1.The Effect of Sub-glottal Convergence Angle on the Degree of Glottal Closure

Objective To study the effect of sub-glottal convergence angle on the degree of glottal closure, and to analyze the potential mechanism of dysphonia by the incomplete glottal closure.Methods Three vocal fold positions (adduction, intermediate and abduction) were evaluated by the degree of glottal closure, and divided into three groups.The neck CT images of the subjects were gathered.The vocal folds were adduction group when the normal subjects made a sustaining phonation /i:/ during CT scanning;the vocal folds were abduction group when the normal subjects made a deep breathing during CT scanning, and the unilateral vocal folds were incomplete closing group when the unilateral vocal fold paralysis subjects made a deep breathing during CT scanning.3D models of the vocal folds and the airway were reconstructed using Mimics software, through which the sub-glottal convergence angle was measured.Using one-way factor analysis of variance, we compared the angle among three groups.Results The angle changed with degree of glottal closure, with statistically significant differences among the three groups(P<0.01).The means of the angle in three groups were 33.49°±3.75°, 55.03°±2.61° and 75.02°±7.32°.Conclusion The 3D model generation from CT data is an effective method of measuring the sub-glottal convergence angle, while the angle changes with the degree of glottis closure.The angle may affect sub-glottal pressure distribution in the sub-glottal shear or normal direction and influence vocal fold vibration, which would make vibration and vocal fold mucosal wave generation more difficult, and might cause dysphonia.
2.Histological observation on the effects of TGF-β3 and dental pulp stem cells in repair of rabbit facial nerve injury

Objective To investigate the feasibility of combing the application of transforming growth factor-β3 (TGF-β3) with concentration of 100 ng/μl and dental pulp stem cells (DPSCs) for recovering rabbit facial nerve transverse trauma.Methods Thirty-six healthy adult Zelanian rabbits of clean-grade were selected and randomly divided into group DPSCs+TGF-β3 (experimental group), group TGF-β3 (control group 1) and group PBS (control group 2) with 12 rabbits in each group.The operations for all three groups were applied at rabbit's left cheek.A model of traumatic transection was set on upper buccal branch, then 100 ng/μl TGF-β3 solution and 0.1 ml of 1 ×108/L DPSCs suspension were added into regeneration chamber for the experimental group, while the same amount of 100 ng/μl TGF-β3 solution was added for group TGF-β3 and the same amount of PBS for group PBS.The recovery of facial nerve regeneration with the prepared animal's specimen was evaluated in the 1st, 4th and 12th week after the operation on sacrificed rabbits.Results The effects on nerve regeneration recovery for the experimental group was superior to that of control groups 1 and 2 with all the 36 models included in the result analysis, and that of control group 1 was superior to that of control group 2, which was getting better with the extension of time.Conclusions The combined application of TGF-β3 and DPSCs can effectively promote the facial nerve regeneration, which is better than that of single application of TGF-β3.Meanwhile, the effect with TGFβ3 application is better than that with application of PBS.
3.Effect of Aging on Activity of H+-K+-ATPase in Gastric Fundal Mucosa

