1.Study on status and influencing factors of anxiety and depression of adolescent students in Weifang city

Objective To investigate the prevalence of anxiety and depression in adolescent children, and to explore influencing factors of anxiety and depression. Methods From May 2016 to September, 435 students at the age of 10~18 were randomly selected from primary school, middle school, high school in Weifang city. The Screen for Child Anxiety Related Emotional Disorders (SCARED) and Depression Self-rating Scale for Children (DSRSC) were used to evaluate the Clinical manifestation of students with anxiety and depression. A survey on the factors influencing anxiety and depression in adolescence was used to evaluate the related factors of students with anxiety and depression. Results The incidence rate of the anxious and depression of adolescent children in Weifang City was 20.6% and the average age was (12.38±3.58). The occurrence rate of anxious and depression was higher in girls than in boys (26.5%vs. 15.7%, P<0.05). Personal unhealthy (OR=0.387, P=0.001), lack of parental accompaniment (OR=0.671, P=0.020), low parental educational levels(OR father=0.338, P=0.001; OR mather=0.262, P=0.001), poor parental relationship (OR=1.049, P=0.008), bad parenting styles (OR=0.471, P=0.016) were risk factors of anxious and depression in adolescent children (P<0.05). Conclusion Adolescent girls are the high incidence population of emotional disorder, especially in early adolescence. Lack of parental accompaniment, low parental educational levels, poor parental rela-tionship, bad parenting styles are closely related to anxiety and depression.
2.Effect of humic acid and soil on the dissolution performance of depleted uranium in water

Objective To study the effect of humic acid (HA) and soil on dissolution performance of depleted uranium (DU).Methods Using the static dissolve-adsorption experiment and inductively coupled plasma-mass spectrometry(ICP-MS) to determine uranium concentration and 235U/238U isotope ratios of the samples at different time and study the dissolution of DU in water.Results The solubility of DU in water was reduced by 90% by adding HA.Soil could increase the solubility of DU in water by nearly 25%,adding an appropriate amount of HA could play a supporting role on the dissolution of DU,in this experiment adding 5% of HA was best.Conclusions Soil and HA could produce positive and negative impact on the solubility of DU in water,and the combined effect of the two relied on the complex absorption and complexation reactions of soil,HA and dissolved uranium ions.
3.Effects of starvation, diabetesand obese conditions on mouse hepatic SOCS2 gene expression

Objective To determine the expression levels of SOCS2 in the mouse livers under starvation, diabetes and obese conditions and to study the effect of SOCS2 on gluconeogenesis.Methods Animals were divided into 3 groups: C57BL/6J mice, the control group was fed ad libtum and the experimental group was fasted for 24 h.Diabetes model db/db and the control db/m mice were fed ad libitum.Obese model ob/ob and the control C57BL/6J mice were fed ad libitum.All the mice above were sacrificed and total RNA was isolated from mouse livers and reverse transcribed to cDNA.The expression of SOCS2 and gluconeogenesis genes in the mouse livers in the 3 groups above were detected by real-time quantitative PCR.SOCS2 was overexpressed in the primary C57BL/6J mouse hepatocytes by the adenovirus system.The effect of SOCS2 on glucose production was measured by glucose output assay.Results C57BL/6J mouse hepatic SOCS2 expression was suppressed by starvation status.The expression of SOCS2 was decreased in the livers of db/db and ob/ob mice.In contrast, the key regulators of gluconeogenesis, PGC-1α, PEPCK and G6Pase exhibited the opposite expression pattern as SOCS2 in the livers underidentical starvation, diabetes and obese conditions.The protein was Mr 23 000 and glucose production was inhibited after SOCS2 being overexpressed in the primary C57BL/6J mouse hepatocytes by adenovirus system.Conclusions SOCS2 may inhibit gluconeogenesis in the C57BL/6J mouse primary hepatocytes, and SOCS2 may be a potential target for the treatment of type Ⅱ diabetes.
4.Protective effect of 17β-estradiol on traumatic brain injury in rats

