Acta Laboratorium Animalis Scientia Sinica 2015;(2):209-215
Parkinson’ s disease ( PD) is a neurodegenerative disorder primarily characterized by resting tremor, muscular rigidity, akinesia and postural reflex impairment.Behavioral tests of PD in animal models are essential for under-standing the pathogenesis of PD as well as for the development and testing of potential therapeutics .Here we mainly use the 6-hydroxydopamine (6-OHDA)-induced rat model, to introduce a review on the research progress in non-drug-induced be-havioral tests of motor function in PD rats.

Chinese Medical Ethics 1996;0(01):-
The "all-free health care" system is being implemented in Shenmu County in Shaanxi Province, whose ethical orientation is during the transitional process from "basic medical insurance" to "meeting residents′ actual healthcare demand".The "all-free health care" system safeguards human health rights, embodies the principle of giving priority to social benefits, and is in accordance with the principle of effectiveness. However, due to the lack of scientific management, the capital flows violate the principle of prevention in the system.

OBJECTIVE To explore the predictive value of human telomerase RNA gene component(hTERC) gene amplification and high-risk human papilomavirus(HR-HPV) testing in cervical intraepithelial neoplasia(CIN) as a marker for early diagnosis of cervix carcinoma.METHODS Fluorescence in situ hybridization(FISH) was used to detect the amplification of hTERC of cervical epithelial cells in 72 cases.By using hybrid capture 2(HC-2),two types of the HR-HPV DNA(HPV16/18) of each case were detected. Then, the results were compared with the pathologic diagnosis. The dual-color probe we used was GLP TERC/CSP 3.HeLa cells and lymphocytes from normal marrow were the positive control, while the cervical specimens from healthy outpatients were the negative control.RESULTS hTERC Gene amplification of specimens was tested in 72 cases, the positive amplification rate of hTERC gene in the cervicitis/CINⅠgroup and normal, compared to the cervical carcinomas(100%) and CIN Ⅱ/Ⅲ(68.75%),which showed a significant difference. The rates in CINⅡ and CINⅢ were 60.00% and 83.33%,respectively,which showed a significant difference compared with normal and CINⅠ/inflammation groups.hTERC gene amplification was positive in both HeLa cells and lymphocytes from normal marrow and HC-2 testing was positive in 32 cases of patients containing 11 cases of CINⅡ/Ⅲ,3 cases of cervical cancer,18 cases of cervicitis/CIN1 diagnosed. The positive predictive value(PPV) and specificity(Sp) of hTERC for the high-grade CIN was significantly higher than the PPV and Sp of HC-2 HR-HPV testing.CONCLUSIONS hTERC Gene involves in the progression and occurrence of cervical intraepithelial neoplasia and cervical squamous carcinoma. As a marker for early diagnosis of cervical intraepithelial neoplasia and cervical squamous carcinoma, the FISH method for hTERC gene is more reliable to differentiate the malignant diseases from the benign ones in cervixes than HC-2 HR-HPV DNA testing. The combined detection of HR-HPV and hTERC gene will provide more effective and suitable management to enhance the early diagnosis rate of cervical intraepithelial neoplasia and cervical squamous carcinoma.

Objective To investigate the inhibiting effects of shRNA(short hairpin RNA,shRNA) on COX-2 gene expression, the cell cycle and growth of gastric carcinoma SGC-7901 cells. Methods Specific shRNA plasmid to COX-2 were constructed, and then transfected into SGC-7901 cells by lipofectamine methods.Tests were divided into three groups: untransfected gastric carcinoma SGC-7901 cells group, negative control HK group and pshRNA-COX-2 group.Gastric carcinoma SGC-7901 cells were transfected with LipofectamineTM 2000.Expression of COX-2mRNA and protein were detected with reverse transcriptional polymerase chain reaction(RT-PCR) and Western blot respectively.Cell cycle analysis and cell growth chart were detected with flow cytometry and cell count respectively. Results Compared with negative control HK group, recombinant expression vector pshRNA-COX-2 resulted in the reduction of COX-2mRNA and protein expresion by 70.1% and 43.2% respectively;cells in G0-G1 phase increased from 61.5%to 70.2%,cells in S phase decreased from 27.3% to 21.7%,and the growth of SGC-7901 cells cells was slowed significantly. Conclusions Recombinant expression vector pshRNA-COX-2 can significantly inhibit the expression of COX-2 gene, result in the increase of cells in G0-G1 phase and decrease of cells in S phase, and suppress proliferation of gastric carcinoma SGC-7901 cells.

Chinese Journal of Tissue Engineering Research 2014;(39):6282-6286
BACKGROUND:A new open“C”-shaped molar band with nanocoating (patent number:ZL201110057699.1) has been invented to overcome some weakness of traditional bands and buccal tubes, which is used for molar fixation. OBJECTIVE:Using three-dimension finite element analysis software to theoretical y prove that the new band comes up to clinic requirement. METHODS:We established a finite element model of alveolar bone-tooth-band, and node domains tensile and shear loading were given. Stress distribution of the new bands through numerical simulation was predicted and calculated. Simulation of 20 N orthodontic force was applied in medial direction, distal direction and vertical direction in the three-dimensional model of a new band buccal tube and traditional band, to analyze the stress state in different directions under the force of the binder. RESULTS AND CONCLUSION:By numerical simulation of three-dimensional finite element, the stress of the new band was higher than that of the traditional band, but it could not reach the clinical cracking limit of resin reinforced glass ionomer cement. So the new band could theoretical y satisfy the clinical requirements.

Objective To explore the effects of total flavonoids from portulaca against liver fibrosis in rats by detecting TGF-β1 gene and protein expressions. Methods A total of 48 SD rats were randomly divided into normal control, model control, glucyrrhizin aqueous, and total flavonoids groups, with 12 rats in each group. Except those in the normal control group, rats in other groups were intraperitoneally injected with 2 mL · kg-1 · d-1 carbon tetrachloride to induce liver fibrosis. Rats in glucyrrhizin aqueous group and total flavonoids ones were intragastrically administered with 15. 75 mg · kg-1 of glycyrrhizin aqueous solution or 35. 6 mg·kg-1 of total flavonoids aqueous solution, respectively. The normal and model control groups were administered with equal volume of aqueous solution. Thirty days later, rats were sacrificed by anesthesia. Livers were obtained to detect TGF-β1 gene and protein expressions by RT-PCR and Western-Blot. Results Relative gene expression of TGF-β1 in the normal control, model control, glucyrrhizin aqueous and flavonoids groups was 0. 725±0. 130,7. 493±1. 410,3. 016±1. 240,and 2. 668±1. 150,respectively. Total flavonoids from portulaca significantly reduced the gene (P<0. 01) and protein (P<0. 01) expressions of TGF-β1 . Conclusion Efficacy of total flavonoids from portulaca in treating hepatic fibrosis may be related to decreased TGF-β1 expression in rats.
