1.A Study on Fingerprint of Citri Reticulatae Decoction Pieces by HPLC

Objective To establish an analytical method for fingerprint of Citri Reticulatae decoction pieces by HPLC. Methods Thermo C18 chromatographic column (4.6 mm×250 mm, 5μm) was used with methanol-0.3%phosphoric acid in gradient elution. The flow rate was 0.8 mL/min, the detection wavelength was set at 280 nm, and the temperature was 40 ℃. The similarities of the 15 batches of samples were compared by similarity evaluation, cluster analysis and principal component analysis. Results Based on the fingerprints of Citri Reticulatae decoction pieces, 8 common peaks were identified. The similarities were all more than 0.9 among all batches. 15 batches can be divided into three categories. Conclusion The method is simple and steady. It provides a basis for the quality control and evaluation of Citri Reticulatae decoction pieces.

3.CONSTRUCTION OF THE MUTANTS OF HUMAN PEPTIDE ANTIBIOTICS hPAB-? AND ITS RECOMBINANT BACULOVIRUS

The aim was to determine the antimicrobial activities of peptide antibiotics hPAB ? and construct the recombinant baculovirus of its mutants. Four mutants of hPAB ? were designed based on molecular autosyndetic modeling and its genes were cloned by PCR. The transfer plasmid and recombinant baculovirus were constructed. The results showed that the hPAB ? cloned previously, is a good peptide antibiotics. Four mutant genes of hPAB ? were inserted into pFAST HTa plasmid and the recombinant pFAST hPAB ? were screened by restriction enzymes analysis and DNA sequencing. The recombinant baculovirus was obtained after transforming pFAST hPAB ? into Escherichia coli DH10Bac. Our work lays a good foundation for further research.
4.Preparation of Compound Anisodamine and Neostigmine Sustained-release Tablet and Study on the in vitro Drug Release Behavior

OBJECTIVE:To prepare the Compound anisodamine and neostigmine sustained-release tablet and study the in vitro drug release behavior. METHODS:Using raceanisodamine and neostigmine methyl sulfate as main medicines,hydroxypropyl meth-yl cellulose as sustained release skeleton material,magnesium stearate as the lubricant,polyvinyl pyrrolidone as the adhesive,pre-gelatinized starch as the thinner,microcrystalline cellulose as the disintegrant and filler,Compound anisodamine and neostigmine sustained-release tablet was prepared by wet granulation method and direct compression method. The in vitro cumulative release rate within 12 h of the 2 main medicines was detected by HPLC method. RESULTS:Compound anisodamine and neostigmine sus-tained-release tablet was successfully prepared,and the in vitro release was basically completed within 12 h,with accumulative re-lease rate of 91.3% for anisodamine and 96.5% for neostigmine. CONCLUSIONS:Compound anisodamine and neostigmine sus-tained-release tablet that can cumulatively release for 12 h is successfully prepared.
5.Transdermal fentanyl in the management of cancer pain

Objective To evaluate the pain control, general condition and well-being of the patients with advanced cancer treated with transdermal fentanyl as well as the effects of transdermal fentanyl on heart rate variability (HRV) and endocrine function. Methods This open prospective study included 40 patients with advanced cancer. The direct conversion ratio from oral morphine to transdermal fentanyl system (TFS) was 100:1 daily. In patients who had never received any opioid before the initial dose of TFS was 25?g/h. The transdermal system was changed every 72h and the dosage was adjusted to the need of the patients according to the VAS scores and the requirement of morphine. 8 hospitalized patients were monitored by 24h Holter for changes in HRV. Blood samples were taken before transdermal fentanyl patch when pain was severe and 24h after TFS for determination of ACTH,?-endorphin, growth hormone(GH) and insulin levels.Results Pain relief during treatment with transdermal fentanyl was significant accoring to VAS scores. Well-being scores showed that patients' quality of life including appetite, sleep, mental state and daily activities was significantly improved after two weeks treatment. 6.The appilication of detachable balloon embolization in carotid cavernous fistula