Objective To explore the protective effect of 17β-estradiol on traumatic brain injury in rats.Methods A total of 45 adult male SD rats were divided into 3 groups using the random digit table, 15 rats in each group:the control group only exposed but not injured the brain, the injury group received traumatic brain injury ( TBI) by Feeney’ s method, and the treatment group received the same handling with injury group, and pretreatment with 17β-estradiol peritoneal injection, 1 mg/kg per day for one week.The other two groups were given the same volume of castor oil.At 6 h, 24 h and 48 h after injury, the brain tissue water content, malondialdehyde ( MDA) content and superoxide dismutase ( SOD) activity were measured.Results At 6 h, 24 h and 48 h after injury, the levels of brain tissue water content in the injury group and treatment group were significantly higher than those in the control group ( P<0.05 ) .At 6 h after injury, the brain tissue water contents in the injury group and treatment group were ( 99.83 ±0.40 )% and ( 99.53 ±0.41 )%,respectively, with a non-significant difference between the two groups (P>0.05).At 24 h and 48 h after injury, the brain tissue water contents in the injury group was ( 105.17 ±0.43 )% and ( 107.54 ±0.39 )%, in the treatment group was (103.26 ±0.42)%and (105.89 ±0.43)%, respectively, showing a significant difference between the two groups ( P<0.05).At 6 h after injury, the levels of MDA in the injury group and treatment group increased and maintained at a higher level, and the levels of SOD decreased.Compared with the control group, there had a significant difference ( P<0.05 ) . The levels of MDA and SOD in the injury group and treatment group at 6 h after injury had a non-significant difference ( P>0.05).But at 24 h and 48 h after injury, the levels of MDA in the treatment group were significantly lower than those in the injury group [(130.39 ±7.02) μmol/g vs.(149.41 ±8.25) μmol/g, (125.41 ±6.59) μmol/g vs.(157.72 ± 8.93) μmol/L] , and the levels of SOD in the treatment group were significantly higher than thoset in the injury group [(88.46 ±7.17) U/g vs.(80.10 ±4.87) U/mg, (97.31 ±7.89) U/g vs.(84.29 ±6.13) U/g], with a significant difference ( P<0.05 ) between the two groups.Conclusions 17β-estradiol has a protective effect on traumatic brain injury.
5.Primary culture and proliferation activity identification of rabbits subchondral bone cell

ObjectiveTo study the method of cell isolation,primary culture and identification of subchondral bone cell of osteoarthritis(OA) rabbits.Methods The rabbit instable knee joint models were made by modified Hulth modeling method.The osteoblasts were harvested from the subchondral bone of rabbits by collagenase and tissue explants attachment.The morphology observation and biological identification were performed by inverted microscope and immunocytochemistry staining,respectively.The proliferative activity of cells were detected by MTT and the expression of Ⅰ-collagen at gene level was detected.ResultsThe cells started to appeared on the 11th day after the attachment.The cells form were fusiformis and triangle,the nucleolus were clear.The cultured cells had typical osteoblast morphological characteristics.The cells obtained from subchondral bone of rabbits were identified to be osteoblast by immunocytochemistry staining.The proliferative activity of cells were equably proliferation which detected by MTT.ConclusionThe modified method provides better way to obtain ideal subchondral osteoblast and the co-culture method is suitable for the study of OA microenvironment,which can simulate interactions of the subchondral osteoblast,synovial cells and chondrocyte.
6.Analysis for low scores in patients with reflux gastroesophageal disease using reflux diagnostic questionnaire

Objective To analyse the clinical features of patients with gastroesophageal reflux disease(GERD)who had scores<12 using reflux diagnostic questionnaires(RDQ).Methods A comparative and retrospective study was carried out in 148 GERD patients.Among whom 22 patients had low RDQ score(<12)and 126 patients had high RDQ score(≥12).The differences in heart burn,substantial chest pain,regurgitation,severity and frequency of food reflux between two groups were compared.Nevertheless,the atypical symptoms such as chest distress.abnormal sensation of regurgitation or food reflux was significantly lower in patients with RDQ<12 than those with RDQ≥accounted for 60.28%of the total RDQ score,whereas the heart burn,regurgitation and food reflux atypical symptom of chest distress was found in 13 patients(59.09%),abnormal sensation of throat in 8 patients(36.36%).cough and asthma in 3 patients(13.63%),headache in 5 patients(22.72%),belching in 9 patients(40.90%),aypnia in 8 patients(36.36%),and anxiety in 6 patients(27.27%).Conclusion RDQ is insensitive to those who have chest pain with no obvious symptom of heart burn,regurgitation or food reflux as well as those with atypical symptom beyond the RDQ.
7.Effect of curcumin on expression of p21 and CD44V_6 in human breast cancer xenografts

