1.Research status of targeted drug and whole brain radiotherapy for non-small cell lung cancer with brain metastasis

Whole brain radiation therapy has become the standard treatment of non-small cell lung cancer (NSCLC) with brain metastasis,because it prolongs the survival times of NSCLC patients with brain metastases.The emergence of molecular targeted drugs is a major innovation in the traditional sense of the cancer treatment.Molecular targeted drugs have many advantages such as convenient dosing,rapid onset,improving the intracranial and extracranial tumor lesions at the same time,mild adverse reactions and good tolerance,which provide a new and better choice for the treatment of NSCLC patients with brain metastases.
2.Recombinant adenovirus-mediated hypoxia inducible factor 1alpha mutant promotes proliferation of adipose-derived mesenchymal stem cells

BACKGROUND:To improve the survival rate of transplanted tissue, most scholars focus on cel therapy, particularly cel-assisted fat grafting.
OBJECTIVE:To analyze the effect of recombinant adenovirus-mediated hypoxia inducible factor 1alpha (HIF1α) mutant on survival rate of transplanted fat particles through transfection of adipose-derived mesenchymal stem cels.
METHODS:Recombinant adenovirus-mediatedtriple-mutantHIF1αwas inserted into an adenovirus pAdEasy-1 system, folowed by viral packaging and titer determination. Human adipose-derived mesenchymal stem cels were cultured, passaged and identified, and subsequently transfected with three kinds of viruses and blank vector (experimental group with transfection of the triple mutantof HIF1α;positive control group; negative control group; blank control group). Transfection efficiency was determined using enhanced green fluorescent protein labeling. Additionaly, MTT assay was used to detect cel proliferation.
RESULTS AND CONCLUSION:The recombinant adenovirus was successfuly constructed and packaged in line with transfection requirements. Moreover, adipose-derived mesenchymal stem cels were successfuly identified by adipogenic, osteogenic and chondrogenic induction and could be used asseed cels for subsequent experiments. RT-PCR results showed that HIF1α mRNA expression in the experimental group and positive control group was significantly higher than that in the other two groups (P< 0.05). Western blot analysis showed that the relative absorbance value in the experimental group was significantly higher than that in the other three groups (P< 0.05). A significant increase in the cel proliferation was found in the experimental group, significantly different from the other three groups (P<0.05). Therefore, our findings indicate that transfection of adenovirus-mediated triple-mutant HIF1α not only can sustain the expression of target protein in transfected adipose-derived mesenchymal stem cels under normoxic conditions, but also can promote the proliferation of transfected adipose-derived mesenchymal stem cels.
3.Effect of lidocaine combined with ropivacaine in penile dorsal nerve block for pediatric circumcision

Objective To explore the effect of lidocaine combined with ropivacaine in penile dorsal nerve block for pediatric circumcision.Methods 130 children with pediatric circumcision surgery were divided into observation group and control group by the digital meter method,65 cases in each group.The control group was taken inhaled sevoflurane composite static note ketamine surgery,the observation group was given ropivacaine combined with intravenous infusion of ketamine for penile dorsal nerve block anesthesia.The anesthesia effects of the two groups were recorded.Results In the observation group,10 min after surgery,the rhythm of the heart was (98.47±10.88)times/min,central arterial pressure was (65.13±7.78)mmHg.In the control group,10min after surgery,the rhythm of the heart was (121.78±15.23)times/min,central arterial pressure was (85.27±12.31)mmHg,the differences between the two groups were statistically significant(t=10.040,11.150,all P<0.05).In the observation group,the used ketamine,waking up time,pain score,incidence rate of pain and irritability were (41.38±11.28)mg,(5.54 ±1.03)min,(2.11±1.13)points,7.69%,respectively,which in the control group were (71.47±16.82)mg,(13.67±2.57)min,(3.89±1.79)points,26.15%,respectively,the differences between the two groups were statistically significant(t=11.978,23.673,6.779,χ2=7.878,all P<0.05).Conclusion The anesthesia effect of lidocaine combined with ropivacaine in penile dorsal nerve block for pediatric circumcision is stable,safe and reliable,it is worthy of popularization and application in clinic.
4.Study on status and influencing factors of anxiety and depression of adolescent students in Weifang city