Background:It has been demonstrated that H + -K + -ATPase expression in human parietal cells had a tendency to increase with aging. However,the effect of aging on activity of H + -K + -ATPase is still unclear. Aims:To investigate effect of aging on activity of H + -K + -ATPase. Methods:Fifteen male Sprague-Dawley(SD)rats were divided into 4,21,24, 27 and 30 months group,and 19 healthy beagle dogs were divided into younger group,junior elderly group and senior elderly group. The activity of H + -K + -ATPase in gastric fundal mucosa was assessed. Results:The activity of H + -K + -ATPase in gastric fundal mucosa in 4,21,24,27 and 30 months old rats were(4. 850 ± 0. 312)μmol·mg - 1 ·h - 1 , (5. 466 ± 0. 379)μmol·mg - 1 ·h - 1 ,(6. 068 ± 0. 228)μmol·mg - 1 ·h - 1 ,(5. 733 ± 0. 767)μmol·mg - 1 ·h - 1 and (6. 223 ± 0. 428)μmol · mg - 1 · h - 1 ,respectively. With aging,H + -K + -ATPase activity in rats had a tendency to increase(F = 4. 519,P = 0. 031). The activity of H + -K + -ATPase in beagle dogs in younger group,junior elderly group and senior elderly group were(11. 087 ± 4. 320)μmol·mg - 1 ·h - 1 ,(8. 549 ± 3. 250)μmol·mg - 1 ·h - 1 ,(12. 071 ± 2. 820)μmol·mg - 1 ·h - 1 ,respectively. There was no statistically significant difference among the three groups(F =1. 339,P = 0. 290). Conclusions:With aging,the activity of H + -K + -ATPase in rats and beagle dogs does not decline, but even has a tendency to increase.
4.Intervention studies on the community health education among patients with metabolic syndrome

Objective To observe the control efficacy of community health education on population with metabolic syndrome (MS).Methods 56 cases of metabolic syndrome patients treated in the community were selected as the research object from June 2012 to 2013,and were randomly divided into the observation group (30 cases) and the control group (26 cases).The patients in the observation group received systematic community health education,and patients in the control group received regular treatment.The blood pressure,BMI Level,abdominal circumference,FPG level,TC,TG,LDL,HDL,2hPG,and HbAlc level of patients from both groups were compared one year later.Results Compared with the result before treatment,the abdominal circumference,blood pressure,blood glucose and lipid levels of patients in the observation group were all superior to the control group; the reasonableness for food taken was compared one year later,the observation group was also superior to the control group.Statistical significancc cxistcd in the differences between both groups.Conclusions The implementation of community health education has a positive significance in promoting the rehabilitation of people with metabolic syndrome,which is worth being promoted.
5.Establishment of cell models for PXR, FXR and LXRαagonists high-throughput screening based on reporter gene assay

Aim To develop an in vitro high throughput drug screening system based on reporter gene assay for identification of novel compounds with PXR, FXR and LXRα agonist activity. Methods The expressions of exogenous PXR, FXR and LXRαgene in HEK293, HepG2 and LS174T cells were examined by Real-Time quantity PCR. pSG5-hPXR and pGL3-XREM-CYP3A4, pEGFP-N3-hFXR and EcRE-TK-Luc, pCMX-FLAG-hLXRα and pGL3-XREM-CYP3A4 were cotransfected into cells and the optimal ratio of three plasmids was determined. The dose-response relationship between the positive drug and the fold induction was determined. The specificity of the model was ex-amined, and the repeatability was also determined by Z′ value. Results ① The PXR, FXR and LXRα mRNA expression in HEK293 cell is low among three different cells. ②reporter gene vector and expression plasmid ratio of 1∶ 1, 2∶ 1 and 2∶ 1 were proved to be suitable for highest relative luciferase activity for PXR, FXR or LXRα agonist screening model. ③ The relative luciferase activity was induced by Rif, CDCA or T0901317 in a dose-dependent manner. ④Only Rif, CDCA or T0901317 could significantly increase the relative luciferase activity in PXR,FXR or LXRα agonist screening model, no effect of other nuclear re-ceptors agonist was observed, and the values of Z′-factor for PXR, FXR and LXRαagonist screening model were 0. 58, 0. 66 and 0. 63, respectively. Conclusion An in vitro PXR, FXR and LXRα agonist high-throughput screening models are devel-oped with acceptable specificity and repeatability, and the mod-els can be used to screen PXR, FXR and LXRα agonist.
6.Design of POCT quality management system based on the Internet of things