Objective To study the clinical appilication of detachable balloon embolization techenique in carotid cavernous fistula. Methods 27 cases consisted of 26 cases of typical TCCF and 1 case of CCF caused by left anterio cerebral artery(A1) aneurysm ruptured . 27 cases were treated by endovascular embolization with detachable balloon. Transcranial Doppler ultrasound(TCD) was used before and after embolization.All the patients were followed-up for more than 6 months. Results 22 cases of TCCF were cured by detachable embolization and their internal carotid artery(ICA) remained patent. The first segement of left anteriol cerebral artery was embolized by two detachable balloons in the patient with CCF caused by ruptured aneurysm. Internal carotid artery was embolized by two balloons in 4 cases. There was no severe complication in this group. Conclusions Detachable balloon embolization is a safe and effective method for internal carotid cavernous fistula.
7.Protective Effect of Tongsaimai Pellets and Butylphthalide to PC12 Cell in Hypoxia/Hypoglycemia Model

Objective To discuss the protective effect of Tongsaimai pellets(TSMP) and butylphthalide(NBP) on PC12 cell in the hypoxia/hypoglycemia model.Methods Two ischemia models including hypoxia,hypoglycemia models were used to assay the anti-ischemic roles of TSMP and NBP in cultured PC12.Results TSMP and NBP possessed obvious protective effects on PC12 cells from two injured models.Both of them could increase the number of living cells and decrease LDH activity significantly,particularly in hypoglycemia injured model.Conclusion TSM and NBP have protective effect on PC12 cells from two injured models effectively in vitro.
8.Application of CO2 injection method in the magnetic resonance imaging preoperative staging diagnosis for stage Ⅰ carcinoma of endometrium

Objective To explore the accuracy rate of CO2 injection method and conventional magnetic resonance imaging (MRI) in the staging diagnosis for stage Ⅰ carcinoma of endometrium.Methods Preoperative staging diagnosis for stage Ⅰ carcinoma of endometrium was done,and the method of injecting CO2 gas into the uterine lumen by catheter was applied for pelvic MRI scan in 38 cases of stage Ⅰ carcinoma of endometrium.The pathological staging result was treated as the gold standard to compare the accuracy rate of CO2 injection method with conventional scan method (43 cases).Results For conventional MRI scan group,the accuracy rate of staging diagnosis in stage Ⅰ carcinoma of endometrium was 81.3 % (35/43),including stage Ⅰ A 75.0 %(6/8),stage Ⅰ B 83.9 %(26/31),stage Ⅰ c 75.0 %(3/4).For CO2 injection group,the accuracy rate was 89.4 %(34/38),including stage Ⅰ A 85.7 %(6/7),stage Ⅰ B 88.9 %(16/18),stage Ⅰ c 84.6 % (12/13).There was a statistical difference between the accuracy rates of two methods (x2=7.81,P < 0.05).Conclusion Compared with conventional scan method,the CO2 injection method with better simplicity,safety and application value,could be more accurate to determine the location of endometrial cancer and the degree of myometrial infiltration.
10.The value of peripheral blood circulating tumor cells in KRAS mutation testing of colorectal cancer patients

Objective To explore the value of peripheral blood samples in KRAS mutation testing of colorectal cancer patients and the correlation between the number of circulating tumor cells and KRAS mutation testing.Methods We detected KRAS mutation using amplification refractory mutation system PCR method in paraffin embedded tissues and matched peripheral blood samples obtained from 112 colorectal cancer patients and 10 proctitic peripheral blood samples in Affiliated Hospital of Jiangnan University between 2013 and 2014.Meanwhile,immunofluorescence in situ hybridization method was used to count the circulating tumor cells in peripheral blood samples and proctitic control samples.Results Among the 112 colorectal cancer samples tested,25 cases of peripheral blood samples found KRAS mutation (41.1%) and which was 46 in formalin fixed paraffin embedded tissues testing (22.3 %),with a significant difference (x2 =40.12,P < 0.001).One case with KRAS wild type in formalin fixed paraffin embedded tissues was mutation type in peripheral samples.In another case,mutation site was different in different kinds of samples.The sensibility of KRAS mutation testing was 73.3%,41.9% and 16.7% when the number of circulating tumor cells was more than 15,5 to 15,and 1 to 5,respectively,with significant differences (x2 =23.70,P < 0.001).No KRAS mutation and no circulating tumor cells were found in 10 proctitic control samples.Conclusion We find high specificity in KRAS mutation testing of peripheral blood samples.but the accurate rate is not satisfying.KRAS mutation testing in peripheral blood samples may be an optional choice to test KRAS mutations for colorectal cancer patients who were not subjected to surgery.The sensibility of KRAS mutation testing in peripheral blood samples has a corretion with the number of circulating tumor cells.