Objective To investigate the prevalence of anxiety and depression in adolescent children, and to explore influencing factors of anxiety and depression. Methods From May 2016 to September, 435 students at the age of 10~18 were randomly selected from primary school, middle school, high school in Weifang city. The Screen for Child Anxiety Related Emotional Disorders (SCARED) and Depression Self-rating Scale for Children (DSRSC) were used to evaluate the Clinical manifestation of students with anxiety and depression. A survey on the factors influencing anxiety and depression in adolescence was used to evaluate the related factors of students with anxiety and depression. Results The incidence rate of the anxious and depression of adolescent children in Weifang City was 20.6% and the average age was (12.38±3.58). The occurrence rate of anxious and depression was higher in girls than in boys (26.5%vs. 15.7%, P<0.05). Personal unhealthy (OR=0.387, P=0.001), lack of parental accompaniment (OR=0.671, P=0.020), low parental educational levels(OR father=0.338, P=0.001; OR mather=0.262, P=0.001), poor parental relationship (OR=1.049, P=0.008), bad parenting styles (OR=0.471, P=0.016) were risk factors of anxious and depression in adolescent children (P<0.05). Conclusion Adolescent girls are the high incidence population of emotional disorder, especially in early adolescence. Lack of parental accompaniment, low parental educational levels, poor parental rela-tionship, bad parenting styles are closely related to anxiety and depression.
5.Characteristics of Phthalate Esters Pollution in Indoor Settled Dust

Objective To investigate the pollution of phthalate ester in indoor environment. Methods Settled dust samples from 10 households, 10 offices and 10 student dormitories in Beijing were collected. Seven kinds of phthalate esters in these samples were determined with solid phase extraction-high performance liquid chromatography (SPE-HPLC) .Dates were analyzed statistically by nonparametric tests. Results The main phthalate esters in dust were di (2-ethylhexyl) phthalate (DEHP), dibutyl phthalate (DBP) and dicyclohexyl phthalate (DCHP). Of phthalate esters, DEHP with the range of 28-6 073 mg/kg had the highest concentration in indoor dust. The total level of phthalate esters in households was the highest, offices followed, student dormitories was the last. 6.Effect of saikogenin d on prostaglandin E_2 (PGE_2) production in vitro in C_6 rat glioma cells

Aim To study the effect of saikogenin d (SGD) on prostaglandin E 2(PGE 2) production in C 6 rat glioma cells. Methods Radioimmunoassay was applied to determine PGE 2 production in the cells. Scintillation counting was used to measure liberation of -arachidonic acid (AA) from the cells labeled with -AA. Results In vitro, SGD alone at 1~20 ?mol?L -1 did not affect PGE 2 release from cells, but inhibited its release induced by A23187, a Ca 2+ ionophore. The inhibition was concentration-dependent, with the IC 50 value of about 3 ?mol?L -1. SGD (2~10 ?mol?L -1 ) had no inhibitory effect on A23187-induced AA release or on the conversion of AA to PGE 2 in microsomal preparations. Conclusion SGD inhibites A23187-induced PGE 2 production in C 6 rat glioma cells in vitro, without either inhibition of free AA liberation or a direct inhibition of cyclooxygenase (COX) activity.
7.Relationship of Radiological Characteristics and Clinical Symptoms of Ossification of Posterior Longitudinal Ligament of Cervical Spine