Objective To design a POCT quality management system based on the Internet of things so as to standardize POCT quality management.Methods The existing problems in POCT quality management were analyzed.Taking portable blood glucose meters as an example,the quality management system of POCT-Internet of things was designed based on the all aspects of data acquisition,transmission,server management and storage,etc.Results The POCT quality management system based on the Internet of things and medical cloud technology was designed.The system could implement the management of POCT operators and devices,monitor the realtime quality control data and reports,and facilitate the administrators to analyze the quality problems in the whole process of POCT management.Conclusion The designed POCT quality management system based on the Internet of things has the theory feasibility.
7.Age-associated histological changes of gastric fundal mucosa and ultrastructure of gastric chief cells in human and Beagle dogs

Objective To investigate the effect of aging on gastric pepsinogen secretion through observing histological changes of gastric fundal mucosa and ultrastructure of gastric chief cells in human and Beagle dogs at different ages.Methods Fifty middle-aged and elderly indigestion patients with gastroscopy were selected as study objects and divided into young and middle age group (age 20-59 years,n=19),junior elderly group (age 60-69 years,n=11),middle elderly group (age 70-79 years,n=10),and senior elderly group (no less than 80 years,n=10).In addition,nineteen healthy Beagle dogs were also selected as study objects and divided into young and middle age group (age between 1 to 5 years,n=8),junior elderly group (age six to eight years,n=5),and senior elderly group (no less than 9 years,n=6).The histology and morphology of gastric fundal mucosa of human and Beagle dogs were observed under light microscope.Then the thickness of lamina propria was measured and the number of gastric chief cells was counted.The ultrastructure of gastric chief cells was observed under electron microscope,and the area percentages of secretory granule(also called mucous granule)area in cytoplasm of chief cell was calculated.The data was analyzed by one-way analysis of variance.Results No significant histological and morphological changes of gastric fundal mucosa were observed in human and Beagle dogs at different ages.There was no significant difference in the thickness of lamina propria among different ages in human and Beagle dogs (all P>0.05).The number of human chief cells of young and middle age group,junior elderly group,middle elderly group and senior elderly group was 71.79±16.85,52.73±16.60,57.10±20.21 and 43.70 ±16.89/high power field (HPF),respectively,which decreased as age increased (F=6.431,P=0.001).The numbers of chief cells of young and middle age group,junior elderly group,and senior elderly group of Beagle dogs were 328.38 ±32.36,341.20 ±42.49 and 225.67 ± 52.19/HPF,respectively,which decreased as age increased (F=13.647,P<0.01).Aging-related degeneration was founded in ultrastructure of gastric chief cells in human and Beagle dogs including rough endoplasmic reticulum dilation,ribosones missing,secretory granule decreasing and so on.The area percentages of secretory granule area in cytoplasm of human chief cells of young and middle age group,junior elderly group,middle elderly group and senior elderly group were (67.28±3.79)%,(66.88±4.84)%,(65.63±7.10)% and (56.25±8.47)%,respectively,which decreased as age increased (F=6.069,P=0.002).The area percentages of secretory granule area in cytoplasm of Beagle dogs' chief cells of young and middle age group,junior elderly group,and senior elderly group were (58.83 ± 3.07) %,(52.34 ± 4.73) % and (39.89 ± 4.46) %,respectively,which decreased as age increased (F=38.837,P<0.01).Condusion As age increased,the ability of pepsinogen secretion of fundal gland decreased,which may be one of the reasons of functional dyspepsia in the eldly.
9.Remote intracerebral hemorrhage after intravenous thrombolysis in acute ischemic stroke

Intracerebral hemorrhage is the most important complication after intravenous thrombolytic therapy for acute ischemic stroke.It often occurs inside the infarct focus,and a considerable proportion occurs outside the infarct focus (i.e.remote intracerebral hemorrhage).Remote intracerebral hemorrhage has different risk factors,pathophysiological mechanisms and prognosis from intralesional hemorrhage.In recent years,more and more research has begun to focus on remote intracerebral hemorrhage.This article reviews the diagnostic typing,clinical features,risk factors and potential pathophysiological mechanisms of remote intracerebral hemorrhage after intravenous thrombolysis in acute ischemic stroke.
