQ-RSC) ,Social Support Rating Scale(SSRS) ,Activities of daily living (ADL) ,Memorial University of Newfound-land Scale of Happiness(MUNSH) .Results 14 .7% aged′s mental health was in the sub-healthy status .Older age ,low education level ,no spouse ,urban elderly people were higher in mental sub-healthy .There was significant difference between mental sub-healthy group and mental healthy group in the aspect of personality 、social support、subjective well-being、activities of daily living (P<0 .05) .Conclusion The elderly psychosomatic health status of the Three Gorges Reservoir area is good ,with low incidence of mental sub-health ,their psychosomatic health more affected by the state of mind .
9.Posterior short-segment instrumentation without fusion for severe thoracolumbar burst fractures

Objective To evaluate the clinical efficacy of treatment of severe thoracolumbar burst fractures by posterior short-segment instrumentation without spinal fusion and assess radiographic imaging and function recovery after surgery.Methods Thirty-eight patients with severe monosegmental thoracolumbar burst fractures treated between July 2011 and March 2013 were analyzed retrospectively.Operation procedures were posterior short-segment pedicle screw distraction reduction and fixation combined with screw insertion to the injured vertebrae and calcium sulphate augmentation.In addition,there was no need for posterolateral interbody fusion.X-ray and CT were performed before and after operation to evaluate local kyphotic angle,anterior fractured vertebral body height and canal encroachment.Visual analogue scale (VAS) and Oswestry disability index (ODI) were assessed before and after operation as well as in follow-up.Results All patients were followed up for average 14 months (range,3-20 months).Local kyphotic angle was (21.2 ±4.3)° before operation,(3.5 ± 1.8)°immediately after operation,and (4.8 ± 2.7) ° in final follow-up.Relative anterior vertebral height was (54.8 ± 14.6)% before operation,(91.7 ± 8.0)% after operation,and (87.2 ± 6.0)% in final follow-up.Mean canal encroachment was (48.0 ± 4.5)% preoperatively,(23.8 ± 7.8)%postoperatively,and (8.8 ± 4.6) % in final follow-up.In final follow-up,six patients with American Spinal Injury Association (ASIA) grade C on admission showed improvement to grade D (n =2) and grade E (n =4) ; 10 patients with ASIA grade E on admission showed improvement to grade E; 22 patients with grade E had no changes.ODI and VAS scored 15.5 ±8.8 and 2.3 ±0.8 in final follow-up with substantial improvement from those before operation (P < 0.01).Complications from internal fixation were not found during follow-up.Conclusion Posterior short-segment fixation without fusion is one of the foremost effective methods for severe thoracolumbar burst fractures,for it can effectively restore the sagittal spinal alignment and the fractured vertebral body height.