AIM: To study the effect of curcumin on the expression of p21 and CD44V6 in breast carcinoma in nude mice.METHODS: Nude mice were xenografted with human breast cancer cell line MCF-7 and randomly divided into 2 groups(n=4 in each group): control group and curcumin group.In latent period,the percentage of tumor development was observed.Tumors were measured and the surface areas were calculated.RT-PCR was performed to detect the expression level of cyclin D1,p21 and CD44V6 mRNA.RESULTS: The tumor surface areas in the curcumin group were significantly lower than those in control group.In curcumin treatment group,the expression of p21 was up-regulated while cyclin D1 was nearly not changed.The expression of CD44V6 was significantly down-regulated in curcumin group.CONCLUSION: Curcumin inhibits the expression of CD44V6 and up-regulates the expression of p21 in nude mice bearing human breast cancer cell line MCF-7.
8.In vitro culture and identification of synovial cell of rabbit knee osteoarthritis

Objective To replicate experimental animal model of knee osteoarthritis and to investigate the method of culture and biological characteristics of rats synoviocytes of gonarthritis in vitro.MethodsAnimal models of knee osteoarthritis were made by the Modified Hulth method.4 weeks after the replicating experiment,synovial tissues were mechanically isolated and enzyme-digested and the growth of the synovial cells was investigated.Results The synovial tissues were obviously hyperplasia in the model made by the Modified Hulth method.The synovial cells were abundant after enzyme-digested cultivation and the cell activity was higher than 98%.Conclusion The study exhibits that the Modified Hulth method apparently promotes the hyperplasia of synovial tissues.The methods of isolation and cultivation of the synovial cells in vitro is proved to be simple and feasible.
9.Role of Fas/FasL signaling pathway in ulinastatin postconditioning-induced attenuation of apoptosis in myocardial cells of patients undergoing cardiac valve replacement with cardiopulmonary bypass

Objective To evaluate the role of Fas/FasL signaling pathway in ulinastatin postconditioning-induced attenuation of apoptosis in the myocardial cells of patients undergoing cardiac valve replacement with cardiopulmonary bypass (CPB).Methods Forty patients of both sexes,aged 21-59 yr,of ASA physical status Ⅱ or Ⅲ (NYHA class Ⅱ or Ⅲ),scheduled for elective cardiac valve replacement with CPB,were randomly divided into 2 groups (n =20 each):control group (group C),and ulinastatin postconditioning group (group U).In group U,ulinastatin 10 000 U/kg was perfused via the aortic root at 4 000-5 000 U·kg-1 ·min-1 starting from 5 min before aortic unclamping.In group C,the equal volume of normal saline was infused instead of ulinastatin.Myocardial specimens were taken from the right auricle at 45 min after aortic unclamping for determination of Fas,Fas ligand (FasL),caspase-8,Bcl-2 and Bax expression and cell apoptosis.The ratio of Bcl-2 expression to/Bax expression (Bcl-2/Bax) and apoptotic index were calculated.Results Fas,FasL,caspase-8 and Bax expression and apoptotic index were significantly lower,and Bcl-2 expression and Bcl-2/Bax were higher in group U than in group C.Conclusion Ulinastatin postconditioning attenuates apoptosis in the myocardial cells through inhibiting Fas/FasL signaling pathway in the patients undergoing cardiac valve replacement with CPB.
10.Correlation between serum interleukin-6 and MRI dynamic changes in patients with acute cerebral infarction

Objective To investigate the function of serum Interleukin-6 (IL-6) in the formation of cerebral edema after acute cerebral infarction through dynamic changes of serum IL-6 and MRI indicators.Methods One hundred and twenty-eight patients with cerebral edema after acute cerebral infarction and 30 healthy were selected as our subjects.The serum IL-6 were measured in at 1,5 and 14 d after acute cerebral infarction.MRI scans were performed in the corresponding time,and then processed synthesis of apparent diffusion coefficients (ADC) graph.Cerebral infarction volume,signal intensity ratio (SIR) of each sequence and relative apparent diffusion coefficient(rADC) were measured and calculated.Results The serum IL-6 in patients with acute cerebral infarction at 1 d after cerebral infarction was significantly higher than that in normal control group and reached the peak level at 5th D after cerebral infarction,significantly decreased at 14th d.The serum IL-6 of each time point were significantly higher than that of normal control group(P =0.000).There were liner positive correlation between the serum IL-6 and the volume of cerebral infarction and SIR of T2 weighted image and fluid attenuation inversion recovery (r =0.750,0.621,0.691 ; P =0.000).The serum of IL-6 and SIR of T1 weighted image was showed negative correlation (r =-0.404,P =0.000).The serum of IL-6 and SIR of diffusion-weighted imaging sequence at 1,5 d after cerebral infarction showed a positive correlation (r =0.678,P =0.000).There was liner negative correlation between the serum IL-6 and rADC at 1 d after cerebral infarction (r =-0.826,P =0.000).Conclusion The rise of serum IL-6 may promote the formation and development of ischemic cerebral edema.
