1.Effects of Rab7 gene silencing on cytokine and MAPK signal pathway activated by R848 in macrophage

Objective:To investigate the effect of Rab7 on cytokine induced by TLR7 (Toll like receptor-7) R848 activated in Raw264.7,and discusses the influence of Rab7 on MAPK signal transduction.Methods: TLR7 downstream cytokines such as TNF-α,IL-6,IFN-α,IFN-β and IP-10 activated by R848 were detected through Q-PCR in Rab7 silenced mouse macrophages,and then analysis of phosphorylation of MAPK determined with Western blot showed the effect of Rab7 on signal transduction of MAPK.Results: Rab7 inhibit production of cytokine activated by TLR7,and also,Rab7 had an inhibitory effect on MAPK signal pathway.Conclusion: The experimental results further illustrate that the Rab7 is the TLR7 signal transduction pathway negative regulatory factor,and to participate in MAPK signaling pathway.
2.Rab5a promotes LPS-induced cytokine expression in macrophages

Objective:To establish cell lines stably expressing Rab5a and its the inactive mutant Rab5aN133I,analyze the effect of Rab5a on the expression of cytokines in LPS-stimulated RAW264.7 cells .Methods:RAW264.7 cells were transfected with Rab5a and its inactive mutant vector Rab5a N133I separately,and then screened by G418.Rab5a stable expressing cell lines were identified by Real time-PCR.The growth of the stable cell lines was analyzed by MTT assay.After the stable cell lines were stimulated by LPS for different time periods,the expression of iNOS,TNF-αand IL-6 was detected.Results:Rab5a and Rab5aN133I transfection resulted in elevated Rab5a mRNA expression compared with the control cells ( P<0.05 ).Rab5a overexpression enhanced the proliferation of RAW264.7 cells.However,the proliferation of Rab5aN133I cells was significantly slower than the control cells ( P<0.05).Overexpression of Rab5a promoted LPS-induced production of iNOS,TNF-αand IL-6 in RAW264.7 cells (P<0.01). Conversely,overexpression of Rab5aN133I abolished the stimulating effects of Rab5a.Conclusion: Rab5a promoted LPS-induced expression of iNOS,TNF-αand IL-6 in RAW264.7 macrophages in a GTP-binding ability-dependent manner.
3.Application and research progress of in situ gel for local treatment of periodontitis

Recently, in situ gel has been widely used as a local delivery system for periodontitis treatment because of its lesion injectability, local drug depot function, and drug sustained-release effect.Different therapeutic purposes can be achieved by loading different types of drugs such as antibiotics, bioactive factors, etc.In this review, different types of in situ gel with temperature-sensitive, ion-sensitive, pH-sensitive and solvent-exchanged characteristics were introduced for their applications and limitations in the delivery of drug for periodontitis;and the elimination of periodontal inflammation, periodontal tissue repair and the long-term role after loading microsphere achieved by the in situ gel system were also reviewed.
4.Application effects of endoscopic laminar fenestration versus percutaneous endoscopic transforaminal decompression in the treatment of degenerative lumbar lateral recess stenosis

Objective To compare the short-term efficacy and occurrence of complication of en-doscopic laminar fenestration(Endo-LOVE)and percutaneous endoscopic transforaminal decompres-sion(PETD)in patients with degenerative lumbar lateral recess stenosis(LSLRS).Methods A to-tal of 160 patients with LSLRS were enrolled as study objects,and were randomly assigned to control group(treated with PETD)and observation group(treated with Endo-LOVE),with 80 patients in each group.Surgical indicators,occurrence of complications,and therapeutic effects were observed and compared between the two groups.Bone lateral recess angle,soft lateral recess angle,Oswestry Disability Index(ODI)score,pain score,and Japanese Orthopaedic Association(JOA)score were compared before and after surgery.Results The observation group had less intraoperative blood loss,and shorter operative and hospital stays than the control group(P<0.05).There were no sta-tistically significant differences in the incidence of complications such as lower limb numbness,fever,and postoperative low back pain between the two groups(P>0.05).There were no statistically significant differences in preoperative bone lateral recess angle or soft lateral recess angle between the two groups(P>0.05).At 72 hours postoperatively,both angles increased in both groups compared with preoperative values,and the observation group had larger angles than the control group(P<0.05).There were no statistically significant differences in preoperative pain score,ODI score,or JOA score between the two groups(P>0.05).At 3 months postoperatively,JOA scores increased and pain scores as well as ODI scores decreased in both groups compared with preoperative values,with the observation group having higher JOA scores and lower pain scores as well as ODI scores than the control group(P<0.05).The excellent and good rate was 96.25%(77/80)in the observation group,which was higher than 87.50%(70/80)in the control group(P<0.05).Conclusion Com-pared with the PETD approach,the Endo-LOVE approach for the treatment of LSLRS offers shorter operative time,less intraoperative blood loss,and better short-term efficacy.