Objective To explore the relationship of the radiological characteristics and clinical symptoms of ossification of the posteriorlongitudinal ligament (OPLL) of cervical spine. Methods 200 OPLL patients were recruited into this study. The data of JOA scores of theOPLL patients were collected. The stenotic rate of the cervical spine canal was calculated by the sagittal plane CT. The sorts and ossificatedcervicle were also identified by the images. Results The stenotic rate of the cervical spine canal was 19.8% to 60.3% with average 42.9%,the JOA scores was 4 to 15, averaged 10. Linear regression showed the stenotic rate of the cervical spine canal was negatively correlatedwith the JOA scores (P<0.01). There were 94 cases for segmental-type, 46 for mixed-type, 23 for continuous-type and 37 for focal-type. Thethird and fourth cervical vertebrae were susceptible to ossify. Conclusion The higher the OPLL occupation ratio, the worse the clinicalOPLL symptoms. The OPLL patients following clinical symptoms with spinal cord oppression are more likely the segmental-type.
8.Absorption, distribution and excretion of 2-fluorine-6-trifl-uoromethylpyridine in rats by radioactivity isotope tracing method

OBJECTIVE To study the absorption,distribution and excretion of 2-fluorine-6-trifluoromethylpyridine (JJBD) in rats.METHODS [14C] Radioactivity isotope tracing method was used.Male SD rats were ig given a single dose of JJBD 10 and 100 mg·kg-1 (radioactivity:3.7 GBq·kg-1).Concentrations of rat plasma,tissue,feces,urine and bile were determined with a liquid scintillation counting (LSC) analyzer.Toxicokinetics (TK) parameters were fitted using WinNonlin.RESULTS TK parameters of JJBD 10 and 100 mg · kg-1 in male SD rats were as follows:area under the curve (AUC(0-t)) was 22 548±1579 and (203 395±27 586) h·iμg Eq.·L-1,half time (t1/2) was 15.8±1.0 and (14.1±0.9) h,peak time (Tmax) was 4.0±3.0 and (6.0±5.0) h,peak concentration (Cmax) was 1450±355 and (7776±1703) μg Eq.·L-1.JJBD was mainly distributed in fat,livers,kidneys,stomachs and intestinal walls.The concentration of JJBD in most of the tissues reached peak values after 4 h.However,JJBD couldn't be detected in the muscle,thymus gland,brain,gonad or spleen.Excretion rate of JJBD was 43.1% in urine,29.7% in feces and 9.97% in cleaning solution within 0-168 h.JJBD could be excreted through bile at a rate of 28.1% within 0-72 h.CONCLUSION JJBD can be absorbed immediately and excreted slowly in SD rat.There is no accumulation risk.The distribution of JJBD in vivo is very extensive,but cannot go through the blood-brain barrier.JJBD is mostly excreted through feces and urine.
9.Treatment of non-small cell lung cancer with brain metastasis

30%-50% patients with non-small cell lung cancer (NSCLC)will occur cerebral metastasis in the course of their illnesses.NSCLC with brain metastases is one of the difficulties in cancer treatment. Treatment measures include surgery,radiotherapy,chemotherapy,biological targeted therapy and so on.The new prognostic scoring system is recommended to predict the prognosis of patients.Individualized treatment methods should be chosen according to prognostic score.
10.Relationship Between Somatosensory Evoked Potential Diversification during Operation and Short Term Outcome of Ossification of Posterior Longitudinal Ligament of Cervical Spine

Objective To explore the relationship between the somatosensory evoked potential (SEP) diversification during operation and the short term outcome of ossification of posterior longitudinal ligament (OPLL) of cervical spine. Methods 70 OPLL inpatients from February 2008 to February 2011were included in this study. All patients received anterior cervical operation. According to the diversification of the SEP during surgery, the patients were divided into 2 groups. The preoperative and postoperative JOA scores of the OPLL patients were collected. Results There were 35 patients in the SEP no change group and 30 in SEP improved group. There was no difference in the preoperative JOA scores between them (P>0.05), but was significantly different in the postoperative JOA scores (P>0.05). Conclusion There is some relationship between the SEP diversification during operation and the short term outcome of OPLL of cervical spine.
